일부의 EGF receptor 에는 EGF 가 세포표면에서 receptor 와 결합할 때 보다 높은 친화력(high affinity)을 보이고 있는데 그 이유를 설명하기 위해서 EGF receptor 의 cytoplasmic 영역을 절단하여 EGF 와의 친화력을 측정하였다. Scatchard plot 의 결과 1022 아미노산 이하로 절단된 receptor 는 high affinity 특성을 상실하였다. Triton X-100로 세포막을 제거하여 cytoskeleton 이 EGF receptor 의 구조에 미치는 영향을 조사한 결과 cytoskeleton과 결합한 receptor 보다 EGF 에 대해서 더 높은 친화력을 보였다. 따라서 cytoskeleton 이 high affinity EGF receptor 를 형성하는데 영향을 미치고 receptor 와 cytoskeleton 의 가능한 결합부위는 1022-1186 아미노산 사이인 것 같다.
This study was attempted to evaluate an agonistic activity to benzodiazepine receptor of several medicinal pants, which have been used as sedatives in oriental medicine. The activities of the methanol extracts of composite preparation of oriental drugs were compared with those of the simple drugs, furthermore, the active fraction was found out from the simple preparation. Inhibitory effects of composite preparations, Cyperus rotundus/Acorus gramineus, Thuja orientalis/Euphoria longan, Thuja orientalis/Albizzia julibrissin, on the binding of ${[^3H]}$Ro15-1788, a selective benszodiazepine receptor antagonist to benzodiazepine receptor of rat cortices, were observed to be lower than those of corresponding simple preparations. These unexpected results suggest that some components of the composite druge may rather act as an obstacle, not to show the sinergistic effect. The methanol extracts of Cyperus rotundus having the highest activity were fractionated using polar and nonpolar solvents to give ethylacetate and hexane fractions, respectively. The ethylacetate fraction containing relatively polar components exhibited much higher activity than the hexane fraction, which consiste of nonpolar agonist, binding to benzodiazepine receptor. However, in the presence of GABA, this fraction inhibited ${[^3H]}$flunitrazepan binding, and these positive GABA shift supported the strong possibility of agonistic activity to benzodiazepine receptro. As a result, it may be concluded that the substance or substances with neurochemical properties as a benzodiazepine receptor agonist may contribute to the sedative property of Cyperus rotundus.
We have cloned a soluble type human folate receptor(hFR type${\gamma}$) from human thymus cDNA library using the PCR amplification technique. To examine whether hFR type${\gamma}$ has a folate transport activity, CHO cells were transfected with the pcDNAhFR${\gamma}$ expression plasmid, and the stable cell line CHO/hFR${\gamma}$ expressing a high level of the hFR type${\gamma}$ was identified by northern and western blot analysis. The CHO/hFR${\gamma}$ cells produced a [$H^3$]folic acid binding protein in the culture medium. However, we couldn't detect any cell surface [$H^3$] folic acid binding and transport activities. The growth of the CHO/hFR${\gamma}$ cells was more rapidly inhibited than the wild type CHO cells in the low concentration folic acid media. These observations indicate that although soluble type human folate receptor can bind [$H^3$]folate, it does not involve in folate transport.
The objective of the present study was to investigate the effect of senescence on the binding properties of muscarinic receptors in the neostriatum of young (3 months), middle-aged (18 months) and aged (33 months) male Fischer 344 x Brown Norway hybrid rats by employing direct binding of selective radiolabeled antagonists. Using the selective M, muscarinic receptor antagonist, $[^3H]$AF-DX384, as the ligand, no significant difference in the maximal receptor density (Bmax) was observed in the neostriatum among any age-groups. In contrast, with the selective M, receptor antagonist, $[^3H]$4-DAMP, a significant increase in the number of muscarinic receptors was observed in neostriatal membrane fractions prepared from the aged animals relative to that observed in the young rats. For each ligand there was no age-related change in its affinity (Kd) for the muscarinic receptors. These results indicate that the observed age-related changes in the muscarinic receptor density may not be necessarily decremuntal and depend upon the muscarinic receptor subtype examined.
The affinity of mequitazine, a non-sedating antihistamine, for muscarinic receptors was evaluated in the guinea-pig ventricle and ileum by in vitro binding techniques and functional studies. In binding studies, [$^3$H]quinuclidinyl benzilate (QNB) identified a single class of muscarinic receptors with similar apparent $K_{D}$ value of about 100 pM in two tissues. Mequitazine inhibited [$^3$H]QNB binding to muscarinic receptors competitively. Analysis of the mequitazine inhibition curve of [$^3$H]QNB binding to ventricular microsome and ileal homogenate indicated the presence of a single homogeneous binding site with Ki value of 25 nM and 18 nM, respectively. In functional studies, mequitazine caused parallel rightward shifts of concentration-response curves for carbachol and histamine in the isolated guinea-pig ileum. The slope values obtained from Schild plot analysis for the antagonistic action of mequitazine on muscarinic and histamine $H_1$-receptors were not significantly different from unity. The p $A_2$values of mequitazine for muscarinic and histamine $H_1$-receptors were about 7.6 ( $K_{M}$= 25.1 nM) and 8.88 ( $K_{H}$= 1.32 nM), respectively. These results indicate that the muscarinic receptor blocking action of mequitazine is 15 times less potent than the $H_1$receptor blocking action, but high concentration of this drug may cause the peripheral muscarinic receptor blocking effect.t.t.t.
The ability of Hep-G2 cells to process $[^{125}I]LDL$ under basal conditions was investigated. The receptor-binding and internalization of $[^{125}I]LDL$ increased with the time of incubation in a saturable manner. After 4 h of incubation, 31.4 ng of $[^{125}I]LDL$ was cell bound. The cells rapidly internalized $[^{125}I]LDL$ via specific, receptor-mediated endocytosis. The amount of internalized $[^{125}I]LDL$ reached a maximun of 96.7 ng at 2 h of incubation and remained constant for the next 2 h. The rate of degradation of internalized $[^{125}I]LDL$ proceeded in a linear manner over the entire 4 h of incubation after an initial lag period. The effects of individial fatty acids (C18:0. C18:1, C18:2. and C18:3), differing in their degree of unsaturation. on the receptor-binding, internalization and degradation of $[^{125}I]LDL$ were also investigated. Inclusion of 1.0 mM of each fatty acid into the culture medium significantly increased $[^{125}I]LDL$ metabolism in Hep-G2 cells. Among the fatty acids tested, stearic acid had the least effect on the receptor-binding activity. There were no significant differences among the unsaturated fatty acids in LDL-receptor binding. The effect of individual fatty acids on the $[^{125}I]LDL$ uptake was similar to that of the receptor-binding. showing a significantly lower effect with stearic acid. The amount of degraded material of internalized $[^{125}I]LDL$ was the lowest with stearic acid when it was compared with unsaturated fatty acids.
몰핀의 투여량, 몰핀을 주사하는 시간, 쥐의 나이에 따라서 몰핀이 food intake에 미치는 효과 유무를 Sprague-Dawley 쥐를 이용해 조사하였다. 또한 $^{3}H-naloxone$을 이용한 opiate receptor binding assay에 의해서 정상상태에서 몰핀이나 식염수를 주사한 쥐의 뇌에서 opiate receptor binding의 변화를 조사하여 아래와 같은 결과를 얻었다. Food intake에 미치는 몰핀의 효과는 2시간에 유의차를 나타냈으며 몰핀 투여량이 증가할수록 효과는 감소한 것으로 나타났다. 또 어릴수록 효과가 많이 나타났으며 female이 male보다 효과가 컸으나 지속적이지 못했다. 몰핀을 주사하는 시간을 달리하는 실험에서 male 이나 female 모두 10:00에 주사한 경우에 몰핀 효과가 16 : 00에 주사한 경우보다 크게 나타났다. Opiate receptor binding assay 결과, 몰핀을 주사한 쥐에서 $^{3}H$ naloxone binding 이 유의있게 감소했으며 오전에 몰핀을 주사한 쥐에서 naloxone binding 이 더 많이 감소한 것으로 나타났다. 이상의 실험에서, 몰핀이 food intake 에 미치는 효과는 몰핀의 opiate receptor binding을 통해서 작용한 것으로 생각된다.
Nonsedating antihistamines do net cause sedation in therapeutic doses because these drugs hardly cross the blood-brain barrier. Since most of the peripheral side dffects of conventional antihistamines are related to their muscarinic receptor blocking action, the present study was performed to investigate whether nonsedating antihistamines interact with the muscarinic receptors and discriminate the muscarinic receptor subtypes in the rat cerebral microsomal fraction which containes both $M_1,{\;}M_2,{\;}M_3{\;}and{\;}M_4$ receptors. Five nonsedating antihistamines at high concentrations inhibited [$^3H$]QNB binding to the muscarinic receptor in a dose-dependent manner. The inhibition curves of these drugs except loratadine which showed positive cooperativity (nH=1.55) were steep (nH=1), indicating interaction with a single homogenous population of the binding sites. Astemizole, clemizole and mequitazine increased the $K_D$ value for [$^3H$]QNB without affecting the binding site concentrations, and this increase in the $K_D$ value resulted from the ability of these drugs to slow [$^3H$]QNB-receptor association. The Ki values of astemizole, clemizole and mequitazine for the inhibition for the inhibition of [$^3H$]QNB binding to muscarinic receptor were 0.58, 5.99 and $0.007{\;}{\mu}M$, respectively. However, loratadine and terfenadine inhibited noncompetitively [$^3H$]QNB binding with the normalized $IC_50$ value of about $2{\;}{\mu}M$. These results demonstrate that; 1) astemizole, clemizole and mequitazine interact directly with the muscarinic receptor at high concentrations; 2) muscarinic receptor blocking potency of these drugs varies widely among drugs; 3) these drugs do not discriminate between muscarinic receptor subtypes.
We re-cloned mouse ${\delt}-Opioid$receptor from NG108-15 cells using RT-PCR, and confirmed it by restriction analysis and by sequencing the beginning and end part of the amplified DNA. When transiently expressed in COS-7 cells, cloned ${\delt}-Opioid$ receptor showed saturable and specific binding to $[^3H]$naloxone with very similar binding parameters to originally reported ones. To make antibodies specific for the ${\delt}-Opioid$ receptor, the carboxy tail of the receptor, which is unique to the ${\delt}-Opioid$ receptor compared with other opioid receptors, was expressed in bacteria as a ufsion proteinwith glutathione S-transferase. Purified fusion protein selective for ${\delt}-Opioid$ receptor when tested by western blotting using membrane proteins prepared from transfected COS-7 cells. Cloned ${\delt}-Opioid$ receptor andl antibodies specific for ${\delt}-Opioid$ receptor are going to be valuable tools for studying pharmacological actions of the ${\delt}-Opioid$ receptor and morphine dependence.
혈청을 비롯해서 extracellular matrix에 존재하는 당단백질인 fibronectin은 근세포의 융합과 밀접한 관계가 있는 것으로 알려져 있다. 본 연구실에서는 최근에 근세포가 분화하는 동안에 fibronectin의 수준이 감소되며, 이러한 감소는 fibronectin의 28 kDa fragment에 대한 수용체으 유용성이 감소하는 결과로 밝혀낸 바 있다. 본 연구에서는 근세포으 융합을 억제하는 물질로 알려진 EGTA를 이용하여 근세포의 융합과 28 kDa fragmen receptor의 관계를 검토하여 보았다.EGTA를 처리한 경우 EGTA를 처리하지 않은 근세포에 비해서 fibronectin의 수준과 28 kDa fragmen binding이 훨씬 적게 감소하였으며, 융합이 봉쇄된 근세포에서 EGTA를 제거하여 융합을 재개시키면 fibronectin의 수준과 28 kDa fragmen의 binding이 정상 근세포 수준으로 환원되었다. 이상의 실험 결과로 볼 때 28 kDa fragmen에 대한 수용체의 감소 또는 변화가 근세포의 분화과정에서 일어나는 fibronectin 수준의 감소와 연관성이 있음을 알 수 있다. 한편, 배양액 내에 trypsin을 처리한 경우에는 처리하지 않은 경우에 비해서 28 kDa fragmen의 binding이 현저하게 감소되었고, gangliosides를 처리한 상태에서는 gangliosides의 농도에 정비례해서 28 kDa fragmen의 binding이 감소되었다. 이 밖에 gel overlay technique을 이용하여 28 kDa fragmen가 SDS-PAGE gel에서 분자량이 약 43kDa인 단백질 및 gangliosides와 binding하는 사실을 알 수 있었다. 이러한 실험 결과를 종합하여 볼 때, 근세포의 융합은 28 kDa fragmen에 대한 receptor의 감소와 관계가 있으며, 그 수용체는 gangliosides와 비슷한 당을 가지고 있는 당단백질일 것으로 추정된다.
