사간마황탕(射干麻黃湯) 및 <석실비록(石室秘錄)> 역의법방(逆醫法方)이 호흡기 점액의 분비에 미치는 영향 (The Effects of Saganmahwang-tang and prescription C on airway mucin secretion)

  • Objectives : This study was done to investigate whether two oriental prescriptions, saganmahwang-tang (SMT) and prescription C (P-C) significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Methods : Cofluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of SMT or P-C to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase (LDH) release. Also, the effects of SMT and P-C on contractility of isolated tracheal smooth muscle were investigated. Results : SMT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity. P-C significantly increased mucin release from cultured HTSE cells, with significant cytotoxicity. SMT inhibited Ach-induced contraction of isolated tracheal smooth muscle. P-C did not affect Ach-induced contraction of isolated tracheal smooth muscle. Conclusion : Results su99est that SMT and P-C have regulating effects on mucin secretion from airway goblet cells. Further investigation is needed, because of the value in finding novel agents to this purpose, and these oriental medical prescription have potential for such a role.

  • The intent of this study is to investigate whether two oriental medical prescriptions named haengsotang(HST) and gami-palmihwan(GPMH) significantly effect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells, Confluent HTSE cells were metabolically radiolabeled with $^3H-glucosamine$ for 24 hrs and chased for 30 min in the presence of HST or GPMH to assess the effect of each agent on $^3H-mucin$ release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of HST and GPMH on contractility of isolated tracheal smooth muscle were investigated. The results are consistant with the following assertions: (1) HST significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) GPMH did not effect mucin release without cytotoxicity; (3) HST and GPMH did not effect contractility of isolated tracheal smooth muscle. These results suggest a need for further investigation of HST and its components, for its potential in oriental medicine prescriptions and novel agents that effectively regulate (inhibit) mucin secretion from airway goblet cells.

아크릴릭 레진에 혼합된 클로르헥시딘의 방출 : 새로운 방법의 약물송달시스템을 위한 예비실험 (EVIDENCE OF SUSTAINED RELEASE OF CHLORHEXIDINE ADDED TO ACRYLIC RESIN : PRELIMINARY INVESTIGATION OF A POTENTIAL DRUG DELIVERY SYSTEM)

  • For more than two decades, many investigators have tried a variety of methods for delivering antimicrobial agents to the oral cavity with the objective of eliminating mutans streptococci. In the belief that the effectiveness of chemotherapy might be improved by a more effective delivery system, the intention of the present study was to exploit a new drug delivery system delivering chlorhexidine to the oral cavity. The vehicle delivering chlorhexidine tested in this study was self-curing acrylic resin(polymethyl methacrylate). The powder of the acrylic resin was polymerized with the 5 different liquid preparations, in which $Chlorzoin^{(R)}$ was mixed with five different monomer/Chlorzoin ratios immediately prior to the polymerization, in a stainless steel mold ($40mm{\times}40mm{\times}2mm$). A total of 50 cured resin specimens were divided into 5 groups according to the different monomer preparations. Every specimen was soaked in an airtight container filled with distilled water (100 ml) and then kept in an incubator at $37^{\circ}C$. The solutions (0.8 ml) were collected from the container at every 24 hours, and the amount of released chlorhexidine in the solutions was measured in an ultraviolet spectrophotometer at 250nm. The container was refilled with distilled water every after measurement. This procedure was repeated for 14 days. It was found that chlorhexidine was continuously released from all of the 50 specimens during the experimental period. And it was noted that the pattern of chlorhexidine release was a type of sustained-release preparation, that is, the amount of the released chlorhexidine at the first day in all 5 groups was high (p<0.0001), and then the release was decreased during the rest of the experimental period (p<0.001).

  • To develop a sustained-release tablet which had the similar dissolution to commercial ibudilast-loaded sustained-release capsule, the tablets were prepared using hydroxypropylmethylcellulose phthalate (HPMCP), ethylcellulose (EC) and hydroxypropylcellulose (HPC), and dissolution test were carried out with paddle method in KP. The tablet prepared only with HPMCP and EC showed sno similar dissolution pattern to the commercial product. As the ratio of HPMCP/HPC in tablet decreased, the dissolution rate of drug decreased in pH 1.2 but increased in pH 6.8. Furthermore, an ibudilast-loaded sustained-release tablet composed of [ibudilast/EC/HPMCP/HPC (10/10/170/10 mg/tab)] gave similar dissolution to commercial product in pH 1.2 for 3 h and in pH 6.8 for 10 h. Thus, it could be a potential candidate for the substitute of commercial capsule.

Effects of Extracellular Electron Shuttles on Microbial Iron Reduction and Heavy Metals Release from Contaminated Soils

  • To test the potential effects of extracellular electron shuttles (EES) on the rate and extent of heavy metal release from contaminated soils during microbial iron reduction, we created anaerobic batch systems with anthraquinone-2,6-disulfonate (AQDS) as a surrogate of EES, and with contaminated soils as mixed iron (hydr)oxides and microbial sources. Two types of soils were tested: Zn-contaminated soil A and As/Pb-contaminated soil B. In soil A, the rate of iron reduction was fastest in the presence of AQDS and > 3500 mg/L of total Fe(II) was produced within 2 d. This suggests that indigenous microorganisms can utilize AQDS as EES to stimulate iron reduction. In the incubations with soil B, the rate and extent of iron reduction did not increase in the presence of AQDS likely because of the low pH (< 5.5). In addition, less than 2000 mg/L of total Fe(II) was produced in soil B within 52 d suggesting that iron reduction by subsurface microorganisms in soil B was not as effective as that in soil A. Relatively high amount of As (~500 mg/L) was released to the aqueous phase during microbial iron reduction in soil B. The release of As might be due to the reduction of As-associated iron (hydr)oxides and/or direct enzymatic reduction of As(V) to As(III) by As-reducing microorganisms. However, given that Pb in liquid phase was < 0.3 mg/L for the entire experiment, the microbial reduction As(V) to As(III) by As-reducing microorganisms has most likely occurred in this system. This study suggests that heavy metal release from contaminated soils can be strongly controlled by subsurface microorganisms, soil pH, presence of EES, and/or nature of heavy metals.

A SYSTEMS ASSESSMENT FOR THE KOREAN ADVANCED NUCLEAR FUEL CYCLE CONCEPT FROM THE PERSPECTIVE OF RADIOLOGICAL IMPACT

  • In this study, we compare the mass release rates of radionuclides(1) from waste forms arising from the KIEP-21 pyroprocessing system with (2) those from the directly-disposed pressurized-water reactor spent fuel, to investigate the potential radiological and environmental impacts. In both cases, most actinides and their daughters have been observed to remain in the vicinity of waste packages as precipitates because of their low solubility. The effects of the waste-form alteration rate on the release of radionuclides from the engineered-barrier boundary have been found to be significant, especially for congruently released radionuclides. the total mass release rate of radionuclides from direct disposal concept is similar to those from the pyroprocessing disposal concept. While the mass release rates for most radionuclides would decrease to negligible levels due to radioactive decay while in the engineered barriers and the surrounding host rock in both cases even without assuming any dilution or dispersal mechanisms during their transport, significant mass release rates for three fission-product radionuclides, $^{129}I$, $^{79}Se$, and $^{36}Cl$, are observed at the 1,000-m location in the host rock. For these three radionuclides, we need to account for dilution/dispersal in the geosphere and the biosphere to confirm finally that the repository would achieve sufficient level of radiological safety. This can be done only after we have known where the repository site would by sited. the footprint of repository for the KIEP-21 system is about one tenth of those for the direct disposal.

Calculation of preliminary site-specific DCGLs for nuclear power plant decommissioning using hybrid scenarios

  • Korea's first commercial nuclear power plant at Kori site was permanently shut down in 2017 and is currently in transition stage. Preparatory activities for decommissioning such as historical site assessment, characterization, and dismantling design are being actively carried out for successful D&D (Dismantling and Decontamination) at Kori site. The ultimate goal of decommissioning will be to ensure the safety of workers and residents that may arise during the decommissioning of nuclear facilities and, thereby finally returning the site to its original status in accordance with the release criteria. Upon completion of decommissioning, the resident's safety at a site released will be assessed from the evaluation of dose caused by radionuclides expected to be present or detected at the site. Although the U.S. commercial nuclear power plants with decommissioning experience use different site release criteria, most of them are 0.25 mSv/y. In Korea, both the unrestricted and restricted release criteria have been set to 0.1 mSv/y by the Nuclear Safety and Security Commission. However, since the dose is difficult to measure, measurable concentration guideline levels for residual radionuclides that result in dose equivalent to the site release criteria should be derived. For this derivation, site reuse scenario, selection of potential radionuclides, and systematic methodology should be developed in planning stage of Kori site decommissioning. In this paper, for calculation of a preliminary site-specific Derived Concentration Guideline Levels (DCGLs) for the Nuclear Power Plant site, a novel approach has been developed which can fully reflect practical reuse plans of the Kori site by taking into account multiple site reuse scenarios sequentially, thereby striking a remarkable distinction with conventional approaches which considers only a single site scenario.

Characterization of alginate/carboxymethyl scleroglucan hydrogels as a delivery system for protein drug

  • The aim of this study was to prepare a hydrogels composed of alginate blended with a carboxymethyl scleroglucan (CMSC) and evaluate the feasibility of the hydrogels as a drug delivery system for a protein. The main advantage of the alginate/CMSC hydrogels is to improve a restricted drug release from alginate hydrogels. The CMSC was chemically synthesized with chloroacetic acid and confirmed using a FT-IR. The alginate/CMSC hydrogels were prepared at distinct compositions by crosslinking with calcium ions. The swelling ratios of these hydrogels increased significantly with increasing the content of CMSC. At pH 7.4, the swelling ratios of the hydrogels increased remarkably as compared to those at pH 1.2. In ovalbumin (OVA) release test, the amount of OVA released from the hydrogels showed higher as compared to those released at pH 1.2. In addition, the release of OVA was improved with increasing the content of CMSC. Thus, the alginate/CMSC hydrogels may be used as a potential system for oral delivery of protein drugs.

A Polymeric Antibacterial Agent with Sustained Anti-bacterial Activity: Cellulose Xanthate-metal-neomycin Complexes

  • Neomycin coupled to a polymer matrix via a metal linker was prepared and evaluated for prolonging antibacterial activity. Microcrystallized cellulose was chemically modified to cellulose xanthate(MCX) to afford metal binding sites. MCX was treated with Cu(II), Fe(III) or Zn(II) followed by reaction with neomycin (Ne). The release of Ne from MCX-Zn(II)-Ne was investigated and its activity duration was measured by ditch plate method. The amount of metal bound to MCX was 0.36 mmol/g matrix, 0.26 mmol/g matrix and 0.56 mmol/g matrix for Cu(II), Zn(II) and Fe(III), respectively. Ne bound to MCX-metal chelates was 0.006 mmol, 0.07 mmol and 0.01 mmol per g MCX for Cu(II), Zn(II) and Fe(III), respectively. The Ne release from MCX-Zn(II)-Ne was sustained even after seven washes, whereas Ne from MC/Zn(II)/Ne mixture was almost completely released in two washes. Antibacterial activity was prolonged with MCX-Zn(II)-Ne and MCX-Fe(III)-Ne, but not with MCX-Cu(II)-Ne when compared with that of free Ne. Taken together, these results suggest that neomycin coupled to MCX via a proper metal linker has a potential as a polymeric antibacterial agent with sustained activity.

The influence of initial stresses on energy release rate and total electro-mechanical potential energy for penny-shaped interface cracks in PZT/Elastic/PZT sandwich circular plate-disc

  • This paper studies the energies and energy release rate (ERR) for the initially rotationally symmetric compressed (or stretched) in the inward (outward) radial direction of the PZT/Elastic/PZT sandwich circular plate with interface penny-shaped cracks. The investigations are made by utilizing the so-called three-dimensional linearized field equations and relations of electro-elasticity for piezoelectric materials. The quantities related to the initial stress state are determined within the scope of the classical linear theory of piezoelectricity. Mathematical formulation of the corresponding problem and determination of the quantities related to the stress-strain state which appear as a result of the action of the uniformly normal additional opening forces acting on the penny-shaped crack's edges are made within the scope of the aforementioned three-dimensional linearized field equations solution which is obtained with the use of the FEM modelling. Numerical results of the energies and ERR and the influence of the problem parameters on these quantities are presented and discussed for the PZT- 5H/Al/PZT-5H, PZT-4/Al/PZT-4, $BaTiO_3/Al/BaTiO_3$ and PZT-5H/StPZT-5H sandwich plates. In particular, it is established that the magnitude of the influence of the piezoelectricity and initial loading on the ERR increases with crack radius length.
