고사리 열수 추출물로부터 보체계 활성화 산성 다당의 분리 및 특성 (Isolation and Characterization of Acidic Polysaccharides Activating Complement System from the Hot Water Extracts of Pteridium aquilinum var. latiusculum)

  • From the hot water extract of bracken(Pteridium aquilinum var. latiusculum), a Korean win edible plant, anti-complementary acidic polysaccharides were Isolated. Crude polysaccharide fraction(HPA-1) was obtain ed by methanol reflux, ethanol precipitation, dialysis, and lyophilization. HPA-1 contained 81.80% of total sugar, 30.40% of uronic acid, and 15.60cA of protein. HPA 1 was purified consecutively by cetavlon fractionation and chromatography including ion exchange nth DEAE-Sepharose CL 6B and gel permeation with Sephadex G-100 and Sepharose CL-6B. HPA-2- IVa and HPA-Va-2 were nearly homogeneous on HPLC and had 500,000 and 560,000 daltons of molecular weights, respectively. HPA-2-Wa consisted of fucose, galacturonic acid, and glucuronic acid at the molar ratio of 1.40 : 0.97 : 1.88. HPA-2-Va 2 was composed of rhamnose, galactose, and galacturonic acid at the molar ratio of 1.00 : 1.38 : 1.39. The polysaccharides were found to activate the C3 component both In the presence and In the absence of Ca2+ through the crossed-immunoelectrophoresis suggesting that those Involved in both classical and alternative complement pathway.

Purification and Characterization of Quercitrin-Hydrolyzing ${\alpha}$-L-Rhamnosidase from Fusobacterium K-60, a Human Intestinal Bacterium

  • An ${\alpha}$-L-rhamnosidase (EC 3.2.1.40.), which transforms quercitrin to quercetin, was purified from Fusobacterium K-60, a human intestinal anaerobic bacterium. The specific activity of the purified ${\alpha}$-L-rhamnosidase was 2.89 mol/min/mg protein. ${\alpha}$-L-Rhamnosidase, whose molecular size was 170 kDa by gel filtration, was composed of four subunits ($M_r$ 41,000 Da) with pI and optimal pH values of 5.2 and 5.5-7.0, respectively. The apparent $K_m$ and $V_{max}$ values for p-nitrophenyl-${\alpha}$-L-rhamnopyranoside and quercitrin were determined to be 0.057 mM and 3.4 mol/min/mg, and 0.077 mM and 5.0 mol/min/mg, respectively. This enzyme was strongly inhibited by $Cu^{2+},\;Mn^{2+}$, L-rhamnose, and p-chlormercuriphenylsulfonic acid. These findings suggest that the biochemical properties and substrate specificity of the purified enzyme are different from those of the previously purified ${\alpha}$-L-rhamnosidase. This is the first reported purification of quercitrin-hydrolyzing ${\alpha}$-L-rhamnosidase from intestinal bacteria.

Electrochemical Sensor for the Selective Determination of Prindopril Based on Phosphotungestic Acid Plastic Membrane

  • A novel PVC membrane sensor for perindopril based on perindopril-phosphotungstate ion pair complex was prepared. The influence of membrane composition (i.e. percent of PVC, plasticizer, ion-pair complex, and kind of plasticizer), inner solution, pH of test solution and foreign cations on the electrode performance was investigated. The optimized membrane demonstrates Nernstian response ($30.9{\pm}1.0$ mV per decade) for perindopril cations over a wide linear range from $9.0{\times}10^{-7}$ to $1{\times}10^{-2}$ M at $25^{\circ}C$. The potentiometric response is independent of the pH in the range of 4.0-9.5. The proposed sensor has the advantages of easy preparation, fast response time. The selectivity coefficients indicate excellent selectivity for perindopril over many common cations (e.g., $Na^+$, $K^+$, $Mg^{2+}$, $Cu^{2+}$, $Ni^{2+}$, rhamnose, maltose, glycine and benzamide. The practical applications of this electrode was demonstrated by measuring the concentrations of perindopril in pure solutions and pharmaceutical preparations with satisfactory results.

경북지방 환돈에서 분리한 Salmonella Typhimurium의 생화학적 성상 및 약제내성 패턴 (Pattern of antimicrobial resistance and biochemical characteristics of Salmonella Typhimurium isolated from diseased pigs in Gyeongbuk province)

  • Salmonella Typhimurium is a virulent pathogen for human and animal. We studied serotypes, biochemical characteristics, and antimicrobial resistance of S. Typhimurium isolated from diseased pigs in Gyeongbuk province over 1998 to 2008. One hundred sixteen isolates were identified as S. Typhimurium by biochemical characteristics and serotypes from 90 farms. The biochemical characteristics of S. Typhimurium isolates was production of $H_2S$, indole-negative, fermentation of mannitol, dulcitol, sorbitol, inositol, rhamnose, and maltose, and ornithine decarboxylase. At antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, cefepime, ciprofloxacin, while were highly resistant streptomycin, cephalothin, enrofloxacin, nalidixic acid, apramycin, chloramphenicol, tetracycline. The isolates were divided into 65 resistant patterns and 47 of the isolates were shown as a DT104 ASSSuT resistant phenotype.

  • This study was conducted to investigate the characteristics of nonstarchy polysaccharides in Korean ginseng, (Panax ginseng C.A. Meyer). The results obtained are as follows. 1. The total sugar content of ginseng roots were decreased with increasing the cultural period. On the other hand, the crude fiber content was increased with that of the ginseng leaves or stems. But the crude fiber in root was much less than that of leaves and stems. 2. The dietary fiber content of ginseng root on 5 years old was 14.20% as neutral detergent fiber, 9.08% as acid detergent fiber, hemicellulose 5.12%, cellulose 7.98% and lignin of 1.10%, respectively. 3. Much more pectin was found in ginseng roots which was cultivated for shooter Period. And it was contained much more in the root than in the leaves and stems. 4. ginseng hemicellulose content in root was 5% to 10%. It was decreased with increasing: cultivated period. Hemicellulose was constituted of xylose, arabinose, glucose, rhamnose and xylose of these sugars was the predominant. 5. X-ray diffraction Pattern of ginseng cellulose showed maximum intensity at tile interplanar angle of 4.1$^{\circ}$.

병계로부터 분리된 Salmonella pullorum의 생화학적 특성과 항균성 약제에 대한 감수성 (Biological Properties and Antimicrobial Drug Susceptibilities of Salmonella pullorum Isolated from Diseased Chickens)

  • The present study was conducted to investigate biochemical properties, antimicrobial drug susceptibilities and epidemiology of 71 stranins of Salmonella pullorum isolated from about 110 diseased chickens of 23 poultry farms located in Cheongjoo, Cheongweon and Koesan county, Chungbuk province, from August 1991 to March 1993. The isolates were identified as S. pullorum by serological and biochemical means. S. pullorum were mostly isolated in chicken under 3weeks of age, and also isolated in 58, 72 days and 23 weeks of age. According to breeds, most of the cultures were isolated in colored broiler chicken (14 to 23 cases), and also variously isolated in native chicken, white broiler chicken, black bone chicken and laying hen. According to organs of diseased chickens, most of the cultures were isolated in liver (37 to 71 strains), and also variously isolated in spleen, lungs, blood, heart, oviduct and brain. According to media used for primary culture from organs, most of the cultures were isolated purely with SS and BHI medium. The majority of biochemical properties of S. pullorum isolated from diseased chickens were identical to those of the standard strains, but in the properties of rhamnose, and arabinose fermentation, some isolates were negative in spite of positive in those of standard S. pullorum. All the isolates were highly susceptible to colistin, amikacin, kanamycin, gentamicin, carbenicillin, ampicillin, sulfamethxazole, cephalothin, tetracycline and piperacillin regardless of isolated years, but no susceptible to penicillin, erythromycin, vancomycin, tylosin and novobiocin.

Characterization of Soluble Dietary Fibers from Wax Gourd (Benincasa hispida) Pulp and Peel

  • The physicochemical and in vitro physiological properties of soluble dietary fiber (SDF) from wax gourd (Benincasa hispida) pulp and peel were investigated. The pulp was composed of 11.4% SDF and 24.3% insoluble dietary fiber (IDF), while the peel contained 3.2% SDF and 43.3% IDE The predominant sugar in the SDF of the wax gourd pulp and peel was uronic acid, followed by galactose and rhamnose. The SDFs from the wax gourd pulp and peel gave similar elution patterns, with 4 main neutral sugar and uronic acid peaks eluted by 0.4, 0.5, 1, and 2 M ammonium acetate buffer. The pulp SDF had a much higher glucose retardation index (GRI) than the peel SDF for all measurement times. The pulp SDF showed strong growth-inhibiting activity against Escherichia coli and Clostridium perfringens, whereas the peel SDF produced strong growth-promoting activity against Bifidobacterium longum, Bifidobacterium infantis, and Lactobacillus brevis when compared to glucose.

동결건조 및 열풍건조 방법에 따른 마의 성분과 물리적 성질 변화 (Changes in Chemical Components and Physical Properties with Freeze Drying and Hot Air-Drying of Dioscorea batatas)

  • Chemical components and some physical proeprties of fresh, defrosted and dried yams(Dioscoreab batatas) were determined to obtain basic data for high quality yam processing. Fresh yam showed 81.79% moisture and 15.24% N-free extract. The cohesiveness and adhesiveness of defrosted yam homogenate were significantly higher than those of fresh yam homogenate. Free sugars of yam were mainly composed of sucrose, rhamnose, fructose and glucose, and the contents of fructose and glucose were apparently decreased by hot air drying. Linoleic(45.64%), oleic(8.32%), and arachidic acids(7.40%) were major fatty acids of yam. Hot air drying caused a decrease in unsaturated fatty acids and an increase in saturated acids. Hot air-dried yam powder showed higher gelatinization properties than freeze-dryed yam powder, such asinitial pasting temperature, temperature at maximum viscosity, and viscosity at different parameters. Hot air-dried yam powder showed higher Hunter parameter b and ΔE values and lower L value than freeze-dried one.

Biological Activities on Phenolic Compounds of Japanese anise (Illicium anisatum L) Extracts

  • In this paper, we have isolated six phenolic compounds, such as (+)-catechin (1), taxifolin (2), taxifolin-3-O-${\beta}$-D-(+)-xylose (3), quercetin (4), quercetin-3-O-${\alpha}$-L(+)-rhamnose (quercitrin) (5), apigenin-8-C-rhamnosyl-(1'''${\rightarrow}$2'')-glucoside (2''-O-rhamnosylvitexin) (6) from the EtOAc(Ethyl Acetate) and $H_2O$ soluble fractions of Japanese anise(Illicium anisatum L) leaves and twigs. Also, we have evaluated antioxidative and antiviral activity for each isolated compound. The antioxidative test was DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. According to the experimental results, all of the isolated compounds indicated the increased radical scavenging activities as the concentration increases and most of the isolated compounds indicated generally good antioxidative values compare to the controls, ascorbic acid and ${\alpha}$-tocopherol. In the antiviral activities, all of the isolated compounds had no potentials in rhinovirus 1B (HRV 1B). But in enterovirus 71 (EV 71) and Influenza virus A/PR/8 (Influenza PR8), only quercetin (4) indicated the good antiviral activity compare to the control. Based on the above results, we found that the phenolic compounds of Japanese anise may be applied for one of the natural biomass sources that can be used as an antioxidant and an antiviral substance.

저장(貯藏)간장의 생화학적(生化學的) 연구(硏究) (The Biochemical Studies on Stored Soy-sauce)

  • 12년(年) 묵은 개량식(改良式) 간장과, 20년(年) 묵은 재래식(在來式) 간장 및 7년(年)묵은 재래식(在來式) 간장에 대(對)하여 화학적(化學的) 조성(組成)을 분석(分析)하고 microflora를 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. 1. 간장 맛에 영향을 주는 일반성분(一般成分)의 분석결과(分析結果)는 다음과 같았다. a) 오래 묵은 간장일수록 재래식(在來式) 및 개량식(改良式) 간장을 불문(不問)하고 맛을 좌우(左右)하는 성분(成分)인 유기산(有機酸), 유리환원당(遊離還元糖) 및 유리(遊離) 아미노산(酸)의 함량(含量)이 증가(增加)되었다. b) 유기산(有機酸)에 있어서 오래 묵은 간장일수록 비휘발성(非揮發性) 유기산(有機酸)은 증가(增加)하고 휘발성(揮發性) 유기산(有機酸)은 감소(減少)하며, 총산(總酸)은 비휘발성(非揮發性) 유기산(有機酸)에 의(依)하여 지배(支配)되어 있음을 알았다. c) 식염농도(食鹽濃度)는 오래 묵은 간장일수록 감소(減少)되었다. 2. 저장(貯藏)간장중(中)의 microflora를 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. a) 일반세균(一般細菌)은 묵은 간장중(中)에서 pH의 강하(降下) 및 식염농도(食鹽濃度)의 영향(影響)으로 생존(生存) 불능(不能) 상태(狀態)였다. b) 내염성(耐鹽性) 유산균(乳酸菌)은 저장(貯藏)함에 따라 식염농도(食鹽濃度)는 강하(降下)하기는 하나 pH의 강하(降下)로 거의 생존(生存)이 불능상태(不能(狀態)였다. c) 내염성효모(耐鹽性酵母)는 묵힐수록 어느 시기 이후는 식염농도(食鹽濃度)의 강하(降下) 및 pH 직강하(直降下)로 생존(生存)됨을 알았다. 3. Paper chromatography 및 colorimetry에 의(依)하여 유리(遊離) 아미노산(酸)을 분석(分析)한 결과(結果)는 다음과 같았다. a) 재래식(在來式) 간장에 있어서 7년(年)묵은 간장에 대하여 20년(年) 묵은 간장중에는 aspartic acid, glutamic acid, serine, valine, leucine, lysine, histidine, methionine이 증가(增加)하였고, alanine, tyrosine phenylalanine, cystine이 감소(減少)하였다. b) 개량식(改良式) 간장에 있어서는 colorimetry로 정량(定量)된 산(酸)의 양적(量的)인 변화(變化)는 햇간장에 대하여 12년(年)된 간장중의 methionine, tyrosine, histidine는 감소(減少)하고, phenylalanine는 증가(增加)하며 cystine는 별차(別差) 없었다. 4. Paper chromatography에 의하여 유리당류(遊離糖類)를 분석(分析)한 결과(結果)는 다음과 같았다. a) 12년(年)묵은 개량식(改良式) 간장중(中)에서 galactose, glucose, arabinose, xylose, rhamnose, maltose, unknown 1를 검출(檢出)하였고, 정량(定量)한 당류(糖類)의 함량(含量)은 maltose와 미지당(未知糖)을 제외(除外)하고 이상(以上)의 순위(順位)와 같았다. b) 20년(年), 7년(年) 묵은 재래식(在來式) 간장에서 galactose, arabinose, xylose, glucose, rhamnose를 다같이 분리(分離)하였으며 그들의 함량(含量)은 이상(以上)의 순위(順位)와 같았다. c) 7년(年) 및 20년(年) 묵은 재래식(在來式) 간장중(中)에서 햇간장중(中)에 별로 없든 glucose가 생성(生成)되었다. 5. Paper chromatograhpy에 의하여 유기산(有機酸)을 분석(分析)한 결과(結果)는 다음과 같았다. a) Volatile acid i) 7년(年) 묵은 재래식(在來式) 간장중(中)에서 acetic acid, propionic acid, butyric acid를 분리(分離)하였으며, acetic acid가 제일 많았다. ii) 20년(年)된 재래식(在來式) 간장과, 12년(年)된 개량식(改良式) 간장중(中)에는 acetic acid가 주로 많았고, propionic acid, butyric acid는 흔적 정도(程度)로 분리(分離)되었다. iii) 간장은 오래 묵힐수록 고린냄새의 한성분(成分)인 propionic acid, butyric acid가 격감(激減)함을 알았다. b) Non-volatile acid i) 재래식(在來式) 간장에 있어서는 묵힘에 따라 생성(生成)된 것은 citric acid이고, 증가(增加)된 것은 lactic, malic, tartaric acid이고 감소(減少)된 것은 succinic, glycolic, fumaric, malonic acid이며 소실(消失)된 것은 glutaric, oxalic acid이다. ii) 개량식(改良式) 간장에 있어서는 묵을수록 생성(生成)된 것은 citric acid이고, 증가(增加)된 것이 latic, tartaric acid이며, succinic, malic, glycolic acid는 감소(減少)하였고 malonic, glutaric acid는 소실(消失)되었다. iii) 묵은 간장중(中)의 citric acid의 생성(生成)은 간장 맛을 좋게하는 인자(因子)가 될 것으로 생각된다. c) ${\alpha}-Keto$ acid i) 모든 묵은 간장중(中)에서 pyruvic, ${\alpha}-ketoglutaric$, acetoacetic(추정(推定)), oxaloacetic(추정(推定)) acid를 분리(分離)하였으며 양적(量的)인 순위(順位)는 pyruvic acid, ${\alpha}-ketoglutaric$ acid이고, 기타는 흔적 정도(程度)였다. ii) 모든 ${\alpha}-Keto$ acid는 간장이 묵을수록 격감(激減)함을 알았다.
