This study was conducted to evaluate the effect of tuberculin test and associated various environmental factors on the quantity and quality of milk production of daily cows. The change of milk quantity followed to tuberculin test was investigated on 109 daily farms in south Gyeonggi, and the change of milk quality was also studied on 48 daily farms. The result of this study showed that the quantity and quality of milk production was decreased after tuberculin test (P<0.05). The amount of loss of the milk production due to tuberculin test was, however, only 0.92 L per cow. In terms of milk quality, the level of total protein was statistically significantly decreased. This study also showed that the milk quality was further decreased when the cows were restraint after veterinarians arrived compared to the case that cows were stayed restraint before the veterinarians visited the farm. In addition, milk quality and quantity were lowered worse when relative humidity of the location of tuberculin test was lower than 50% or higher than 70%, or when the milk production of a farm is higher than its quarter. This study showed that stress on daily cows and the farmers induces the decrease of milk yield and milk quality, and these losses can be minimized by regulating various environmental factors to the direction to maximize productivity.
Beef producers are trying to produce not only better quality but also greater quantity of beef in order to meet the preferences of some consumers at a lower cost. This can be accomplished if we understand the factors regulating lipid deposition in intramuscular adipose tissue and the tenderness of meat. Propylene glycol (PG) might be used as a precursor of intramuscular fat synthesis especially in the late period of fattening because adipose tissue in ruminants is thought to mature sequentially in abdominal, intermuscular, subcutaneous and intramuscular depots. The action of cholecalciferol supplementation has been verified in producing more tender meat through the enhancement of calpain activity over the postmortem ageing period. A synergistic effect can be expected if the dietary cation and anion difference (DCAD) technique is used in combination with dietary supplementation of cholecalciferol. In another approach, the optimization of hormonal implant use also may provide similarly marbled beef at a much lower cost.
A model is developed for estimating optimal cultivation areas for apples to maximize the total profit of apple farming by region, focusing on Gyeongsangbuk-do, Chungchungbuk-do, Gyeongsangnam-do, and Jeonllabuk-do in Korea. Comparing the current cultivation areas to the optimal areas according to the model estimation during the period 1999 - 2019, the former has exceeded the latter since 2015 in all regions except for Jeonllabuk-do. This result stems from a lack of the regulation of production quantity among apple producers' regional organizations. Accordingly, the decreasing rate of the market price was greater than the increasing rate of the production quantity, and the total profit of apple farming has fallen in conjunction with increasing agricultural wage rates. Therefore, in order to reverse the current decreasing trend in the profits of apple farming, it is necessary to regulate nationwide apple cultivation areas through regional producers' associations. Ex-ante forecasting for 2019 posits the following regional optimal cultivation areas for maximizing the total income from apple farming. The Gyeongbuk apple producers' association needs to reduce its current cultivation area by 1,089 ha and to maintain 18,373 ha. In the Chungbuk region, current cultivated area should be reduced by 1,027 ha to maintain 2,722 ha, and in the Gyeongnam region, the current cultivated area should be reduced by 582 ha to maintain 2,730 ha. In contrast, the Jeonbuk region needs to increase its current cultivation area by 174 ha and to maintain at a level of 2,872 ha.

Objective: The number of bovine mammary epithelial cells (BMECs) is closely associated with the quantity of milk production in dairy cows; however, the optimal levels and the combined effects of hormones and essential amino acids (EAAs) on cell proliferation are not completely understood. Thus, the purpose of this study was to determine the optimal combination of individual hormones and EAAs for cell proliferation and related signaling pathways in BMECs. Methods: Immortalized BMECs (MAC-T) were treated with six hormones (insulin, cortisol, progesterone, estrone, 17β-estradiol, and epidermal growth factor) and ten EAAs (arginine, histidine, leucine, isoleucine, threonine, tryptophan, lysine, methionine, phenylalanine, and valine) for 24 h. Results: Cells were cultured in a medium containing 10% fetal bovine serum (FBS) as FBS supplemented at a concentration of 10% to 50% showed a comparable increase in cell proliferation rate. The optimized combination of four hormones (insulin, cortisol, progesterone, and 17β-estradiol) and 20% of a mixture of ten EAAs led to the highest cell proliferation rate, which led to a significant increase in cell cycle progression at the S and G2/M phases, in the protein levels of proliferating cell nuclear antigen and cyclin B1, cell nucleus staining, and in cell numbers. Conclusion: The optimal combination of hormones and EAAs increased BMEC proliferation by enhancing cell cycle progression in the S and G/2M phases. Our findings indicate that optimizing hormone and amino acid levels has the potential to enhance milk production, both in cell culture settings by promoting increased cell numbers, and in dairy cows by regulating feed intake.
Myogenic factor 5 (MYF5) plays an important role in regulating skeletal muscle fiber characteristics, consequently affecting meat production and quality. We identified a novel p.A41P mutation in exon1 of the porcine MYF5 gene by direct sequencing. The mutation was predicted to be destabilizing in protein structure based on the resultant amino acid substitution. We estimated the significant substitution effect of p.A41P on the energy stabilization of Myf5 protein structure. Then, we demonstrated that the mutation in Yorkshire population significantly affected muscle fiber type I composition (p<0.05), loin-eye area of lean meat content (p<0.05) and filter-fluid uptake of meat quality (p<0.01). Furthermore, dominant effects significantly influenced total muscle fiber number (p<0.05). This study suggests that the novel p.A41P mutation in porcine MYF5 may be a valuable genetic marker to affect the muscle fiber characteristics and consequently improve meat production quality and quantity.
This study attempted to clarify the difference in eggshell pigmentation between blue-shelled ducks (BSD) and whiteshelled ducks (WSD). The eggshell pigmentation deposition process is discussed. Ultraviolet spectro-photometer and HPLC were used to determine the biliverdin concentration in the shell gland, uterus liquid and eggshell at 6, 12, 18, 20, 23.5 h post-oviposition. The biliverdin concentration in the eggshell and uterus fluid showed significant differences between BSD and WSD, but not in the shell gland. The heme oxygenase activity in the shell gland of both kinds of ducks remained mostly constant during the ovulatory cycle with no variation. The assay of exogenous biliverdin injection into the shell gland antrum in the WSD indicated that exogenous biliverdin could be deposited continuously into the eggshell until the source was exhausted. A layer-by-layer dissolution assay was used to examine the eggshell pigment deposition process. The biliverdin concentration in the first to sixth layers of the eggshell in the BSD was significantly higher than that in the white-shelled counterpart. The blue pigment concentration increased persistently from the 6th layer to the $1^{st}$ layer. The BSD eggshells did not accumulate a large quantity of biliverdin in the most external layer. They tended to increase the deposition layer by layer. Our results demonstrated that different BSD and WSD eggshell colors were influenced by the amount of biliverdin in the uterus fluid and not determined by the amount of biliverdin in the shell gland. This implies the existence of a mechanism that controls biliverdin transportation from the shell gland into the uterus fluid, thereby playing a key role in regulating duck eggshell color.
Objective: Meat quality including muscle color in chickens is an important trait and continuous selective pressures for fast growth and high yield have negatively impacted this trait. This study was conducted to investigate genetic variations responsible for regulating muscle color. Methods: Whole genome re-sequencing analysis using Illumina HiSeq paired end read method was performed with pooled DNA samples isolated from two broiler chicken lines divergently selected for muscle color (high muscle color [HMC] and low muscle color [LMC]) along with their random bred control line (RAN). Sequencing read data was aligned to the chicken reference genome sequence for Red Jungle Fowl (Galgal4) using reference based genome alignment with NGen program of the Lasergene software package. The potential causal single nucleotide polymorphisms (SNPs) showing non-synonymous changes in coding DNA sequence regions were chosen in each line. Bioinformatic analyses to interpret functions of genes retaining SNPs were performed using the ingenuity pathways analysis (IPA). Results: Millions of SNPs were identified and totally 2,884 SNPs (1,307 for HMC and 1,577 for LMC) showing >75% SNP rates could induce non-synonymous mutations in amino acid sequences. Of those, SNPs showing over 10 read depths yielded 15 more reliable SNPs including 1 for HMC and 14 for LMC. The IPA analyses suggested that meat color in chickens appeared to be associated with chromosomal DNA stability, the functions of ubiquitylation (UBC) and quality and quantity of various subtypes of collagens. Conclusion: In this study, various potential genetic markers showing amino acid changes were identified in differential meat color lines, that can be used for further animal selection strategy.
The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.
This study aimed at targeting fixed increases in body weight (100 g/wk) by quantitatively regulating energy allowances (ME) in broiler breeders from 5 to 20 wks of age. Four energy regimes were tested: 1. The energy required for maintenance, activity and growth was calculated for 100 g increases in body weight/wk and a measured quantity of grower diet (160 g protein and 2,600 kcal ME/kg) was offered to the control group (ME-100) to achieve the anticipated weight gain. The energy allowances increased with age from 132 to 294 kcal/d. 2. Additionally, three energy regimes were considered, quantitatively reducing ME by 10% (ME-90) or 20% (ME-80) and increasing by10% (ME-110) over the control group. Each test group had 23 replicates5 female chicks housed in cages. The influence of energy regimes and age on growth, nutrient digestibility, carcass attributes, bone parameters and stress was evaluated at 4 wk intervals. Quantitative ME restriction by 10% (119-265 kcal/d) produced an average weight gain of 98.1 g/wk, which was closer to the targeted increase of 100 g/wk, whereas the control group attained it nine days earlier. Restriction of energy by 10 or 20% produced better conversion efficiency of feed, energy and protein and apparent digestibility of protein, Ca and P than 10% excess ME. Energy regimes did not influence eviscerated meat yield, but higher energy allowances (ME-110) significantly increased abdominal fat pad and liver weights and decreased giblet weight, percent muscle protein and tibia ash. Relatively higher stress was recorded in ME-restricted groups, as reflected by wider heterophil and lymphocyte ratios and increased bursa weight. Early age (5-12 wk) significantly influenced bone mineralization, conversion efficiency of feed, energy and protein and apparent digestibility of protein, Ca and P, while later ages (13-20 wk) increased eviscerated meat yield, abdominal fat, tibia weight and muscle protein and reduced stress. Energy regime x age interactions were significant and are discussed. In conclusion, the synthetic broiler line used in our study responded positively to controlled energy feeding during the rearing period. Breeders offered 119-265 kcal/d, a reduction of 10% energy over the control group, were more effective in regulating grower performance than the latter. In addition to energy regimes, age intervals also exhibited significant influence on specific parameters during the grower phase.
