DNA double-strand break (DSB) is a serious treat for the cells including mutations, chromosome rearrangements, and even cell death if not repaired or misrepaired. Ku heterodimer regulatory DNA binding subunits (Ku70/Ku80) bound to double strand DNA breaks are able to interact with 470-kDa DNA-dependent protein kinase catalytic subunit (DNA-PKcs), and the interaction is essential for DNA-dependent protein kinase (DNA-PK) activity. The Ku80 mutants were designed to bind Ku70 but not DNA end binding activity and the peptides were treated in breast cancer cells for co-therapy strategy to see whether the targeted inhibition of DNA-dependent protein kinase (DNA-PK) activity sensitized breast cancer cells to ionizing irradiation or chemotherapy drug to develop a treatment of breast tumors by targeting proteins involved in damage-signaling pathway and/or DNA repair. We designed domains of Ku80 mutants, 26 residues of amino acids (HN-26) as a control peptide or 38 (HNI-38) residues of amino acids which contain domains of the membrane-translocation hydrophobic signal sequence and the nuclear localization sequence, but HNI-38 has additional twelve residues of peptide inhibitor region. We observed that the synthesized peptide (HNI-38) prevented DNA-PKcs from binding to Ku70/Ku80, resulting in inactivation of DNA-PK complex activity in breast cancer cells (MDA-465 and MDA-468). Consequently, the peptide treated cells exhibited poor to no DNA repair, and became highly sensitive to irradiation or chemotherapy drugs. The growth of breast cancer cells was also inhibited. These results demonstrate the possibility of synthetic peptide to apply breast cancer therapy to induce apoptosis of cancer cells.

A new set of functional group interaction energy descriptors relevant to the ACE (Angiotensin-Converting Enzyme) inhibitory peptide, QSAR (Quantitative Structure Activity Relationships), is presented. The functional group interaction energies approximate the charged interactions and distances between functional groups in molecules. The effective energies of the computationally derived geometries are useful parameters for deriving 3D-QSAR models, especially in the absence of experimentally known active site conformation. ACE is a regulatory zinc protease in the renin-angiotensin system. Therapeutic inhibition of this enzyme has proven to be a very effective treatment for the management of hypertension. The non bond interaction energy values among functional groups of six-feature of ACE inhibitory peptides were used as descriptor terms and analyzed for multivariate correlation with ACE inhibition activity. The functional group interaction energy descriptors used in the regression analysis were obtained by a series of inhibitor structures derived from molecular mechanics and semi-empirical calculations. The descriptors calculated using electrostatic and steric fields from the precisely defined functional group were sufficient to explain the biological activity of inhibitor. Application of the descriptors to the inhibition of ACE indicates that the derived QSAR has good predicting ability and provides insight into the mechanism of enzyme inhibition. The method, functional group interaction energy analysis, is expected to be applicable to predict enzyme inhibitory activity of the rationally designed inhibitors.

Preproenkephalin (PPE) is a precursor molecule for multiple endogenous opioid peptides Leu-enkephalin (ENK) and Met-ENK, which are involved in a wide variety of modulatory functions in the nervous system. Despite the functional importance of ENK in the brain, the effect of brain-derived factor(s) on PPE expression is unknown. We report the dual effect of neural epidermal growth factor (EGF)-like-like 2 (NELL2) on PPE gene expression. In cultured NIH3T3 cells, transfection of NELL2 expression vectors induced an inhibition of PPE transcription intracellularly, in parallel with downregulation of protein kinase C signaling pathways and extracellular signal-regulated kinase. Interestingly, these phenomena were reversed when synthetic NELL2 was administered extracellularly. The in vivo disruption of NELL2 synthesis resulted in an increase in PPE mRNA level in the rat brain, suggesting that the inhibitory action of intracellular NELL2 predominates the activation effect of extracellular NELL2 on PPE gene expression in the brain. Biochemical and molecular studies with mutant NELL2 structures further demonstrated the critical role of EGF-like repeat domains in NELL2 for regulation of PPE transcription. These are the first results to reveal the spatio-specific role of NELL2 in the homeostatic regulation of PPE gene expression.
방선균은 토양속에 서식하는 그람 양성 세균으로 세포성 장의 어느 시기에 영양세포가 이어져 연쇄상의 기균사를 형성하고 그 끝에 포자를 형성하는 동시에 생리학적 분화로 표현되는 다양한 이차대사물질을 생산한다. 이들의 복잡한 생활사에 따른 분화에는 진핵생물의 ser/thr protein kinase와 원핵생물의 his/asp acid protein kinase 등과 같은 다양한 신호전달 단백질들이 조절을 담당하고 있다. Akt kinase는 진핵생물에서 보고된 ser/thr kinase로.세포내의 다양한 신호전달기구를 조절하고 있으며, 세포내의 Akt kinase의 활성화 또는 불활성화가 세포 증식, 분화, 생존, 세포사등의 신호전달에 결정적인 역할을 담당한다. 방선균으로부터 Akt kinase와 유사한 기능을 갖는 신호전달 단백질을 규명하기 위하여, Akt kinae의 target단백질들의 인산화 부위 보존영역으로부터 나타나는 아미노산의 consensus sequence를 기초로 하여 형광물질로 라벨시킨 합성 peptide(FITC-TRRSRfESIT)를 제작하였다 제작한 기질 peptide에 인산화가 일어나면 아가 로스 전기영동상에서의 운동성에 차이가 나타나고, 이를 자외선하에서 형광 peptide를 관찰하는 방법으로 인산화 assay를 실시하였다. S. griseus IFO 13350을 배양한 cell-free extract로부터 ammonium sulfate fractionation과 DEAE-Sepharose, Mono Q, Resource Phenyl-Superose, Gel permeation 등 수 단계의 column chromatography를 통하여 Akt 유사 단백질을 정제하였다. 그 결과 방선균에도 고등생 물의 Akt와 유사한 기질특이성을 갖는 인산화 단백질이 존재하는 것으로 판단되었으며, 그 중의 하나는 분자량이 39 kDa 정도의 크기를 갖는 단백질로 판명되었다. 지금까지의 인산화 단백질 연구는 활성측정법이 어려워 연구자들에게 많은 제한을 주어 왔지만, 본 연구에서 사용한 합성 peptide를 이용하는 방법을 보다 다양한 인산화 단백질에 대하여 적용한다면, 인산화 단백질 및 조절물질 개발에 많은 도움이 될 수 있을 것으로 예상된다.
Bacillus subtilis strains produce a broad spectrum of bioactive peptides. The lipopeptide surfactin belongs to one well-known class, which includes amphiphilic membrane-active biosurfactants and peptide antibiotics. Both the srfA promoter and the ComP-ComA signal transduction system are an important part of the factor that results in the production of surfactin. Bs-M49, obtained by means of low-energy ion implantation in wild-type Bs-916, produced significantly lower levels of surfactin, and had no obvious effects against R. solani. Occasionally, we found strain Bs-M49 decreased spore formation and the development of competence. Blast comparison of the sequences from Bs-916 and M49 indicate that there is no difference in the srfA operon promoter PsrfA, but there are differences in the coding sequence of the comA gene. These differences result in three missense mutations within the M49 ComA protein. RT-PCR analyses results showed that the expression levels of selected genes involved in competence and sporulation in both the wild-type Bs-916 and mutant M49 strains were significantly different. When we integrated the comA ORF into the chromosome of M49 at the amyE locus, M49 restored hemolytic activity and antifungal activity. Then, HPLC analyses results also showed the comA-complemented strain had a similar ability to produce surf actin with wild-type strain Bs-916. These data suggested that the mutation of three key amino acids in ComA greatly affected the biological activity of Bacillus subtilis. ComA protein 3D structure prediction and motif search prediction indicated that ComA has two obvious motifs common to response regulator proteins, which are the N-terminal response regulator receiver motif and the C-terminal helix-turn-helix motif. The three residues in the ComA N-terminal portion may be involved in phosphorylation activation mechanism. These structural prediction results implicate that three mutated residues in the ComA protein may play an important role in the formation of a salt-bridge to the phosphoryl group keeping active conformation to subsequent regulation of the expression of downstream genes.
CCK and cholinergic agonist stimulate enzyme release from the pancreatic acini via G-protein-mediated activation of phospholipase C, In contrast secretin and related peptides increase the level of cAMP and activate cAMP-dependent protein kinase. Camostat, a synthetic protease inhibitor, causes pancreatic hypertrophy and hyperplasia by increasing the CCK release. In this study, the secretagogue-induced changes of intracellular proteins were examined in the dispersed pancreatic acini of rats with or without camostat treatment. Camostat(FOY-305, 200 mg/kg, p.o.) was given for 4 days twice daily and the dispersed acini were prepared at 12 bouts after last treatment. The profiles of Intracellular phosphoproteins were analyzed by two-dimensional gel electrophoresis after incubating the acini with $^{32}P$. The amylase release from the dispersed acini was measured. The pancreatic weight was increased to 126% of control, while amylase activity per mg acinar protein decreased to 41% of control, The maximum response of amylase release from dispersed acini to CCK-8 or carbachol was markedly decreased(65% or 46% of control, respectively). The group of intracellular proteins(24 kD, pI $4.5{\sim}8.5$) was increased in quantity by camostat. CCK-8 or secretin increased phosphorylation of a protein(34 kD, pI 4.7) in camostat-treated as well as control rats. CCK-8 increased tyrosine phosphoryiation in the acini of control rats. However, in camostat-treated rats, the basal level of tyrosine phosphorylation was increased and it was rather decreased by CCK-8. Secretin had no effect on the level of tyrosine phosphorylation in acini. These results indicate that both phospholipase C and adenylate cyclase induce phosphorylation of an intracellular acinar protein(34 kD, pI 4.7) and camostat treatment increases the basal level of tyrosine phosphorylation in acinar cells. And these results suggest that not only serine/threonine protein kinase but also protein tyrosine kinase/phosphatase are involved in the process of CCK receptor mediated stimulation-secrelion coupling.
돼지의 난소 조직내에 존재하는 심방이뇨 호르몬 수용체 분포를 알아 보기 위하여 자가방사법을 통해 125I로 표지한 rANP(1-28)의 특이적 결합 부위를 관찰하였다. 난소 조직 중 125I-rANP(1-28)의 강한 결합 부위는 난포의 과립막 세포층이었으며, 외난포막층에서도 125I-rANP(1-28)의 결합 부위가 관찰되었다. 그러나 내난포막층을 포함한 난소내의 다른 조직과 특히 황체에서는 125I-rANP(1-28)의 결합 부위가 나타나지 않았다. 난포의 과립막 세포층과 외난포막층에서의 이러한 125I-rANP(1-28)의 결합은 다량의 rANP(1-28)에 의하여 완전히 전위되었지만, 펩티드 호르몬인 angiotensin II 및 arginine vasopressin에 의해서는 과량의 농도에서도 전위되지 않아 125I-rANP(1-28)의 결합이 특이적임을 확인하였다. 또한 이러한 특이적 결하은 심방이뇨 호르몬의 생물학적 수용체 외에, 다른 기능을 담당하는 clearance 수용체의 특이적 ligand인 C-ANF에 의해서도 전이되었다. 이상의 결과는 돼지의 난소에 있어서 난포의 과립막 세포층 및 외난포막에 심방이뇨 호르몬의 생물학적 또는 coearance 수용체가 존재함을 보여주며, 이는 심방이뇨 호르몬의 수용체가 난자의 성숙에 관련된 난포의 발달과정에 관여할 수 있음을 시사한다.
여포와 난포의 성숙, 배란과 착상, 임신의 유지와 태아의 성장 발달, 분만 및 유선발육과 비유 등 일련의 생식현상에서 있어서 성선자극호르몬과 스테로이드 호르몬의 작용이 중추적인 역할을 한다는 사실은 오래 전부터 알려져왔다. 그러나 이러한 일련의 현상에 고전적인 호르몬 외에도 다수의 성장인자가 관여되고 있음이 최근의 연구 결과 밝혀지고 있다. 생식기관에서 성장인자들은 대부분 autocrine/paracrine mode로 작용하여, 성선자극호르몬과 스테로이드 호르몬의 작용을 매개하거나 이들 호르몬 등과 교호적인 작용(synergy)을 한다. 생식기관 내 insulin-like growth factor(IGF) system은 최근 가장 활발히 연구된 분야 중의 하나로 생식현상의 전반에 걸쳐 중요한 역할을 하고 있음이 밝혀졌다. 본 지면에서는 IGF system에 관한 개괄적인 정보를 소개하고 현재까지 보고된 intrauterine IGF system에 관한 연구를 요약하고자 한다. IGF family는 IGF-I과 IGF-II ligands, 두종류의 IGF receptors(수용체), 그리고 지금까지 발견된 6종류의 IGF-binding proteins(IGFBPs)로 이루어져 있다. IGF-I과 IGF-II는 proinsulin과 상동한 구조를 가진 peptide로서 포도당과 아미노산 운반을 자극하는 등 insulin과 유사한 작용을 한다. 이 외에도 IGFs는 세포분열촉진제(mitogens)로서 여러 형태의 세포에 걸쳐 세포증식을 자극하고, 세포의 분화(differentiation)과 세포기능의 발현에 관여한다. IGFs는 간과 주로 messenchymal cells에서 발현되어 endocrine mode는 물론 autocrine/paracrine mode로 거의 모든 조직에 작용한다. IGF 수용체는 두종류가 알려져 있는데 type I IGF receptor는 tyrosine kinase로서 IGF-I과 IGF-II에 공히 high-affinity를 나타내고, 상기한 대부분의 IGFs의 작용을 매개한다. Type II IGF receptor 혹은 IGF-II/mannose-6-phosphate receptor는 두개의 서로 다른 binding sites를 가지고 있는데 IGF-II binding site는 IGF-II에만 high-affinity를 나타낸다. Type II IGF receptor의 주요 역할은 IGF-II를 lysosomal targeting하여 ligand를 파괴하는데 있다. 체액 속의 IGFs는 대부분 IGFBP에 결합되어 있다. IGFBPs는 IGF의 저장/운반체 혹은 IGF 작용을 조절하는 역할을 하는 것으로 알려져 있으나 개개 IGFBP의 역할에 대해서는 지극히 제한된 정보만이 알려져 있다. IGFBPs의 IGF ligands에의 affinity는 IGF receptors의 IGFs에의 affinity보다 크기 때문에 대부분의 in vitro 상황 하에서 IGFBPs는 IGF 작용을 억제한다. IGFBP에 결합되어 있는 IGF가 어떤 기작에 의해 IGFBP로부터 분리되어 IGF receptor에 도달하는지는 알려지지 않고 있으나, 혈액과 조직액에 들어있는 불특정 IGFBP protease activity는 IGF의 방출과정에서 일역을 하는 것으로 믿어지고 있다. 최근 연구보고에 의하면 특정 in vitro 상황 하에서 IGFBP-1, -3, -5 등은 IGF와 무관한 작용도 있다는 증빙이 있어 IGF system의 또 다른 차원을 예고하고 있다. IGF family members의 mRNAs & proteins는 영장류, 설치류 및 가축의 자궁조직과 수태물(conceptus)에서 발현되어 자궁과 태아의 성장 발달에 중요한 역할을 한다. 자궁조직의 IGF system의 발현은 성선호르몬, 국소 생리조절인자 등에 의해 발현시기와 장소의 특이성이 결정되며, 발현된 IGFs와 IGFBPs는 autocrine/paracrine mode로 자궁조직에 작용하기도 하고, 자궁강에 분비되어 수태물의 성장 발달에 관여한다. 착상을 전후하여 수태물에서도 IGF system이 발현되는데 개개 IGF family member의 발현 시기는 모체로부터 유래된 mRNA의 유무, 수태물 자체의 genetic programming, 모체와의 상호작용 등에 의해 결정되고, IGFs의 작용 부위 역시 시간(생리적 상태)과 장소의 특이성이 있다. 이와같이 conceptus IGF system의 발현이 시간적, 공간적으로 조절되고있다는 사실은 IGFs가 수태물의 성장 발달에 일역을 한다는 가설을 간접적으로 지지해 준다. 자궁조직과 수태물에서 발현된 IGFs는 세포의 증식과 분화, 포도당과 아미노산의 운반과 단백질합성, placental lactogen과 prolactin 등과 같은 유선자극호르몬의 생성을 자극하고 스테로이드 호르몬의 합성에도 관여한다. 태아의 성장 발달에 있어서 IGFs의 역할은 embryo의 IGF-I, IGF-II, 혹은 IGF receptor gene을 homologous recombination technique에 의해 파괴(gene targeting)하여을 때의 결과로써 입증되었다. 생쥐의 IGF and/or IGF-II gene 혹은 IGF receptor gene을 파괴했을 때 출생 전후 모두 성장 발달이 지연되며 출생시 무게는 정상치의 30-60% 수준에 머물고, 특히 type I IGF receptor gene 혹은 IGF-I과 IGF-II genes를 모두 파괴했을 경우에는 출생 후 곧 치사한다. 자궁 내의 IGFBPs는 IGF ligands를 자궁 내에 제한시키거나 IGFs의 receptor binding을 억제하는 negative regulators 역할을 하는 것으로 믿어지고 있다. 그러나 영장류의 자궁에서 IGFBP-I과 같은 특정 IGFBP는 IGFs보다 월등히 많은 양이 발현 분비되고 있는 것으로 추산되고 있으며, 또한 이 단백질은 모체탈락막세포에서 분비되는 주요 단백질 중의 하나라는 점을 감안할 때 IGFBP-I이 IGF과 무관한 작용이 있을 가능성도 배제할 수 없다. 따라서 IGFBPs의 역할 규명은 IGF system을 이해하는데 중요한 부분을 차지하고 있어 향후 이 분야의 연구에 많은 기대와 촛점이 모여지고 있다.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
