New Fused Quinoxalines : Synthesis and Reactions of Pyrimidothienoquinoxaline and Oxadizolylthienoquinoxalines

  • Diazotization of 3-amino-2-ethoxycarbonylthieno[2,3-b]quinoxaline 1 gave the diazonium salt 2 which was reacted with SO2 and N-methylaniline to give sulfamoylquinoxaline derivatives 3-5. Imidazothienoquinoxaline 8 was obtained from the reaction of carboxylic acid hydrazide 6 with nitrous acid and followed by boilling the carboazide 7 in dry xylene. Also, compound 6 react with CH(OEt)3 to give aminopyrimidine 9 which was reacted with arylidene malonodinitrile, furfural and/or dimethoxy-tetrahydrofuran to afford compounds 10, 11 and/or 12 respectively. Refluxing of 6 with CS2 gave oxadiazolylthienoquinoxaline 13, reaction of 13 with hyrazine hydrate, CH(OEt)3, nitrous acid, CS2 and a-halocompounds to give 14-19.

Analysis of Phenolic Acids in Tobacco Leaf by GC/MS (GC/MS에 의한 잎담배중 Phenolic Acid의 분석)

  • A GC/MS method was developed to analyze phenolic acid extract from tobacco leaf. Extracted acids were converted to their methyl esters by refluxing with 3M hydrogen chloride in methanol, and the esters were reacted with his (trimethylsilyl) trifluoroacetamide plus 10% trimethylchlorosilane to silylate the phenolic groups. Derivatives of standard salicylic, p-hydroxybenzoic, vanillic, gentisic, p-coumaric, syringic, ferulic, and sinapic acids prepared by this procedure were analyzed by GC/MS on $20m{\times}0.2mm$ column of SE-54 glass capillary. GC/MS analysis of the extract from tobacco leaf revealed the presence of salicylic, p-hydtoxybenzoic, vanillic, gentisic, protocatechuic, p-coumaric, syringic, gallic, ferulic, caffeic, sinapic, and quinic acids, respectively. The quantitative analysis of these phenolic acids were achieved by using multiple ion selection technique.

Synthesis of Some New Condensed Pyrimidine Derivatives

  • Cyclodehydration of 6-amino-5-cyano pyrimidine derivative (2) afforded pyrimidoisoindole derivatives (3). Compound (3) reacted with carbethoxymethylene derivative to give pyridopyrimidine derivatives (5a,b). Compound (3) was also reacted with formamide to give the corresponding pyrimidopyrimdine derivatives (6) that condensed with benzaldehyde to give Schiff's base (7). Refluxing of compound (3) with triethyl orthoformate afforded compound (8) that cyclized with ammonium hydroxide giving the same compound (6). Compound (8) cyclized with hydrazine hydrate giving compound (9) which also cyclized with triethyl orthoformate affording compound (10). Diazotization of compound (3) led to the formation of triazinopyrimidine derivative (11). Cyclization of compound (11) upon treatment with hydrazine hydrate afforded compound (12). Compound (15) was prepared from reaction of compound (3) and ethylenediamine in presence of carbon disulfide. The behaviour of compound (15) toward benzoyl chloride, triethyl orthoformate, nitrous acid and/or carbon disulfide was also described. All proposed structures were supported by elemental analyses, spectroscopic data and some of the new products showed antimicrobial activity.

Synthesis and Antitumor Evaluation of 3-(2-Chloroethyl)-hydantoins from Some Amino Acids (아미노산으로부터 3-(2-Chloroethyl) hydantoin들의 합성과 그들의 항암작용 평가)

  • Six hydantoin derivatives, 3-(2-chloroethyl) hydantoin (6a), 3-(2-chloroethyl)-5-isopropylhydantoin (6b), 3-(2-chloroethyl)-5-isobutylhydantoin (6c), 3-(2-chloroetbyl)5-(2-butyl) hydantoin (6d), 3-(2-chloroethyl)-5-benzylhydantoin (6e), 3-(2-chloroethyl)-5-(indolyl-3-methyl) hydantoin (6f), were prepared by the treatment of the corresponding salt of amino acids with 2-chloroethyl isocyanate in cold water, followed by refluxing in concentrated HCl solution. Anticancer activity of the synthesized hydantoin derivatives were examined on murine leukemia L1210 cells growing in Fischer medium. Among them, 3-(2-chloroethy)-5-isobutyl-hydantoin (6c) showed substantially low $ED_{50}$ value of $9.6{\mu}g/ml$.

  • It was confirmed that korean scabby barley during the summer of 1963 was infected with Gibberella zeae. Feeding experiments showed that the diseased grain was toxic and induced emesis in pigs. The concentrated extracts which contained the emetic principle were prepared by refluxing the grain with methanol or water for 5 hours and then concentrating the filtrates and the washing in vacuum. Animal tests proved that methanol extraction for the emetic principle was more effective than extraction with water.

  • We synthesized new N-substituted 3-amino-6-chloropyridazine derivatives which were expected to retain biological activity. All synthetic process from pyridazine to 3-aminopyridazines could be carried out conveniently in high yield. N-Substituted 3-amino-6-chloropyridazine derivatives were prepared through amination and acylation from 3,6-dichloropyridazine. 3-Amino-6-chloropyridazine was prepared from the reaction of 3,6-dichloropyridazine with liquid ammonia under autoclave for 6 hrs. The refluxing of 3-amino-6-chloropyridazine and the corresponding acid anhydride for $1{\sim}2$ hrs afforded the N-substituted 3-amino-6-chloropyridazines. Alkyl chain of N-substituent was prolonged to six carbon (hexanoic acid).

  • For the development of new synthetic method for unnatural amino acid esters, N-aryl phenylglycine Ο-alkyl esters 4a∼i were synthesized through esterification, bromination, C-N bond formation from commercially available phenylacetic acids. An efficient and practical reaction condition for esters 2a∼c was that the starting materials 1a∼c were refluxed in absolute methanol for 3 hours with catalytic concentrated hydrosulfuric acid. In addition, bromines 3a∼c were formated for 3h in dichloromethane at rt with N-bromosuccinimide. Bromines 3a∼c were also converted to 4a∼i through substitution of arylamines during refluxing for 24 hours in ethanol with triethylamine. Interestingly, ethyl esters 5a∼c were formed via transesterification reaction when the p-sulfamylanilino group was used as a nucleophile in ethanol solvent.

Synthesis of Zinc Oxide Nano Rods, Sheet and Flower at $80^{\circ}C$ by the Sol-gel Method

  • Synthesis of zinc oxide nanorods, sheets and flower like structure were done by the sol-gel method using zinc acetate dihydrate and sodium hydroxide at $80^{\circ}C$ with 12 hours refluxing time nanorods, in case of as synthesized powder, with diameter of 20-60nm. Annealing at higher temperature (300 and $500^{\circ}C$,) in air ambient changes the morphology to sheet and flower like structure. The standard peak of zinc oxide was observed in IR at $523cm^{-1}$. The UV-VIS spectroscopy of zinc oxide shows a characteristic peak at 375nm.

  • 2,4,6-Triamino-5-nitrosopyrimidine was prepared using malononitrile and guanidine carbonate, and acetylated refluxing in acetic acid with acetic anhydride in order to activate the nitroso group for nucleophilic attack. Nucleophilic attack of phenylpyrimidium bromide on the nitroso group of 2,4,6-triacetamido-5-nitrosopyrimidine gave the intermediate, which lost pyrdidine to give the nitrone derivative. Addition of the methanethiol anion to nitrone gave 2,4-diacetamido-7-phenyl-6-methylthiopteridine which was hydrolyzed to give 2,4-diamino-7-phenyl-6-methylthiopteridine. Spectral data (IR, M.S, NMR) were provided to identify the reaction products during synthesis.

  • The synthesis of bisphenol A-derived calixarenes has been studied by changing the protecting group of the phe-nol moiety and reaction conditions. Depending on the protecting groups,the corresponding calix[6,7,8]arenes are obtained in different rat ios. For example, whcn mono-p-tert-butyldimethylsilyl-protected bisphenol A is treated with paraformaldehyde and a catalytic amount ofaqueous KOH in refluxing p-xylene with a Dean-Stark water collector for 36 h, the corresponding calix[8]arene, calix[7]arene, and calix[6]arene are producedand separated in the ratio of about 3 : 2 : l and with overall 63% yield. The calixarenes are characterized by NMR spectroscopy and mass analysis. The X-raycrystal structure of the calix[8]arene shows a pleated loop confor-mation, stabilized by intramoIecular hydrogen bonding between the inner phenolic hydroxy groups.
