Induction of RNA-mediated Resistance to Papaya Ringspot Virus Type W

  • Transformation of cantaloupes with the coat protein (cp) gene of papaya ringspot virus type W (PRSV-W), Thai isolate, was used to introduce virus resistance. Binary vectors containing either the full length coat protein coding region under control of the 35S CaMV promoter(pSA1175), or the inverted-repeat of a coat protein coding region (pSA1304), were constructed and used for Agrobacteriummediated transformation of cotyledonary explants of the cantaloupe cultivar Sun Lady. Four independent transgenic lines were obtained using pSA1304 and one using pSA1175. Integration of the PRSV-W cp gene into the genome of these transgenic lines was verified by PCR amplification, GUS assays and Southern blot hybridization. In vitro inoculation of these lines with PRSV-W revealed that whereas the line containing pSA1175 remained sensitive, the four lines containing pSA1304 were resistant. The presence of small RNA species, presumably siRNA, corresponding to regions of the viral cp gene in transgenic lines resistant to PRSV-W supports the involvement of post-transcriptional gene silencing in the establishment of resistance.

New Variety 'Sanggang' of Coix lachryma-jobi L. with Early Maturity and Resistance to Leaf Blight (율무 조숙 내병 다수성 신품종 '상강')

  • 'Sanggang' was a new adlay cultivar which was developed from the three way crosses of $(Hukuhane\;{\times}\;Suwon-6)\;{\times}$ Suwon-6 at the Northern Agricultural Research Station, Gyunggido ARES. This cultivar had green sheaf, dark-green leaf, green culm, milky white silk and darkbrown seedcoat. Sanggang shortened 10 days of heading date and 12 days of ripening time compared with Yulmoo 1. In the yield's conponent of Sanggang, the number of grains was by 56 grains less but ripening rate and 1,000 grain weight were higher by 6.5% and 20.1 g compared with Yulmoo 1, respectively. This cultivar showed moderate resistance to leaf blight. The yield of Sanggang was about 313 kg/10a of unpolished grain at ordinary cultivation. This cutivar showed about 17% and 27% higher yield in Yonchon Gyeonggi province and Cheongju Chungbuk province compared with the check variety respectively. This cultivar was adaptable to mono cropping in central and northern region of Korea.

A New Medium-late Maturity Rice Cultivar, "Dongjin2" with Direct Seeding Adaptability and Multiple Disease Resistances (벼 중만생 복합내병 직파적응성 "동진2호")

  • "Dongjin2" is a new japonica rice cultivar developed from the cross between Milyang165 with short culm and lodging resistance and $F_1$ plant of Iksan438, HR14018-B-1-1 and Iksan435 with high palatability at Honam Agricultural Research Institute (HARI), NICS, RDA in 2005. This cultivar has short grain shape and about 143 days growth duration from direct seeding to harvesting under Korean climate condition. The milled kernels of "Dongjin2" is translucent with non-glutinous endosperm. It has about 19.3% of amylose content and better palatability of cooked rice compared with "Nampyeongbyeo". This cultivar shows high resistance reaction to the bacterial blight pathogene race from $K_1$ to $K_3$, blast and stripe virus but susceptible to insect pests. "Dongjin2" yields about 5.71 and 5.74 MT/ha under the wet direct seeding and the transplanting at standard fertilizer level. "Dongjin2" would be adaptable for the southern plain area of Korea.

  • A new peanut cultivar "Jopyeong" (Arachis hypogaea ssp. hypogaea L.) was developed at the Yeongnam Agricultural Research Institute, NICS, in Milyang in 2006. It was developed from the cross between the very short stem cultivar "CUP brittle" and the high-yielding cultivar "Daekwang". "Jopyeong" which is Virginia plant type has 23 branch number per plant with early maturing and ellipse-shaped large kernel. Each pod has two grains with brown testa and 100 seed weight was 87g in the regional yield trials (RYT). This variety also showed more resistant to late leaf spots compared with check one. Especially it has resistance to lodging owing to short stem and erect plant type. In the regional yield trials "Jopyeong" was out-yielded than check variety by 11% with 8.37 ton/ha for fresh pod and by 4% with 3.95 ton/ha for grain.

A New Sprout-soybean Cultivar, "Hoseo" with Early maturity, Small Seed Size and High Sprout yielding (극소립·조숙 고수율 나물콩 신품종 "호서")

  • A new sprout-soybean cultivar, "Hoseo" was developed at the Honam Agricultural Research Institute (HARI) in 2007. Hoseo was selected from a cross between Camp and Nattosan. The preliminary, advanced, and regional yield trials to evaluate the performance of Iksan 57 were carried out from 2003 to 2007. This cultivar has a determinate growth habit with purple flower, grayish brown pubescence, yellow seed coat, grayish brown hilum, rhomboid leaflet shape and small seed size (7.4 g/100 seeds). The maturity date of "Hoseo" is 12 days earlier than the check variety, "Pungsan". It has good seed quality for soybean-sprout and resistance to lodging. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N). The average yield of "Hoseo" was 2.51 ton per hectare in the regional yield trials (RYT) for double cropping carried out for three years from 2005 to 2007.

Selection and Characterization of Horticultural Traits of Tomato leaf curl virus (TYLCV)-resistant Tomato Cultivars (토마토 황화잎말림바이러스(TYLCV) 저항성 품종 선발 및 원예특성 분석)

  • This study was conducted to evaluate imported tomato $F_1$ cultivars as breeding materials for the resistance to Tomato yellow leaf curl virus (TYLCV) by molecular markers and bioassay. From marker genotyping and disease evaluation of 40 $F_1$ cultivars, most of the cultivars declared as TYLCV-resistance carried heterozygous marker genotype for the TYLCV resistance genes Ty-1, Ty-3, or Ty-3a, and showed low disease rates. Whereas, 4 of 5 $F_1$ cultivars declared as intermediate resistance showed marker genotype for susceptibility and disease rates ranged 18.1-33.3%. However, the xx cultivars showed inconsistency in marker genotype and disease rate. Characterization of horticultural traits of the $F_1$ cultivars with TYLCV-resistance indicated that large-size fruit cultivars were higher in yield and similar in sugar contents and solid-acid ratio compared to a control cultivar preferred in the domestic market, although hardness remained to be a problem. On the other hand, cherry tomato cultivars showed lower yield and brix, but longer internode compared to a control cultivar, indicating that breeding for TYLCV-resistance using these cultivars will require more efforts and time compared to large-sized.

Evaluation of the Qualitative Characteristics of Creeping Bentgrass(Agrostis palustris Huds.) Cultivars Using NTEP Data (미농무성 NTEP(Nat'l Turfgrass Evaluation Program) 자료를 이용한 Creeping Bentgrass(Agrostis palustris Huds.) 품종의 특성 평가)

  • This study was initiated to evaluate the qualitative characteristics of creeping bentgrass(Agrostis palustris Huds.) cultivars for the climate In Korea through the NTEP(Nat'l Turfgrass Evaluation Program) data. 'L-93' showed the highest rating in overall mean visual quality. It was also the most prominent cultivar in seedling vigors, ground cover, and genetic color especially in summer. In case of turf texture, 'Penn A-1' and 'A-2' were the finest group, but the poorest group in cold tolerance. Leaf density and thatch accumulation were lower with 'Penncross', 'Pennlinks', 'Crenshaw', and 'L-93' as compared with 'Penn A'-type and 'G'-type cultivars. Resistance to moss invasion was greater with 'Penn A'-type and 'G'-type cultivars, but 'Penncross' was the least. These observations indicated that leaf density was considered to associate with the characters of turf quality, thatch accumulation and resistance to moss invasion. 'Penn A'-type cultivars were highly resistant to snow mold. Greater resistance to brown patch was associated with 'Penn A' and 'Penncross'. Higher resistance to pythium blight was found with 'Penncross' and 'Pennlinks'. 'L-93' showed higher resistance to dollar spot, but not to pythium. Therefore, these results demonstrated that turf maintenance program for the new bentgrass cultivars should be different from a conventional management for the cultivar of 'Penncross'.

Resistance Evaluation of Commmercial Tomato Cultivars against Tomato yellow leaf curl virus (토마토품종의 토마토황화잎말림바이러스병에 대한 저항성 평가)

  • Tomato yellow leaf curl virus (TYLCV) is a viral disease causing severe economic losses on tomato. Practical prevention of the TYLCV disease is to control tabacco whitefly (Bemisia tabaci) or to cultivate TYLCV-resistant tomato cultivars, because no agrochemical products are available to control TYLCV. In this study, TYLCV resistance of the commercial tomato cultivars were evaluated using the DNA markers tightly linked to TYLCV resistance genes Ty-1 and Ty-3 and infection with the TYLCV clones mediated by Agrobacterium. In marker genotyping, resistance alleles were detected from 4 oval type tomato cultivars (Titichal, TY tinny, TY saengsaeng II, TY sense Q). Four cheery type cultiavrs (TY endorphin, TY smartsama, Tiara TY, Olleh TY) and 6 round type cultivars (TY kingdom, TY ace, TY homerun, TY altorang, Dotaerang TY winner, Styx TY). The seedling bioassay indicated that tomato cultivars of the oval type and cherry type showed consistancy in marker genotype and phenotype while slight disease symptom was observed from some round type cultivras (TY ace, TY homerun, Styx TY) with resistance marker genotype. For fruit yields, TY tinny was greater than its control cultivar Titichal in oval types, TY smartsama was greater than its control Smile in cherry type, and TY ace and TY kingdom were greater than their control Dabok. These cutliavrs can be a good choice for high-yielding TYLCV-resistant tomato cultivars.

Agronomic characteristics and field resistance to bacterial soft rot of transgenic potato overexpressing the soybean calmodulin 4 gene (SCaM4) (SCaM4 과발현 형질전환 감자의 농업적 특성 및 감자 무름병 저항성 평가)

  • We performed in vitro assay and field trials to assess levels of changes in intrinsic properties and resistance against soft rot of the potato cv. Dejima upon the introduction of a soybean calmodulin 4 gene (SCaM4). Field trials with four lines overexpressing SCaM4 gene were conducted over two seasons, and harvested tubers were evaluated in bioassay for resistance to Pectobacterium carotovorum ssp. carotovorum. The SCaM4 transgenic potato lines inoculated with $10^8$ CFU/ml of P. carotovorum ssp. carotovorum showed enhanced resistance compared to control. Among the SCaM4 transgenic lines, the transgenic line SCaM4-4 exhibited the highest tolerance to soft rot in vitro assays, so did in field trials. In the field trial, the soft rot resistance of SCaM4-4 line was more than 5 times higher compared to that of control cultivar, Dejima. The major agronomic characteristics of the SCaM4 transgenic lines were not different from those of the nontransgenic 'Dejima'. The result demonstrated that the transformation of a calmodulin 4 gene was a successful strategy in development of potato cultivar enhanced to soft rot.

Growth Characteristics and Productivity of New Orchardgrass (Dactylis glomerata L.) Cultivar, 'Luckyone 2ho' (오차드그라스 신품종 '럭키원 2호'의 생육특성과 수량성)

  • 'Luckyone 2ho' is a new orchardgrass (Dactylis glomerata L.) cultivar developed by the National Institue of Animal Science (NIAS) in 2017. To develope the new variety of orchardgrass, 5 superior clones were selected and polycrossed for seed production. The agronomic growth characteristics and forage productivity of 'Luckyone 2ho' were examined at Cheonan from 2012 to 2014, and regional trials were conducted in Cheonan, Pheonchang, Jinju and Jeju from 2015 to 2017, respectively. 'Luckyone 2ho' showed medium type growth habit in fall, and medium in length of flag leaf and very long upper internode. Plant height of 'Luckyone 2ho' was 3 cm less than that of standard cultivar, 'Potomac' and heading date was 1 days later than 5th May compared to standard cultivar, 'Potomac'. Characteristics such as waterlogging and disease resistance of 'Luckyone 2ho' were stronger or better than those of standard cultivar, 'Potomac', Especially, dry matter yield of 'Luckyone 2ho'(15,980 kg/ha) increased 9 % compared to that of standard cultivar, 'Potomac'(14,702 kg/ha). Nutritive values were appeared to be similar in both varieties except in Vitro dry matter digestibility(IVDMD) and crude protein and total digestible nutrients (TDN).
