Effect of Ethanol Concentration on Saponin Composition of Red Ginseng Extract (추출용매(抽出溶媒) 에탄올의 농도(濃度)가 홍삼(紅蔘)엑기스의 사포닌조성(組成)에 미치는 영향)

  • The effect of ethanol concentration on saponin composition of red ginseng extract was studied during extraction at $80^{\circ}C$ for 5 times of 8 hours. The increase in ethanol concentration from 0% to 90% resulted a gradual reduction in solids yield and an increase in the recovery of total ginsenosides. All of the ginsenosides determined were also significantly increased, but ginsenoside-$Rb_1.$-$Rb_2$ and -Rd were relatively decreased a little by raising the concentration 70% to 90%. The yield ratio of protopanaxadiol/protopanaxatriol saponin were in the range of 1.69${\sim}$1.95. No significant improvement in pure saponin yield was observed between 70% and 90% ethanol. Extraction with 70% ethanol was suggested for preparation of red ginseng extract from the result of this work.

  • Saponin is the most pharmaceutical active ingredients of the ginseng plant, it was called "Ginsenoside" which means the Glycoside of ginseng that composed glycosides and aglycones. The human body will absorb the saponin easily if these substrate was decomposed by active microorganism. Fermentation is the most convenient technique to decompose this active ingredients. The purpose of this research was to study the sugar content, pH and acidity development during the ginseng fermentation process. Fresh Korean ginseng and red ginseng extract was used as the main ingredient. The concentrated of pure ginseng extract was added to increase the saponin extract. Furthermore, the mix microbial powder was added as starter to increase the fermentation efficiency. The ginseng was fermented in fermentation chamber at temperature $37^{\circ}C$ during 70 days. In the end of experiment the sugar content was decreased from 24% to 7.65%, The pH was decreased from 6.5 to 3.4, and the acidity level was incresed from 0% to 1.2%.

Effect of Ginseng on the Relaxation of the Penile Corpus Cavernosal Smooth Muscle in Rabbits (인삼이 토끼 음경해면체 평활근의 이완작용에 미치는 영향)

  • The effect of ginseng on the reactivity of penile corpus cavernosal smooth muscle strips of rabbits was investigated to support the clinical application of ginseng for the treatment of erectile dysfunction. New Zealand white rabbits were randomly divided into two groups, normal diet group (n=9) and ginseng saponin diet group (n=5). Then, each group was fed normal diet and Korean red ginseng saponin diet (50 mg/kg/day) for 10 weeks. We measured concentration dependent relaxation of corporal smooth muscle to acetylcholine (10-8 M to 10-4M) in organ chamber. The degree of relaxation was expressed as percentage of maximal relaxation obtained by papaverine (10-4M). Dose dependent relaxation of corpus cavernosal smooth muscle to acetylcholine, at the concentration of 10-8 M to 10-4M by half log increment, was 4.06$\pm$0.00, 4.30$\pm$1.30, 5.32$\pm$0.68, 11.64$\pm$1.74, 16.24$\pm$1.61, 23.33$\pm$ 2.29, 26.45$\pm$2.25, 30.43$\pm$2.40 and 33.41 $\pm$2.48 (%), respectively in normal diet group and 9. 83$\pm$4.15, 20.60$\pm$4.62, 24.1815.12, 35.75$\pm$5.71, 43.35$\pm$6.11, 51.30$\pm$6.22, 56.33$\pm$6.22, 54.30$\pm$4.17 and 51.98$\pm$3.92 (Vc), respectively in ginseng group. These data suggest that ginseng enhances ondothelium-dependent acetylcholine-induced relaxation of penile corpus cal.ernosal smooth muscle in rabbits.

Stabilizing Effect of Ginseng Saponin and Water Extract on Malate Dehydrogenase from Pigeon Breast Muscle (인삼사포닌 및 인삼수용성 추출물이 비둘기 가슴근육으로부터 분리된 Malate Dehydrogenase에 미치는 안정화효과)

  • Studies were carried out to elucidate the protein stabilizing effect of ginseng. Malate dehydrogenase (EC 1.1.1.37) was used as a protein and the rate constant of the enzyme inactivation was determined under the heat denaturation condition. There was an optimum pH for the enzyme stability, the rate constant of the enzyme inactivation was minimum at BH 8.8. The rate constant was increased at lower and higher pH regions than the optimum pH. The inactivation reaction followed the Arrehnius law and the activation energy was measured as 36.8kcal/mole. The reaction rate was not affected by the enzyme concentration and thus it was assumed to be unimolecular first order reaction. The water extract of red ginseng decreased the rate constant of Malate dehydrogenate under heat inactivation condition to stabilize the enzyme activity. Purified ginseng saponin also stabilized the enzyme against heat inactivation.

Effects of Red-ginseng Extracts on the Activation of Dendritic Cells (고려홍삼의 수지상세포 활성화 효과)

  • Ginseng is a medicinal herb widely used in Asian countries. Dendritic cells(DCs) play a pivotal role in the initiation of T cell-mediated immune responses, making them an attractive cellular adjuvant for use in cancer vaccines. In this study, we examined the effects of Red-ginseng(water extract, edible and fermented ethyl alcohol extract, crude saponin) on the DCs phenotypic and functional maturation. Immature DCs were cultured in the presence of GM-CSF and IL-4, and the generated immature DCs were stimulated by water extract, edible and fermented ethyl alcohol extract, crude saponin and LPS, respectively, for 24hours. The expression of surface co-stimulatory molecules, including MHC(major histocompatibility complex) class II, CD40, CD80 and CD86, was increased on DCs that were stimulated with crude saponin, but antigen-uptake capacity was decreased. The antigen-presenting capacity of Red-ginseng extracts-treated DCs as analyzed by allogeneic T cells proliferation and IL-2, $IFN-{\gamma}$ production was increased. Furthermore, $CD4^+$ and $CD8^+$ syngeneic T cell(OVA-specific) proliferation and $IFN-{\gamma}$ production was significantly increased. However, $CD4^+$ syngeneic T cell secreted higher levels of IL-2 in responding but not $CD8^+$ syngeneic T cell. These results indicate the immunomodulatory properties of Red-ginseng extracts, which might be therapeutically useful in the control of cancers and immunodeficient diseases through the up-regulation of DCs maturation.

Red Ginseng Saponin Fraction A Isolated from Korean Red Ginseng by Ultrafiltration on the Porcine Coronary Artery

  • Red ginseng saponin fraction-A (RGSF-A) contains a high percentage of panaxadiol saponins that were isolated from Korean red ginseng by ultrafiltration. The aim of this study was to elucidate the effects of RGSF-A on the porcine distal left anterior descending (LAD) coronary artery. The relaxant responses to RGSF-A were examined during contractions induced by 100 nM U46619 (9,11-dideoxy-9a,11a-methanoepoxy-prostaglandin F2a), a stable analogue of thromboxane A2. RGSF-A dose-dependently induced biphasic (fast- and slow-) relaxation in the distal LAD coronary artery in the presence of an intact endothelium. The fast-relaxation was quickly achieved in a minute, and then the slow-relaxation was slowly developed and sustained for more than thirty minutes after the administration of RGSF-A. The slow-relaxation had a tendency to be bigger than the fast-relaxation. Fast relaxation induced by RGSF-A was almost blocked by $N_{\omega}$-Nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase synthase inhibitor and 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (ODQ), a guanylate cyclase inhibitor. However slow relaxation induced by RGSF-A was only partially inhibited by L-NAME and ODQ. In the endothelium-removed ring, RGSF-A evoked only slowrelaxation to a certain extent. These data suggest that RGSF-A induced both endothelium dependent fast- and slow-relaxation and endothelium independent slow-relaxation in the porcine distal LAD coronary artery. The endothelium dependent fast-relaxation is mediated by the nitric oxide (NO)-cGMP pathway, and the endothelium dependent slow-relaxation is at least partially mediated by the NO-cGMP pathway. However, the endothelium-independent slow-relaxation remains to be elucidated.

Study on Stability of Red and White Ginseng by Storage Years (홍삼(紅蔘)과 백삼(白蔘)의 저장기간(貯藏期間)에 따른 안정성(安定性) 비교(比較) 연구(硏究))

  • Ginseng is one of the most important plant drugs which has been used as a medicine in Asia. Main effective components of ginseng have been recently believed to be saponins which have been isolated and identified from Panax ginseng C. A. Meyer. Decrease in total saponin content of white ginseng by storage years was remarkable in contrast with red ginseng. In storage for three years, decrease in red and white ginsengs were 12% and 27% for total saponin content, and 16.8% and 42.2% for major saponin content, respectively. The PT/PD ratios of red and white ginseng saponins were ranged 0.646 to 0.694% and 0.414 to 0.428%, respectively. According to the result of PT/PD ratios, red ginseng was greater than 50% in compare with white ginseng. The change of PT/PD ratio would greatly influence on the biochemical and pharmacological effects of ginseng.

  • A genuine dammarane-glycoside, named as ginsenoside $ Rs_3$, was isolated from the MeOH extracts of Korean red ginseng (Panax ginseng C.A. Meyer) through repeated silica gel column chromatographies and its chemical structure was determined as (20S)-protopanaxadiol $3-O-[6^{11}-O-acetyl-{\beta}-D-glucopyranosyl (1{\rightarrow2)-{\beta}-D-$glucopyranoside on the basis of several spectral and physical evidences including HMBC and FAB-MS.

  • Acidic polysaccharide from Korean red ginseng was found to inhibit pancreatic lipase activity and cause reduction of plasma triglyceride level after oral administration of corn oil emulsion to rats. Thus acidic polysaccharide may reduce plasma triglyceride through its inhibitory action on pancreatic lipase and successive inhibition of intestinal absorption of fat due to reduction of lipolysis. In the course of this experiment, we found an unknown ninhydrin positive substance in Korean red ginseng. The unknown substance was identified to be arginyl-fructosyl glucose(Arg - Fru - Glc). Coment of this new compound was $5.37\%$ in Korean red ginseng powder. Sucrase and maltase activities in mucous layer of rat jejunum were found to be inhibited by Arg-Fru-Glc. Physiological significance of the new compound was discussed based on these experimental results.

Screening of Antioxidative Components from Red Ginseng Saponin (홍삼 사포닌의 항산화활성 성분 Screening)

  • Aerobic cells are normally protected from the damage of free radicals by antioxidative on , zymes such as superoxide dismutase (SOD), catalase, glutathione (GSH) peroxidase, GSH S- transferase and GSH reductase which scavenge free radicals as well as nonenzymatic antioxidants such as ceruloplasmin, albumin and nonprotein-SH including GSH. The effects of each component (ginsenoside $Rb_1$, $Rb_2$, Rc, Rd, Re, $Rb_1$, Rf, $Rh_1$ and $Rh_2$) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. Ginsenoside $Rb_1$ and Rc showed a tendency to increase GSH peroxidase activity, while ginsenoside Rc significantly decreased Cu,Zn-SOD activity. Especially, ginsenoside $Rh_2$ significantly increased catalase activity. These results suggest that ginsenoside $Rh_2$ is an important active component among total saponins of red ginseng.
