  • A total of 187 endophytic fungi were isolated from 11 plant species, which were collected from 11 locations in Korea. Their antifungal activities were screened in vivo by antifungal bioassays after they were cultured in potato dextrose broth and rice solid media. Antifungal activity against plant pathogenic fungi such as Magnaporthe grisea(rice blast), Corticium sasaki(rice sheath blight), Botrytis cinerea(tomato gray mold), Phytophthora infestans(tomato late blight), Puccinia recondita(wheat leaf rust), and Blumeria graminis f. sp. hordei(barley powdery mildew) was determined in vivo by observing the inhibition of plant disease development. Twenty(11.7%) endophytic fungi fermentation broths were able to control, by more than 90%, at least one of the six plant diseases tested. Among 187 liquid broths, the F0010 strain isolated from Abies holophylla had the most potent disease control activity; it showed control values of more than 90% against five plant diseases, except for tomato late blight. On the other hand, fourteen(7.5%) solid culture extracts exhibited potent disease control values of more than 90% against one of six plant diseases. The screening results of this study strongly suggested that metabolites of plant endophytic fungi could be good potential sources for screening programs of bioactive natural products.

  • The mitogen-activated protein kinase (MAPK) signaling cascade is critical for regulating plant defense systems against various kinds of pathogen and environmental stresses. One component of this cascade, the MAP kinase kinases (MAPKK), has not yet been shown to be induced in plants following biotic attacks, such as those by insects and fungi. We describe here a gene coding for a blast (Magnaporthe grisea)- and insect (Nilaparvata lugens)-responsive putative MAPK kinase, OmMKK1 (Oryza minuta MAPKK 1), which was identified in a library of O. minuta expressed sequence tags (ESTs). Two copies of OmMKK1 are present in the O. minuta genome. They encode a predicted protein with molecular mass 39 kDa and pI of 6.2. Transcript patterns following imbibition of plant hormones such as methyl jasmonic acid (MeJA), ethephone, salicylic acid (SA) and abscisic acid (ABA), as well as exposure to methyl viologen (MV), revealed that the expression of OmMKK1 is related to defense response signaling pathways. A comparative analysis of OmMKK1 and its O. sativa ortholog OsMKK1 showed that both were induced by stress-related hormones and biotic stresses, but that the kinetics of their responses differed despite their high amino acid sequence identity (96%).

  • A wide variety of pathogens are known io be seedborne, carried either as infectious mycelium internally or as contaminants on the seed coat. When seed is infected with a pathogen, the seed nay be rendered nonviable or it may remain viable but produce weak seedling. In some cases, the Infected seedling nay not be severely weakened, but nay serve as a source of primary inoculum within a community of plants. A recent problem nay be the dissemination of seedborne pathogens occurring as a result of the massive movements of seed, as a part of the 'Green revolution' Disease of great danger to agriculture may be introduced with seed from other parts of world. Seed treatment with organic mercury compounds in liquid form had become popular since about 1955. Organic mercury compounds contributed considerably to the increase in production of many crops and vegetables. In 1975, however, the use of organic mercury compound was forbidden because of doubts regarding their residual mammalian toxicity in agricultural products. Benomyl-thiram mixture, thiophanate methyl-;hiram mixture and TCMB have now been registered as seed disinfectants for the use of rice blast, brown spot and Bakanae disease. Oxathiinsthiram mixture has been registered as seed disinfectant for barley and wheat loose smut and leaf stripe of barley. Agricultural techniques have made such rapid progress that the nursery methods changed from the use of paddy nursery to box nursery designed for machine-transplanting. The spread of rice transplanting machines has caused increase of seedborne diseases. Among seedborne diseases, Bakanae disease has remarkably increased and causes much damage recently. In order to counter this trend, seed disinfectants must also be diversified. First, effective non-selective disinfectants need to be developed, and second, appropriate control methods always need to be prepared in parallel with the development of new techniques for cultivation.

  • Various microorganisms live in soil, of which those colonizing rhizosphere interact with nearby plants and tend to develop unique microbial communities. In this study, we isolated diverse actinomycetes from rhizosphere of rice (Oryza sativa L.) cultivated in fertilized (APK) and non-fertilized (NF) paddy soils, and investigated the diversity and antimicrobial activity of them. Using four kinds of selective media, 152 isolates were obtained from the soil samples and identified by determining 16S rRNA gene sequence. All of the isolates showed 99.0%~100.0% similarities with type strains and were classified into six genera: Dactylosporangium, Micromonospora, Kitasatospora, Promicromonospora, Streptomyces and Streptosporangium. Most of the isolates, 143 isolates, were classified into the genus Streptomyces. Additionally, many isolates had antimicrobial activity against plant pathogens, especially Magnaporthe oryzae (rice blast pathogen) in fungi. These findings demonstrated that rice rhizosphere can be a rich source of antagonistic actinomycetes producing diverse bioactive compounds.
