CLINICO-HAEMATOLOGICAL AND BIOCHEMICAL ALTERATIONS IN ETHYLENE GLYCOL INDUCED ACUTE NEPHROTOXICITY IN COW CALVES

  • Ethylene glycol was given orally in 6 crossbred male cow calves @ 12 ml/kg b wt for 2 days continuously to develop acute nephrotoxicity and monitor blood chemicals profile in affected calves. Progressive depression, hypersalivation, ataxia, incoordination, staggering gait, grinding of teeth, recumbency, coma, convulsions and death were prominent symptoms in affected calves. Respiration and pulse rates were increased whereas body temperature and rumen movements were low. Haematological investigations revealed increase in total erythrocyte count, platelets count and packed cell volume till death and total leukocyte count up to day 3 which decreased on day 4 and 5. These calves revealed azotaemia, reduction in calcium, chloride and potassium and rise in sodium and AST, ALT and alkaline phosphatase enzymes activity.

  • Hemorrhagic shock is a common cause of death in emergency rooms. Since the symptoms of hemorrhagic shock occur after shock has considerably progressed, it is difficult to diagnose shock early. The purpose of this study was to improve early diagnosis of hemorrhagic shock using a survival prediction model in rats. We measured ECG, blood pressure, respiration and temperature in 45 Sprague-Dawley rats, and then obtained a logistic regression equation predicting survival rates. Area under the ROC curves was 0.99. The Hosmer-Lemeshow goodness-of-fit chi-square was 0.86(degree of freedom=8, p=0.999). Applying the determined optimal boundary value of 0.25, the accuracy of survival prediction was 94.7%

  • Disulfiram (tetraethylthiuram disulphid) is used in the treatment of chronic alcoholism since it causes an unpleasant aversive reaction to alcohol. It works by inactivating hepatic aldehyde dehydrogenase, leading to pronounced rise in the acetaldehyde concentration when ethanol is metabolized. Acetaldehyde causes alcohol sensitivity, which involve vasodilation associated with feeling of hotness and facial flushing, increased heart rate and respiration rates, lowered blood pressure, nausea, headache. One of its metabolites, diethyldithiocarbamate (DDC) can inhibit the enzyme dopamine $\beta$-hydroxylase (DBH), this may account for the profound refractory hypotension and hypothermia seen with the disulfiram-ethanol reaction (DER), resulting from norepinephrine depletion. This report is presents the case of a patient we met, who presented with hypothermia caused by the disulfiram-ethanol reaction, and along with a brief review of the subject.

$^{13}C$ NMR Studies of Metabolic Pathways Regulated by HSP104 in Saccharomyces cerevisiae

  • HSP104 protein in Saccharomyces cerevisiae is known to provide thermotolerance when induced by various kinds of stresses, such as a mild heat shock, ethanol, and hypoxia. It helps cells survive at an otherwise lethal temperature. Mechanisms by which HSP104 protein works are yet to be elucidated. In order to understand a molecular basis of thermotolerance due to HSP104 protein induced by a mild heat shock, studies on respiratory pathways were carried out in the wild type as well as in the hsp104 deleted mutant. Especially the degree of 13C-acetate incorporation into glutamate-C4 was examined for both strains using 13C-13C homonuclear spin coupling measurements, since glutamate is in a rapid equilibrium with α-ketoglutarate in the TCA cycle. In addition, the temperature effects on the rate of 13C incorporation are compared with or without HSP104 protein expressed. Finally, the inhibitory effect of HSP104 on the respiration pathway was confirmed by the measurements of oxygen consumption rates for both strains.

Evaluation of the Accuracy and usability of Trigger mode in Respiratory Gated Radiation Therapy (호흡동조방사선치료를 위한 Trigger mode 투시영상 획득 시 호흡 속도에 따른 정확성 평가 - Phantom Study)

  • Purpose : The purpose of this study is to evaluate the accuracy and usefulness of the Trigger mode for the Respiratory Gated Radiation Therapy (RGRT) Materials and methods : A QUASAR respiratory phantom that inserted a 3 mm fiducial marker (a gold marker) was used to estimate the accuracy of the Trigger mode. And the 20 bpm was used as reference respiration rate in this study. The marker that placed at the center of the phantom was contoured, and the lower threshold of a gating window was fixed at 2.0 mm using an OBI with Truebeam STxTM. The upper threshold was measured every 0.5 mm from 1.0 mm to 3.0 mm. The respiration rates were changed every 10 bpm from 10 bpm to 60 bpm. We repeatedly measured five times to check the error rate of the trigger mode in the same condition. Result : The differences of a distance from a peak phase to upper threshold, 1.0 to 3.0 mm at a 20 bpm as a reference for 3 days in a row were 0.68±0.05 mm, 0.91±0.03 mm, 1.23±0.03 mm, 1.42±0.04 mm, and 1.66±0.06 mm, respectively. Measurement result of changes in respiratory rate compared to baseline respiratory rate in maximum absolute difference. The coefficient of determination (R2) to estimate the correlation between the respiration velocity and variation of absolute difference was on average 0.838, 0.887, 0.770, 0.850, and 0.906. The p-values of all the variables were below 0.05. Conclusion : Using Trigger mode during respiratory gated radiation therapy (RGRT), accuracy and usefulness of trigger mode at reference breathing rate were confirmed. However, inaccuracies depending on the rate of breathing it could be uncertain in case of respiration rate is faster than 20 bpm as a standard respiration rate compared to slower than 20 bpm. Consequently, when conducting a RGRT using the trigger mode, real time monitoring is required with well educated respiration.

Determination of Oxygen Transfer Characteristics (α, β and R) in an Aeration Basin at a Conventional Activated Sludge Wastewater Treatment Plant (활성슬러지 하수처리장 포기조내 산소 전달특성(α, β와 R)의 효율적 산정방안 개발)

  • The major objective of this study is to develop an effective method to estimate ${\alpha}$ and ${\beta}$ in an aeration basin at a conventional activated sludge wastewater treatment plant. A series of unsteady state batch tests were simultaneously performed with clean water and mixed liquor in two batch reactors under identical operational conditions. Oxygen uptake rates (OURs) of the mixed liquor were measured during the tests. The results show that the OURs due to synthesis respiration and endogenous respiration were averaged about $17.96 mg/(l{\cdot}hr)$ and about $12.29mg/(l{\cdot}hr)$, respectively. The corresponding ${\alpha}$ and ${\beta}$ were ranged between 0.65 to 0.95, and between 0.88 to 0.93, respectively. Based on the overall experimental results, the proposed experimental test method and the proposed method for determination of ${\alpha}$ and ${\beta}$ are found to be relatively simple and easy to use in evaluating the characteristics of aeration systems.

Studies on the Decomposition of Leaf Litter Containing Heavy Metals in Andong Serpentine Area, Korea I. Microcosm Experiment (사문암지대의 중금속 함유 낙엽의 분해에 관한 연구 I. Microcosm 실험)

  • This study attempted to compare the litter decomposition rate of Arundinella hirta and Miscanthus sinensis var. purpurascens which collected from serpentine soil acting potentially toxic concentration of heavy metals and non-serpentine soil by using the microcosm method for 192 days under constant humidity and $23^{\circ}C$. The contents of Ni, Fe, Mg and Cr in the serpentine and nonserpentine soil originated litter showed high differences between them. The litter samples from serpentine site have lower C/N than non-serpentine litter, but the soluble carbohydrate content was shown almost similar between two plant litter. The mass loss rates of leaf litter from serpentine area were slower than those from non-serpentine site. During the experimental period, the remained dry weight of A. hirta and M. sinensis var. purpurascens litter collected from serpentine site were 64.7%, 65.0% of initial dry weight and litter samples from non-serpentine site showed 54.2%, 50.7%, respectively. K and Na were leached rapidly at the initial decomposition periods, but Ca showed immobilization and other metal elements reserved at the decomposing litter for a long time. The decomposing A. hirta litter from non-serpentine soil showed higher values of $CO_2$ evolution, microbial biomass-C, and microbial biomass-N than those in serpentine soil originated litter acting nutrient stresses and exhibited rapid decay rate. The microbial biomass and microbial respiration of decaying litter were positively correlated with litter decomposition rate, and these relationships showed more rapid slope in non-serpentine soil originated litter than that in serpentine soil.

Acute Toxicity and General Pharmacological Action of QGC EXT

  • It has been shown that QGC isolated and purified from Rumecis folium found protective effects of gastritis and esophagitis which EXT is an ethanol extract of it. We examined acute toxicity and the general pharmacological action of QGC EXT to search for any side effects of it in rats, mice, guinea pigs, and cats. In a single dose toxicity study, QGC EXT didn't show toxicological effects in rats and mice, and the $LD_{50}$ was over 5 g/kg in both animals, and there were also no changes in weight, feed and water intake during these toxicological experimental periods. We examined the general pharmacological action on central controlled behavior responses, and peripheral organs including blood pressure, heart rate, respiration and gastrointestinal system, We found that there were no significant changes in body temperature, locomotors activity, stereotyped behaviors, sleeping time, and convulsion. In other studies, writhing reaction, normal body temperature, there did not appear to be any changes. The large intestine movement and electrical field stimulation-induced contraction was not changes by its EXT. In addition, the influences on blood pressure, heart rates, and respiration by QGC EXT were not found. These results indicate that QGC EXT may be very safe as a new drug, since its $LD_{50}$ was very high over 5 g/kg and any side effects were not found.

Effects of Dietary Protein Sources and Levels on Heat Production and Thermoregulatory Responses of Sheep Exposed to a High Ambient Temperature

  • Four Suffolk ewes were used in Latin Square switch over design to study the effects of varying levels and sources of protein on heat production and thermoregulatory responses at daytime high ($33^{\circ}C$ temperature. They were fed Italian ryegrass hay supplemented with fishmeal and/or urea, providing three different levels of crude protein (CP) (low/unsupplemented: 7.9, medium: 11.6, and high: 15.8%) at $1.5{\times}maintenance$. Feeds were distributed at 0900 (30%) and 1700 (70%). Urea diet caused higher heat production and increased vaginal temperature compared to fishmeal and fishmeal-urea mix diets. Time spent standing, skin temperature and respiration rate of sheep fed urea were similar with those of sheep fed fishmeal. Sheep fed diet with low CP level had higher heat production, increased vaginal and skin temperature than sheep fed diet with medium CP content. Sheep on high CP diet produced significantly more heat than sheep fed medium CP diets. Their vaginal temperatures were similar with those of sheep fed medium CP diet but lower than those of sheep fed low CP diet. Respiration rates of sheep and time spent by them for standing on all diets did not differ significantly. These results suggest that urea is inferior protein supplement for thermoregulation of animal at hot environment, as it induced higher heat production than fishmeal and fishmeal-urea mix. Thermoregulatory response on fishmeal-urea mix diet was similar to fishmeal diet. Increasing CP of the diet from low to medium gives advantage for thermoregulation of animal. Increasing CP further to high level was not beneficial as it resulted in the responses of sheep similar to those on low protein diet.

  • To clarify the anesthetic effect of acupoint injection(aquapupuncture) using general anasthetics in dogs, 18 mongrel dogs were divided into control and two experimental groups(Tian-ping+Bai-hui : Tian-ping group and San-yang-luo+Gong-sun group : San -yang- lux group). Control group was intramuscularly injected with ketamine hydrochloride, 22 mg/ kg of body weight into the thigh and experimental groups were injected into each acupoint with half volume of dosage, respectively. Clinical findings(recumbency time, induction time of anesthesia, time of head lift and standing time) and changes of vital sign(temperature, heart rate and respiration rate) were investigates at pre-anesthesia, during anesthesia and poststanding, respectively. In recumbency time San-yang-luo groupui<0.05) and Tian-ping group (p<0.05) were faster than that of control, respectively and Tian-ping group was the fastest.
