Administration Duration Dependent Effect of Ginseng Radix Alba Extract Solution on the Reproductive Capacities in the Mice (기간별(期間別) 인삼(人蔘) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Purpose : This study was undertaken to evaluate the effects of the different administration duration of Ginseng Radix Alba extract solution on the spermatogenic abilities. Methods : We used the 2-month-old mice and administered Ginseng Radix Alba extract solution 0.3mg/g/day for 30, 60, 90 and 120 days. The control group was administered the normal saline in the same way. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase. Also we observed changes of isolated testis before and after administration of Ginseng Radix Alba extract solutions in the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant differences were observed in the concentration of total sperm and normality of spermatozoa of the Ginseng Radix Alba extract solution administered groups compared to the control group in 60, 90 and 120days groups. The significant differences were observed the motility of the Ginseng Radix Alba extract solution administered groups compared to the control group in 90 days group respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Ginseng Radix Alba extract solution administered groups compared to the control group, respectively. but the activity of hyaluronidase was not significantly increased in the Ginseng Radix Alba extract solution administered groups Conclusion : This study shows that Ginseng Radix Alba has the beneficial effect on the concentration, morphology and motility of sperm especially in 90 days administration group. We can suggest that Ginseng Radix Alba extract solution be useful for the treatment of male sexual dysfunctions and infertility.

  • Objective: The present study was conducted to evaluate the viability of germ cells from the adult and fetal ovarian tissues after vitrification followed by xenografting. Method: The human adult ovarian tissues were obtained from 33 years old patient, and the fetal ovarian tissues were obtained from 22 weeks and 25 weeks in gestation. Ovarian tissues were cryopreserved by vitrification with 5.5 M ethylene glycol (EG 5.5) and 1.0 M sucrose as cryoprotectants. Adult and fetal ovarian tissues were pre-equilibrated with EG 5.5 at room temperature for 10 and 5 minutes, respectively and plunged into liquid nitrogen immediately. Frozen-thawed tissues were xenografted into NOD-SCID mice to evaluate the viability and capacity for further growth of the primordial follicles. Grafts were recovered from the recipients 4 weeks after transplantation and histological analysis was accomplished. Result and Conclusion: Grafts recovered 4 weeks after transplantation contained less number of oocytes and primordial follicles compared to that of the fresh tissues. Survived follicles were mainly primordial and intermediary with larger diameter and more granulosa cells. It is confirmed that 1) the ovarian tissues were healthy and the germ cells were survived after vitrification, and 2) the survived fetal primordial follicles after vitrification resumed the growth in the xenografts.

The Effects of Oocyte Preparation on the Developing Capacity of Human Oocytes at Intracytoplasmic Sperm Injection (ICSI) (난자내 정자 직접주입술에서 난자의 처리방법이 난자의 발생능력에 미치는 영향)

  • Objective: In the preparation of ICSI, cumulus and corona cells should be removed from the oocytes by using a combination of enzymatic (hyaluronidase) and mechanical (pipetting) methods. But little is known about the effects of different degrees of oocyte denudation and incubation time between denudation and sperm injection on the outcomes of ICSI. The aim of this study was to evaluate the effects of varying the degrees of oocyte denudation and the lengths of incubation time from denudation to sperm injection on the outcomes of ICSI. Methods: In experiment 1, patients (oocytes) were grouped into group A and B according to the degree of denudation, complete and partial, respectively. In experiment 2, patients (oocytes) were grouped into group I, II and III according to the length of incubation time of denuded oocytes until sperm injection as < 1, $1{\sim}2$ and >2 hours, respectively. Results: There was no significant difference between the degree of oocyte denudation on the survival, fertilization and development rates after ICSI procedure. In case of the incubation time of denuded oocytes until ICSI, survival rates was higher in group III (83.1 %) than in group I (61.5%, p<0.05) or group II (64.3%). However no statistically significant differences were found between incubation time and fertilization or development rates. Conclusions: This study reveals that the outcomes of ICSI are not affected by the degree (complete or partial) of oocyte denudation. However, the denuded oocytes with incubation period of more than 2 hours show better outcomes of ICSI than those with the incubation period of less than 2 hours.

  • To increase fertilization rate in vitro, separation of viable spermatozoa from the seminal plasma and its other components may be a useful procedure. Ejaculates from healthy men, whose semen analysis findings were normal in 19, and abnormal in 10, were filtered using the glass-wool filtration technique to yield a concentrated, viable sperm samples for IVF, and the usefulness and safety of this method were evaluated. The recovery rate of motile sperm in abnormal groups was 46.2% and 54.5% in normal group. The % motility was increased significantly compared with original sample after filtration, and the grade motility was improved, too. The sperm population with normal morphology was also increased significantly in both group. Using transmission electron microscopy, the ultrastructural integrity of acrosomal segment was examined in order to evaluate the potentially hazardous effect of glass-wool filtration to sperm head, however, sperm population with normal ultrastructure was increased compared with that of original ejaculate after separation. The filtered sperm was then processed for IVF, as the fertilizing capacity is the ultimate parameter of the sperm function. In abnormal group, the fertilization rate(41.5 %) and the ET rate per stimulated cycle were much lower than that of mormal group(69.6%). However, the cleavage rate and the number of embryos transfered per ET cycle were comparable with those of nomal group. The results suggest that the glass-wool filtration of sperm, particularly in oligo-asthenozoospsrmia, may be useful and safe method in the preparation of sperm for IVF.

Estrus Synchronization and Pregnancy Rate Using Ovsynch Method in Uganda Dairy Farms (우간다 낙농가에서 Ovsynch 방법에 의한 발정동기화 및 수태율)

  • The artificial insemination (AI) is one of the best assisted reproductive technologies for increasing reproductive capacity and facilitating the genetic improvement in farm animals. AI has been used in Uganda for over 60 years, but a small population of the total herd has been used. This study was conducted to investigate the efficacy of AI with estrus synchronization technique and to propose ways of improving the productivity of dairy farms through AI services in Uganda. In total, 78 cows from 11 dairy farms were selected for timed-AI. Synchronization was performed according to the ovsynch programs followed by AI using frozen semen from Korean Holstein (0.5 ml straws). Pregnancy rate was varying among farms (0-50%) and the overall pregnancy rate was 28.2%. Cows in luteal phase at the time of treatment was 40.0% whereas that in follicular phase was 20.8%. After treatment, cows that showed normal estrus signal were 45.5% (25/55). Abnormal estrus was categorized into pre-estrus (9.1%), cystic ovaries (21.8%), anestrus (18.2%) and delayed ovulation (5.5%), respectively. These results imply that an assured protocol for timed-AI should be developed to improve the productivity of dairy farms through AI services in Uganda.

  • Pituitary gonadotropes, as target cells, exhibit cyclic changes in terms of LH and FSH release in synchrony with the estradiol levels. The ultimate release is determined by the relative size of the two pools of gonadotropins, which is regulated by the two controllers: LH-RH and estradiol. LH-RH appears to serve as a primary drive on the gonadotrope, stimulating gonadotropin synthesis, storage, and release. Estradiol amplifies the action of LH-RH and induces the development of a self-priming effect of LH-RH except that it impedes LH-RH mediated gonadotropin release. Negative and positive feedback action of estradiol is revealed to operate by different mechanisms. The pituitary capacity increases severalfold from early to late follicular phase, which is considered to be prerequisite for the development of mid-cycle surge. CNS-hypothalamic dopamine, norepinephrine, and prostaglandins, as well as LH-RH, are involved in the negative and positive feedback effects of estradiol. The possible mechanisms in the triggering of LH-RH release for the initiation of midcycle LH-RH surge are considered.

  • Objective: To investigate effects of cryoprotectant and cryopreservation on the chromosome of the human immature oocytes. Design: Intact cumulus-enclosed immature oocytes were collected from unstimulated ovaries and divided into three groups, such as no treatment as control (group 1), only 1,2-propanediol (PROH)-treated (group 2), and cryopreserved oocytes (group 3). Oocytes in group 1, 2, and survived oocytes after cryopreservation in group 3 were cultured for 48 hours. Setting: Infertility Medical Center at the CHA General Hospital, Seoul, Korea. Patients: Oocytes were obtained from Patients undergoing gynecological surgery. Main Outcome Measures: Maturation rate, abnormality in chromosomes by fluorescence in situ hybridization (FISH). Results: There was no effect of PROH only treatment on the chromosomal abnormalities in group 2 compared to control oocytes (41.4% and 31.8%, respectively). Whereas significantly increased abnormalities in chromosome (77.8%) were found in group 3. Conclusions: Human oocytes matured in vitro after cryopreservation at the germinal vesicle (GV) stage showed increased incidence of chromosomal abnormalities. These abnormalities may impair the capacity for further development of the embryos derived from frozen-thawed oocytes.

Comparative Transcriptome Analysis Reveals Differential Response of Phytohormone Biosynthesis Genes in Glumous Flowers of Cold-Tolerant and Cold-Sensitive Rice Varieties Upon Cold Stress at Booting Stage

  • Low temperature stress is one of the major negative factors affecting vegetative and reproductive growth of rice. To better understand responses of rice plants to low temperature we analyzed transcriptome expression patterns in glumous flower of cold-tolerant japonica rice variety, Stejaree45, and cold-susceptible variety, HR19621-AC6 at booting stage under cold water irrigation. A total of 2,411 probes were differentially expressed by low temperature in glumous flowers of the two varieties. Some important genes involved in hormone biosynthesis showed variety-specific regulation. Expression of GA20ox3 and GA2ox, among the genes involved in GA biosynthesis, was regulated differentially in the two varieties. Among the genes involved in IAA biosynthesis, YUCCA1 and TAA1:1 showed variety-specific regulation. Among the genes involved in cytokinin biosynthsis and signaling, expression of LOG, HK1 and HK3 was significantly down-regulated only in the cold-susceptible variety. Among the genes involved in ABA biosynthesis, NSY and AAO3 were down-regulated only in the cold-tolerant variety. In general, genes involved in GA, IAA and cytokinin biosynthesis responded to cold temperature in such a way that capacity of those bioactive hormones is maintained at relatively higher levels under cold temperature in the cold-tolerant variety, which can help minimize cold stress imposed to developing reproductive organs in the cold-tolerant variety.

The impact of two doses of coenzyme Q10 on semen parameters and antioxidant status in men with idiopathic oligoasthenoteratozoospermia

  • Objective: Oxidative stress contributes to male infertility, and antioxidants have been recommended for treating idiopathic oligoasthenoteratozoospermia (OAT). There is, however, a lack of agreement on the type, dosing, and use of individual antioxidants or combinations thereof. The purpose of this study was to compare the effects of two doses of coenzyme Q10 (CoQ10) on semen parameters and antioxidant status in men with idiopathic OAT. Methods: In this prospective study, patients with idiopathic OAT received 200 mg/day (n = 35) or 400 mg/day (n = 30) of CoQ10 orally for 3 months. All patients underwent semen analysis according to the fifth editions of the World Health Organization criteria. Total antioxidant capacity (TAC), catalase (CAT) activity, and superoxide dismutase (SOD) activity were measured both before and after treatment. Results: Treatment with CoQ10 (200 mg/day or 400 mg/day) resulted in a significant increase in sperm concentration from baseline ($8.22{\pm}6.88$ to $12.53{\pm}8.11million/mL$, p= 0.019; $7.58{\pm}5.41$ to $12.33{\pm}6.1million/mL$, p= 0.002, respectively), progressive motility ($16.54%{\pm}9.26%$ to $22.58%{\pm}10.15%$, p=0.011; $14.22%{\pm}12.85%$ to $26.1%{\pm}14.52%$, p= 0.001, respectively), and total motility ($25.68%{\pm}6.41%$ to $29.96%{\pm}8.09%$, p= 0.016; $23.46%{\pm}12.59%$ to $34.82%{\pm}14.17%$, p= 0.001, respectively). CoQ10 therapy also increased TAC (p= 0.009, p= 0.001, respectively), SOD activity (p= 0.004, p= 0.001, respectively), and CAT activity (p= 0.039, p= 0.024, respectively). Furthermore, antioxidant measures correlated significantly with seminal fluid parameters (r = 0.36-0.76). Conclusion: CoQ10 supplementation improved semen parameters and antioxidant status in men with idiopathic OAT, with a greater improvement shown in men who took 400 mg/day than in those who took 200 mg/day.

Identification of sperm motility subpopulations in Gyr falcon (Falco rusticolus) ejaculate: a tool for investigating between subject variation

  • Subgroups of sperm which share similar motility features documented in mammals indicate between-subject variations that might be related to fertilizing potential of the respective ejaculates. The objectives of this study were to define subpopulations of motile sperm in Gyr falcon semen using kinematic parameters driven by Computer Assisted Semen Analysis (CASA) and to investigate the subject-related variations in these subpopulations. A total of 24 fresh ejaculates from 6 falcons were used to assign each of the 20473 sperms into 3 subpopulations by a multivariate cluster analysis. The proportion of sperms in different sub-populations were compared among subjects by a generalized linear model and repeatability of sperm frequency in different subpopulations was investigated by corelation analysis. The resulting 3 categories of sperm indicated significant differences in all kinematic parameters (p < 0.05). Subpopulation 1 (15.91%) contained sperms with the highest velocity and progressiveness of movement trajectory while subpopulation 3 (6.4%) included the least progressively motile sperms. Proportion of rapid and medium progressive sperm were consistently higher in the ejaculate of three falcons compared to the two other birds which also had the highest proportion of slow non-progressive sperms (p < 0.05). Respective proportion of sperms in each subpopulations indicated significant repeatability over multiple measurements (p < 0.05). In conclusion, subpopulations of motile sperm in Gyr falcon can be identified using kinematic parameters generated by CASA. Individual differences in the proportion of these subpopulations might have potential application for identifying the males with higher fertilizing capacity.
