Non-destructive Method for Selection of Soybean Lines Contained High Protein and Oil by Near Infrared Reflectance Spectroscopy

  • The applicability of non-destructive near infrared reflectance spectroscopic (NIRS) method was tested to determine the protein and oil contents of intact soybean [Glycine max (L.) Merr.] seeds. A total of 198 soybean calibration samples and 101 validation samples were used for NIRS equation development and validation, respectively. In the developed non-destructive NIRS equation for analysis of protein and oil contents, the most accurate equation was obtained at 2, 8, 6, 1(2nd derivative, 8 nm gap, 6 points smoothing, and 1 point second smoothing) and 2, 1, 20, 10 math treatment conditions with Standard Normal Variate and Detrend (SNVD) scatter correction method and entire spectrum (400-2500 nm) by using Modified Partial Least Squares (MPLS) regression, respectively. Validation of these non-destructive NIRS equations showed very low bias (protein: 0.060%, oil: -0.017%) and standard error of prediction (SEP, protein: 0.568 %, oil : 0.451 %) as well as high coefficient of determination ($R^2$, protein: 0.927, oil: 0.906). Therefore, these non-destructive NIRS equations can be applicable and reliable for determination of protein and oil content of intact soybean seeds, and non-destructive NIRS method could be used as a mass screening technique for selection of high protein and oil soybean in breeding programs.

Algorithm Development of a Visibility Monitoring Technique Using Digital Image Analysis

  • Atmospheric visibility is one of the indicators used to evaluate the status of air quality. Based on a conceptual definition of visibility as the maximum distance at which the outline of the selected target can be recognized, an image analysis technique is introduced here and an algorithm is developed for visibility monitoring. Although there are various measurement techniques, ranging from bulk and precise instruments to naked eye observation techniques, each has their own limitations. In this study, a series of image analysis techniques were introduced and examined for in-situ application. An imaging system was built up using a digital camera and was installed on the study sites in Incheon and Seoul separately. Visual range was also monitored by using a dual technology visibility sensor in Incheon and transmissometer in Seoul simultaneously. The Sobel mask filter was applied to detect the edge lines of objects by extracting the high frequency from the digital image. The root mean square (RMS) index of variation among the pixels in the image was substantially correlated with the visual ranges in Incheon and Seoul with correlations of $R^2$=0.88 and $R^2$=0.71, respectively. The regression line equations between the visual range and the RMS index in Incheon and Seoul were VR=$2.36e^{0.46{\times}(RMS)}$ and VR=$3.18e^{0.15{\times}(RMS)}$, respectively. It was also confirmed that the fine particles ($PM_{2.5}$) have more impacts to the impairment of visibility than coarse particles.

Prediction of Postpneumonectomy Pulmonary Function by Lung Scan in Lung Cancer Patient (폐관류스캔을 이용한 폐암환자의 일측 전폐절제술후의 폐기능예측)

  • If the postoperative pulmonary function can be predicted in the patients undergoing pneumonectomy for lung cancer preoperatively, it will be helpful for identifying them as high or low risk candidates. Perfusion lung scan and spirometry were performed in 12 patients with lung cancer pre-operatively and the predicted postoperative Vital Capacity, FVC, FEV1.0, FEF25 - 75% and MVV were estimated by multiplying the preoperative values by the percentage of perfusion of the nonsurgical lung. Three months postoperation the patients were reinvestigated with spirometry and the obtained values were compared with the predicted values. The linear regression lines derived from the correlation between predicted values [X] and observed values [Y] were as follows; VC; R=0.532, Y=0.48X+1.28, P=0.075 FVC; R=0.566, Y=O 54X+1.15, P=0.055 FEV1.0; R=0.832, Y=0.68X+0.70, P=0.001 FEF25 ~ 75%; R=0.781, Y=0.68X+0.54, P=0.003 MVV; R=0.718, Y=0.67X+34.75, P=0.009 The prediction of postoperative FEV1.0, FEF25 ~ 75% and MVV in lung cancer patients undergoing pneumonectomy appear to be valid for evaluating preoperative pulmonary function.

Detection and Molecular Identification of Human Enteric Viruses in Urban Rivers in Korea

  • We monitored the occurrence of human enteric viruses in urban rivers by cell culture-PCR and RT-nested PCR. Water samples were collected monthly or semimonthly between May 2002 and March 2003 in four urban tributaries. Enteric viruses were detected by RT-nested PCR and cell culture-PCR based on a combination of Buffalo Green monkey kidney (BGMK) and A549 cell lines, followed by phylogenetic analysis of amplicons. By RT-nested PCR analysis, 45 (77.6%), 32 (55.2%), 32 (55.2%), 26 (44.8%), 12 (20.7%), 2 (3.4%), 4 (6.9%), and 4 (6.9%) of 58 samples showed positive results with adenoviruses, enteroviruses, noroviruses (NV) genogroup I (GI) and II (GII), reoviruses, hepatitis A viruses, rotaviruses and sapoviruses, respectively. Adenoviruses were most often detected and only eight (13.8%) samples were negative for adenoviruses and positive for other enteric viruses in the studied sites. Thirty-one (77.5%) of the 40 samples were positive for infectious adenoviruses and/or enteroviruses based on cell culture-PCR, and the frequency of positive samples grown on A549 and BGMK (65.0%) was higher than that grown on BGMK alone (47.5%). The occurrence of each enteric virus, except reoviruses and hepatitis A viruses was not statistically correlated with the water temperature and levels of fecal coliforms according to Binary logistic regression model. By sequence analysis, most strains of adenoviruses and enteroviruses detected in this study are similar to the causative agent of viral diseases in Korea and most NV GI- and GII-grouped strains were closely related to the reference strains from China and Japan, and GII/4-related strains had similar sequences to strains recognized as a worldwide epidemic outbreak. Our results suggested that monitoring human enteric viruses is necessary to improve microbial quality and cell culture-PCR using the combination of A549 and BGMK cells and the adenovirus detection by PCR could be useful for monitoring viral contamination in the aquatic environment.

A Study on Split Grading Methods for Women's Pants and Increase Rate of Body Size of Adult Women (성인 여성의 연령대별 인체 부위 간 치수증감률을 반영한 바지 그레이딩에 관한 연구)

  • This study investigated the location of grading lines and grading amount defined by 17 women's wear brands for a pants item by target age groups (20-30, 30-40, and 40-50). This study utilized 6th Size Korea data and a script-based on a 3D scan measurement program to analyze the increase rate of body size in order to suggest a grading deviation distribution ratio for the pants using regression analysis. This study found that most brands appropriately divided grading amount at front thigh girth and back hip girth into the side and center by 1:1. Most brands divided the grading amount at the front hip girth into the side and center by 1:1; however, the ratio found from Size Korea is 0.8:1.2 for the 20-30 age group, 0.7:1.3 for the 30-40 age group, and 0.6:1.4 for the 40-50 age group. Regarding the back thigh girth, the brands targeting 20-30s, 30-40s, and 40-50s respectively assigned the grading amount into the side and center by 1:1, 1:1.2, and 1:1.3. However, the ratio found from Size Korea is 1.4:0.6 for the 20-30 age group, 1.7:0.3 for the 30-40 age group, and 1.3:0.7 for the 40-50 age group. The results can be utilized in improving the grading system of the pants item.

Sexual Maturity and Growth Characteristics of Octopus minor (낙지 (Octopus minor)의 성 성숙과 성장 특성)

  • Sexual maturity and growth characteristics of Octopus minor were investigated in 796 individuals from mud flats in Goorori, Muan-Goon, Korea. Gonadosomatic index (GSI) peaked between June and July in females and between November and January in males. The discrepancy in the index peaks between females and males might result from the earlier sexual maturation of males. The sex ratio was biased toward females (68%) in April and toward males (78%) in September, although the differences were not significant (p>0.05). In females, about 50% of individuals with a mantle length of 70.6 mm were sexually mature versus 100% of individuals with mantle lengths over 80 mm. In females, fecundity was also related to size, ranging from 44 eggs in individuals with a mantle length of 54 mm to 179 eggs in the female with a mantle length of 100.5 mm. The relationship between mantle length (ML) an body weight (BW) was BW=0.008 ML$^{2.2797}$ (n=389, r$^2$=0.83, p<0.01) in males and BW=0.029 ML$^{2.2797}$ (n=407, r$^2$=0.74, p<0.01) in females. Analysis of co-variation showed that the difference in the slopes of the two regression lines was significant (p<0.01). Analysis of mantle length-frequency for each month revealed 1 to 3 normal distribution modes. The growth parameters obtained by fitting the modal progression to the seasonalized von Bertalanffy growth function (VBGF) were ML$\infty$= 112.38 mm, K=1.9, C=0.90, and WP=0.1. The results indicate that the seasonal growth oscillation of the stock is very strong and winter is the season when growth is the slowest.

Antitumor Effects of Water Extracts of Panax notoginseng on NCI-H460 Tumor Regression Model

  • Objective: This study aimed to investigate the antitumor effects of water extracts of Panax notoginseng (WEPN) in NCI-H460 human lung cancer cell xenografted nude mice. Materials and Methods: We cultured NCI-H460 cell lines and xenografted them to nude mice. The mice were divided into 3 groups; positive control group, NCI-H460+150 mg/kg WEPN-treated group, and NCI-H460+300 mg/kg WEPN-treated group. They had been raised and treated in 28 days. We checked their body weight and tumor weight and volumes twice a week and their absolute organ weight and microhistological observation at the final day. We also calculated their tumor inhibition rate (I.R.), mean survival time and percent increase in life span (% ILS). Results: Body weight of WEPN (300 mg/kg) treated mice tended to slightly greater increase than those of the positive control group, but had no significance. Tumor volume (measurement with a caliper) of WEPN-treated mice tended to be lower than that of the positive control group. Inhibition rate (I.R.) of the WEPN group decreased more than the positive control group, but had no significance. Results of tumor weights and volume (plethysmography) had no significance. Mean survival time and percent increase in life span (% ILS) in the WEPN 300 mg/kg treatment group were higher than those of any other group (p<0.05). In absolute organ weights, the WEPN (150-300 mg/kg) treatment group decreased liver weights (p<0.05). Liver tissue of mice treated with WEPN (300 mg/kg) did not show any specific lesions. Conclusion: We suggest that WEPN may have potential as a growth inhibitor of solid tumors induced by NCI-H460 without any side effects. However, this study has limitations in proving anti-tumor effects of WEPN, so further studies to overcome those limitations will be needed.

High Performance Size Exclusion Chromatographic Analysis of Polymerization Products in Used Frying Oil (High Performance Size Exclusion Chromatography 를 이용한 튀김유의 중합체 분석)

  • A simple and rapid method, based on separation of a flying oil into monomeric, dimeric and oligomeric triglyceride by means of high performance size exclusion chromatography on Ultrastyragel column (500 A), is proposed for evaluation of the quality of used frying oil. The relative area of the monomeric triglyceride was decreased with frying time increase, however, the decrease rate was significantly reduced by treatment of a composited powder (porous rhyorite/citric acid 40/56/4). This measurement showed good linear relationship with change in polar component measurement. There were no significant differences between the slopes of regression lines in both treated and non-treated frying oil system for relationship between monomeric triglyceride and polar component. By this method, it was found that a frying oil habe to be discarded if the content monomeric triglyceride decreased to 71%, which was corresponed to 27% polar component.

The effect of movement of the neck on the body surface variation (목운동에 따른 목과 어깨부위의 체표변화에 관한 연구)

  • With the wide range of movement, the movement of neck accompanies the body surface change of neck and shoulders. And neck corresponds to the collar of garment, meaning that the body surface change of this part affects the construction of patterns for collars. So the purpose of this study was to manifest the changes of body surface in the neck and shoulder accompanied with the movement of the neck and to draw up the facts worth consideration when constructing the collar pattern. In this study, the draft of body surface of neck and shoulder was drawn up by gypsum method according to the 5 movements (standing at attention, neck bending - front, back, right, left). The length of body surface was measured and analyzed by ANOVA, post hoc test, correlation and regression analysis using SPSS 10.0 for Windows. The variation of the surface of neck was remarkable in the vertical lines than the horizontal ones. So the height of collar should be established considering the range of movement of the neck. It was the raising amount of c.f(center front) of neck and girth of neckbase (back) that were proved to have significantly varied after movement. With correlation analysis done, in every movement, the raising amount of side and the girth of neckbase had remarkably positive relation. The movement of the neck accompanied the variation of body surface in the shoulder as well. It was the part of scapula and side of neckbase that the variation was notable, suggesting that the surplus is needed in these parts.

A Study on the Correction of Spectral Line Overlaps for the Determination of Rare Earth Elements by X-Ray Fluorescence Spectrometry (X-선 형광분광법에 의한 희토류원소 분석에서 스펙트럼선 겹침 방해의 보정에 관한 연구)

  • The 8 rare earth elements were determined in the monazite concentrates by X-ray fluorescence spectrometry. The spectral line overlapping interferences from the other elements were investigated and the correction methods of interferences were studied using line overlap coefficients. The coefficients were calculated from the ratios of the intensities measured at the diffracted angle (2${\theta}$) of the analytical lines of other elements to the intensity of pure rare earth oxide. The coefficients were used to correct the line overlaps by a regression analysis. The linearities of calibration curves from the corrected intensities were remarkably improved, and their standard deviations were decreased. The analytical results agreed with that of the inductively coupled plasma(ICP) spectrometry within an allowable error range.
