Cultural and Rainfall Factors Involved in Disease Development of Fusarium Wilt of Sweet Potato

  • Environmental factors such as soil moisture, land management, and weather conditions affecting Fusarium wilt of sweet potato were investigated in major sweet potato cultivation regions in Korea. Fusarium wilt occurred mainly in reclaimed terracing lands, which are flattened and located in hilly to mountainous areas at the base of the mountain, in early seasonal cultivation regions. Disease severity was lower in reclaimed fields with natural slope. The development of Fusarium wilt in the fields was highly correlated with precipitation during planting period (r=-0.96**). Fusarium wilt was more severe in fields with less than 20 cm of available soil depth than in fields with over 20 cm of available soil depth. Greenhouse studies were consistent with field studies that less soil moisture content caused severe Fusarium wilt of sweet potato. These results indicate that low rainfall and moisture of soil with low effective soil depth during planting period are important environmental factors influencing the development of Fusarium wilt.

Estimation of Regional Future Agricultural Available Groundwater Supply in Jeju Island Using Water Balance Method (물수지 분석법을 이용한 제주도 권역별 미래 농업용 지하수 공급 가능량 추정)

  • To evaluate the available groundwater supply to the agricultural water demand in the future with the climate change scenarios for 40 sub-regions in Jeju Island, groundwater recharge and the available groundwater supply were estimated using water balance analysis method. Groundwater recharge was calculated by subtracting the actual evapotranspiration and direct runoff from the total amount of water resources and available groundwater supply was set at 43.6% from the ratio of the sustainable groundwater capacity to the groundwater recharge. According to the RCP 4.5 scenario, the available groundwater supply to the agricultural water demand is estimated to be insufficient in 2020 and 2025, especially in the western and eastern regions of the island. However, such a water shortage problem is alleviated in 2030. When applying the RCP 8.5 scenario, available groundwater supply can't meet the water demand over the entire decade.

A study on the vulnerability of field water supply using public groundwater wells as irrigation in drought-vulnerable areas with a focus on the Dangjin-si, Yesan-gun, Cheongyang-gun, and Goesan-gun regions in South Korea

  • The severe effects of climate change, such as global warming and the El Niño phenomenon, have become more prevalent. In recent years, natural disasters such as drought, heavy rain, and typhoons have taken place, resulting in noticeable damage. Korea is affected by droughts that cause damage to rice fields and crops. Societal interest in droughts is growing, and measures are urgently needed to address their impacts. As the demand for high-quality agricultural products expands, farmers have become more interested in water management, and the demand for field irrigation is increasing. Therefore, we investigated water demand in the irrigation of drought-vulnerable crops. Specifically, we determined the water requirements for crops including cabbage, red pepper, apple, and bean in four regions by calculating the consumptive water use (evapotranspiration), effective rainfall, and irrigation capacity. The total consumptive water use (crop evapotranspiration) estimates for Dangjin-si (cabbage), Yesan-gun (apple), Cheongyang-gun (pepper) in Chungnam, and Goesan-gun (bean) in Chungbuk were 33.5, 206.4, 86.1, and 204.5 mm, respectively. The volumes of groundwater available in the four regions were determined to be the following: Dangjin-si, 4,968,000 m3; Yesan-gun, 4,300,000 m3; Cheongyang-gun, 1,114,000 m3, and Goesan-gun, 3,794,000 m3. The annual amounts available for the representative crops, compared to the amount of evapotranspiration, were 313.9% in Dangjin-si, 29.5% in Yesan-gun, 56.1% in Cheongyang-gun, and 20.1% in Goesan-gun.

Estimation of Spatial Accumulation and transportation of Chl-$\alpha$ by the Numerical Modeling in Red Tide of Chinhae Bay (진해만 적조에 있어서 수치모델링에 의한 Chl-$\alpha$의 공간적 집적과 확산 평가)

  • The summer distribution of $Cha-{alpha}$ and physical processes for simulating outbreak region of red tide were estimated by the Eco-Hydrodynamic model in Chinhae Bay. As a result of simulation of surface residual currents, the southward flow come in contact with the northward flow at the inlet and western part of bay in case of windlessness and below wind velocity 2 m/sec. As wind velocity increases, the velocity and direction of currents were fairly shifted. The predicted concentration of $Cha-{alpha}$ exceeded 20 mg/㎥ in Masan and Haengam Bays, and most regions were over 10 mg/㎥, which meant the possibility of red tide outbreak. From the results of the contributed physical processes to $Cha-{alpha}$, accumulation sites were distributed at the northern part of Kadok channel, around the Chilcheon island, the western part of Kajo island and some area of Chindong Bay. On the other hand, inner parts of the study area such as Masan Bay were estimated as the sites of strong algal activities. Masan and Haengam Bay are considered as the initial outbreak region of red tide by the modeling and observed data, and then red tide expanded to other areas such as physical accumulation region and western inner bay, as depending on environmental variation. The increase of wind velocity led to decrease of $Cha-{alpha}$ and enlargement of accumulation region. The variation of intensity of radiation and sunshine duration caused to rapidly fluctuation of $Cha-{alpha}$: however, it was not largely affected by the variation of pollutant loads from the land only.

Establishment of Functional Cells for Vascular Defect Disease from Human Embryonic Stem Cell via Region Sorting Depending on Cell Volume (세포 크기 차이를 이용한 유세포 분석을 통한 인간배아줄기세포 유래 기능성 혈관세포의 확립)

  • Human embryonic stem cells have been highlighted as a valuable cellular source in the regenerative medicine field, due to their pluripotency. However, there is the challenge of the establishment of specific functional cell type forms of undifferentiated human embryonic stem cells (hESC). To establish and purify functional cell types from hESCs, we differentiated undifferentiated hESCs into vascular lineage cells and sorted the specific cell population from the whole cell population, depending on their cell volume, and compared them with the non-sorted cell population. We observed that about 10% of the PECAM positive population existed in the VEGF induced differentiating human embryoid body (hEB), and differentiated hEBs were made into single cells for cell transplantation. After making single cells, we performed cell sorting using a fluorescence-activated cell sorter (FACs), according to their cell volume on the basis of FSC region gating, and compared their therapeutic capacity with the non-sorted cell population through cell transplantation into hindlimb ischemic disease model mice. 4 Weeks after cell transplantation, the recovery rate of blood perfusion reached 54% and 17% in the FSC regions of sorted cells- and non-sorted cells, respectively. This result suggests that derivation of a functional cell population from hESCs can be performed through cell sorting on the basis of cell volume after preliminary differentiation induction. This approach may then greatly contribute to overcoming the limitations of marker sorting.

Isolation of Leuconostoc mesenteroides subsp. mesenteroides DU-0608 with Antibacterial Activity from Kimchi and Characterization of Its Bacteriocin

  • A bacteriocin-producing strain, DU-0608, was isolated from Kimchi and identified as Leuconostoc mesenteroides subsp. mesenteroides. The bacteriocin from isolate was inhibitory against Listeria monocytogenes, Micrococcus luteus and several strains of lactic acid bacteria. The bacteriocin was inactivated by pepsin, trypsin, $\alpha$-chymotrypsin, protease, $\alpha$-amylase and lipase, but not by catalase or by heating at $100^{\circ}C$ for 60 min. The molecular weight of the bacteriocin was estimated approximately 6 kDa. The inhibitory effect was bactericidal and rapid. Following treatment with isolate bacteriocin, cells of indicator strain (Lactobacillus sake JCM 1157) were damaged at the end regions of the cell wall, whereas the cells treated with nisin were damaged at many places around the cell wall.

  • To analyze subcellular localization of betanodavirus protein B2, a plasmid expressing Betanodavirus protein B2 fused to enhanced green fluorescent protein (EGFP-Nl) was constructed. The transient expression of full-length B2 fused to EGFP in GF cells confirmed the equal distribution of protein B2 between cytoplasm and nucleus. However, transfection of N-terminal half of the B2 revealed that this truncated form predominantly localized to the cytoplasm. By using several deletion mutants and point mutants, we determined the regions and/or motif responsible for the subcellular localization of betanodavirus.

Non-Invasive Environmental Detection using Heat Shock Gene-Green Fluorescent Protein Fusions

  • Three 'stress probe' plasmids were constructed and characterized which utilize a green fluorescent protein (CFP) as a non-invasive reporter to elucidate Escherichia coli cellular stress responses in quiescent or 'resting' cells. Facile detection of cellular stress levels was achieved by fusion of three heat shock stress protein promoter elements, those of the heat shock transcription factor ${\sigma}^{32}$, pretense subunit ClpB, and chaperone DnaK, to the reporter gene $gfp_{uv}$. When perturbed by chemical or physical stress (such as heat shock, nutrient (amino acid) limitation, addition of IPTG, acetic acid, ethanol, phenol, antifoam, and salt (osmotic shock), the E. coli cells produced GFPuv which was easily detected from within the cells as emitted green fluorescence. A temporal and amplitudinal mapping of these responses was performed, demonstrating regions where quantitative delineation of cell stress was afforded.

  • Single-walled carbon nanotubes (SWNT) exhibit strong Raman signals as well as fluorescence emissions in the near infrared regions where most biomolecules are transparent. Such signals do not blink or photobleach under prolonged excitation. which is advantageous to optical nano-bio marker applications. In this paper, single walled carbon nanotubes are conjugated with specific types of single-stranded DNA in order to detect oligonucleotides of corresponding complimentary sequences. Dot blotting experiments and comparative Raman spectroscopy observations demonstrated excellent sensitivity and specificity of carbon nanotube-DNA probes. The results show the possibility of using SWNT as generic nano-bio markers for the precise detection of specific kinds of genes.

  • With the recent development of technologies to manage vast amounts of data, data mining technology has had a major impact on all industries.. Data mining is the process of discovering useful correlations hidden in data, extracting executable information for the future, and using it for decision making. In other words, it is a core process of Knowledge Discovery in data base(KDD) that transforms input data and derives useful information. It extracts information that we did not know until now from a large data base. In the decision tree, c4.5 algorithm was used. In addition, the C4.5 algorithm was used in the decision tree to analyze the difference between frequency and mortality in the region. In this paper, the frequency and mortality of percutaneous coronary intervention for patients with heart disease were divided into regions.
