The dorsal guard hair identification key of Korean small mammals (Rodentia and Lagomorpha)

  • We analyzed the hair microstructure of Korean small terrestrial mammals, such as Rodentia and Lagomorpha, to classify the characteristics of hair morphology. Micromys minutus showed a unicellular irregular type of medulla structures; on the other hand, the other mammals showed multicellular structures. Regarding the cuticular scale structures, the Rodentia species exhibited a narrow and broad diamond petal type, while the Lagomorpha species exhibited an elongated petal type. Interestingly, the hair cross-sections showed quite unique characteristics. We constructed hair identification keys to distinguish species with a single hair. The dichotomous key of Rodentia and Lagomorpha can be used for their behavioral ecology and dietary analysis of upper predators to serve as the basis for ecological research.

Development of SCAR Markers for the Authentication of Acori Rhizoma Based on the Analysis of RAPD and Multiplex-PCR (RAPD 분석과 multiplex-PCR을 이용한 석창포 감별용 SCAR 마커 개발)

  • The rhizomes and herbal medicines originating from Acorus gramineus, A. calamus, A. tatarinowii, and A. gramineus var. pusilus, show significant similarity, and the correct identification of species is very difficult. Random Amplified Polymorphic DNA (RAPD) and Sequence Characterized Amplified Region (SCAR) were used to develop a reliable method for identification of these four species. Several distinct SCAR markers were developed from species-specific RAPD amplicons for each species. Furthermore, a useful molecular marker was established for multiplex-PCR, in order to the four species could be distinguished concurrently. These markers allow efficient and rapid identification of closely-related Acorus species and will be useful for standardization of herbal medicines.

A Fluorescence-based cDNA-AFLP Method for Identification of Differentially Expressed Genes

  • Identification of differently expressed genes under specific tissues and/or environments provides insights into the nature and underlying mechanisms of cellular processes. Although cDNA-AFLP (Amplified Fragment Length Polymorphism) is a powerful method for analyzing differentially expressed genes, its use has been limited to the requirement of radioactive isotope use and the difficulty of isolating the bands of interest from a gel. Here, we describe a modified method for cDNA-AFLP that uses a fluorescence dye for detection and isolation of bands directly from a small size polyacrylamide gel. This method involves three steps: (i) preparation of cDNA templates, (ii) PCR amplification and differential display, and (iii) identification of differentially expressed genes. To demonstrate its utility and efficiency, differentially expressed genes during vegetative growth and appressorial development of Magnaporthe oryzae were analyzed. This method could be applied to compare gene expression profiles in a diverse array of organisms.

  • In this paper, a real-coded genetic algorithm is proposed for identification of time delay systems from step responses. FOPDT(First-Order Plus Dead-Time) and SOPDT(Second-Order Plus Dead-Time) systems, which are the most useful processes in this field, but are difficult for system identification because of a long dead-time problem and a model mismatch problem. Genetic algorithms have been successfully applied to a variety of complex optimization problems where other techniques have often failed. Thus, the modified crossover operator of a real-code genetic algorithm is proposed to effectively search the system parameters. The proposed method, using a real-coding genetic algorithm, shows better performance characteristics when compared to the usual area-based identification method and the directed identification method that uses step responses.

Rapid Identification of Vibrio Species Isolated from the Southern Coastal Regions of Korea by MALDI-TOF Mass Spectrometry and Comparison of MALDI Sample Preparation Methods

  • Vibrio species are generally recognized as pathogens predominant in seafood along coastal areas. The food industry has sought to develop efficient microbial detection methods. Owing to the limits of conventional methods, this study aimed to establish a rapid identification method for Vibrio isolated from Korea, based on matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Four different preparation procedures were compared to determine the appropriate means to pretreat Vibrio species, using 17 isolates and five reference strains. Extended direct transfer and full formic acid extraction methods using bacterial colonies on agar plates revealed very low identification rates. Formic acid and trifluoroacetic acid (TFA) extractions using bacterial broth cultures were also performed. All Vibrio isolates and reference strains prepared by TFA extraction were successfully identified to the species level (17/22, 77.3%) and to the genus level (5/22, 22.7%). Thus, TFA extraction was considered the most appropriate method to pretreat Vibrio species for MALDI-TOF MS. The remaining 33 isolates and two reference strains were prepared by TFA extraction and analyzed by MALDI-TOF MS. Overall, 50 isolates were identified to the species level (40/50, 80%) and to the genus level (10/50, 20%). All isolates were identified as 43 V. alginolyticus, six V. parahaemolyticus, and one V. vulnificus species. V. alginolyticus and V. parahaemolyticus were isolated from fish offal (87.5% and 12.5%, respectively), seawater (91.3%, 8.7%), and shellfish (62.5%, 37.5%), whereas V. alginolyticus and V. vulnificus were isolated from sediment (90.9% and 9.1%, respectively). This study established a reliable system of MALDI-TOF MS preparation and analysis for Vibrio identification.

Pyricularia oryzae의 성장을 억제하는 물질을 생산하는 Streptomyces sp. NA-52의 분리 및 동정

  • The aim of the present research program was to isolate a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among isolates, a strain (NA -52) producing antifungal substance against Pyricularia oryzae was selected. Chemotaxonomic and numerical identification were carried out for the isolate. Fifty taxonomic unit characters were tested and the data were analyzed numerically using TAXON program. The isolate was identified as a synonym of streptomyces diastaticus belong to cluster No. 19 (Streptomyces diastaticus). But it showed a low similarity to S. diastaticus in simple matching coefficients, hence it was considered as one new species in Streptomyces.
