Biological Activities of Larix kaempferi Needles

  • The needles of L. kaempferi was extracted with 95% ethanol and successively partitioned with n-hexane, $CH_2Cl_2$ and EtOAc. Repeated column chromatography on the EtOAc and $H_2O$ soluble fractions gave three flavan-3-ols, one flavone glycoside, six flavonol glycosides and one lignan derivative. Their structures were elucidated on the basis of chemical and spectroscopic evidences. The antioxidant activities of the isolated compounds were evaluated by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method. Flavan compounds indicated good antioxidative potentials compared with BHT (butylated hydroxytoluene) and ${\alpha}$-tocopherol as controls. In the anti-inflammatory test on most of the isolated compounds, NO (nitric oxide) assay against the RAW 264.7 (Mouse Macrophage) showed similar inhibitory potentials to NO production of the control. The cytotoxicity was determined by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay and most of the isolated compounds indicated no toxicity in various concentration.

Effects of Psoralen and Angelicin on Hepatic Drug-Metabolizing Enzyme Activities

  • The effects of psoralen and angelicin on hepatic microsomal drug-metabolizing enzyme (DME) activities were investigated to elucidate the mode of the interaction of furanocoumarins with DME system. A single administration (30 mg/kg,i. p.) of both coumarins to mice cased a significant prolonagation of hexobarbital-induced hypnosis as well as an increase in strychnine toxicity. The inhibitory potencies of both coumarins as measured by rat hepatic microsomal aminopyrine N-demethylase and hexobarbital hydroxylase activities in vitro were considerably weaker than those of other furanocoumarins which possess a side chain moiety. Both coumarins were found to have significant inducing effects of DME system, with repeated treatments of them. The activities of an angular coumarin were stronger than those of a linear coumarin.

Sprague-Dawley Rats을 이용한 결명자 물 추출물의 26주 반복 경구투여 독성시험 및 4주 회복시험 (A 26-Week Repeated Oral Dose Toxicity Test and a 4-Week Recovery Test of Cassia tora L. Water Extract in Sprague-Dawley Rats)

  • Background: Cassia tora L., an annual or perennial plant of the Fabaceae family, is traditional medicine with various biological activities, including anti-constipation and, anti-inflammation. Chemical compounds such as anthraquinone glycoside and naphthalene derivatives have been isolated from this plant. Cassia tora L. is a common contaminant of agricultural commodities, but is toxic to cattle and poultry. Methods and Results: To investigate the potential toxicity, Cassia tora L. aqueous extract (CO) was administered orally to rats for 26 weeks at 0 (control), 300, 1,500 and 3,000 mg/kg/day (n = 10 for male rats for each dose). The positive control comprised animals orally administered anthraquinone 100 mg/kg/day. There was no treatment-related mortality. An increase in the kidney weight was observed at 3,000 mg/kg/day of CO and anthraquinone 100 mg/kg/day. Macrophage infiltration in the colon was observed at CO 1,500 and 3,000 mg/kg/day and anthraquinone 100 mg/kg/day, but there were no significant toxicological changes in the incidence and severity of the finding. Conclusions: The oral no-observed-adverse-effect level (NOAEL) of CO was 3,000 mg/kg/day in male rats and no target organs were identified. In addition, 300 mg/kg was found to be the no-observed-effect level (NOEL) for systemic toxicity under the conditions of the study.

백서의 척수강 내로 장기간 투여한 Cannabinoids의 진통 효과 및 독성에 관한 연구 (Study for the Antinociceptive Effect and Toxicity of Chronic Intrathecal Infusion of Cannabinoids in Rats)

  • Background: Cannabinoids have shown antinociceptive action. The aims of this study were to examine the effect of chronic infusion of a cannabinoids receptors agonist (WIN 55,212-2) for thermal nociception at the spinal level, and to also observe the development of toxicity. Methods: Male Sprague-Dawley rats were implanted with lumbar intrathecal catheters with the nociceptive response (withdrawal response latency) determined by exposing the plantar surface of the hindpaw to radiant heat. Initially, the effect of intrathecal WIN 55,212-2 was evaluated followed by the change in the effect at 1, 2, 3 and 4 weeks after repeated infusion. Finally, the histopathological findings were assessed 1 and 4 weeks following the infusion of WIN 55,212-2. Results: Intrathecal WIN 55,212-2 was found to produce a limited antinociception during the thermal test. %MPE of WIN 55,212-2 at 1, 2, 3, and 4 weeks after infusion was not different from each other. No abnormal pathological findings were observed following a chronic intrathecal infusion of WIN 55,212-2. Conclusions: WIN 55,212-2, a cannabinoids receptors agonist, may be useful in the management of thermal nociception, without changing the effectiveness or causing the toxicity following a chronic infusion at the spinal level.
