Benfuresate와 Oxolinic Acid의 작물체중 잔류량 평가 (Assessment of the Residues of Benfuresate and Oxolinic Acid in Crops)

  • 본 실험은 수도체중의 benfuresate 잔류량과 배추중의 oxolinic acid 잔류량을 정량하여 이들 농약의 안전사용 기준설정및 최대 잔류 허용량 설정을 위한 기초자료를 얻기위하여 수행하였다. 논 토양에 benfuresate 입제를 0.6 kg a.i./ha의 수준으로 처리한 후 수도체를 재배, 수확한 후 현미와 볏짚 시료 중 benfuresate 잔류량을 정량하였으며, 배추 중 oxolinic acid의 잔류량 측정을 위해 각 처리구 당 15 kg a.i./ha의 수준으로 수확전 45, 30, 21, 15, 7, 3일에 배추에 직접 경엽 살포하였다. 본 실험에서 얻은 결과를 요약하면 다음과 같다. 1. 수도체와 배추 중의 benfuresate와 oxolinic acid의 회수율은 각각 $87{\sim}89%$, $90{\sim}951%$이었다. 2. 수도체 중 benfuresate의 잔류량은 $0.27{\sim}0.46mg/kg$이었다. 배추중의 oxolinic acid 의 잔류량은 $0.23{\sim}1.53mg/kg$의 범위였다. 3. 두 가지 공시약제의 잔류량은 처리횟수가 많을수록, 또 처리일수가 수확일에 가까울수록 증가하는 경향을 보여주었다. 4. 배추 중에 잔류하는 oxolinic acid는 first-order kinetics에 따랐으며, 잔류량은 시간이 경과함에 따라 감소하는 경향이었다. 5. 배추 중에 잔류하는 oxolinic acid의 잔류량은 6회 처리 또는 수확 3일전에 처리하였을 경우에도 FAO/WHO에서 설정한 최대 잔류 허용량인 5mg/kg에 미치지 않았다.

Changes of Soil Physical Properties by Manured Sorghum Residues Incorporation

  • Although sorghum residue production was estimated to be $8{\sim}10Mg\;ha^{-1}$, most sorghum straw was used to be burnt or removed after harvest. This experiment was conducted to evaluate the effect of the incorporation of manured sorghum residues on soil physical properties from 2010 to 2012 in the converted paddy field. Five treatment with 3 replication consisted of control, inorganic fertilizer (NPK), manured residues, sorghum stover and sawdust manure. The incorporation level of organic source was $10Mg\;ha^{-1}$ without inorganic fertilizer NPK. The investigated physical parameters were bulk density (BD), porosity, water stability aggregate (WSA), water infiltration rater (WIR), penetration resistance (PR) and soil water retention characteristics (WRC) with soil depth. The incorporation of manured sorghum residues improved WIR significantly over inorganic fertilizer (NPK), sorghum residues and sawdust manure. The BD by incorporating with manured residues on sorghum was reduced significantly with crop residue over plot of NPK and sawdust. Significant increase in WSA was observed with stubble incorporation alone or manured sorghum residues. Soil WRC were significantly enhanced with manured sorghum residue incorporated without NPK. The average PR at 0~15 cm was 0.54 MPa for manured sorghum residues. For sawdust manure plot it was 0.42 MPa, for Sawdust manure plot 0.39 MPa and for NPK plot 0.54 MPa.

Effect of bio-char application combined with straw residue mulching on soil soluble nutrient loss in sloping arable land

  • We assessed the effects of combining bio-char with straw residue mulching on the loss of soil soluble nutrients and citrus yield in sloping land. The two-year study showed that straw residue mulching (ST) and bio-char application combined with straw residue (ST+BC) can significantly reduce soil soluble nutrient loss when compared with the control treatment (CK). The comparative volume of the soil surface runoff after each of the treatments was as follows: CK > ST > ST + BC. Compared with the CK, the runoff volume of the ST was reduced by 13.6 % and 8.5 % in 2014 and 2015, respectively. Compared with the CK, combining bio-char with the ST application reduced the loss of soluble nitrogen and improved the soil total nitrogen content reaching a significant level in 2015. It dramatically increased the soil organic matter content over the two year period (36.3% in 2014, 50.6% in 2015) as well as the carbon/nitrogen ratio (C/N) (16.6% in 2014 and 39.3% in 2015). Straw mulching combined with bio-char showed a trend for increasing the citrus yield.

논 토양 및 현미중 Imidacloprid의 잔류성 (Residue of Imidacloprid in Hulled Rice and Paddy Soil)

  • 논 토양과 현미 중 살충제 imidacloprid의 잔류성을 조사한 결과는 다음과 같다. 실내 조건의 토양 중 imidacloprid의 분해속도는 1차 반응식에 따랐으며 토양온도 및 토양 종류에 크게 영향을 받았다. $18-23^{\circ}C$의 토양중 imidacloprid의 반감기는 증식토에서 66.7-96.3일, 식양토에서 56.8-117.5일이었다. Arrhenius activation energy는 증식토에서 25.5KJ/mol, 식양토에서 50.3KJ/mol이었다. 포장조건하의 토양 중 imidacloprid의 분해는 처리 초기에는 매우 빨랐으나 점점 분해속도가 느려 1차반감기는 약 10일 정도이었으나 2차, 3차 반감기는 점점 길어져 처리량의 90%까지 분해되는데는 약 120일이 소요되었다. 현미 중 imidacloprid의 잔류량은 검출한계 0.01ppm이하이었다.

배추중(中) Metaldehyde의 잔류량평가 (Evaluation of Metaldehyde Residues on Chinese Cabbage)

  • 달팽이류등 연체동물을 방제하기 위하여 배추에 사용한 metaldehyde 입제(6%)를 0.3kg AI/10a 수준으로 처리한 후 잎과 뿌리에 이행한 metaldehyde의 잔류량을 측정하였다. 처리는 관행, 수확전 7, 15, 30일로 하였다. 잎은 안쪽부분과 바깥쪽부분으로 나누어 분석 하였으며 뿌리에 대한 잔류량도 아울러 실시 하였다. 본 실험에서 얻은 결과는 다음과 같다. 1. 배추내에 존재하고 있는 free acetaldehyde의 양은 0.04-0.18ppm 이었다. 2. 배추잎과 뿌리에 대한 metaldehyde의 잔류량은 처리횟수가 증가할 수록 그리고 수확전 처리일수가 짧을 수록 증가함을 보여 주었다. 3. 잎에서의 metaldehyde 잔류범위는 0.11-1.4ppm 이었다. 4. 배추잎을 두부분으로 나누어서 분석한 결과, 바깥쪽부분(outer part) 보다는 안쪽부분(innerpart)에서 0.01-0.6ppm 정도로 잔류량이 다소 많았다. 5. 뿌리의 metaldehyde 잔류범위는 0.04-0.63ppm이었으며, 잎에 비해서는 낮은 경향이었다.

Molecular cloning of a novel cecropin-like peptide gene from the swallowtail butterfly, Papilio xuthus

  • A new cecropin-like antimicrobial peptide (Px-CLP) gene was isolated from the immunechallenged larvae of the swallowtail butterfly, Papilio xuthus, by employing annealing control primer (ACP)-based GeneFishing PCR. The full-length cDNA of Px-CLP is 310 nucleotides encoding a 70 amino acid precursor that contains a putative 22-residue signal peptide, a 4-residue propeptide, a presumed 37-residue mature peptide, and an uncommon 7-residue acidic pro-region at the C-terminus. The deduced amino acid sequence of Px-CLP showed significant identities with other Lepidopteran cecropin D type peptides. RT-PCR revealed that the Px-CLP transcript was detected at significant level after injection with bacterial lipopolysaccharide (LPS). The peptides with or without C-terminal acidic sequence region were synthesized on-solid phage and submitted to antibacterial activity assay. The synthetic 37-mer peptide (Px-CLPa), which removed C-terminal acidic sequence region, was showed exclusively antibacterial activity against E. coli ML35; meanwhile, a 44-mer peptide (Px-CLPb) with C-terminal acidic peptide region was not active. This result suggests that Px-CLP is produced as a larger precursor containing a C-terminal pro-region that is subsequently removed by C-terminal modification.

Gas Chromatography Residue Analysis of Bifenthrin in Pears Treated with 2% Wettable Powder

  • This study was conducted to monitor the level of bifenthrin residues in pear sprayed with 2% bifenthrin wettable powder (WP) at the recommended rate at four different schedules prior to harvest. The target analyte was extracted with acetone, partitioned into dichloromethane, and then purified by florisil chromatographic column. The residue determination was performed on a DB-5 capillary column using GC with electron capture detector (ECD). Linearity of this method was quite good ($r^2$ = 0.9951) in the concentration ranged from 0.2 mg/kg to 10 mg/kg. Recovery test was carried out at two concentration levels, 0.2 mg/kg and 1.0 mg/kg, in three replicates, and their rates were from 82.9% to 107.2%. No quantitative bifenthrin was detected in pear of all kinds of treatments including the treatment sprayed 4 times until 7 days before harvest. This sensitive and selective method can be used to monitor the trace residual amounts of bifenthrin in pear in a quite low concentration level.

수출용 단감에 대한 Difenoconazole과 Thiamethoxam의 잔류특성 연구 (Residue Studies of Difenoconazole and Thiamethoxam during Cultivation of Sweet Persimmon for Export)

  • 국내 유망 수출 식품 중 잔류농약으로 인하여 수출 장애가 있는 식품을 대상으로 문제가 되는 농약에 대해 CAC 및 수출국의 농약잔류 허용기준 제안을 위한 작물잔류 시험을 수행하였다. 대상 작물은 수출용 단감이었고, difenoconazole과 thiamethoxam을 대상으로 시험을 수행하였다. Difenoconazole의 잔류량 범위는 0.2~0.56 mg/kg였고, thiamethoxam의 잔류량 범위는 0.08~0.28 mg/kg으로 두 약제 모두 수확 1일차 시료부터 MRL 농도 이하로 잔류량이 감소하였다. 따라서 안전사용기준에 따라 약제를 살포하고 수확한다면 국내에 설정된 MRL 기준을 충분히 만족시키는 것으로 확인되었다. Difenoconazole의 각 포장별 반감기는 각각 13.6, 19.4, 16.3일 이었고, thiamethoxam의 각 포장별 반감기는 각각 10.0, 15.3, 14.0일 이었다. Clothianidin의 잔류농도는 1개 지역에서 14, 28일 시료에서 검출되었으나, 농도는 0.03 mg/kg 이하였다.

  • 유기인제 농약을 70% acetone으로 추출한 다음 dlchloromethane 층으로 옮겨 추출물을 florisil로 충진된 open-column에서 chromatography를 행하였다. 마지막 추출물을 nitrogen-phosphorus detector(GC/NPD)을 가진 GC로 분석하였다. Narrow-bore capillary GC(Ultra-2)에 대해 18 가지의 유기인제 농약의 회수율이 88.7%에서 100.0%에 달하였다. 본 분석 방법의 최소 검출 준위는 0.019 - 0.035 mg/kg 이었으며 sample throughput(추출, open-column chro-matography 및 GC 분석)도 상당히 개선되었다.(sample당 8시간)

붕어고음 잔사분말을 첨가한 쇠고기 Patty의 단백질 품질 평가 (Protein Nutritional Qualities of Beef Patties Added with Crucian Carp Extraction Residue)

  • The know the possibility in development of the low-fat beef patty models using crucian carp9 extraction residues (CCER, freeze dried powder : 5%, 10%, 15%), those protein nutritional quality, texture, color and sensory properties were investigated. About 13∼23% (on dry basis) of lipid in control was reduced in cooked beef patties with the higher addition ratios of CCER. In vitro protein digestibility was not changed in raw patties before cooking but 2∼4% higher digestibility was revealed in cooked patties. Computed protein efficiency ratio (C-PER) and discriminant computed protein efficiency ratio (DC-PER) of beef patties containing CCER were almost same as control. Lightness and red color value of both (raw and cooked) beef patties were decreased with the higher CCER addition ratios but brown color value of cooked samples were similar to control. Stronger hardness was noted in all beef patties containing CCER significantly (p<0.05). Consumer's acceptability were generally decreased by addition of CCER, but 10% level could be recommendable in beef patty processing.
