Synergistic Effects of Catechin or Ascorbic Acid on Antioxidative Activities of Hexane and Methanol Extracts from Rosemary, Sage, Oregano, and Ginger (Rosemary, Sage, Oregano와 Ginger의 메탄올과 헥산 추출물의 항산화 작용에 대한 카테킨과 아스코르브산의 상승 효과)

  • This study was peformed to investigate the synergistic effects of catechin and ascorbic acid on antioxidative activities of methanol and hexane extracts (500 ppm) from rosemary, sage, oregano, and ginger. Ascorbic acid (200 ppm) and (-)-catechin (200 ppm) could be solubilized in a rice bran oil via a reverse micelles using small amount of water and dioctyl sulfosuccinate as the sufactant. Methanol extracts from rosemary, sage, oregano, and ginger showed the synergistic effects by (-)-catechin. However, methanol extracts showed the synergistic effect by ascorbic acid except that of ginger. The synergistic effects of (-)-catechin on methanol extracts were higher than those of ascorbic acid. Hexane extracts of oregano and ginger showed the synergistic effects by (-)-catechin, and no synergistic effects by ascorbic acid. On the other hand, rosemary and sage showed the synergistic effects by ascorbic acid and no synergistic effects by (-)-catechin.

  • Present expriments were conducted to determine the possibility of cultivation of Coriolus versicolor selected among the higher fungi growing in Korea. Dried apple sawdust used in this study consisted of 31.5% C, 0.54% N, 2.41 % CaO, 0.05% $P_2O_5$, 0.48% MgO and 0.81% $K_2O$(pH 5.6). Coriolus versicolor mushrooms were cultivated on the sawdust media; apple sawdust; rice bran=80:20 in 850cc polypropylene bottles. The isolate of Coriolus versicolor used was YCV collected from an Apple farm in Youngchun district. It took 34 days to make fruitbody from spawning. The fruit bodies produced the total fresh weight 28 g in a bottles, and converted to 64% of fresh weight.

Comparison of Fermentation Characteristics of Korean Traditional Alcoholic Beverages Prepared by Different Brewing Methods and Their Quality Changes after Aging (전통주의 제조방법별 발효 특성 및 숙성후 품질변화 비교)

  • Fermentation characteristics and quality changes of the typical Korean traditional alcoholic beverages (Kwahaju, Sokokju, Baikhaju, Samhaeju and Hosanchun) were investigated during fermentation and after aging, respectively. They were prepared by their own brewing methods with respect to the same contents of rice and Nuruk (Korean-style bran koji). The consumed total sugar content of the alcoholic beverages (Sokokju, Baikhaju, Samhaeju and Hosanchun) which were brewed with the multiple input steps of seed mash and raw material (ABMIS) was higher than that of the alcoholic beverage (Kwahaju) which was brewed with the single input step (ABSIS). The pH values of all the beverages were similar and were ranged from 3.2 to 3.6. The acidity of ABMIS were lower than that of ABSIS, but alcohol concentration of ABMIS were higher. The fermentation period of the alcoholic beverages with triple input steps was the longest among them. The inner temperature and microbial growth in all the beverages increased at each of input steps of the raw materials. Fusel oil content of ABMIS was lower than that of ABSIS. Major components of the fusel oil were iso-amyl alcohol, iso-butanol, n-propanol, ethyl acetate and acetaldehyde, and its minor components were methanol and n-butanol. The results on quality changes of the alcoholic beverages after aging showed increase of alcohol in ABMIS, but decrease of sugar and acidity. Fusel oil contents of Kwahaju and Samhaeju decreased to be ranged from 3.1% to 13.9% after aging, but those of Sokokju, Baikhaju and Hosanchun increased to be ranged from 10.9% to 23.0%. Sensory evaluation of ABMIS showed better scores than that of ABSIS. It was suggested that the brewing method with multiple input steps of seed mash and raw material was one of the methods to increase fermentation efficiency for brewing the Korean traditional alcoholic beverages.

Effects of Dietary n-3/n-6 Fatty Acid Ratio on In Vitro Fermentation Characteristics and Fatty Acid Profiles

  • This study was conducted to examine the effect of dietary n-3/n-6 fatty acid (FA) ratio on in vitro dry matter digestibility (IVDMD), fermentation indices and FA profile. Rice bran was mixed with oil sources (cotton seed oil and linseed oil) to make the diets at 0.02, 0.29 and 0.61 of dietary n-3/n-6 FA ratio. These diets (0.5g) were placed into the incubation bottles with 40 ml of anaerobic culture medium, which contained rumen fluid and Van Soest medium at 1:2 ratio. Five replicates of each diet and two blanks were incubated at $39^{\circ}C$ for 48 hours. After incubation, the incubated contents were centrifuged. The residues were freeze-dried for DMD and FA analyses. The supernatant was used for pH, $NH_3-N$ and volatile fatty acid analyses. The concentrations of lactate (p<0.001) and iso-valerate (p<0.001) decreased linearly with increasing dietary n-3/n-6 FA ratio, but acetate concentration (p=0.056) and the ratio of acetate to propionate (p=0.005) was increased linearly. The concentrations of n-3, n-6 FA and the ratio of n-3/n-6 FA in residues increased (p<0.001) linearly with increasing dietary n-3/n-6 FA ratio, but C18:1n-9 FA concentration was decreased (p<0.001) linearly. With these results, it could affect fermentation characteristics and FA profile of rumen content by dietary n-3/n-6 FA ratio.

  • To develop an efficient mass rearing system for Cadra cautella(Walker) and Orius laevigatus(Fieber) were investigated under the conditions of photoperiod 16L : 8D, $70{\pm}5%$ RH and $23{\pm}2^{\circ}C$. The highest efficiency rate(multiply 28.6) of C. cautella was achieved when using the rearing cage of $20{\ell}$ than $3{\ell}$. The highest efficiency rate(multiply 16.3) of O. laevigatus was achieved when using the rearing cage of $5{\ell}$ (adult 1,500, 10 pcs(10 cm) of plant of egg taking). The moth could be successfully reared with all food substrates tested, of which rice bran(43%) + chick feed(43%) + yeast(14%) assumed to be proper for mass-rearing in view of cost.

  • This study was carried out to select the suitable for bag cultivation of Lentinula edodes. We investigated the optimal major materials and its mixing ratio in bag cultivation of L. edodes, 'Sanjo701ho' and 'Nongjingo'. The Suitable substrates for L. edodes bag cultivation were oak sawdust and douglas fir sawdust and rice bran mixed ratio 40:40:20(v/v). At the result, the period mycelial incubation shortened up to 13~18 days compared to the control. And Yield of commercial fruiting bodies was 17~19% higher than that in control. We expect that the cultivation period of L. edodes will be shortened, and the yield increased in the medium replaced half of oak sawdust by douglas fir sawdust.

  • The purpose of this study was to produce Pleurotus ostreatus using fermenter with bag cultivation. These results are as follows. 1. While mushroom composts were being fermented in a fermenter, the physical property of the fermented composts was getting better when there isn't any screw or revolving flies in the fermenter and the strength of pressing the composts was getting less. 2. The composts were fermented well as slaked lime of 1% density added to the composts. 3. According to the result of examining our fermenting ways, composts were in the best condition after being fermented for 48 hours since the temperature in a fermenter has come to 60℃, which could be reached by heating the fermenter by 40℃ after putting compost materials and water into it. 4. The good condition of fermenting could be maintained by controlling the speed of revolving flies, therefore the speed be down when the temperature is above 60℃ and up bellow 60℃. 5. Since the composts had been added with 1.5~2% of cottonseed meal or rice bran, the fermented composts were in good condition and also the quantity and quality of the mushroom produced on the fermented composts were satisfied. 6. There were needed 7 hours of labour for 3days from the first day of putting composts into a fermenter for fermenting 3.5M/T(10,000~12,000bags of 750~800g per bag) of composts to the third day of finishing the fermenting work, and also the cost was 112,066₩(130$) including 52,066₩(60$) of electric charge and fuel expense.

Studies on Artificial Cultivation of Pleurotus eryngii (De Canolle ex Fries) Quel. (큰느타리(Pleurotus eryngii)의 인공재배에 관한 연구)

  • This study was carried out to investigate the artificial cultivation of Pleurotus eryngii on the optimal medium vessel, periods of cultivation and the optimal method of pinheading for both yield and quality of fruiting body were also performed. The optimal composition of sawdust medium in polypropylene(PP) bottle was combination of sawdust(70%) and corncob(30%) but increased amount of corncob delayed the period of mycelial growth. The mycelial growth and the yield of fruiting body in the medium with beat pulp were worse than that without beat pulp. The optimal composition of nutrients for both yield and quality of fruit body tuned out to be a combination of rice bran(12%), wheat bran(12%) and cottonseed cake(6%). Additions of zeolite, shell lime and bean curd dregs were not effective in mycelial growth and yield of fruit body. When testing size of PP bottle for cultivation, the larger of bottle mouth is, the more pinheading number found, but the number of available fruit body is not significantly different. The culture in $1100\;ml-{\phi}75\;mm$ bottle is the best in the yield and quality of fruit body than those in $555\;ml-{\phi}50\;mm,\;850\;ml-{\phi}58\;mm,\;850\;ml-{\phi}65\;mm\;and\;1100\;ml-{\phi}65\;mm$ bottle. Using the PP bag for cultivation, a square shaped bag was better than a round shaped and black square shaped in mycelial growth and yield of fruit body. The most suitable period of incubation was 35 days after inoculation at $22{\pm}2^{\circ}C$. When the incubation periods was decreased less than 35 days, the pileus formation and yields were very bad but a pinheading condition looked similar, For an optimal pinheading, turning upside down was better than standing and covering.

Effect of Rice Straw Heated in Water or 0.25N-NaClO$_2$ on the Nutrient Utilization of Diets in Chicks (증류수 혹은 0.25N NaClO$_2$에서 가열한 볏짚의 가금에서의 영양소이용성에 미치는 영향)

  • In order to observe an effect of the components of rice straw on the utilization of nutrient in chicks, the rice straw of 100g were digested in 800$m\ell$ of distilled water or 0.25N NaClO$_2$ at 135 C and in the pressure of 3.2kg/$\textrm{cm}^2$ by autoclave during 30, 60 and 120 minutes (water or NaClO$_2$-30, 60 and 120- RS). The contents of neutral detergent fiber(NDF), acid detergent fiber (ADF) and lignin were analysed in the washed and dried rice straw meal. Hatched single comb white Leghorn male chicks were fed with a commercial chick mash for the first 10 days and five kinds of experimetal diets for the next 8 days which contained 17.0% of wheat bran (basal), cellulose(cotton meal), nontreated RS, water-30-RS and NaClO$_2$ 30-RS, respectively. The water-30, 60 and 120-RS baa leased 9.7, 12.1 and 13.3% of dry matter, respectively, while NaClO$_2$-30-RS had similar contents of dry matter loss with those of water-30-RS, and NaClO$_2$-60 and 120-RS had tossed 1.5 times of dry matter comparing with those of water-60 and 120-RS, respectively. And the dry matter loss of the water-RS or NaClO$_2$-RS was mainly originated front the extractable cell contents and hemicellulose of the non-treated RS. Birds fed water-30-RS diets had higher body weight gain and lower feed conversion than those of birds fed non-treated and NaClO$_2$-30-RS diets during 8 days of experimental feeding. Also nitrogen balance and retention rate of birds fed water -30-RS was higher comparing with those of birds non-treated and NaClO$_2$-30-RS. And digestibility of crude fat had been shown a highering trend in birds water-30-RS. The rate of metabolizable energy (MEn) to gross energy (GE) of birds fed non-treated RS, water-30-RS and NaClO$_2$30-RS diets were 71.9, 72.9 and 70.4%, respectively, and energy intake per metaboic body size (kg 0.75) were reached to 307.3, 296.2 and 291.4 kcal per day, respectively. And daily protein retention per kg 0.75 were 1.647, 1.969 and 1.560g, respectively. Then 30.56kcal of MEn required for 1 g of protein retention in birds fed water-30-RS, which was lower thu 36.90 and 37.56 kcal of birds fed non-treated and NaClO$_2$-30- RS, respectively. The results seems to indicate that non-treated rice straw had a substance or characters which affect the energy unilization or protein retention of diets and which will be eliminated by boiling in water.

  • Anticarcinogenicity of 12 kinds of colored rice cultivars was investigated in vitro with Ilpumbyeo as a control. Pigment and hexane fractions were prepared from the rice brans through extraction with 80% ethanol containing 0.5% TFA or n-hexane to use subsequent evaluation of their anticarcinogenic effect by determining inhibitory activity against DNA strand scission and tumor promotion, as well as antimutagenic activity using SOS chromotest system. Antimutagenic activity of the pigment fractions was revealed to decrease in order of LK1B4-12-1-1 > Sanghaehanghyulla > HP833-1-3-1-1 > Chokoto14 > Jagwangdo > Jajin > Kilmheugmi > Tiwan-Tsi-C > Heugjinmi > Linsia-Shoea-Dau > Suweon425 > Suweon415. The activity of the hexane fractions decreased in order of LK1B4-12-1-1 > Sanghaehyanghyulla>Ilpunibyeo > HP833-1-3-1-1 > Chokoto14 > Jagwangdo > Jajin > Linsia-Shoea-Dau >Tiwan-Tsi-C > Heugjinmi > Suweon425, Kilimheugmi > Suweon415. Results showed that inhibitory activity against DNA strand scission in the pigment fractions decreased in order of HP833-1-3-1-1 > Tiwan-Tsi-C > Jajin > Chokoto14 > Suweon425 > Heugjinmi > LK1B4-12-1-1 > Suweon415 > Jagwangdo > Kilimheugmi > Linsia-Shoea-Dau > Sanghaehyanghyulla. On the other hand, the activity of the hexane fractions decreased in order of LK1B4-12-1-1 > Suweon 415 > Kilimheugmi > Chokoto14 > Heugjinmi > Jajin > Suweon425 > Tiwan-Tsi-C > Sanghaehyanghyulla a Linsia-Shoea-Dau > Jagwangdo > HP833-1-3-1-1 > Ilpumbyeo. Experiment using the short-term assay for promotor substance showed that inhibitory activity contained in the pigment fractions against rumor promotor TPA decreased in order of Linsia-Shoea-Dau > Kilimheugmi > HP833-1-3-1-1, Suweon425 > Heugjinmi > Jajin > Chokoto14 > Sanghaehyanghylla > Suweon 415 > LK1B4-12-1-1 > Tiwan-Tsi-C > Jagwangdo.
