Optimization of Ingredient Mixing Ratio for Preparation of Chinese Radish (Raphanus sativus L.) Jam (무 잼 재료 혼합비율의 최적화)

  • This study was performed to find the optimum ratio of ingredients in the Chinese radish jam. The experiment was designed according to the RSM (response surface methodology), which included 18 experimental points with 4 replicates for three independent variables (sugar $45{\sim}70%$, pectin $0.5{\sim}2.0.%$, citric acid $0.2{\sim}0.5%$). The compositional and functional properties of the prepared products were measured, and these values were applied to the mathematical models. By use of F-test, sweetness, color values (L, a, b), and firmness were expressed by a linear model, while the sensory characteristics (color, smell, taste, texture and overall acceptance) were by a quadratic model. In the numeric optimization, the optimal ingredient amounts were 53.7% sugar, 1.0% pectin, and 0.3% citric acid. And in the graphical optimization, 53.9% sugar, 1.0% pectin, and 0.3% citric acid; these data were equivalent to 0.6985 desirability, indicating that the values were almost equivalent to the numerical optimization points. The above results demonstrate the feasibility of Chinese radish jam, and therefore, the commercialization of a Chinese radish jam marketed as a functional food is deemed possible.

Acute Hypoxia Activates an ENaC-like Channel in Rat Pheochromocytoma (PC12) Cells

  • Cells can resist and even recover from stress induced by acute hypoxia, whereas chronic hypoxia often leads to irreversible damage and eventually death. Although little is known about the response(s) to acute hypoxia in neuronal cells, alterations in ion channel activity could be preferential. This study aimed to elucidate which channel type is involved in the response to acute hypoxia in rat pheochromocytomal (PC12) cells as a neuronal cell model. Using perfusing solution saturated with 95% $N_2$ and 5% $CO_2$, induction of cell hypoxia was confirmed based on increased intracellular $Ca^{2+}$ with diminished oxygen content in the perfusate. During acute hypoxia, one channel type with a conductance of about 30 pS (2.5 pA at -80 mV) was activated within the first 2~3 min following onset of hypoxia and was long-lived for more than 300 ms with high open probability ($P_o$, up to 0.8). This channel was permeable to $Na^+$ ions, but not to $K^+$, $Ca^+$, and $Cl^-$ ions, and was sensitively blocked by amiloride (200 nM). These characteristics and behaviors were quite similar to those of epithelial sodium channel (ENaC). RT-PCR and Western blot analyses confirmed that ENaC channel was endogenously expressed in PC12 cells. Taken together, a 30-pS ENaC-like channel was activated in response to acute hypoxia in PC12 cells. This is the first evidence of an acute hypoxia-activated $Na^+$ channel that can contribute to depolarization of the cell.

Dose-Response Relationship of Avian Influenza Virus Based on Feeding Trials in Humans and Chickens (조류인플루엔자 바이러스의 양-반응 모형)

  • This study aimed to determine dose-response (DR) curve of avian influenza (AI) virus to predict the probability of illness or adverse health effects that may result from exposure to a pathogenic microorganism in a quantitative microbial risk assessment. To determine the parametric DR relationship of several strains of AI virus, 7 feeding trial data sets challenging humans (5 sets) and chickens (2 sets) for strains of H3N2 (4 sets), H5N1 (2 sets) and H1N1 (1 set) from the published literatures. Except for one data set (study with intra-tracheal inoculation for data set no. 6), all were obtained from the studies with intranasal inoculation. The data were analyzed using three types of DR model as the basis of heterogeneity in infectivity of AI strains in humans and chickens: exponential, beta-binomial and beta-Poisson. We fitted to the data using maximum likelihood estimation to get the parameter estimates of each model. The alpha and beta values of the beta-Poisson DR model ranged 0.06-0.19 and 1.7-48.8, respectively for H3N2 strain. Corresponding values for H5N1 ranged 0.464-0.563 and 97.3-99.4, respectively. For H1N1 the parameter values were 0.103 and 12.7, respectively. Using the exponential model, r (infectivity parameter) ranged from $1.6{\times}10^{-8}$ to $1.2{\times}10^{-5}$ for H3N2 and from $7.5{\times}10^{-3}$ to $4.0{\times}10^{-2}$ for H5N1, while the value was $1.6{\times}10^{-8}$ for H1N1. The beta-Poisson DR model provided the best fit to five of 7 data sets tested, and the estimated parameter values in betabinomial model were very close to those of beta-Poisson. Our study indicated that beta-binomial or beta-Poisson model could be the choice for DR modeling of AI, even though DR relationship varied depending on the virus strains studied, as indicated in prior studies. Further DR modeling should be conducted to quantify the differences among AI virus strains.

A Comparative Study on Geotechnical System Response Probabilities of Fill Dams in Korea Using Internal Erosion Toolbox (내부 침식 평가 도구를 이용한 국내 필댐의 지반공학적 시스템 응답 확률에 대한 비교 연구)

  • In this study the geotechnical system response probabilities (SRP) of the failure modes caused by the interal erosion in three dams with different histories of defects in Korea were analyzed via the internal erosion toolbox developed by USACE. The calculated values of SRP were compared with the values reported for other dams in worldwide and also the statistics on the past failures of dams and embankments in Korea. The calculated values of SRP related to the internal erosion adjacent to the conduits or spillway structures are as high as in the order of $10^{-5}$ that is significantly greater than the SRP values related to other mechanisms of internal erosion. The statistical data showing that the interal erosion adjacent to the structure is the most frequent cause of the failure of the dam in Korea could be a collateral evidence of this finding. In contrast, the values of SRP related to the internal erosion through foundation, one of the major causes of dam failure in worldwide, are relatively as low as in the order of $10^{-7}$. This result could be comparable with the rareness of the failure cases of dam caused by the internal erosion through foundation in Korea.

Optimal design of nonlinear damping system for seismically-excited adjacent structures using multi-objective genetic algorithm integrated with stochastic linearization method (추계학적 선형화 방법 및 다목적 유전자 알고리즘을 이용한 지진하중을 받는 인접 구조물에 대한 비선형 감쇠시스템의 최적 설계)

  • Optimal design method of nonlinear damping system for seismic response control of adjacent structures is studied in this paper. The objective functions of the optimal design are defined by structural response and total amount of the dampers. In order to obtain a solution minimizing two mutually conflicting objective functions simultaneously, multi-objective optimization technique based on genetic algorithm is adopted. In addition, stochastic linearization method is embedded into the multi-objective framework to efficiently estimate the seismic responses of the adjacent structures interconnected by nonlinear hysteretic dampers without performing nonlinear time-history analyses. As a numerical example to demonstrate the effectiveness of the proposed technique, 20-story and 10-story buildings are considered and MR dampers of which hysteretic behaviors vary with the magnitude of the input voltage are considered as nonlinear hysteretic damper interconnecting two adjacent buildings. The proposed approach can provide the optimal number and capacities of the MR dampers, which turned out to be more economical than the uniform distribution system while maintaining similar control performance. The proposed damper system is verified to show more stable performance in terms of the pounding probability between two adjacent buildings. The applicability of the proposed method to the design problem for optimally placing semi-active control system is examined as well.

Seismic Evaluation of Steel Moment Frame Buildings based on Different Response Modification Factors and Fundamental Periods (반응수정계수와 주기의 영향에 대한 철골모멘트저항골조 건물의 내진성능평가)

  • This study was performed to evaluate the effect of Response modification factors (R-factor) in 3-, 9- and 20- story steel Moment Resisting Frame (MRF) buildings. Each structure was designed using a R-factor of 8, as tabulated in the 2000 International Building Code provision (IBC 2000) and Korea Building Code (KBC) 2008. In order to evaluate the maximum and minimum performance expected for such structures, an upper bound and lower bound design were adopted for each model. Next, each analytical model was designed using different R-factors (8, 9, 10, 11, 12) and four different structural periods with the original fundamental period. For a detailed case study, a total of 150 analytical models were subjected to 20 ground motions representing a hazard level with a 2% probability of being exceeded in 50 years. In order to evaluate the performance of the structures, static push-over and non-linear time history analysis (NTHA) were performed, and displacement ductility demand was investigated to consider the ductility capacity of the structures. The results show that the dynamic behaviors for the 3- and 9-story buildings are relatively stable and conservative, while the 20-story buildings show a large displacement ductility demand due to dynamic instability factors.   • Due to man-made climate change, global abnormal weather phenomena have occurred, increasing disasters. Major developed countries(military) are preparing for disasters caused by extreme weather appearances. However, currently, disaster prevention plans and facilities have been implemented based on the frequency and intensity method based on statistical data, it is not enough to prepare for disasters caused by frequent extreme weather based on probability basis. The U.S. and British forces have been the fastest to take research and policy approaches related to climate change and the threat of disaster change, and are considering both climate change mitigation and adaptation. The South Korean military regards the perception of disasters to be storm and flood damage, and there is a lack of discussion on extreme weather and disasters due to climate change. In this study, the process of establishing disaster management systems in developed countries(the United States and the United Kingdom) was examined, and the response policies of each country(military) were analyzed using literature analysis techniques. In order to maintain tight security, our military should establish a response policy focusing on sustainability and resilience, and the following three policy approaches are needed. First, it is necessary to analyze the future operational environment of the Korean Peninsula in preparation for the environment that will change due to climate change. Second, it is necessary to discuss climate change 'adaptation policy' for sustainability. Third, it is necessary to prepare for future disasters that may occur due to climate change.

Probabilistic Risk Assessment of a Cable-Stayed Bridge Based on the Prediction Method for the Combination of Failure Modes (붕괴모드 조합 예측법에 의한 PSC사장교의 위험도평가)

  • Probabilistic Risk Assessment considering statistically random variables is performed for the preliminary design of a Cable Stayed Bridge, which is Prestressed Concrete Bridge consisted of cable and plate girders, based on the method of Working Stress Design and Strength Design. Component reliabilities of cables and girders have been evaluated using the response surface of the design variables at the selected critical sections based on the maximum shear, positive and negative moment locations. Response Surface Method (RSM) is successfully applied for reliability analyses for this relatively small probability of failure of the complex structure, which is hard to obtain through Monte-Carlo Simulations. or through First Order Second Moment Method that can not easily calculate the derivative terms of implicit limit state functions. For the analysis of system reliability, parallel resistance system consisting of cables and plate girder is changed into series connection system and the result of system reliability of total structure is presented. As a system reliability, the upper and lower probabilities of failure for the structural system have been evaluated and compared with the suggested prediction method for the combination of failure modes. The suggested prediction method for the combination of failure modes reveals the unexpected combinations of element failures in significantly reduced time and efforts compared with the previous permutation method or system reliability analysis method, which calculates upper and lower bound failure probabilities.

  • Lithium Fluoride (LiF; TLD-100) crystal chips are normally used as thermolu minescence dosimeters (abbreviated as NC-100) for estimating the absorbed dose to the skin of a patient or in a solid water phantom undergoing radiotherapy with megavoltage photon (6 and 15MV) beams. In general, investigation has revealed a reduction in the sensitivity of NC-100 chips after many runs through heating cycles. A TLD-100 chip laminated with gold plate (140${\mu}{\textrm}{m}$) on the upper surface layer of its face toward the photon beam (abbreviated as GC-100) has properties different from that of a NC-100 chip activated by incident photons and contaminant electrons with various lower energies coming from the gantry head and air. Activation of the valence band electrons of GC-100 chips by incident photons, positrons and electrons-which come from the gold plate by mainly pair production process and partly from Compton scattering-results in more enhanced signal intensity, higher response per monitor unit, as well as a good linearity with monitor units and independence of dose rate. Since the electron beams (6 and 15 MeV) do not have the probability of pair production process with gold plate, there is only a small difference (about a 3.3% increase for 15 MeV) in the signal gaps in the TL readout for electron beams between GC- and NC-100 chips. The 3.3% increase is entirely due to the buildup caused by the 140 m gold plate. The sensitivity of GC-100 chips is much more susceptible to high energy photon beams than electron one because of pair production. The interaction of high energy photon with a material of high atomic number, such as the good plate in this case, results in a considerably significant probability of pair production. The gold plate on the NC-100 chips acts as not only an intensifier of their signals but also acts as a filter of contaminant electrons in therapeutic high energy X-ray beams.

Application of Rainfall Runoff Model with Rainfall Uncertainty (강우자료의 불확실성을 고려한 강우 유출 모형의 적용)

  • The effects of rainfall input uncertainty on predictions of stream flow are studied based extended GLUE (Generalized Likelihood Uncertainty Estimation) approach. The uncertainty in the rainfall data is implemented by systematic/non-systematic rainfall measurement analysis in Weida catchment, Germany. PDM (Probability Distribution Model) rainfall runoff model is selected for hydrological representation of the catchment. Using general correction procedure and DUE(Data Uncertainty Engine), feasible rainfall time series are generated. These series are applied to PDM in MC(Monte Carlo) and GLUE method; Posterior distributions of the model parameters are examined and behavioural model parameters are selected for simplified GLUE prediction of stream flow. All predictions are combined to develop ensemble prediction and 90 percentile of ensemble prediction, which are used to show the effects of uncertainty sources of input data and model parameters. The results show acceptable performances in all flow regime, except underestimation of the peak flows. These results are not definite proof of the effects of rainfall uncertainty on parameter estimation; however, extended GLUE approach in this study is a potential method which can include major uncertainty in the rainfall-runoff modelling.
