Reliable experimental data as a key factor for design of mechanical structures

  • The experimentally determined mechanical behavior of the material under the prescribed service conditions is the basis of advanced engineering optimum design. To allow experimental data on the behavior of the material considered, uniaxial stress tests were made. The aforementioned tests have enabled the determination of mechanical properties of material at different temperatures, then, the material's resistance to creep at various temperatures and stress levels, and finally, insight into the uniaxial high cyclic fatigue of the material under different applied stresses for prescribed stress ratio. Based on fatigue tests, using modified staircase method, fatigue limit was determined. All these data contributes the reliability of the use of material in mechanical structures. Data representing mechanical properties are shown in the form of engineering stress-strain diagrams; creep behavior is displayed in the form of creep curves while fatigue of the material is presented in the form of S-N (maximum applied stress versus number of the cycles to failure) curve. Material under consideration was 18CrNi8 (1.5920) steel. Ultimate tensile strength and yield strength at room temperature and at temperature of $600^{\circ}C$: [${\sigma}_{m,20/600}=(613/156)MPa$; ${\sigma}_{0.2,20/600}=(458/141)MPa$], as well as endurance (fatigue) limit at room temperature and stress ratio of R = -1 : (${\sigma}_{f,20,R=-1}=285.1MPa$).

Analysis of Thermal Oxide Behavior with Isothermal Degradation of TBC Systems Applied to Single Crystal Superalloy (단결정 초내열합금에 적용된 열차폐코팅의 등온열화에 따른 산화물 거동분석)

  • In the field of combined cycle power generation, thermal barrier coating(TBC) protects the super-heat-resistant alloy, which forms the core component of the gas turbine, from high temperature exposure. As the turbine inlet temperature(TIT) increases, TBC is more important and durability performance is also important when considering maintenance cost and safety. Therefore, studies have been made on the fabrication method of TBC and super-heat-resistant alloy in order to improve the performance of the TBC. In recent years, due to excellent properties such as high temperature creep resistance and high temperature strength, turbine blade material have been replaced by a single crystal superalloy, however there is a lack of research on TBC applied to single crystal superalloy. In this study, to understand the isothermal degradation performance of the TBC applied to the single crystal superalloy, isothermal exposure test was conducted at various temperature to derive the delamination life. The growth curve of thermally grown oxide(TGO) layer was predicted to evaluate the isothermal degradation performance. Also, microstructural analysis was performed by scanning electron microscope(SEM) and energy dispersive X-ray spectroscopy (EDS) to determine the effect of mixed oxide formation on the delamination life.

Formation Behavior of Anodic Oxide Films on Al 6061 Alloy in Sulfuric Acid Solution (황산 용액에서 Al6061 합금의 아노다이징 피막 형성거동)

  • Formation behavior of aluminum anodic oxide (AAO) films on Al6061 alloy was studied in view of thickness, morphology and defects in the anodic films in 20 vol.% sulfuric acid solution at a constant current density of $40mA/cm^2$, using voltage-time curve, observation of anodized specimen colors and surface and cross-sectional morphologies of anodic films with anodization time. With increasing anodizing time, voltage for film formation increased exponentially after about 12 min and its increasing rate decreased after 25 min, followed by a rapid decrease of the voltage after about 28 min. Surface color of anodized specimen became darker with increasing anodizing time up to about 20 min, while it appeared to be brighter with increasing anodizing time after 20 min. The darkened and brightened surfaces with anodizing time are attributed to an increase in thickness of porous anodic oxide film and a chemical damage of the films due to heat generated by increased resistance of the film, respectively. Cross-sectional observation of AAO films revealed the formation of defects of crack shape at the metal/oxide interface after 15 min which prevents the growth of AAO films. Width and length of the crack-like defect increased with anodizing time up to 25 min of anodizing, and finally the outer part of AAO films was partly dissolved or detached after 30 min of anodizing, resulting in non-uniform surface structures of the AAO films.

Autumn olive (Elaeagnus umbellata Thunb.) berry reduces fasting and postprandial glucose levels in mice

  • BACKGROUND/OBJECTIVES: Fasting and postprandial hyperglycemia should be controlled to avoid complications of diabetes mellitus. This study investigated the effects of autumn olive (Elaeagnus umbellata Thunb.) berry (AOB) on fasting and postprandial hyperglycemia in mice. MATERIALS/METHODS: In vitro ${\alpha}$-glucosidase inhibitory effect of AOB was determined. Maltose solution (2 g/kg) with and without AOB extract at 500 mg/kg or acarbose at 50 mg/kg was orally administered to normal mice after overnight fasting and glucose levels were measured. To study the effects of chronic consumption of AOB, db/db mice received the basal diet or a diet containing AOB extract at 0.4% or 0.8%, or acarbose at 0.04% for 7 weeks. Blood glycated hemoglobin and serum glucose and insulin levels were measured. Expression of adiponectin protein in epididymal white adipose tissue was determined by Western blotting. RESULTS: In vitro inhibitory effect of AOB extract on ${\alpha}$-glucosidase was 92% as strong as that of acarbose. The AOB extract (500 mg/kg) or acarbose (50 mg/kg) significantly suppressed the postprandial rise of blood glucose after maltose challenge and the area under the glycemic response curve in normal mice. The AOB extract at 0.4% or 0.8% of diet or acarbose at 0.04% of diet significantly lowered levels of serum glucose and blood glycated hemoglobin and homeostasis model assessment for insulin resistance values in db/db mice. The expression of adiponectin protein in adipose tissue was significantly elevated by the consumption of AOB at 0.8% of diet. CONCLUSIONS: Autumn olive (E. umbellata Thunb.) berry may reduce postprandial hyperglycemia by inhibiting ${\alpha}$-glucosidase in normal mice. Chronic consumption of AOB may alleviate fasting hyperglycemia in db/db mice partly by inhibiting ${\alpha}$-glucosidase and upregulating adiponectin expression.

Experimental Investigation of Concave and Convex Micro-Textures for Improving Anti-Adhesion Property of Cutting Tool in Dry Finish Cutting

  • Tool-chip adhesion impacts on cutting performance significantly, especially in finish cutting process. To promote cutting tools' anti-adhesion property, the concave micro-grooves texture (MGT) and convex volcano-like texture (VLT) were fabricated separately on lathe tools' rake faces by laser surface texturing (LST). Various orientations of MGT and different area densities (9% and 48%) and regions (partial and full) of VLT were considered in textured patterns designing. The following orthogonal cutting experiments, machining of aluminum alloy 5038, analyzed tools' performances including cutting force, cutting stability, chip shape, rake face adhesion and abrasion. It indicated that under dry finish cutting conditions, MGT contributed to cutting stability and low cutting forces, meanwhile friction and normal force reduced by around 15% and 10%, respectively with a weak correlation to the grooves' orientation. High density VLT tools, on the other hand, presented an obvious anti-adhesion property. A $5{\mu}m$ reduction of crater wear's depth can be observed on textured rake faces after long length cutting and textured rake faces presented half size of BUE regions comparing to the flat tool, however, once the texture morphologies were filled or worn, the anti-adhesion effect could be invalid. The bearing ratio curve was employed to analysis tool-chip contact and durability of textured surfaces contributing to a better understanding of anti-adhesion and enhanced durability of the textured tools.

Development of a Label-Free LC-MS/MS-Based Glucosylceramide Synthase Assay and Its Application to Inhibitors Screening for Ceramide-Related Diseases

  • Ceramide metabolism is known to be an essential etiology for various diseases, such as atopic dermatitis and Gaucher disease. Glucosylceramide synthase (GCS) is a key enzyme for the synthesis of glucosylceramide (GlcCer), which is a main ceramide metabolism pathway in mammalian cells. In this article, we developed a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to determine GCS activity using synthetic non-natural sphingolipid C8-ceramide as a substrate. The reaction products, C8-GlcCer for GCS, could be separated on a C18 column by reverse-phase high-performance liquid chromatography (HPLC). Quantification was conducted using the multiple reaction monitoring (MRM) mode to monitor the precursor-to-product ion transitions of m/z $588.6{\rightarrow}264.4$ for C8-GlcCer at positive ionization mode. The calibration curve was established over the range of 0.625-160 ng/mL, and the correlation coefficient was larger than 0.999. This method was successfully applied to detect GCS in the human hepatocellular carcinoma cell line (HepG2 cells) and mouse peripheral blood mononuclear cells. We also evaluated the inhibition degree of a known GCS inhibitor 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) on GCS enzymatic activity and proved that this method could be successfully applied to GCS inhibitor screening of preventive and therapeutic drugs for ceramide metabolism diseases, such as atopic dermatitis and Gaucher disease.

Analysis of Growth Characteristics Using Plant Height and NDVI of Four Waxy Corn Varieties Based on UAV Imagery

  • Although waxy corn varieties developed after the 1980s show differences depending on development stages and conditions, studies on the characteristics of waxy corn during the growth stage are rare. The subject of this study was a field survey and unmanned aerial vehicle (UAV) image acquisition of four waxy corn varieties cultivated in Idam-ri, Gammul-myeon, Goesan-gun, Korea. The study was conducted in four stages at intervals of two weeks after planting in 2019. The growth characteristics of each of the four varieties were analyzed using growth curves obtained based on field survey and UAV imagery data. The characteristics of each growth stage of the four varieties of corn, as assessed using normalized difference vegetation index (NDVI) and plant height (P.H.) values, were as follows. The growth model was identified as a model in which three-parameter logistic (3PL) curves reflect the growth characteristics of corn well. In particular, it was found that the variations in growth rate shown by P.H. and NDVI values clearly explain the differences between corn varieties. Among the four cultivars, growth and development first occurred at the early vegetative stage in Daehakchal, followed by Mibaek 2, Miheukchal, and finally Hwanggeummatchal. The variationsin P.H. and NDVI were achieved quickly and earlier in Daehakchal, followed by Mibaek 2, Hwanggeummatchal, and Miheukchal. It was confirmed that these results reflected the characteristics of the fast white-type varieties, while the black-type varieties were delayed, as in a previous study. These results reflect the resistance to lodging that affects the cultivation environment and the response characteristics to nutrients and moisture. It was confirmed that UAV accurately provides growth information that is very useful for analyzing the growth characteristics of each corn variety.

Effect of Zn/Al Cation Ratio on Corrosion Inhibition Capabilities of Hydrotalcites Containing Benzoate Against Carbon Steel

  • Corrosion inhibitors based on Zn-Al hydrotalcites containing benzoate (ZnAlHB) with different molar ratios of Zn/Al were prepared with a co-precipitation process. Compositions and structures of the resulting hydrotalcites were studied with suitable spectroscopic methods such as inductively coupled plasma mass spectrometry (ICP-MS), ultraviolet-visible spectrophotometry (UV-Vis), scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), and surface zeta potential measurements, respectively. Results of physico-chemical studies showed that crystallite sizes, compositions of products, and surface electrical properties were significantly changed when the molar ratio of Zn/Al was increased. The release of benzoate from hydrotalcites also differed slightly among samples. Anticorrosion abilities of hydrotalcites intercalated with benzoate at a concentration of 3 g/L on carbon steel were analyzed using electrochemical impedance spectroscopy (EIS), polarization curve, energy-dispersive X-ray spectroscopy (EDX), and SEM. Corrosion inhibition abilities of benzoate modified hydrotalcites in 0.1 M NaCl showed an upward trend with increasing Zn/Al ratio. The reason for the dependence of corrosion resistance on the Zn/Al ratio was discussed, including changes in the microstructure of hydrotalcites such as crystal size, density, uniformity, and formation of ZnO.

A Quantum Resistant Lattice-based Blind Signature Scheme for Blockchain (블록체인을 위한 양자 내성의 격자 기반 블라인드 서명 기법)

  • In the 4th industrial revolution, the blockchain that distributes and manages data through a P2P network is used as a new decentralized networking paradigm in various fields such as manufacturing, culture, and public service. However, with the advent of quantum computers, quantum algorithms that are able to break existing cryptosystems such as hash function, symmetric key, and public key cryptography have been introduced. Currently, because most major blockchain systems use an elliptic curve cryptography to generate signatures for transactions, they are insecure against the quantum adversary. For this reason, the research on the quantum-resistant blockchain that utilizes lattice-based cryptography for transaction signatures is needed. Therefore, in this paper, we propose a blind signature scheme for the blockchain in which the contents of the signature can be verified later, as well as signing by hiding the contents to be signed using lattice-based cryptography with the property of quantum resistance. In addition, we prove the security of the proposed scheme using a random oracle model.

Stability Evaluation of Weathered Gneiss Soil Slopes according to Clay Content (점토함유량에 따른 편마풍화토 비탈면의 안정성 평가)

  • In this study, the infiltration behavior of slopes composed of mixed soils with clay contents of 0%, 5%, and 10% in weathered Gneiss soil, which is a representative weathered soil in Korea, was investigated, and the stability of unsaturated slopes due to rainfall infiltration was examined. For this, in this study, the soil water characteristic curve was obtained through the water retention test, and the strength constant was obtained through the triaxial compression test. Based on the obtained results, the influence of clay content and antecedent rainfall effect (i.e., initial suction) on the formation of saturated zone (i.e., wetting band) and slope stability due to rainfall infiltration was examined through infiltration and stability analyses. As a result, it was found that the hig her the initial suction, the slower the formation of the saturated zone on the slope. In addition, it was found that as the clay content increases, the shear strength of the ground increases and the resistance to rainfall infiltration increases, and eventually the slope stability is greatly improved.
