The Selection Effect of Native Gold and Magnetite by Microwave-nitric Acid Leaching and Magnetic/hydro Separation (마이크로웨이브-질산용출과자력/수력선별에의한자연금및자철석의선별효과)

  • This study aimed to recover native gold from the concentrate using microwave-nitric acid leaching and magnetic/hydro separation experiments. The insoluble-residue was filtered from leaching solution through microwave-nitric acid leaching experiment. As a result of the atomic absorption spectrometer(AAS) analysis of the filtered leaching solution, it was discovered that Au content was not eluted at all and it was observed from the back scattered electron(BSE) image that native gold was liberated in the insoluble-residue. When magnetic/hydro separation experiments were applied for the insoluble-residue, magnetic and non magnetic minerals were separtated from insoluble-residue. Magnetite was recovered from the magnetic minerals and as a result of applying the hydro separation experiment again for the non-magnetic mineral, native gold was recovered. The native gold was identified through the X-ray diffraction(XRD) analysis and BSE image.

Establishment of Pre-Harvest Residue Limits (PHRLs) of Insecticide Clothianidin and Fungicide Fluquinconazole on Peaches during Cultivation Period (복숭아의 재배기간 중 살충제 Clothianidin 및 살균제 Fluquinconazole의 생산단계 농약잔류허용기준의 설정)

  • BACKGROUND: It is hard to control the agricultural products exceeding MRL (maximum residue limit) before forwarding. Therefore, NAQS (National Agricultural Products Quality Management Service, South Korea) established PHRL (pre-harvest residue limit) on agricultural products during their cultivation periods. This study was performed to set the PHRLs of peach during cultivation period, and also to estimate biological half-lives for residues of clothianidin and fluquinconazole. METHODS AND RESULTS: Two groups of peach were treated under Korean GAP (Good Agricultural Practices) with application time, single and triple treatments. Sample was collected over 14days (each after 0, 2, 4, 6, 8, 10, 12, 14 days. 8times), and clothianidin and fluquinconazole were analyzed by HPLC/DAD and GC/ECD, respectively. CONCLUSION: The biological half-life of clothianidin in single treatment and triple treatment was 5.2days and 7.0days. That of fluquinconazole was also 3.9days and 4.1days, respectively. The PHRL of peach on 10days before harvest was 1.4 mg/kg in clothianidin and 1.8 mg/kg in fluquinconazole.

Identification of operating parameters in auto-discharging filter system for treatment of urban storm water (자동방류가 가능한 여과형 비점오염처리장치의 운전인자 도출)

  • To identify operating parameters of the up-flow filtering system, which is available to discharge filtering residue after the rain, developed for treatment of urban storm runoff, lab scale test was carried out. Removal efficiency of SS was 68.7%, 62.2%, and 58.6% at the area roading rate of 2.46m/h, 4.68m/h, and 10m/h, respectively, filtering device is desirable to operate at the lower than 4.68m/h of area roading rate to get higher level of 60% SS removal efficiency. The removal efficiency of SS was 57.1% ~ 68.7% at the raw water SS of 100mg/L ~ 600mg/L, and the SS in treated water was maintained at the constant level through the elapsed time. It is indicate that filtering device can guarantee a certain level of effluent water quality at various raw water quality. The removal efficiency of SS to the depth of filter media was 68.3%, 78.6% at the filter depth of 10 cm, 20cm respectively. The final treated water quality was showed 30.2mg/L of CODMn, 1.60mg/L of TN and 0.25mg/L of TP. The average removal efficiencies by filtering device developed in this research were recorded slightly lower levels than other research. The main reason of these results were the first, the filter depth of the media used in this test was shallow, the second, the kind of filter media in discharge port of residue. More research to kind of filter media, filter packing rate, select of media for residue discharge port should be go on to produce optimum operating condition. The result of this study would be valuable for the application of filtration device to control of urban storm water.

Effect of fermented biogas residue on growth performance, serum biochemical parameters, and meat quality in pigs

  • Objective: This study investigated the effect of fermented biogas residue (FBR) of wheat on the performance, serum biochemical parameters, and meat quality in pigs. Methods: We selected 128 pigs (the mean initial body weight was $40.24{\pm}3.08kg$) and randomly allocated them to 4 groups (1 control group and 3 treatment groups) with 4 replicates per group and 8 pigs per pen in a randomized complete block design based on initial body weight and sex. The control group received a corn-soybean meal-based diet, the treatment group fed diets containing 5%, 10%, and 15% FBR, respectively (abbreviated as FBR5, FBR10, and FBR15, respectively). Every group received equivalent-energy and nitrogen diets. The test lasted 60 days and was divided into early and late stages. Blood and carcass samples were obtained on 60 d. Meat quality was collected from two pigs per pen. Results: During the late stage, the average daily feed intake and average daily gain of the treatment groups was greater than that of the control group (p<0.05). During the entire experiment, the average daily gain of the treatment groups was higher than that of the control group (p<0.05). Fermented biomass residue did not significantly affect serum biochemical parameters or meat quality, but did affect amino acid profiles in pork. The contents of Asp, Arg, Tyr, Phe, Leu, Thr, Ser, Lys, Pro, Ala, essential amino acids, non-essential amino acids, and total amino acids in pork of FBR5 and FBR10 were greater than those of the control group (p<0.05). Conclusion: These combined results suggest that feeding FBR could increase the average daily gain and average daily feed intake in pigs and the content of several flavor-promoting amino acids.

  • Fumigant, methyl bromide which is used in most countries for plant quarantine, has been designated and under control as ozone depleting substance. For developing alternative chemicals for methyl bromide, many countries have been intensifying their capacity. This study was carried out to investigate the residue patterns of hydrogen cyanide which is commonly used for plant quarantine. Hydrogen cyanide was treated onto the orange, banana, and pineapple at recommended and double doses and then sampling was done at 1 and 3 day after fumigation treatment. Residue of hydrogen cyanide was found safe because average residue levels on orange, banana, and pineapple after 3 days of fumigation treatment were $0.57{\pm}0.05$, $0.21{\pm}0.17$, and $0.41{\pm}0.08$ ppm, which were lower than the MRLs of Korea (5 ppm), Japan (5 ppm), USA (50 ppm), and Canada (25 ppm). Hydrogen cyanide are expected to be used as alternative chemicals for methyl bromide fumigant for orange, banana, and pineapple.

Assessment of Oncogenicity from Pesticide Residues in Korean Foods (한국식품 중 잔류농약의 종양유발성 평가)

  • Dietary intake and oncogenic risk of 12 pesticides used in Korea whose oncogenic potency was known were assessed from published data. Dietary oncogenic risk (excess tumor incidence for a 70-year human life span) for Korean population was estimated to be $2.17{\times}10^{-3}$ on the basis of legal maximum residue limit, $4.33{\times}10^5$ on the basis of maximum practical residue level and $5.10{\times}10^{-6}$ on the basis of mean practical residue level of examined pesticides, all of which exceeded the negligible risk standard $1{\times}10^{-6}$ of US EPA. A systematic follow-up study on those oncogenic pesticides should be undertaken in order to mitigate the people's worry about the cancer risk by the abuse of pesticides in food production.

Structural Analysis of the Streptomyces avermitilis CYP107W1-Oligomycin A Complex and Role of the Tryptophan 178 Residue

  • CYP107W1 from Streptomyces avermitilis is a cytochrome P450 enzyme involved in the biosynthesis of macrolide oligomycin A. A previous study reported that CYP107W1 regioselectively hydroxylated C12 of oligomycin C to produce oligomycin A, and the crystal structure of ligand free CYP107W1 was determined. Here, we analyzed the structural properties of the CYP107W1-oligomycin A complex and characterized the functional role of the Trp178 residue in CYP107W1. The crystal structure of the CYP107W1 complex with oligomycin A was determined at a resolution of $2.6{\AA}$. Oligomycin A is bound in the substrate access channel on the upper side of the prosthetic heme mainly by hydrophobic interactions. In particular, the Trp178 residue in the active site intercalates into the large macrolide ring, thereby guiding the substrate into the correct binding orientation for a productive P450 reaction. A Trp178 to Gly mutation resulted in the distortion of binding titration spectra with oligomycin A, whereas binding spectra with azoles were not affected. The Gly178 mutant's catalytic turnover number for the 12-hydroxylation reaction of oligomycin C was highly reduced. These results indicate that Trp178, located in the open pocket of the active site, may be a critical residue for the productive binding conformation of large macrolide substrates.

Analytical Method for Triazine Herbicide Cyanazine Residues in Major Medicinal Crops (주요 약용작물에 대한 Cyanazine 제초제의 잔류 분석법)

  • Background: Cyanazine is used as a pre-emergent herbicide once during the growing season to control weeds of many upland crops worldwide. This study aimed to establish a method to determined cyanazine residue levels in major medicinal crops by using high performance liquid chromatography-UV detection/mass spectometry (HPLC-UVD/MS). Methods and Results: Cyanazine residue was extracted with acetone from the raw products of four representative medicinal plants - Scutellaria baicalensis, Paeonia lactiflora, Platycodon grandiflorum and Angelica gigas. The extract was diluted with a large volume of saline water and directly partitioned into dichloromethane to remove polar co-extractives in the aqueous phase. It was then purifined using optimized Florisil column chromatography. HPLC analysis conducted using an octadecylsilyl column allowed the successful separation of cyanazine from co-extractives of the samples, and the amount was sensitively quantified by ultraviolet absorption at 225 nm with no interference. The accuracy and precision of the proposed method were validated by conducting recovery experiments on each medicinal crop sample fortified with cyanazine at two concentration levels per crop in triplicate. Conclusions: The mean recoveries ranged from 91.2% to 105.3% for the four representative medicinal crops. The coefficients of variation were less than 10%, irrespective of the sample types and fortification levels. The limit of quantification of cyanazine was 0.02 mg/kg as verified by the recovery experiment. A confirmatory method was performed by liquid chromatography/MS using selected-ion monitoring technique to clearly identify the suspected residue.

Analysis of Optimal Mixture Ratio for Extrudate of the Soymilk Residue and Corn Grits by Mixture Design (혼합물 실험 계획법에 의한 두유박과 옥분 압출성형물의 최적 혼합비 분석)

  • Experimental designs were applied to optimize the mixture ratio for the extrudate made by soymilk residue and corn grits. Nine candidate points were examined for their significance on extrudate using the modified distance design. Bending force, expansion ratio, bulk density, water solubility index (WSI), water absorption index (WAI) and color $(L^*,\;a^*,\;b^*)$ were the significant factors improving the extruded cereal production, and these values were applied to the mathematical models. Results showed that bending force, expansion ratio WSI, WAI and color $(L^*,\;b^*)$ increased with increasing the corn grits, whereas bulk density tended to decrease. The statistical study showed that the fitted models were adequate to describe the contour plot and all responses. Optimum mixture ratio allowing to maximize the two responses (expansion ratio and $b^*$) and minimize the response (WAI) were examined with a numerical optimization methods. The numerical optimization method was obtained as 53.18% : 46.19% (corn grits : soymilk residue).

Dissipation pattern of pesticide residues in/on different varieties of lettuce applied with foliar spraying under greenhouse condition (시설재배에서의 상추품종별 살포농약의 소실양상)

  • The study was carried out to investigate dissipation pattern of pesticide residue in/on different varieties of lettuce applied with foliar spraying under greenhouse. The initial deposited amount of alpha-cypermethrin and imidacloprid in/on the crop was entirely corresponded with shape of the crop. Dissipation of deposited pesticide residue was supposed to be related with degradation by sunlight and wash off by watering rather than dilution effect by biomass increase. The crop grouping in leaf vegetables has to be carefully considered even in lettuce for dissipation pattern of pesticide residue; head type and leafy type.
