Streptococcus pneumoniae가 생산하는 pneumolysin의 EGFP 융합으로 인한 용혈활성 변화 (C-terminal Fusion of EGFP to Pneumolysin from Streptococcus pneumoniae modified its Hemolytic Activity)

  • Streptococcus pneumoniae는 pneumolysin과 같은 병원성 인자를 가지고 있으며, 지역사회에서 전파되는 심각한 병원성균에 포함된다. Pneumolysin (PLY)은 콜레스테롤 의존적으로 세포막에 구멍을 형성하는 세포독소로서 백신의 주요한 표적 항원이다. PLY의 연구를 위하여 Streptococcus pneumoniae D39 균주에서 추출한 genomic DNA를 주형으로 하여 PCR을 시행하였다. 합성된 PLY 유전자 DNA를 pQE-30 vector에 삽입하고, E. coli M15에 형질전환 시킨 후 LB 배지에 IPTG를 첨가하여 PLY 단백질을 생산하였다. 재조합 단백질은 $Ni^{2+}$-agarose column을 사용하여 정제하였다. 또한, EGFP를 PLY C-말단에 부착한 융합단백질도 동일한 방법으로 클로닝하여 재조합 단백질을 생산하였다. 500 ng/ml 농도의 재조합 PLY는 1.0% 적혈구 현탁액을 100% 용혈시켰으며, 240 ng/ml 농도는 50% 용혈을 나타내었다. 그러나 재조합 PLY-EGFP는 용혈 활성이 전혀 나타나지 않았으나 형광현미경으로 관찰하였을 때 적혈구 막에 결합되어 있었다. 즉, EGFP의 PLY C-말단 부착은 PLY의 세포막 결합능은 유지시켰으나 용혈기능은 방해하였다. PLY C-말단은 용혈기능에 아주 중요한 영역이며, 세포막 결합은 PLY의 다른 영역이 보다 중요하게 작용할 것으로 추측된다. 따라서, 육안으로 관찰이 가능한 결합능은 가졌으나 용혈 기능이 결여된 PLY-EGFP를 대조군으로 활용함으로써 두 재조합 단백질은 폐렴 유발에 있어서 PLY 작용 연구에 사용될 수 있을 것으로 기대된다.

Methylotrophic Yeast, Pichia pastoris에서 사람 락토페린의 발현 및 항균성 연구 (Expression and Antibacterial Activity of Recombinant Human Lactoferrin in Methylotrophic Yeast, Pichia pastoris)

  • 사람의 모유에 많이 함유된 human lactoferrin(hLF)은 항균 및 항 바이러스 작용이 있는 것으로 보고되고 있다. 본 연구에서는 hLf를 메탄올자화 효모인 Pichia pastoris에 cloning하고 그 발현을 RT-PCR, Northern blotting, SDS-PAGE및 Western blotting으로 확인하였다. 그 결과 2.1 kb의 hLf 유전자가 P.pastoris의 염색체 DNA로 끼어들어가 안정적으로 hLf를 발현하였다. 이 재조합 P.pastotis로부터 hLf를 포함하는 세포 추출액을 얻어 항균 작용을 연구하였다. 발현된 재조합 hLf는 Staphylococcus aureus, Micrococcus flavus 등의 그람 양성균에 대해 강력한 항균작용을 보일 뿐만 아니라 그람 음성 동물성 병원균인 Pseudomonas fluorescens ID 9631, E. coli ATCC8739, 25922,35 등과 Salmonella typhimurium 114,115 등 다양한 균에 대해서도 강력한 항균작용을 보였다. 이는 재조합 hLf가 생물학적 활성이 있다는 것을 보여준다.

Preparation and Antioxidant Activities In Vitro of a Designed Antioxidant Peptide from Pinctada fucata by Recombinant Escherichia coli

  • An antioxidant peptide derived from Pinctada fucata meat using an Alcalase2.4L enzymatic hydrolysis method (named AOP) and identified by LC-TOF-MS has promising clinical potential for generating cosmetic products that protect skin from sunshine. To date, there have been few published studies investigating the structure-activity relationship in these peptides. To prepare antioxidant peptides better and improve their stability, the design and expression of an antioxidant peptide from Pinctada fucata (named DSAOP) was studied. The peptide contains a common precursor of an expression vector containing an ${\alpha}$-helix tandemly linked according to the BamHI restriction sites. The DNA fragments encoding DSAOP were synthesized and subcloned into the expression vector pET-30a (+), and the peptide was expressed mostly as soluble protein in recombinant Escherichia coli. Meanwhile, the DPPH radical scavenging activity, superoxide radical scavenging activity, and hydroxyl radical scavenging activity of DSAOP $IC_{50}$ values were $0.136{\pm}0.006$, $0.625{\pm}0.025$, and $0.306{\pm}0.015mg/ml$, respectively, with 2-fold higher DPPH radical scavenging activity compared with chemosynthesized AOP (p < 0.05), as well as higher superoxide radical scavenging activity compared with natural AOP (p < 0.05). This preparation method was at the international advanced level. Furthermore, pilot-scale production results showed that DSAOP was expressed successfully in fermenter cultures, which indicated that the design strategy and expression methods would be useful for obtaining substantial amounts of stable peptides at low costs. These results showed that DSAOP produced with recombinant Escherichia coli could be useful in cosmetic skin care products, health foods, and pharmaceuticals.

Cloning and Expression of a Novel Chitosanase Gene (choK) from $\beta$-Proteobacterium KNU3 by Double Inverse PCR

  • The DNA sequence of the chitosanase gene (choK) from $\beta$-Proteobacterium KNU3 showed an 1,158-bp open reading frame that encodes a protein of 386 amino acids with a novel 74 signal peptide. The degenerated primers based on the partial deduced amino acid sequences from MALDI- TOF MS analyses yielded the 820 bp of the PCR product. Based on this information, double inverse PCR cloning experiments, which use the two specific sets of PCR primers rather than single set primers, identified the unknown 1.2 kb of the choK gene. Subsequently, a 1.8 kb of full choK gene was cloned from another PCR cloning experiment and it was then subcloned into pGEM T-easy and pUC18 vectors. The recombinant E. coli clone harboring recombinant pUC18 vector produced a clear halo around the colony in the glycol chitosan plates. The recombinant ChoK protein was secreted into medium in a mature form while the intracellular ChoK was produced without signal peptide cleavage. The activity staining of PAGE showed that the recombinant ChoK protein was identical to the chitosanase of wild-type. The comparison of deduced amino acid sequences of choK revealed that there is 92% identity with that of Sphingobacterium multivorum chitosanase. Judging from the conserved module in other bacterial chitosanases, chitosanase of KNU3 strain (ChoK) belongs to the family 80 of glycoside hydrolases.

Generation and Expression in Plants of a Single-Chain Variable Fragment Antibody Against the Immunodominant Membrane Protein of Candidatus Phytoplasma Aurantifolia

  • Witches' broom of lime is a disease caused by Candidatus Phytoplasma aurantifolia, which represents the most significant global threat to the production of lime trees (Citrus aurantifolia). Conventional disease management strategies have shown little success, and new approaches based on genetic engineering need to be considered. The expression of recombinant antibodies and fragments thereof in plant cells is a powerful approach that can be used to suppress plant pathogens. We have developed a single-chain variable fragment antibody (scFvIMP6) against the immunodominant membrane protein (IMP) of witches' broom phytoplasma and expressed it in different plant cell compartments. We isolated scFvIMP6 from a naïve scFv phage display library and expressed it in bacteria to demonstrate its binding activity against both recombinant IMP and intact phytoplasma cells. The expression of scFvIMP6 in plants was evaluated by transferring the scFvIMP6 cDNA to plant expression vectors featuring constitutive or phloem specific promoters in cassettes with or without secretion signals, therefore causing the protein to accumulate either in the cytosol or apoplast. All constructs were transiently expressed in Nicotiana benthamiana by agroinfiltration, and antibodies of the anticipated size were detected by immunoblotting. Plant-derived scFvIMP6 was purified by affinity chromatography, and specific binding to recombinant IMP was demonstrated by enzyme-linked immunosorbent assay. Our results indicate that scFvIMP6 binds with high activity and can be used for the detection of Ca. Phytoplasma aurantifolia and is also a suitable candidate for stable expression in lime trees to suppress witches' broom of lime.

Adenoviral Vector Mediates High Expression Levels of Human Lactoferrin in the Milk of Rabbits

  • The limitations in current technology for generating transgenic animals, such as the time and the expense, hampered its extensive use in recombinant protein production for therapeutic purpose. In this report, we present a simple and less expensive alternative by directly infusing a recombinant adenovirus vector carrying human lactoferrin cDNA into rabbit mammary glands. The milk serum was collected from the infected mammary gland 48 h post-infection and subjected to a 10% SDS-PAGE and Western blotting. An 80-kDa protein was visualized after viral vector infection. With this method, we obtained a high level of expressed human lactoferrin of up to 2.3mg/ml in the milk. Taken together, the method is useful for the transient high-level expression recombinant proteins, and the approach established here is probably one of the most economical and efficient ways for large-scale production of recombinant proteins of biopharmaceutical interest.

High-Level Expression of an Aspergillus niger Endo-$\beta$-1,4-Glucanase in Pichia pastoris Through Gene Codon Optimization and Synthesis

  • To improve the expression efficiency of recombinant endo-$\beta$-1,4-glucanase in P. pastoris, the endo-$\beta$-1,4-glucanase (egI) gene from Aspergillus niger was synthesized using optimized codons. Fourteen pairs of oligonucleotides with 15 bp overlap were designed and the full-length syn-egI gene was generated by two-step PCR-based DNA synthesis. In the synthesized endo-$\beta$-1,4-glucanase gene syn-egI, 193 nucleotides were changed, and the G+C content was decreased from 54% to 44.2%. The syn-egI gene was inserted into pPIC9K and transformed into P. pastoris GS115 by electroporation. The enzyme activity of recombinant P. pastoris stain 2-7# reached 20.3 U/ml with 1% barley $\beta$-glucan and 3.3 U/ml with 1% carboxymethylcellulose (CMC) as substrates in shake flasks versus 1,270.3 U/ml and 220.7 U/ml for the same substrates in 50-1 fermentors. The molecular mass of the recombinant protein was approximately 40 kDa as determined by SDS-PAGE analysis, the optimal temperature for recombinant enzyme activity was $70^{\circ}C$, and the optimal pH was 5.0 when CMC was used as the substrate.

Production of the polyclonal subunit C protein antibody against Aggregatibacter actinomycetemcomitans cytolethal distending toxin

  • Purpose: Cytolethal distending toxin (CDT) considered as a key factor of localized aggressive periodontitis, endocarditis, meningitis, and osteomyelitis is composed of five open reading frames (ORFs). Among of them, the individual role of CdtA and CdtC is not clear; several reports presents that CDT is an AB2 toxin and they enters the host cell via clathrin-coated pits or through the interaction with GM3 ganglioside. So, CdtA, CdtC, or both seem to be required for the delivery of the CdtB protein into the host cell. Moreover, recombinant CDT was suggested as good vaccine material and antibody against CDT can be used for neutralization or for a detection kit. Materials and Methods: We constructed the pET28a-cdtC plasmid from Aggregatibacter actinomycetemcomitans Y4 by genomic DNA PCR and expressed in BL21 (DE3) Escherichia coli system. We obtained the antibody against the recombinant CdtC in mice system. Using the anti-CdtC antibody, we test the native CdtC detection by ELISA and Western Blotting and confirm the expression time of native CdtC protein during the growth phase of A. actinomycetemcomitans. Results: In this study we reconstructed CdtC subunit of A. actinomycetemcomitans Y4 and generated the anti CdtC antibody against recombinant CdtC subunit expressed in E. coli system. Our anti CdtC antibody can be interacting with recombinant CdtC and native CDT in ELISA and Western system. Also, CDT holotoxin existed at 24h but not at 48h meaning that CDT holotoxin was assembled at specific time during the bacterial growth. Conclusion: In conclusion, we thought that our anti CdtC antibody could be used mucosal adjuvant or detection kit development, because it could interact with native CDT holotoxin.

재조합 사람 적혈구 조혈인자 DA-3585의 랫드에 대한 단회 및 4주반복 정맥투여 독성시험 (Single and Four-week Intravenous Toxicity Studies of DA-3585, a Recombinant Human Erythropoietin, in Rats)

  • DA-3585 is a recombinant human erythropoietin produced by Dong-A pharmaceutical Co. Ltd. using recombinant DNA technique. Recently, recombinant human erythropoietin (rHu-EPO) has been used to treat various types of anemia. In this study, we examined acute and subacute toxicity of DA-3585 in rats. DA-3585 was intravenously administered to rats at dose levels of 0, 6,250, 12,500 and 25,000 lU/kg for single dose toxicity study and at dose levels of 0,100,500 and 2,500IU/kg daily for 4 week-repeated dose toxicity study. In the single dose toxicity study, there were no death, clinical signs and changes in body weight gain related to the treatment. Necropsy revealed no evidence of toxicity related to DA-3585, In the repeated dose toxicity study, all the rats survived throughout the study. There were no treatment-related changes in clinical signs, food and water intake, and body weight. Hematological examination showed increases in the number of erythrocytes, hemoglobin concentration, hematocrit value and mean corpuscular volume, and decrease in the number of platelet in 500 and 2,500 lU/kg dosed groups. Extramedullary hematopoiesis in the spleen and erythroid hyperplasia in the bone marrow were noted as treatment-related histological changes. Toxicologically significant changes were not observed in blood biochemistry, urinalysis, organ weights and in any other examinations. The treatment-related changes observed in this study were hematological or histological changes associated with pharmacological effects of DA-3585. On the basis of the results of this study, LD5n value of DA-3585 was above 25,000 lU/kg and the no-observed-adverse-effect-level was estimated to be 100 lU/kg.

형질전환 담배 식물체에서 재조합 erythropoietin 유전자의 발현 (Expression of Recombinant Erythropoietin Gene in Transgenic Tobacco Plant)

  • Erythropoietin(EPO)은 적혈구 모세포의 분화와 성장을 중재하는 당단백질이며 담배 식물체에서 재조합 사람 EPO를 생산하기 위해 CaMV 35S promoter를 갖는 발현 vector인 pBI$\Delta$GUS121, pBD$\Delta$ GUS121, pPEV-1을 이용하여 5.4kb의 EPO genomic DNA를 cloning 하였고 Agrobacterium tumefaciens에 의한 형질전환에 의해 Nicotiana tabacum (var. Xanthi)으로 도입되었다. Kanamycin을 포함하는 MS 배지에서 각각의 construct에 대하여 10 km 저항성 식물체들이 얻어졌다. 형질전환된 식물체의 게놈에 EPO genomic DNA의 정확한 결합은 polymerase chain reaction에 의해 332bP의 DNA 조각에 의해 확인되었으며 Northern blot 결과 1.8 kb의 전사체들이 식물체 잎에서 발현 축적되는 것이 확인되었다. Promoter의 수나 5'-UTS 서열에 의한 mRNA 양에는 변화가 없었지만 식물체 게놈에 결합된 위치 및 copy number에 의해 mRNA 수준에 영향을 주는 것으로 밝혀졌다. EPO 항체를 이용한 Western blot 결과 식물체에서 발현된 EPO 단백질의 크기는 동물세포에서 발현된 37kDa 보다 작은 30 kDa 이었다. 이는 식물체에서 modification(glycosylation) system은 동물세포에서와는 다르다는 것을 보여준다.
