Agarase의 분류, 기원, 확보, 활성파악, 분리정제, 생산 및 응용 (The Classification, Origin, Collection, Determination of Activity, Purification, Production, and Application of Agarases)

  • 한천은 거대 홍조류의 세포벽 성분으로 agarase에 의해 가수분해된다. Agarase는 분해 양상에 따라 ${\alpha}$-agarase (E.C. 3.2.1.158)와 ${\beta}$-agarase (E.C. 3.2.1.81)로 나눌 수 있으며 아미노산 서열 유사성에 따라 GH (glycoside hydrolase) -16, -58, -86, -96, -118 로 나눌 수 있다. 많은 agarase들이나 그들의 유전자가 바다나 육지 환경의 세균 혹은 메타게놈에서 검출, 분리, 재조합 발현되었다. Agarase의 산물인 한천올리고당(agarooligosaccharides)과 네오한 천올리고당(neoagarooligosaccharides)은 항암, 면역활성, 항산화, 장내유용세균활성화, 간보호, 항균, 미백, 보습의 효과 등 다양한 기능성이 보고되어 식품, 화장품, 의학 영역에서 광범위하게 이용될 수 있다. 또한 agarase는 연구영역에서 이용될 수 있다. 이 논문은 agarase의 검출원, 정제법, 검출법 그리고 응용영역에 대해 검토하였고, 한천대사에서 agarase의 역할, 대사산물의 기능 등도 고찰하였다.

Dietary Transformation of Lipid in the Rumen Microbial Ecosystem

  • Dietary lipids are rapidly hydrolysed and biohydrogenated in the rumen resulting in meat and milk characterised by a high content of saturated fatty acids and low polyunsaturated fatty acids (PUFA), which contributes to increases in the risk of diseases including cardiovascular disease and cancer. There has been considerable interest in altering the fatty acid composition of ruminant products with the overall aim of improving the long-term health of consumers. Metabolism of dietary lipids in the rumen (lipolysis and biohydrogenation) is a major critical control point in determining the fatty acid composition of ruminant lipids. Our understanding of the pathways involved and metabolically important intermediates has advanced considerably in recent years. Advances in molecular microbial technology based on 16S rRNA genes have helped to further advance our knowledge of the key organisms responsible for ruminal lipid transformation. Attention has focused on ruminal biohydrogenation of lipids in forages, plant oils and oilseeds, fish oil, marine algae and fat supplements as important dietary strategies which impact on fatty acid composition of ruminant lipids. Forages, such as grass and legumes, are rich in omega-3 PUFA and are a useful natural strategy in improving nutritional value of ruminant products. Specifically this review targets two key areas in relation to forages: i) what is the fate of the lipid-rich plant chloroplast in the rumen and ii) the role of the enzyme polyphenol oxidase in red clover as a natural plant-based protection mechanism of dietary lipids in the rumen. The review also addresses major pathways and micro-organisms involved in lipolysis and biohydrogenation.

미세조류 Dunaliella bardawil의 고농도 세포배양 (High Cell Density Culture of Micro-algal Dunaliella bardawil)

  • 본 연구에서는 ($\beta$-carotene 생산균주인 미세조류 Dunaliella bardawil을 사용하여 batch flask에서 미세조류의 고농도세포에 관한 최적배양조건(미량원소, pH, agitation speed, nitrate, phosphate, carbon source)을 확립하고자 하였다. 미량원소는 5X 배지에서 교반하였을 때 비생장속도는 $0.0l3hr^{-l}$와 세포농도는 $4.9{\times}10^6$ cells/mL로서 IX. 3X, lOX 배지에서 배양한 것보다. 약 46%, 18%, 69% 높은 세포수율을 얻었으며 세포배양시 교반한 경우, pH는 80에서 최대 세포농도를 얻었다. 초기 nitrate ($KNO_3$)와 phosphate($KH_2PO_4$)의 영향을 조사한 결과 미세조류 생장에 중요한 영양분으로서 질소원의 주입은 매우 효과적임을 확인하였다. 또힌 탄소원으로서 250mM의 $NaHCO_3$$CO_2$ 가스를 동시에 사용한 배양조건이 500mM $NaHCO_3$만을 탄소원으로 사용한 실험에 비하여 32% 증가된 세포농도를 나타내었다. light는 white light의 경우 blue light보다 세포생장에 적합하였다. 질소원을 이용한 유기배양시 2회의 nitrate주입만으로써 배양 198hr에 $8.955{\times}10^6$cell/mL의 고농도의 세포를 얻었다.

Anti-hyperlipidemic Effect of Polyphenol Extract (SeapolynolTM) and Dieckol Isolated from Ecklonia cava in in vivo and in vitro Models

  • The inhibitory effect of polyphenol extracts (Seapolynol$^{TM}$, SPN) of the marine brown algae Ecklonia cava and dieckol, a major component of SPN, on hyperlipidemia was investigated in ICR mice fed a high-fat diet (HFD) for five weeks. For analysis of the anti-hyperlipidemic effects of SPN and dieckol, these two agents were given orally on a daily basis to HFD-fed mice for four weeks, starting one week after the beginning of HFD feeding. Groups administered with SPN as well as dieckol showed lower body weight gains than the HFD only group. Administration of SPN and dieckol also resulted in a significant reduction of the level of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein (LDL) cholesterol in the serum of HFD-fed mice. In Oil Red O staining using 3T3-L1 preadipocytes, it was shown that both SPN and dieckol markedly inhibited lipid accumulation of 3T3-L1 cells. Furthermore, SPN and dieckol (50 ${\mu}g$/mL) significantly inhibited 3-hydroxyl-methyl glutaryl coenzyme A (HMGCoA) reductase activity in vitro. Taken together, these results suggest that polyphenols of Ecklonia cava (SPN) and dieckol reduce body weight gain and fat accumulation in HFD-induced obese mice, and that their hypolipidemic effect is related to the inhibition of adipogenesis of adipocytes and HMGCoA reductase activity.

Characterization of Amylolytic Activity by a Marine-Derived Yeast Sporidiobolus pararoseus PH-Gra1

  • Marine yeasts have tremendous potential in industrial applications but have received less attention than terrestrial yeasts and marine filamentous fungi. In this study, we have screened marine yeasts for amylolytic activity and identified an amylase-producing strain PH-Gra1 isolated from sea algae. PH-Gra1 formed as a coral-red colony on yeast-peptone-dextrose (YPD) agar; the maximum radial growth was observed at 22 ℃, pH 6.5 without addition of NaCl to the media. Based on the morphology and phylogenetic analyses derived from sequences of internal transcribed spacer (ITS) and a D1/D2 domain of large subunit of ribosomal DNA, PH-Gra1 was designated Sporidiobolus pararoseus. S. pararoseus is frequently isolated from marine environments and known to produce lipids, carotenoids, and several enzymes. However, its amylolytic activity, particularly the optimum conditions for enzyme activity and stability, has not been previously characterized in detail. The extracellular crude enzyme of PH-Gra1 displayed its maximum amylolytic activity at 55 ℃, pH 6.5, and 0%-3.0% (w/v) NaCl under the tested conditions, and the activity increased with time over the 180-min incubation period. In addition, the crude enzyme hydrolyzed potato starch more actively than corn and wheat starch, and was stable at temperatures ranging from 15 ℃ to 45 ℃ for 2 h. This report provides a basis for additional studies of marine yeasts that will facilitate industrial applications.

한국 남해안 한려해상국립공원의 계절별 해조상 및 군집구조 변화 (Seasonal Variations in the Macroalgal Flora and Community Structure in Hallyeohaesang National Park on the South Coast of Korea)

  • Seasonal variability in the marine seaweed community structure was examined in the intertidal zones at nine study sites in Hallyeohaesang National Park, on the southern coast of Korea from March to October 2014. A total of 145 seaweeds were indentified, comprising 15 green, 41 brown and 89 red algae. Coarsely branched seaweeds were the dominant functional group, comprising 58.95% in species number, whereas filamentous, sheet, thick leathery, crustose and jointed calcareous forms comprised 2.63-17.72% each. The seaweed biomass averaged 358.00 g dry wt/m2 and it was maximal at Somaemuldo (847.64 g dry wt/m2) and minimal at Gamam (56.51 g dry wt/m2). Based on biomass, the dominant and subdominant seaweeds were Ulva australis at Gamam, Sargassum thunbergii at Sangju, Ulva australis and S.fulvellum at Neukdo, S.horneri at Dala-Bijindo-Somaemuldo, S. thunbergii at Dapo, and Corallina pilulifera at Songdo. Community indices were as follows: dominance index (DI), 0.43-0.71; richness index (R), 8.26-16.50; evenness index (J'), 0.36-0.54; and diversity index (H'), 1.57-2.19. In conclusion, we found that both biomass and the community structure of seaweeds in Hallyeohaesang National Park were similar to those in other studies of the Southern Sea along the Korean peninsula, and that Hallyeohaesang National Park is a relatively favorable habitat for seaweeds. Future studies should examine the changes in seaweed composition and biomass as they relate to climate change and environmental pollution.

김(Pyropia spp.) 가공식품에 포함된 녹조 파래류(Ulva spp.) 동정을 위한 분자마커 개발 및 적용 (Development and Application of Molecular Markers for Identifying Ulva species in Commercial Pyropia Seafoods)

  • Pyropia, economic red algae species, have been cultivated in Korea (referred to as 'gim'), Japan ('nori'), and China ('zicai') for over 300 years. Vegetable seaweed Pyropia species are sold in the public markets in various forms as commercial seafoods. In Korea, two kinds of Pyropia seafood made with species of Pyropia and Ulva (sea lettuce, referred to as 'parae') are also sold. These are referred to as 'parae-gim' (with Pyropia spp. and U. linza) and 'gamtaegim' (with Pyropia spp. and U. prolifera). There is currently no method for identifying the seaweed species that comprise Pyropia seafood products. Therefore, we developed novel molecular markers to identify Ulva species in commercial Pyropia seafoods. Based on rbcL molecular markers, we identified informative characteristics to discriminate U. linza and U. prolifera as seafood ingredients. Moreover, PCR with 3'-end mismatch primers successfully isolated the specific rbcL sequences of U. linza and U. prolifera from Pyropia seafoods. Therefore, our novel molecular markers will be useful for identifying the ingredient species of commercial seafoods.

변산반도 해조류 군집구조의 계절적 변동 (Seasonal Variation in Macroalgal Community Structure around the Byeonsan Peninsula, Korea)

  • We examined seasonal variation in the structure of marine macroalgal communities at five sites around the Byeonsan Peninsula, Korea, from January to October 2011. Sixty eight species were identified, including 11 green, 17 brown and 40 red algae. Species richness was highest at Gyeokpo (51 species) followed by Sambal (47), Gosapo (34), Byeonsan (33), and Habgoo (18). Seaweed biomass ranged from 12.98 to 145.33 g dry $wt/m^2$, with maximum and minimum biomasses at Gyeokpo and Habgoo, respectively. Sargassum thunbergii was the representative alga species occupying 47.89% of the total biomass. The dominant seaweeds were S. thunbergii, Corallina pilulifera, Ulva spp, Gracilaria vermiculophylla, and Carpopeltis affinis. No distinctive vertical distribution of seaweeds was found because S. thunbergii, Ulva spp, and G. vermiculophylla were all distributed from high to low shore. Community indices were as follows: dominance index (DI), 0.44-0.61; richness index (R), 6.27-10.35; evenness index (J'), 0.38-0.59; and diversity index (H'), 1.48-1.71. Gyeokpo had the most biomass and highest species richness, and the lowest percentage of Ulva species. However, species richness declined at Gyeokpo, from 78 to 51 species within 10 years, implying that eutrophication and sedimentation from Mankyeong and Dongjin rivers have reduced the stability of seaweed communities.

한국 남서해안 흑산도와 홍도의 조간대 해조류 생물량과 군집구조의 계절적 변동 (Seasonal Variation in Biomass and Community Structure of Intertidal Seaweeds at Heuksando and Hongdo, Southwestern Coast of Korea)

  • Seasonal variation in marine macroalgal community structure was examined at the intertidal zones of Heuksando and Hongdo, Shinan, Korea, from July 2008 to May 2009. In total, 86 macroalgal species were identified, including 12 green, 19 brown, and 55 red algae; 67 species at Heuksando and 70 species at Hongdo, were observed. Annual seaweed biomass was 252.44 g dry $wt/m^2$ at Heuksando and 217.67 g dry $wt/m^2$ at Hongdo. The dominant seaweed in importance value (IV > 15) differed between the sites: Sargassum thunbergii at Heuksando and Corallina pilulifera at Hongdo. The vertical distribution pattern of seaweeds from the upper to lower intertidal zones at Heuksando was Gloiopeltis spp., Ulva spp. - S. thunbergii, S. fusiforme, Hildenbrandia rubra - S. thunbergii, C. pilulifera. On the rocky shore of Hongdo, seaweed zonation was distinct: Porphyra yezoensis, Gloiopeltis spp., Ulva spp. - C. pilulifera, S. fusiforme, Myelophycus simplex - Chondrus ocellatus, C. pilulifera, and Carpopeltis affinis. Annual seaweed coverage, richness index (R), evenness index (J'), and diversity index (H') values were greater at Hongdo (41.35%, 12.82, 0.59, and 2.50 respectively) than at Heuksando (31.54%, 11.93, 0.44, and 1.87 respectively), which may indicate that the seaweed community at Hongdo is more stable relative to the one at Heuksando.

한국 남서해안 관매도와 영산도의 해조상 및 군집구조 (Marine Algal Flora and Community Structure at Gwanmaedo and Yeongsando, Korea)

  • Seasonal variations in seaweed communities were examined at Gwanmaedo and Yeongsando, Korea, from May 2014 to February 2015. Eighty-nine species were identified, including 11 green, 20 brown and 58 red algae. Seventy-three and 74 species were identified at Gwanmaedo and at Yeongsando, respectively. Sargassum thunbergii and Myelophycus simplex were the dominant species, comprising 60.89 and 39.50% of total biomass, respectively, and S. fusiforme was subdominant at the two sites. Of six functional seaweed forms, the coarsely-branched form was the most dominant, forming about 43% of the species number at Gwanmaedo and Yeongsando. Seasonal seaweed biomasses ranged between 53.10 and 172.85 g/m2 (average 93.57 g dry wt./m2) and between 83.11 and 176.20 g (138.21 g/m2) at Gwanmaedo and Yeongsando, respectively. The vertical distribution from the high to low intertidal zone was S. thunbergii and Gloiopeltis furcata; M. simplex and S. thunbergii; and S. fusiforme at Gwanmaedo. Seaweed zonation was distinct at Yeongsando, with S. thunbergii and Gelidiophycus freshwateri; M. simplex and S. thunbergii; and S. thunbergii and S. fusiforme. Seaweed biomass, evenness index (J'), and diversity index (H') values were greater at Yeongsando (138.21 g/m2, 0.51, 2.18 respectively) than at Gwanmaedo (93.57 g/m2, 0.48, 2.04), indicating that the seaweed community at Yeongsando is more stable than that at Gwanmaedo.
