Calpains are a class of proteins that belong to the calcium-dependent, non-lysosomal cysteine proteases. There are three major types of calpains expressed in the skeletal muscle, namely, ${\mu}$-calpain, m-calpain, and calpain 3, which show proteolytic activities. Skeletal muscle fibers possess all three calpains, and they are $Ca^{2+}$-dependent proteases. The functional role of calpains was found to be associated with apoptosis and myogenesis. However, calpain 3 is likely to be involved in sarcomeric remodeling. A defect in the expression of calpain 3 leads to limb-girdle muscular dystrophy type 2A. Calpain 3 is found in skeletal muscle fibers at the N2A line of the large elastic protein, titin. A substantial proportion of calpain 3 is activated 24 h following a single bout of eccentric exercise. In vitro studies indicated that calpain 3 can be activated 2-4 fold higher than normal resting cytoplasmic [$Ca^{2+}$]. Characterization of the calpain system in the developing muscle is essential to explain which calpain isoforms are present and whether both ${\mu}$-calpain and m-calpain exist in differentiating myoblasts. Information from such studies is needed to clarify the role of the calpain system in skeletal muscle growth. It has been demonstrated that the activation of ubiquitous calpains and calpain 3 in skeletal muscle is very well regulated in the presence of huge and rapid changes in intracellular [$Ca^{2+}$].
Hesperidin, the most abundant polyphenolic compound found in citrus fruits, has been known to possess neuroprotective, sedative, and anticonvulsive effects on the nervous system. In a recent electrophysiological study, it was reported that hesperidin induced biphasic change in population spike amplitude in hippocampal CA1 neurons in response to both single spike stimuli and theta-burst stimulation depending on its concentration. However, the precise mechanism by which hesperidin acts on neuronal functions has not been fully elucidated. Here, using whole-cell patch-clamp recording, we revealed that hesperidin did not affect excitatory synaptic activities such as basal synaptic transmission and theta-burst LTP. Moreover, in a current injection experiment, spike number, resting membrane potential and action potential threshold also remained unchanged. Taken together, these results indicate that the effects of hesperidin on the neuronal functions such as spiking activity might not be attributable to either modification of excitatory synaptic transmissions or changes in membrane excitability in hippocampal CA1 neuron.
The playas of the Southern High Plains (SHP) of Texas, USA are an important habitat for over one million wintering waterfowl. However, the recent trend toward the modification of playas for agricultural use is threatening winter habitat of waterfowl in this region. Diurnal activity budgets of wintering mallards (Anas platyrhynchos) were conducted from 1 October to 31 March, 1983-1984, and 1984-1985) at three habitat types; steep-sided pits, terraced pits, and open lakes. All seven activity patterns (feeding, locomotion, resting, comfort, courtship, alert, and agonistic) were different (P<0.05) among the three habitat types for wintering mallards on the SHP of Texas. Terraced pits supported more feeding activity (27.8%) (P<0.001) than steep-sided pits (11.2%) or open lakes (2.6%) due to their abundance of natural seeds and aquatic invertebrates. Hens (17.5%) fed more than drakes (11.7%) (P<0.05). Locomotion (32.2%) and alert (2.8%) behavior across the three habitat types showed the highest level during the early morning (6:00-9:00 AM). Paired mallards rested more (37.9%) than unpaired mallards (25.8%) (P<0.05). Agonistic activity was highest (2.4%) in terraced pits throughout the season.
The metabolic pathway of eugenol degradation by thermophilic Geobacillus sp. AY 946034 strain was analyzed based on the lack of data about eugenol degradation by thermophiles. TLC, GC-MS, and biotransformation with resting cells showed that eugenol was oxidized through coniferyl alcohol, and ferulic and vanillic acids to protocatechuic acid before the aromatic ring was cleaved. The cell-free extract of Geobacillus sp. AY 946034 strain grown on eugenol showed a high activity of eugenol hydroxylase, feruloyl-CoA synthetase, vanillate-O-demethylase, and protocatechuate 3,4-dioxygenase. The key enzyme, protocatechuate 3,4-dioxygenase, which plays a crucial role in the degradation of various aromatic compounds, was purified 135-fold to homogeneity with a 34% overall recovery from Geobacillus sp. AY 946034. The relative molecular mass of the native enzyme was about $450{\pm}10$ kDa and was composed of the non-identical subunits. The pH and temperature optima for enzyme activity were 8 and $60^{\circ}C$, respectively. The half-life of protocatechuate 3,4-dioxygenase at the optimum temperature was 50 min.
Since conflicting results have been reported on non-specific immune response in type 1 diabetes, this study evaluates polymorphonuclear neutrophil (PMN) functions in the infection free Long Evan diabetic rats (type 1) by using tests that include: polarization assay, phagocytosis of baker's yeasts (Saccharomyces cerevisiae) and nitroblue tetrazolium (NBT) dye reduction. Polarization assay showed that neutrophils from diabetic rats were significantly activated at the basal level compared to those from the controls (p < 0.001). After PMN activation with N-formyl-methionyl-leucyl-phenylalanine (FMLP), control neutrophils were found to be more polarized than those of the diabetic neutrophils and the highest proportions of polarization were found to be 67% and 57% at $10^{-7}\;M$ FMLP, respectively. In the resting state, neutrophils from the diabetic rats reduced significantly more NBT dye than that of the controls (p < 0.001). The percentages of phagocytosis of opsonized yeast cells by the neutrophils from control and diabetic rats were 87% and 61%, respectively and the difference was statistically significant (p < 0.001). Evaluation of the phagocytic efficiency of PMNs revealed that control neutrophils could phagocytose $381{\pm}17$ whereas those from the diabetic rats phagocytosed $282{\pm}16$ yeast cells, and the efficiency of phagocytosis varied significantly (p < 0.001). Further, both the percentages of phagocytosis and the efficiency of phagocytosis by the diabetic neutrophils were inversely related with the levels of their corresponding plasma glucose (p = 0.02; r = -0.498 and p < 0.05; r = -0.43, respectively), which indicated that increased plasma glucose reduced the phagocytic ability of neutrophils. Such relationship was not observed with the control neutrophils. These data clearly indicate that PMN functions are altered in the streptozotocin (STZ) - induced diabetic rats, and hyperglycemia may be the cause for the impairment of their functions leading to many infectious episodes.
The SCBFC was composed of bilayered cathode, the outside of which was modified with $Fe^{3+}$ (graphite-Fe(III) cathode) and the inside of which was porcelain membrane, and of an anode which was modified with $Mn^{4+}$ (graphiteMn(lV) anode). The graphite-Fe(III), graphite-Mn(IV), and porcelain membrane were designed to have micropores. The outside of the cathode was exposed to the atmosphere and the inside was contacted with porcelain membrane. In all SCBFCS the graphite-Fe(III) was used as a cathode, and graphite-Mn(IV) and normal graphite were used as anodes, for comparison of the function between normal graphite and graphite-Mn(IV) anode. The potential difference between graphite-Mn(IV) anode and graphite-Fe(III) cathode was about 0.3 volt, which is the source for the electron driving force from anode to cathode. In chemical fuel cells composed of the graphite-Mn(IV) anode and graphite-Fe(III) cathode, a current of maximal 13 mA was produced coupled to oxidation of NADH to $NAD^{+}$ the current was not produced in SCBFC with normal graphite anode. When growing and resting cells of E. coli were applied to the SCBFC with graphite-Mn(IV) anode, the electricity production and substrate consumption were 6 to 7 times higher than in the SCBFC with normal graphite anode, and when we applied anaerobic sewage sludge to SCBFC with graphite-Mn(IV) anode, the electricity production and substrate consumption were 3 to 5 times higher than in the SCBFC with normal graphite anode. These results suggest that useful electric energy might possibly be produced from SCBFC without electron mediators, electrode-active bacteria, and extra energy consumption for the aeration of catholyte, but with wastewater as a fuel.
Objectives : Hindered barrier function of vascular endothelium has been implicated in the initiation and progression of degenerative vascular diseases such as atherosclerosis. In this study, the effect of Sunghyangchungisan(SHCS) as a protectant against oxidant-induced destruction of endothelial barrier function was assessed. Methods : Toward this end, endothelial cells derived from the human umbilical vein were cultured as monolayers on permeable membrane filters. Endothelial permeability was monitored by measuring transendothelial electrical resistance and movement of low density lipoprotein (LDL) across the endothelial monolayer. Results : Along with increased movement of LDL, $H_2O_2$-induced increase in endothelial permeability was paralleled by a decrease in transendotheliaI electrical resistance. The effect of $H_2O_2$ was mimicked by phorbol 12-myristate 13-acetate (PMA), a potent activator of proteinkinase C. Calphostin-C, a protein kinase C inhibitor, effectively blocked the increase in endothelial permeability induced by $H_2O_2$ or PMA, indicating that activation of protein kinase C is associated with the $H_2O_2-induced$ permeability change. SHCS effectively protected the endothelial monolayer against $H_2O_2-induced$ increase in permeability, whereas, it did not affect PMA-induced change. Forskolin, a potent activator of adenylyl cyclase, antagonized $H_2O_2$ to increase endothelial permeability. In addition, in ${H_2O_2}-treated$ cens, intracenular cAMP concentration was significantly decreased, indicating that impaired cAMP production as well as activation of proteinkinase C is a mechanism underlying ${H_2O_2}>-induced$$H_2O_2$ with regard to its effect on intracellular cAMP content. However, SHCS itself did not affect resting cAMP concentration in endothelial cells. Conclusions : These results suggest that SHCS might operate as an effective protectant against oxidant-induced destruction of endothelial barrier function. The mechanism does not appear to involve direct interaction with protein kinase C- or cAMP-associated signaling mechanism.
By using DL-acetonitrile as enzyme inducer, 90 bacteria were isolated from a field soil. Among the isolated strains, the strain WJ-003 showed the highest activity for production of D-lactic acid from DL-lactonitrile, and was partially identified as Pseudomonas sp. The production condition of D-lactic acid from DL-lactonitrile using resting cells as an enzyme source was optimized as follows: the reaction mixture contained 10 mM of DL-lactonitrile, 20 g of wet cells in 11 of 20 mM potassium phosphate buffer (pH 7.0) and the reaction was carried out at $30^{\circ}C$. After 18 h of reaction, 0.843 g/l of D-lactic acid was produced which corresponded to a conversion ratio of 93.7% and an optical purity of 99.8%. Additionally, when 10 mM of DL-lactonitrile was added once more to the reaction mixture at 14 h, 1.64 g/1 of D-lactic acid was produced after 28 h. In this experiment, the conversion ratio was 91.1% and optical purity 99.8%.

The primary cilium is a non-motile microtubule-based organelle that protrudes from the surface of most human cells and works as a cellular antenna to accept extracellular signals. Primary cilia assemble from the basal body during the resting stage ($G_0$ phase) and simultaneously disassemble with cell cycle re-entry. Defective control of assembly or disassembly causes diverse human diseases including ciliopathy and cancer. To identify the effective compounds for studying primary cilium disassembly, we have screened 297 natural compounds and identified 18 and 17 primary cilium assembly and disassembly inhibitors, respectively. Among them, the application of KY-0120, identified as Brefeldin A, disturbed Dvl2-Plk1-mediated cilium disassembly via repression of the interaction of $CK1{\varepsilon}-Dvl2$ and the expression of Plk1 mRNA. Therefore, our study may suggest useful compounds for studying the cellular mechanism of primary cilium disassembly to prevent ciliopathy and cancer.
Some blue-green algal species such as those in the genus Anabaena causing severe algal blooms can produce akinetes, resting spores, in aquatic ecosystems. Germinated akinetes staying in the sediment as "seed banks" grow into vegetative cells under favorable conditions of light intensity, nutrient, and temperature. Therefore, akinete plays an important role in forming the nuisance bloom. However, little information is available in the ecological study of akinetes compared to that of vegetative cells in Korea. This review reports ecological and physiological characteristics of akinetes, especially of the blue-green algal genus Anabaena. We also suggest the feasible area of akinetes in the freshwater ecosystems. We expect that the suggested studies associated with akinetes will contribute to further understanding the life cycle and ecology of Anabaena and other algae.
