Effects of Mixture of Yukgunja-tang and Bambusae Caulis in Liquamen on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (육군자탕(六君子湯)과 죽력(竹瀝) 혼합물이 국소 뇌혈류량 및 평균혈압에 미치는 효과)

  • The study was designed to investigate the effects of Mixture of Yukgunja-tang and Bambusae Caulis in Liquamen (YTBCL) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats, and further to determine the mechanism of action of YTBCL. The results in rats were as follows ; YTBCL 25 ${\mu}l$ significantly decreased rCBF and MABP compared with basal condition. YTBCL 100 ${\mu}l$ significantly increased rCBF compared with basal condition, but decreased MABP compared with basal condition. YTBCL 50 ${\mu}l$ significantly increased rCBF compared with basal condition, but MABP was somewhat decreased compared with basal condition. The VTBCL 50 ${\mu}l$-induced increase in rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase and indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. The YTBCL 50 ${\mu}l$-induced decreased MABP significantly increased by pretreatment with methylene blue but was inhibited by indomethacin. This results were suggested that the mechanism of YTBCL was mediated Dy guanylate cyclase.

  • This experimental studies were done to investigate the effects of Sambutang(SBT) and consitituent herbs on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(BP) in rats. The results are as follows: SBT and consitituent herbs(SBT omitting Zingiberis Rhizoma Recens, Zingiberis Rhizoma Recens. Ginseng Radix and Aconiti iateralis preparata Radix) was increased rCBF significantly in a dose-dependent manner. SBT omitting Zingiberis Rhizoma Recens and Ginseng Radix were decreased significantly BP in rats. But, SBT, Aconiti iateralis preparata Radix and Zingiberis Rhizoma Recens were not affected BP in rats. According to the above findings. it is suggested that SBT causes a diverse response of rCBF, and specially the rCBF by increasing SBT is mediated action of Zingiberis Rhizoma Recens.

  • This study was designed to investigate the effects of Samul-tang Extract and Bambusae caulis in liquamen(SEB) on the changes in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. The results were as follows ; SBE significantly increased rCBF in a dose-dependent manner, and increased MABP in a dose-dependent manner. This result suggests that SBE significantly increased rCBF by dilating pial arterial diameter. Increase of SBE-induced rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase and increase of SBE-induced rCBF was inhibited by pretreatment with methylene blue ($10\;{\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase. Increase of SBE-induced MABP was significantly increased by pretreatment with methylene blue but increase of SBE-induced MABP was decreased by pretreatment with indomethacin. These results suggested that the action of SBE was mediated by cyclooxygenase.

  • The study was designed to investigate the effects of Leonuri herba extract (LHE) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and further to determine the mechanism of action of LHE. The results in normal rats were as follows, LHE significantly increased rCBF in a dose-dependent manner, and MABP did not change in a dose-dependent manner. This results were suggested that LHE significantly increased rCBF by dilating pial arterial diameter.   • The study was designed to investigate the effects of MokhwyangJoki-san Extract (MJSE) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and further to determine the mechanism of MJSE. The results in normal rats were as follows ; MJSE significantly increased rCBF in a dose-dependent manner, and MABP did not change in a dose-dependent manner. This results were suggested that MJSE significantly increased rCBF by dilating pial arterial diameter.   • Objective : Carthami Flos has been used as a herb to promote blood circulation to remove blood stasis in oriental medicine for many centuries. To investigate treatment of cerebral vascular disease(CVA) by promoting blood circulation and removing blood stasis(活血化瘀法), we observed the experimental Effects of Aqua-Acupuncture of Carthami Flos on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats. Method : When aqua-acupuncture of Carthami Flos(ACF) was injected into LI4, Liv3, B23, B62, GV16, experimental effects of ACF on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) is not known. Therefor, this study was designed to investigate the effects of ACF on the change of cerebral hemodynamics. The changes of rCBF and MABP were tested by Laser Doppler Flowmetry(LDF). Results : When ACF was injected into LI4, change of rCBF and MABP were decreased in a injected time-dependent manner. When ACF was injected into Liv3, change of rCBF and MABP were decreased in a injected time-dependent manner. When ACF was injected into B23, change of rCBF was significantly increased in a injected time-dependent manner, MABP did not change. When ACF was injected into B62, change of rCBF was increased in a injected time-dependent manner, change of MABP was significantly decreased after injection 60min. When ACF was injected into GV16, change of rCBF and MABP were similar to the change of normal. Conclusions : In conclusion, I suggested that ACF(LI4, Liv3) has an effect that depress the blood pressure & cerebral hemodynamic acceleration and ACF(B23, B62) has an anti-ischemic effect through the improvement of crebral hemodynamics.

Experimental Effects of Sibjeondaebo-tang and Gamy-Sibjeondaebo-tang on Cerebral Hemodynamics in Cerebral Ischemia Rats (십전대보탕(十全大補湯)과 가미십전대보탕(加味十全大補湯)이 뇌허혈 흰쥐의 뇌혈류역학에 미치는 실험적 영향)

  • This Study was designed to investigate the effects of Sibjeondaebo-tang (SDT) and Gamy-Sibjeondaebo-tang (GST, Sibjeondaebo-tang adding Cervi Pantotrichum Cornu) on the improvement in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. And, It was to investigate the effects of the SDT and GST with the change of histologic examination through the BDNF in the hippocampus CA1. In changes of cerebral hemodynamics, SDT and GST significantly increased rCBF in a dose-dependent manner but decreased MABP in normal rats. In mechanism of cerebral hemodynamics, Increase of GST-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and Decrease of GST-induced MABP was significantly increased by pretreatment with methylene. These results suggested that the action of GST was mediated by guantlate cyclase pathway. In cerebral ischemics, the rCBF was stably improved by SDT (10 mg/kg, i.p.) significantly and stably increased by GST (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrast with the findings of rapid and marked increase in Control group. These results suggested that GST had anti-ischemic action in cerebral ischemic state. In histological examination through TTC stain, Sample A group and Sample B group decreased discoloration in the cortical part at $28^{th}$ day after MCAO induction. In immunohistochemistric response of BDNF, Sample A group and Sample B group increased respondent effect at $28^{th}$ day after MCAO induction. These results suggest that GST can Increase rCBF in normal state, as well as improve the stability of rCBF in cerebral ischemic state. Furthermore, methylene blue inhibitor study suggested the mechanism of blood flow enhancement by GST may be mediated by guanylate cyclase pathway.

Effects of Acanthopanax Sessiliflorus $S_{EEM}$ Ethyl Alcohol Extract on the Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal & Ischemic Rats (오갈피 에탄올 추출물이 뇌혈류 및 혈압에 미치는 효과)

  • This study was designed to investigate the effects of Acanthopanax sessiliflorus 5.,M (ASS) on the regional cerebral blood flow (rCBF) and mean arterial blood pressure (BP) in normal rats and ischemic rats. Experimental groups of rCBF and BP in normal and ischemic rats as follows ; ALE was injected ASS leaves 45% ethyl alcohol extract, ASE was injected ASS stems 45% ethyl alcohol extract, ARE was injected ASS roots 45% ethyl alcohol extract. The results were as follows ; In normal rats, ALE and ARE significantly increased rCBF in a dose-dependent manner, but ASE significantly decreased rCBF and MABP in a dose-dependent manner, ALE increased MABP In ischemic rats, rCBF was significantly and staDly improved by ARE (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in control group. From the above results, it was thought that all part of Acanthopanax sessiliflorus $S_{EEM}$ was effective for hemodynamics and especially ARE was more effective than other parts.

  • Cheonghunhwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CTG was CHT adding Aurantii FructusㆍGastrodae Rhizoma. CTG was significantly increased regional cerebral blood f1ow(rCBF) in a dose-dependent, and CTG was decreased mean arterial blood pressure(MABP) compared with normal MABP(100%) in a low dosage, but was increased MABP in a dose-dependent, was aliked with normal MABP in a high-dosage. Therefore, purpose of this Study was to investigate experimental mechanism of CHTAG on the cerebral hemodynamics(rCBF, MABP) in rats. The changes of rCBF and MABP were determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; Pretreatment with indomethacin(3㎎/㎏, i.v.) was significantly inhibited CTG induced increase of rCBF and pretreated with propranolol(3㎎/㎏, i.v.) was inhibited CTG induced increase of rCBF, but pretreatment with methylene blue(10㎍/㎏, i.v) was increased CTG induced increase of rCBF. Pretreatment with indomethacin was decreased CTG induced increase of MABP, but pretreatment with propranolol and methylene blue were increased CTG induced increase of MABP. This results suggest that the mechanism of CTG is mediated by cyclooxygenase.

Effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the Regional Cerebral Blood Flow and Mean Blood Pressure in Rats (대나무 추출액과 죽력의 혼합물이 국소 뇌혈류량 및 평균 혈압에 미치는 영향)

  • This Study was designed to investigate the effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal and cerebral ischemic rats. Experimental materials were as follows ; BE- 1 was Bamboo Extract (BE) extracted with 70% ethyl alcohol, BE-11 was BE extracted with distilled water at $121^{\circ}C$ for 30 min, BE-111 was BE extracted with distilled water at $121^{\circ}C$ for 3 hrs, MLC was mixture of Bambusae Caulis in Liquamen (BCL) and BE-111 mixed at the ratio of 1 to 100 (MLC100), 1 to 50 (MLC50), 1 to 20 (MLC20), 1 to 10 (MLC10), 1 to 5 (MLC5). The results were as follows , The Changes of BE- 1 on the rCBF and MABP in normal rats were not showed, BE- 11 significantly decreased rCBF in a dose-dependent manner Dut increased MABP in a dose-dependent manner. BE-111 increased rCBF in a dose-dependent manner, MLC significantly increased rCBF in a dose-dependent manner and increased MABP in a dose-dependent manner. rCBF was significantly and stably increased by MLC5 (1 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. As results above ; The present author thought that BE- 111 and MLC increased rCBF by dilating pial arterial diameter.
