Association of Endocrine Factors (Insulin-Like Growth Factor-II and Binding Protein-3) with Litter Size in Pigs

  • Litter size has been one of the important economic traits in porcine reproduction. The insulin-like growth factor (IGF) system has been shown to mediate actions of the steroid hormone or to synergize with other endocrine factors so that it consequently plays roles in reproductive processes, including ovulation, implantation, maintenance of pregnancy, and fetal development. However, the effect of the serum IGF system on porcine litter size has not been deeply studied. Therefore, this study was conducted to relate serum IFG-II concentration and IGF binding protein-3 (IGFBP-3) expression with porcine litter size. Moreover, the possible association of those with estrogen receptor (ER) as a candidate gene for litter size was investigated. Swine were separated into two groups showing high and low litter sizes, and sera were collected from sows in the estrous cycle to postnatal growth of their female progeny. Serum IFG-II concentration was measured by radioimmunoassay and IGFBP-3 expression was detected by Western ligand blotting. During the estrous cycle, IGFBP-3 expression in both groups decreased moderately from metestrus to estrus, but IFG-II concentration showed a reverse pattern. Also, IFG-II concentration and IGFBP-3 expression decreased gradually as pregnancy proceeded. Unlike IGFBP-3, IFG-II decreased moderately as newborn pigs grew. Significant differences in serum IFG-II amount between the two groups were detected at 60 (p<0.01), 75, 90, and 105 d (p<0.05) of pregnancy and at 60 (p<0.01), 45, and 105 d (p<0.05) of postnatal growth. Furthermore, based on ER genotypes, a high litter size group with genotypes AB and BB showed lower IFG-II concentration than a low litter size group with a genotype AA during pregnancy. Taken together, the results indicate that the serum IFG-II and IGFBP-3 are correlated with the litter size in pigs.

Differential Expression of miR-34c and Its Predicted Target Genes in Testicular Tissue at Different Development Stages of Swine

  • To verified the target genes of miR-34c, bioinformatics software was used to predict the targets of miR-34c. Three possible target genes of miR-34c related to spermatogenesis and male reproductive development: zinc finger protein 148 (ZNF148), kruppel-like factor 4 (KLF4), and platelet-derived growth factor receptor alpha (PDGFRA) were predicted. Then, the expression of miR-34c and its target genes were detected in swine testicular tissue at different developmental stages by quantitative polymerase chain reaction. The results suggested that the expression of PDGFRA has the highest negative correlation with miR-34c. Then immunohistochemical staining was done to observe the morphology of swine testicular tissue at 2-days and 3, 4, 5-months of age, which indicated that PDGFRA was mainly expressed in the support cells near the basement membrane during the early development stages of testicular tissue, but that the expression of PDGFRA was gradually reduced in later stages. Therefore, western blot analyzed that the highest expression of PDGFRA was generated in 2-days old testicular tissues and the expression levels reduced at 3 and 4-months old, which correlated with the results of immunohistochemical staining. In conclusion, PDGFRA is a target gene of miR-34c.

Passive Smoking and Cervical Cancer Risk: A Meta-analysis Based on 3,230 Cases and 2,982 Controls

  • Objective: Passive smoking has been considered as a risk factor of many cancers. To examine whether it might also pose a risk for cervical cancer, we performed a meta-analysis based on published case-control studies. Methods: We searched the PubMed database and references of included studies up to February 10th, 2012 for relevant studies. After two authors independently assessed the methodological quality and extracted data, a meta-analysis was conducted using CMA v2 software. Publication bias was evaluated by funnel plot, using Egger's and Begg's tests. Results: Finally 11 eligible studies yielded, involving 3,230 cases and 2,982 controls. The results showed that women who never smoke but exposed to smoking experience a 73% increase in risk of cervical cancer compared with non-exposed women (OR = 1.73, 95% CI = 1.35 - 2.21, p<0.001). Subgroup and sensitivity analyses indicated this result to be robust. Moderate publication bias was detected by visualing funnel plot, Egger's and Begg's tests. Conclusion: Based on currently available evidence, the findings of this meta-analysis suggests that passive smoking significantly and independently increases the risk of cervical cancer.

Outcomes of Malignant Ovarian Germ-Cell Tumors Treated in Chiang Mai University Hospital over a Nine Year Period

  • Malignant ovarian germ cell tumors (MOGCT) are rare neoplasms that most frequently occur in women at a young reproductive age. There have been limited data regarding this disease from Southeast Asian countries. We therefore conducted a retrospective study to analyze the clinical characteristics and the treatment outcomes of MOGCT treated at our institute between January, 2003 and December, 2012. Seventy-six patients were recruited from this period with the mean age of 21.6 years and 11.8% were pre-puberty. The two most common symptoms were pelvic mass and pelvic pain. Two-thirds of the studied patients presented at an early stage. The most common histology was immature teratoma (34.2%) followed by endodermal sinus tumor (28.9%), dysgerminoma (25%), mixed type (10.5%) and choriocarcinoma (1.3%). Over 80% of these patients received fertility sparing surgery and about 70% received adjuvant chemotherapy with the complete response rate at 73.3% and partial response at 11.1%. The most frequent chemotherapy was BEP regimen (bleomycin, etoposide, cisplatin). With the mean follow up time at 56.0 months, 12 patients (15.8%) developed recurrence and only an advanced stage was the independent prognostic factor. The ten year progression free survival (PFS) and overall survival rate of our study were 81.9% and 86.2%, respectively. In conclusion, MOGCT often occurs at a young age. Treatment with fertility sparing operations and adjuvant chemotherapy with a BEP regimen showed a good outcome. An advanced stage is a significant prognostic factor for recurrence.

Development and Application of Sexual Rights Awareness Scale for Female Adults (성인여성 대상 성 권리 인식 도구개발 및 적용)

  • Purpose: This study aimed to develop a sexual rights awareness scale for female adults and to further examine the differences after educational sessions for sexual rights awareness with female adults. Methods: Convenient sampling was employed to recruit 152 participants. A self administered questionnaire was developed to explore adult females' understanding of sexual rights awareness. In the next phase, education was provided over 3 consecutive sessions to 28 female university students. Factor analysis and Cronbach's alpha were performed in order to test validity and reliability of the educational sessions. The Wilcoxon rank test was used to identify the differences between pretesting of sexual rights awareness knowledge and post testing after educational sessions was provided. Results: After conducting factor analysis, 5 factors explained 63.3% of the total variance, namely: I) sexuality education, II) prejudice & biasaberration, III) sexual satisfaction, IV) safe & equal sexuality, and V) sexual autonomy. A Cronbach's alpha of 17 items was 0.80. After the education sessions, there was a significant increase in sexual rights awareness. Conclusion: The Sexual rights awareness questionnaire is acceptable as a tool for measuring the level of education in studies. Future research should aim at conducting comparison studies of sexual rights awareness between sexes and cultural differences with larger populations which will further help to develop and to strengthen the rigor with in methodologies.

Multiparity and Breast Cancer Risk Factor among Women in Burkina Faso

  • The relative lack of information on breast cancer etiology in Burkina Faso led us to undertake the present work to highlight risk factors. This prospective study was conducted using a questionnaire between January 2015 and February 2016 on women admitted to Yalgado OUEDRAOGO hospital, for consultation or supervision. The characteristics of multiparous breast cancer patients (n = 44) were compared with their non-multiparous counterparts (n = 36). The study found that increased risk of breast cancer among non-multiparous cases was related to body mass index (BMI) (p <0.001), age at menopause (p <0.004) and use of oral contraception (p <0.021) while abortion (p <0.002) was a risk factor among multiparous cases. These results suggest that even if multiparity is associated with a decreased risk in some women, avoidance of abortion during reproductive life should be recommended. The results provide preliminary information, which now need to be supplemented by survey of a larger sample in the national territory.

Control of MPF Activity of Recipient Oocytes and Subsequent Development and DNA Methylation of Somatic Cell Nuclear Transfer Bovine Embryos

  • We attempted to control the maturation promoting factor (MPF) activity and investigated the subsequent reprogramming of bovine somatic cell nuclear transfer (SCNT) embryos. Serum-starved adult skin fibroblasts were fused to enucleated oocytes treated with 2.5 mM caffeine or $150\;{\mu}M$ roscovitine. The MPF activity, nuclear remodeling patterns, chromosome constitutions and development of SCNT embryos were evaluated. Methylated DNA of embryos was detected at various developmental stages. The MPF activity was increased by caffeine treatment or reduced by roscovitine treatment (p<0.05). Blastocyst development was higher in the caffeine-treated groups (27.6%) than that of the roscovitine-treated group (8.3%, p<0.05). There was no difference in the apoptotic cell index among the three groups. However, the mean cell number of blastocysts was increased in the caffeine-treated group (p<0.05). Higher methylation levels were observed in the Day 3 embryos of the roscovitine-treated group (50.8%), whereas lower methylation levels were noted at Day 5 in the caffeine-treated group (12.5%, p<0.05). These results reveal that the increase in MPF activity via a caffeine-treatment creates a more suitable condition for nuclear reprogramming after SCNT.

Gene Expression Altered in Endometrium of Korean Cattle with Endometritis (한우 자궁내막염에서 발현 변화를 보이는 유전자)

  • This study was carried out to examine gene expression altered in endometrium of Korean cattle (Hanwoo) with endometritis using microarray. In this study, 4,560 diferentially expressed genes (DEGs) were identified in the endometrium of Hanwoo. Of 4,560 DEGs, 2,026 genes were up-regulated, while 2,536 genes were down-regulated in endometritis. Of them, top 10 regulated genes were listed. Filamin A, pancreatic anionic trypsinogen, Rho GDP dissociation inhibitor alpha, collagen type VI alpha 1, butyrate response factor 2, aggrecanses-2, annexin 14, aminopeptidease A, orphan transporter v7-3, and epithelial stromal interaction 1 were up-regulated, while MHC class II antigen, integrin-binding sialoprotein, uterine milk protein precursor, down-regulated in colon cancer 1, glycoprotein 330, dickkopf-1, cfh protein, $Ca^{2+}-dependent$ secretion activator, UL16 binding protein 3, and proenkephalin were down-regulated in the endometritis. Our results suggest that these genes could be useful biomarkers for diagnosis Hanwoo's endometritis.

Population Dynamics of Zacco platypus in Gap-Stream and Its Relation with Water Quality

  • This study was to provide basic data for aquatic ecosystem research using fishes. Field sampling was carried out at five selected sites of Gap Stream, and fish samples, especially for a selection of sentinel species were collected three times in June, September, and October 2007. We analyzed total length distribution of Zacco platypus in relation with the season and the sampling sites, and then compared with total body weight, condition factor (K), and age distribution of the fish. The fish population data were compared with physico-chemical water quality, obtained from the Ministry of Environment, Korea. Water quality analysis showed a significant nutrient enrichment, based on total nitrogen (TN) and total phosphorus (TP), and organic matter pollution, based on biological oxygen demand (BOD) and chemical oxygen demand (COD) in the Site 5, which is directly influenced by wastewater disposal plant (WDP). Population analysis of the sentinel species showed that the total number of individuals, age distribution, and the population size-structure were influenced by the effluents from the WDP, and that reproductive failure of young-age population were evident in Site 5. According to the relation analysis of total weight to K, the disturbed population was mainly attributed to combined effects of habitat modifications and chemical degradations. Regression analysis of K values against water quality parameters showed significant (p<0.05) positive relations with nutrient and organic matter contents. Our data suggest that the population structure using a sentinel fish species reflected the ambient water quality in the stream and that diagnosis of aquatic ecosystem health using Z. platypus population may be practical for water resource and ecosystem conservations.

Effects of Shingihwanhapchangbudodam-Tang on the Estradiol Valerate-induced Polycystic Ovary in Rats (심기환합창부도담탕(腎氣丸合蒼附導痰湯)이 Estradiol Valerate로 유도된 흰쥐의 다낭성 난소에 미치는 영향)

  • Purpose : Polycystic ovarian syndrome(PCOS) is a endocrine disorder and still remains as one of the common causes of anovulation in women of reproductive age. There are some evidences that nerve growth factor(NGF) is involved in the pathogenesis of PCOS. In this research, the effects of ShingihwanhapChangbudodam- Tang(SCT) on the estradiol valerate(EV)-induced polycystic ovary(PCO) were investigated in rats. Methods : PCO was induced by single intramuscular injection with EV(4mg) in female rats. Rats in PCO control group(n = 8) were EV injected and orally administrated distilled water for sixty days. Rats in SCT-treated group(n = 8) were EV injected and orally administrated SCT for same duration. The weights of body, ovary and adrenal gland were measured. And also, ovarian histopathology and NGF immunohistochemistry were performed. Results : The weights of ovaries in SCT-treated group were significantly increased compared with control group. The numbers of secondly and mature follicles, and corpora lutea in SCT-treated group were significantly increased compared with control group. The numbers of atretic follicles were significantly decreased compared with control group. The expression of NGF positive reaction in the ovarian granulosa cells of SCT-treated group were lesser observed than control group. Conclusion : From the these results, we concluded that ShingihwanhapChangbudodam-Tang(SCT) has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect may be related with decreased NGF activities in the ovary.
