Molecular and Morphological Identification of Fungal Species Isolated from Bealmijang Meju

  • Bealmijang is a short-term aged paste made from meju, which is a brick of fermented soybeans and other ingredients. Different types of bealmijang are available depending on the geographic region or ingredients used. However, no study has clarified the microbial diversity of these types. We identified 17 and 14 fungal species from black soybean meju (BSM) and buckwheat meju (BWM), respectively, on the basis of morphology, culture characteristics, and internal transcribed spacer and ${\beta}$-tubulin gene sequencing. In both meju, Aspergillus oryzae, Rhizopus oryzae, Penicillium polonicum, P. steckii, Cladosporium tenuissimum, C. cladosporioides, C. uredinicola, and yeast species Pichia burtonii were commonly found. Moreover, A. flavus, A. niger, P. crustosum, P. citrinum, Eurotium niveoglaucum, Absidia corymbifera, Setomelanomma holmii, Cladosporium spp. and unclassified species were identified from BSM. A. clavatus, Mucor circinelloides, M. racemosus, P. brevicompactum, Davidiella tassiana, and Cladosporium spp. were isolated from BWM. Fast growing Zygomycetous fungi is considered important for the early stage of meju fermentation, and A. oryae and A. niger might play a pivotal role in meju fermentation owing to their excellent enzyme productive activities. It is supposed that Penicillium sp. and Pichia burtonii could contribute to the flavor of the final food products. Identification of this fungal diversity will be useful for understanding the microbiota that participate in meju fermentation, and these fungal isolates can be utilized in the fermented foods and biotechnology industries.

  • The genus Prunus (family: Rosaceae) consists of over 400 plant species and exhibits vast biodiversity worldwide. Given the wide distribution of this genus, its taxonomic classification is important. Anatomical characteristics are conserved and stable and can therefore be used as an important tool for the taxonomic characterization of plants. Therefore, this study aimed to assess and document the anatomical characteristics of the leaf, stem, and seed of P. africana using micrographs and photographs for possible use in the identification, quality control, and phylogenetic analysis of the species. The anatomical sections of a young stem revealed a cortex consisting of isodiametric parenchyma cells, druse crystals, primary vascular bundles, and pith. The mature stem bark majorly consisted of the rhytidome, with the periderm densely arranged in multiple layers; a cluster of stone cells; and sclerenchyma. The leaf sections were hypostomatic, with stomata sizes ranging from 18.90-(22.34)-26.90 × 15.41-(18.40)-21.22 ㎛. The leaf sections showed the presence of characteristic druse crystals, vascular bundles, and mesophyll layers. The pericarp contained the epicarp, mesocarp, and endocarp, with their thickness being approximately 350-400, 300-350, and 30-50 ㎛, respectively. In addition, it contained a seed testa with a thickness of approximately 50-60 ㎛. The morphological and anatomical characteristics observed in P. africana leaves, stems, and seeds in this study could serve as useful data for the taxonomic identification of this species.

Occurrence of longtail tuna (Thunnus tonggol) juvenile in Korean waters (백다랑어 (Thunnus tonggol) 치어의 국내 출현)

  • A total of 24 juvenile specimens of Thunnus tonggol (5.45~7.26mm in total length) of the Sombridae were collected from the southeast sea of Jeju Island during 26~30 July, 2013. Twenty-four specimens identified T. tonggol have melanophores distributed on the 1st dorsal-fin rays, the dorsal of head and vetral side. Three individulas were identified using mitochondrial DNA cytochrome oxidase submit 1 (CO1) sequences (452 base pairs). All were identified as T. tonggol, their mtCO1 sequences being consistent with those of Thunnus tonggol (d=0.000), followed by Thunnus albacares (d=0.002) and Thunnus obesus (d=0.007).

Characterization of Streptomyces sp. AMLK-135 Producing Anti- MRSA Antibiotics

  • The present research program was conducted to characterize a strain of actinomycetes producing an anti methicillin-resistant Staphylococcus aureus (MRSA) antibiotic. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 400 isolates, a strain (AMLK-135) producing anti-MRSA antibiotic against S. aureus TK 784 was selected. According to the morphological and physiological characteristics, the strain AMLK-135 was confirmed to belong to the genus Streptomyces. From the results of species identification with the TAXON program, the strain AMLK-135 was shown to belong to major cluster 5 (Streptomyces exfoliatus), but it had a low simple matching coefficient ($S_{SM}$ SM/) value to member organisms of major cluster 5. Percentage ($\%$) of strain further away of the strain AMLK-135 was low (1.9400) and it was placed further away than the outer-most members in major cluster 5. Therefore, the strain AMLK-135 was identified as a new species of the genus Streptomyces.

Rapid Identification of Lactobacillus and Bifidobacterium in Probiotic Products Using Multiplex PCR

  • Lactic acid bacteria (LAB) are beneficial for the gastrointestinal tract and reinforce immunity in human health. Recently, many functional products using the lactic acid bacteria have been developed. Among these LAB, Lactobacillus acidophilus, Lactobacillus rhamnosus, Bifidobacterium longum, and Bifidobacterium bifidum are frequently used for probiotic products. In order to monitor these LAB in commercial probiotic products, a multiplex PCR method was developed. We designed four species-specific primer pairs for multiplex PCR from the 16S rRNA, 16S-23S rRNA intergenic spacer region, and 23S rRNA genes in Lactobacillus acidophilus, Lactobacillus rhamnosus, Bifidobacterium longum, and Bifidobacterium bifidum. Using these primer pairs, 4 different LAB were detected with high specificity in functional foods. We suggest that the multiplex PCR method developed in this study would be an efficient tool for simple, rapid, and reliable identification of LAB used as probiotic strains.

Development of a Microsatellite Marker Set for the Individual Identification and Parentage Verification of Korean Native Black Goats (재래흑염소 개체식별과 친자확인을 위한 Microsatellite Marker Set 개발)

  • The Korean native black goat (Capra hircus coreanae) is the goat species to be officially registered in Korea under the Food and Agriculture Organization. The object of this study is to establish a set of microsatellite (MS) markers for the individual identification and parentage verification of goats. In this study, we analyzed alleles of MS markers in crosses between Korean native black goats and crossbred goats (n=304 animals), and, based on the diversity of alleles for each marker, we selected 11 MS markers for individual identification and parentage verification. Using these 11 MS markers, the probabilities of different individuals with the same genotype being found within random and half-sib mating populations were 5.58×10-10 and 1.15×10-7, respectively. The parentage verification accuracy was 0.999996 when information about the parents was available and 0.999833 with no information. Thus, even given the total rearing population of 576,150 animals in South Korea, we concluded that these markers could be used for the individual identification and parentage verification of goats. Moreover, by analyzing the genetic relationships between the four lines of Korean native black goats and the crossbred goats, we verified the genetic characteristics of Korean native black goats, confirming their conservation value as a unique genetic resource.

Fractal behavior identification for monitoring data of dam safety

  • Under the interaction between dam body, dam foundation and external environment, the dam structural behavior presents the time-varying nonlinear characteristics. According to the prototypical observations, the correct identification on above nonlinear characteristics is very important for dam safety control. It is difficult to implement the description, analysis and diagnosis for dam structural behavior by use of any linear method. Based on the rescaled range analysis approach, the algorithm is proposed to identify and extract the fractal feature on observed dam structural behavior. The displacement behavior of one actual dam is taken as an example. The fractal long-range correlation for observed displacement behavior is analyzed and revealed. The feasibility and validity of the proposed method is verified. It is indicated that the mechanism evidence can be provided for the prediction and diagnosis of dam structural behavior by using the fractal identification method. The proposed approach has a high potential for other similar applications.

  • System identification is a major component for a control system. In biosystems, which is nonlinear and dynamic, precise identification would be very helpful for implementing a control system. It is difficult to precisely identify such non-linear systems. The measurable data on products from 2,3-butanediol fermentation could not be included in a process model based on kinetic approach. Meanwhile, a predictive capability is required in developing a control system. A neural network (NN) dynamic identifier with a by/(1+ t ) transfer function was therefore designed being able to predict this fermentation. This modified inverse NN identifier differs from traditional models in which it is not only able to see but also able to predict the system. A moving window, with a dimension of 11 and a fixed data size of seven, was properly designed. One-step ahead identification/prediction by an 11-3-1 BPNN is demonstrated. Even under process fault, this neural network is still able to perform several-step ahead prediction.

  • Currently for exploring marine resources, After confirming the location of the resources by controlling search equipment and sensor module in research vessel, the method which collects sample of searched resources and analyzes the quality and a contents of the resources in research vessel is been applying. This search method is structure which analyzes an ingredient in ship and decides the quality of the resources. And real-time processing for an effective resources search is been demanding. Therefore in this paper, we have developed a resources identification algorithm that can display the type feature of resources by real-time 3D Graphic.
