An experimental study was carried out in two laboratory-scale reactors to investigate the effect of heating rate on the behavior of flame front in a pulverized coal flame. Each. reactors had different heating mechanisms. For reactor A losing large heat through transparent quartz wall. pulverized coal particles were ignited by secondary air of 1050K. Flame front could be visualized through the transparent wall. Reactor B was insulated with castable refractory to minimize the heat loss through the reactor wall and accompanied with secondary air of 573K. Flame front was estimated from the gas temperature and species concentration measured using R-type thermocouple(Pt-Pt/Rh 13%) and gas chromatograph at various coal-air ratios and swirl intensities. The flame front position was closely related with the magnitude of heating rate. The heating rate for lifted flame was of the order of $10^4$ to $10^5K/s$ and for coal Ignition at least over $10^4K/s$. The heating mechanism had little impact on the extinction limits. The weak swirl number of 0.68 forced the flame front to move toward the upstream by the rapid mixing of coal and air. The primary/secondary momentum ratio was an inappropriate variable to distinct the liftoff of flame.

Cudrania tricuspidata Bureau is a widely-used, medicinal, perennial and woody plant. Obtaining information about the genetic diversity of plant populations is highly important with regard toconservation and germplasm utilization. Although C. tricuspidata is an important medicinal plant species registered in South Korea, no molecular markers are currently available to distinguish Korean-specific ecotypes from other ecotypes from different countries. In this study, we developed single nucleotide polymorphism (SNP) markers derived from the chloroplast and nuclear genomic sequences, which serve to to identify distinct Korean-specific ecotypes of C. tricuspidata via amplification refractory mutation system (ARMS)-PCR and high resolution melting (HRM) curve analyses. We performed molecular authentication of twelve C. tricuspidata ecotypes from different regions using DNA sequences in the maturaseK (MatK) chloroplast intergenic region and nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The SNP markers developed in this study are useful for rapidly identifying specific C. tricuspidata ecotypes from different regions.
A sensitive reverse transcriptase-PCR (RT-PCR) method to detect viable Escherichia coli O157:H7 in milk was established. The primer sets were designed based on the nucleotide sequences of the rfbE (per) and wbdN genes in the O157 antigen gene cluster of E. coli O157:H7. RT-PCR using five different primer sets yielded DNA with sizes of 655, 518, 450, and 149-bp, respectively. All five of the E. coli O157:H7 strains were detected by RT-PCR, but 11 other bacterial species were not. The sensitivity of RT-PCR was improved by adding yeast tRNA as a carrier to the crude RNA extract. The RT-PCR amplifying the 149-bp DNA fragment was the most sensitive for detecting E. coli O157:H7 and the most refractory to the bactericidal treatments. Heat treatment at $65^{\circ}C$ for 30 min was the least inhibitory of all bactericidal treatments. Treatment with RNase A strongly inhibited the RT-PCR of heated milk but not unheated milk. This study described RT-PCR methods that are specific and sensitive with a detection limit of 10 E. coli O157:H7 cells, and showed that pre-treating milk samples with RNase A improved the specificity to detect viable bacteria by RT-PCR.

We investigate the chemical differentiation in F, G, K type stars with and without planets to extend the work by Kang et al. (2011) to various spectral types. Since the primordial chemical composition has been preserved in the stellar atmosphere, stellar metallicity can provide the information on the primordial material, which is the potential building block of planets. Therefore, we can explore the favored conditions for planet formation through the comparison of chemical compositions between planet-host stars (PHSs) and stars without planets. In this work, we analyze 19 F, G, and K type stars. In each spectrum, we measure equivalent widths (EWs) of Fe, Na, Mg, Al, Si, Ca, Sc, Ti, V, Cr, Mn, Co, and Ni using TAME (Tools for Automatic Measurement of Equivalent width). The abundances of these species can be derived with the measured EWs and MOOG code (Sneden 1973). Like results by precedent studies, we find that planet-host stars have abundances higher than stars without planets. The typical difference in the abundances of Na, Mn, Co and Ni is $0.4{\pm}0.2dex$. In addition, as found in Kang et al. (2011), Mn is the most different element between PHSs and comparison stars.
Organophosphorus (OP) pesticides, particularly malathion, parathion, diazinon, and chlorpyrifos, are widely used in both agricultural and residential contexts. This refractory quality is shared by certain organ phosphorus insecticides, and it may have unintended consequences for certain non-target soil species. Bioremediation cleans organic and inorganic contaminants using microbes and plants. Organophosphate-hydrolyzing enzymes can transform pesticide residues into non-hazardous byproducts and are increasingly being considered viable solutions to the problem of decontamination. When coupled with system analysis, the multi-omics technique produces important data for functional validation and genetic manipulation, both of which may be used to boost the efficiency of bioremediation systems. RNA-guided nucleases and RNA-guided base editors include zinc-finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats (CRISPR), which are used to alter genes and edit genomes. The review sheds light on key knowledge gaps and suggests approaches to pesticide cleanup using a variety of microbe-assisted methods. Researches, ecologists, and decision-makers can all benefit from having a better understanding of the usefulness and application of systems biology and gene editing in bioremediation evaluations.
To determine the depth of preservative penetration in ACQ treated wood, the degree of penetration of Cu was measured. In this study, we developed a DDAC coloring method to investigate the penetration depth of DDAC, which is one of the active ingredient of ACQ, into wood. The following conclusions were obtained. The DDAC component reacts with a 2', 7'-dichlorofluorescein indicator and results in a deep orange color. This orange coloring reaction appears not only in DDAC solutions but also in ACQ treated wood tissues in which DDAC is present. It is possible to visually verify that DDAC has better wood penetration than Cu in the spruce, which is an refractory wood species, by the DDAC coloring method developed this study. In addition to the results, it is necessary to investigate the difference in penetration of Cu and DDAC for other wood species with poor preservative penetration.
This study was designed to investigate the effect of treatment variables on CCA treatment of Japanese red pine (Pinus densiflora), Japanese larch (Larix leptolepis), and ezo spruce (Picea jezoensis) round posts. Variables included were duration of initial vacuum (30, 45, or 60 minutes) and maximum pressure applied (8.5, 10.5, or 14.0 kg/$cm^2$), and maximum pressure was maintained until refusal point was reached. Regardless of wood species, extending the duration of initial vacuum more than 30 minutes did not affect treatability. Increasing pressure did not affect preservative penetration; however, preservative retention was affected by pressure levels, particularly at higher level (14.0 kg/$cm^2$). Preservative penetration depth of Japanese red pine met a minimum requirement specified by Notification of Korean Forestry Administration (No. 1999-8) for hazard class H5 of CCA-treated wood. Penetration of preservative in both Japanese larch and ezo spruce was not deep because of shallow sapwood thickness of these species, so pretreatment such as incising should be considered if these species are treated with preservatives. Although retention in Japanese red pine was not significantly increased even with 14.0 kg/$cm^2$, that of refractory Japanese larch and ezo spruce was significantly increased with the application of 14.0 kg/$cm^2$. Effect of treatment variables on refusal time was unclear; however, it is cleared that the refusal time was shortened with the increase in sapwood thickness.

The antiproliferative effect of the water extract of the branch and root bark of Ulmi Pumilae Cortex(WEUPC) was investigated on the p53-negative human leukemia cell line (HL-60). A dose- and time-dependent inhibition of cell growth was observed; this effect appears to be due to induction of apoptosis. Involvement of oxidative stress is indicated by a dose-dependent increase in intracellular reactive oxygen species levels. In addition. anti-apoptic effect was observed in the cells simultaneously treated with WEUPC and the anti-oxidant N-acetylcysteine. WEUPC did not affect the anti-apoptotic Bcl-2 and the pro-apoptotic Bax, whereas p21/sup WAF1/CIPl/ was enhanced in a dose- and time-dependent fashion; this effect was partially inhibited by N-acetylcysteine. The increase in p21/sup WAF1/CIPl/ was accompanied by a parallel accumulation of cells in the G1 phase of the cycle. These results suggest that the p53-independent induction of p21/sup WAF1/CIP/ and the induction of apoptosis may mediate the anti proliferative effect of WEUPC at least in this study; on the basis of this observation, WEUPC could be proposed as an useful adjunct to the treatment of p53-deficient tumors, which are often refractory to standard chemotherapy.
P. intermedia are considered an important pathogen in adult periodontitis, rapidly progressing periodontitis, refractory periodontitis, pregnancy gingivitis, acute necrotizing ulcerative gingivitis, pubertal gingivitis. So far 2 DNA homology groups and 3 serotypes of P. intermedia have been reported but there is no data available as yet regarding genetic diversity for the species P. intermedia. The purpose of this study is to investigate, using bacterial DNA restriction endonuclease analysis, genetic diversity between individual strains of P. intermedia which are indistinguishable by serotyping and biotyping, occurrence of an intrafamilial transmission and genetic heterogeneity between P. intermedia strains isolated within a patient and within the same serotypes. The families who have had no systemic disease, no experience of periodontal treatment for the previous 1 year and no experience of antibiotics for the previous 6 months were selected and subgingival plaque was collected at 4 sites in each person and incubated in the anaerobic chamber. P. intermedia were identified by colony shape, gram stain, biochemical test, SK-I03(Sunstar Inc.) test and IIF using monoclonal antibody was perfomed for the determination of serotypes. P. intermedia strains were grown in BHI broth and whole genomic DNA was extracted and digested by restriction endonuclease. The resulting DNA fragments were separated by agarose gel electrophoresis, stained and photographed under UV. As the results of this study, intrafamilial vertial transmissions could be assessed in 2 families and horizintal transmissions in another 2 families. There were different DNA digest patterns within a patient, so P. intermedia showed that individuals could be colonized by multiple clonal types at anyone time. And different serotypes could be found within a patient and in the same serotype within a patient, obvius genetic heterogeneity could not be assessed. But in the same serotype in different famies, there were differences in the DNA digest patterns.

To date many kinds of compounds have been obtained from plants kingdom as antineoplastic and anti-cancerous agents. However, there is no special type of compounds for ncancer therapy. Various types of substances are effective for various types of cancers and tumors: for instance, alkaloids, lignans, terpenes and steroids etc. Curcumol obtained from Curcuma aromatica was tested and noticed to be effective against cancer of the uterine cervix clinically Oridonin isolated from Rabdosia ssp.is now investigated for clinical trials in China. Moreover, camptothecine isolated from Camptotheca acuminata is also antineoplastic alkaloid, but is very toxic. Chemical modification has been tried to decrease its toxicity. This compound is now using as clinical agent. Harringtonin was investigated as an anticancerous drug in China. Taxol, a compound with a taxane ring isolated from the bark of Taxus brevifolia, has been demonstrated to have substantial anticancer activity in patients with solid tumors refractory standard chemotherapy. Supply of this drug has severely limited full exploration of its antineoplastic potential. Some efforts are continued in National Cancer Institute NCI) Washington for surveying various Taxus species for optimal taxol content, improvement in semi-synthesis from baccatin III, improvement in method of extraction, and development of alternative renewable resources. Further, there are many compounds which have been reported as antineoplastic agents. On the other hand, we have screened on higher plants collected in Japan, China, Korea, Southeast Asia and South America for antineoplastic activity, which has been done using Sarcoma 180 ascites in mice, P388 Iymphocytic leukemia in mice, Chinese hamster lung V-79 cells, P388 cells and nasopharynx carcinoma (KB) cells in our laboratory, as primary screening. In this meeting, I will present on antitumor and cytotoxic substances of the higher plants (Rubia cordifolia, Ailanfhus Vilmoriniana, Aster tataricus, Taxus cuspidata var. nana, Cephalotaxus harringtonia var drupacea, etc.) selected from above screening tests.
