벼 엽록체 DNA내의 151 bp 반복염기서열에 의한 유전자 재배열 (Gene Reangement through 151 bp Repeated Sequence in Rice Chloroplast DNA)

  • 엽록체 DNA 내에서 반복 염기서열의 존재와 이들에 의한 유전자 재배열 현상을 고찰하기 위하여 151bp Repeated Sequence 갖는 이질적인 유전인자군의 존재를 여러가지 품종의 벼 엽록체 DNA에서 관찰 하였다. 또한 쌀 DNA를 벼의 생장과 조직부위에 따라 분리하고, rp12 probe를 이용하여 Southern blot 분석하여 엽록체의 발달에 따르는 엽록체 DNA의 재배열 현상을 관찰하였다. 아울러 유전자 재배열 현상을 유발하는 반복염기서열을 database로부터 검색하여 유전자의 상호 비교 분석하였다. 그 결과 151bp Repeated Sequence와 유사한 염기 서열을 같는 rp123유전자를 포함하는 이질적인 유전인자군은 어느 특정한 품종의 벼에 국한되는것이 아니고 본 실험에 사용된 다양한 품종의 벼에 일반적으로 나타나는 현상임이 확인되었으며 또한 이들의 양상은 벼의 조직 부위에 따라 다르게 나타나고 있음을 확인하였다. 이러한 실험적 결과와 함께 엽록체 유전자 database의 검색과 유전자의 상호비교분석을 통하여 151bp 반복 염기 저열에 의한 벼 엽록체 DNA의 유전자 재배열현상은 식물 특히 단자엽 식물의 진화와 함께 발달된 현상으로 특히 151bp반복 염기 서열은 매우 다양한 유전자 재배열을 유발하는 변이유발 위치로 발달되어 왔음을 확인할 수 있었다. 따라서 이러한 반복염기서열에 의한 유전자 재배열 현상은 특히 벼에 있어서 plastid의 발달에 밀접하게 관여하고 있음을 제시하고 있다.

Identification of a Tandemly Repented DNA Sequence Using Combined RAPD and FISH in Welsh Onion (Allium fistulosum)

  • A tandemly repeated DNA sequence was identified and characterized y the combined RAPD and FISH data from a total genomic DNA of Welsh onion (Allium fistulosum). A clone containing this repeating sequence was selected and sequenced. This repeating unit of 314 bp inserted into pAf 072 contained 54.1% adenine and thymine residues, and showed the primer sequence used, 5'-GAAACGGGTG-3', in both terminals of the sequence. Fluorescence in situ hybridization using this tandemly repeated sequence as a probe indicated that the detected sites were coincident with the major C-banded constitutive heterochromatin in the terminal regions of both arms of all 6 chromosomes.

  • 변종 악성코드는 그 기능에 있어 차이가 없으나 구조적인 차이가 존재하는 악성코드로, 같은 그룹으로 분류하여 처리하는 것이 유용하다. 변종 악성코드 분석을 위해 본 논문에서는 바이오인포매틱스 분야에서 사용하는 서열 정렬 기법을 사용하여 악성코드들의 API 호출 정보 간의 공통부분을 찾고자 하였다. 서열 정렬 기법은 API 호출 정보의 길이에 대해 의존적인 성능을 가지며, API 호출 정보의 길이가 커짐에 따라 성능이 매우 떨어진다. 따라서 본 논문에서는 서열 정렬 기법 적용 이전에 API 호출 정보에서 발견되는 반복 패턴을 제거하는 방법을 적용함으로써 성능이 보장될 수 있도록 하였다. 최종적으로 서열 정렬 기법을 통한 악성코드 간의 유사도를 구하는 방법에 대하여 논하였다. 또한 실제 악성코드 샘플에 대한 실험 결과를 제시하였다.

기주가 다른 Magnaporthe grisea 균주간의 Polymorphism과 유전적 유연관계 분석 (Polymorphism and Genetic Relationships Among Magnaporthe grisea Isolates Obtained from Various Hosts by Using Repetitive DNA Sequences)

  • 도열병균, Magnaporthe grisea, 균주간의 유전적 유연관계를 분석하고 그들의 유전에 관한 '기본 정보를 얻고자 DNA polymorphism 분석을 실시하였다. 기주가 다른 도열병 균주들이 공시되었고 cloning에 의해 벼 도열병균 KJ201레이스 균주로부터 생성된 임의 선발 genomic clone들이 공시균주들간의 polymorphism을 밝히기 위해 사용되었던 바 그중 repectitive sequence를 보유한 repeated copy clone 하나가 선발되었다. Clone pMJ6에 의해 밝혀진 repetitive sequence는 Southern hybridization시 벼 분리균주에는 약 30개, 다른 기주 분리균에도 20∼33개의 밴드를 형성하였다. 반면 피 분리균주에는 단지 두 개의 밴드만을 나타내 분리기주가 다른 균주간에 뚜렷한 polymorphism이 존재하였으며 parsimony 분석에서도 역시 아주 먼 cluster를 형성하여 피 분리균은 다른 기주 분리균과 유전적으로 상당히 먼 것으로 추정되었다. 공시균의 genomic DNA를 HindIII로 처리했을 때 pMJ6에 의한 밴드양상은 공시균을 EcoRI으로 처리했을 때의 MGR probe의 밴드 양상과 유사하여 이 repeated copy clone이 도열병균주간의 유전적 유연관계를 분석하는데 MGR 못지않게 유용할 것으로 보인다.

  • DNA computing based DNA sequence Is operated through the biology experiment. Biology experiment used as operator causes illegal reactions through shifted hybridization, mismatched hybridization, undesired hybridization of the DNA sequence. So, it is essential to design DNA sequence to minimize the potential errors. This paper proposes method of the DNA sequence generation based evolutionary operation processor. Genetic algorithm was used for evolutionary operation and extra hardware, namely genetic algorithm processor was implemented for solving repeated evolutionary process that causes much computation time. To show efficiency of the Proposed processor, excellent result is confirmed by comparing between fitness of the DNA sequence formed randomly and DNA sequence formed by genetic algorithm processor. Proposed genetic algorithm processor can reduce the time and expense for preparing DNA sequence that is essential in DNA computing. Also it can apply design of the oligomer for development of the DNA chip or oligo chip.

Enhanced Coupling of $M_1$ Muscarinic Receptors to Activation of Phospholipase C upon Mutation of a Transposed Amino Acid Triplet Repeat

  • The C-terminus ends of the second putative transmembrane domains of both $M_1$ and $M_2$ muscarinic receptors contain a triplet of amino acid residues consisting of leucine (L), tyrosine (Y) and threonine (T). This triplet is repeated as LYT-TYL in $M_1$ receptors at the interface between the second transmembrane domain and the first extracellular loop. Interestingly, however, it is repeated in a transposedfashion (LYT-LYT) in the sequence of $M_2$ receptors. In our previous work, we investigated the possible significance of this unique sequence diversity for determining the distinct differential receptor function at the two receptor subtypes. However, we found mutation of the LYTTYL sequence of $M_1$ receptors to the corresponding $M_2$ receptor LYTLYT sequence demonstrated markedly enhanced the stimulation of phosphoinositide (PI) hydrolysis by carbachol without a change in its coupling to increased cyclic AMP formation. In this work, thus, the enhanced stimulation of PI hydrolysis in the LYTLYT $M_1$ receptor mutant was further investigated. The stimulation of PI hydrolysis by carbachol was enhanced in the mutant $M_1$ receptor, and this change was not due to alterations in the rate of receptor desensitization or sequestration. The observed larger response to carbachol at mutant $M_1$ receptors was also not due to an artifact resulting from selection of CHO cells which express higher levels of G-proteins or phospholipase C. Our data suggest that although the LYTTYL sequence in $M_1$ muscarinic receptors is not involved in determining receptor pharmacology, mutation of the sequence enhanced the coupling of $M_1$ receptors to the stimulation of phospholipase C.

다중충돌 피닝잔류응력 평가를 위한 소성숏이 포함된 3차원 유한요소해석 모델 (A 3D FEA Model with Plastic Shots for Evaluation of Peening Residual Stress due to Multi-Impacts)

  • In this paper, we propose a 3-D finite element (FE) analysis model with combined physical behavior and kinematical impact factors for evaluation of residual stress in multi-impact shot peening. The FE model considers both physical behavior of material and characteristics of kinematical impact. The physical parameters include elastic-plastic FE modeling of shot ball, material damping coefficient, dynamic friction coefficient. The kinematical parameters include impact velocity and diameter of shot ball. Multi-impact FE model consists of 3-D symmetry-cell. We can describe a certain repeated area of peened specimen under equibiaxial residual stress by the cell. With the cell model, we investigate the FE peening coverage, dependency on the impact sequence, effect of repeated cycle. The proposed FE model provides converged and unique solution of surface stress, maximum compressive residual stress and deformation depth at four impact positions. Further, in contrast to the rigid and elastic shots, plastically deformable shot produces residual stresses closer to experimental solutions by X-ray diffraction. Consequently, it is confirmed that the FE model with peening factors and plastic shot is valid for multi-shot peening analyses.

Moment resisting steel frames under repeated earthquakes

  • In this study, a systematic investigation is carried out on the seismic behaviour of plane moment resisting steel frames (MRF) to repeated strong ground motions. Such a sequence of earthquakes results in a significant damage accumulation in a structure because any rehabilitation action between any two successive seismic motions cannot be practically materialised due to lack of time. In this work, thirty-six MRF which have been designed for seismic and vertical loads according to European codes are first subjected to five real seismic sequences which are recorded at the same station, in the same direction and in a short period of time, up to three days. Furthermore, the examined frames are also subjected to sixty artificial seismic sequences. This investigation shows that the sequences of ground motions have a significant effect on the response and, hence, on the design of MRF. Additionally, it is concluded that ductility demands, behaviour factor and seismic damage of the repeated ground motions can be satisfactorily estimated using appropriate combinations of the corresponding demands of single ground motions.

Mutation of a Transposed Amino Acid Triplet Repeat Enhances Coupling of m1 Muscarinic Receptor to Activation of Phospholipase C

Interestingly, however, it is repeated in a transposed fashion (LYT-TYL) in the sequence of m1 receptors. In this work we employed site-directed mutagenesis to investigate the possible significance of this unique sequence diversity for determining the distinct differential drug-receptor interaction and cellular function at m1 muscarinic receptor. Mutation of the LYTTYL sequence of m1 receptors to the corresponding m2 receptor LYTLYT sequence, however, did not result in a significant change in the binding affinity of the agonist carbachol or in the affinity of the majority of a series of receptor antagonists which are able to discriminate between wild-type m1 and m2 receptors. Surprisingly, the LYTLYT ml receptor mutant demonstrated markedly enhanced coupling to activation of phospholipase C without a change in its coupling to increased cyclic AMP formation. There was also an enhanced receptor sensitivity in transducing elevation of intracellular Ca$\^$2+/. These changes were not due to alterations in the rate of receptor. desensitization or sequestration, On the other hand, the reverse LYTLYT-LYTTYL mutation in the m2 receptor did not alter its coupling to inhibition of adenylate cyclase, but slightly enhanced its coupling to stimulation of PI hydrolysis, Our data suggest that the LYTTYL/LYTLYT sequence difference between ml and n12 muscarinic receptors is not involved in determining receptor pharmacology. On the other hand, while these differences might play a role in the modulation of muscarinic receptor coupling to PI hydrolysis, they are not important for specifying coupling of various subtypes of muscarinic receptors to different cellular signaling pathways.

  • A bag is a set-like entity which can contain repeated elements. Fuzzy bags have been studied by Yager, who defined their basic relations and operations. However, his definitions of the basic relations and operations are inconsistent with the corresponding relations and operations for ordinary fuzzy sets. The present paper presents new basic relations and operations of fuzzy bags using a grade sequence for each element of the universal set. Moreover the .alpha.-cut, t-norms, the extension principle, and the composition of fuzzy bag relations are described.
