Antimutagenic Effect of the Extracts of Comfrey (컴프리 추출액에 의한 항돌연변이효과)

  • This study was carried out to investigate the antimutagenic affect of crude and heated comfrey extract on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), benzo${(\alpha)}$pyrene (B${(\alpha)}$P) and 3-amino-1, 4-dimethyl-5H-pyri-do [4,3-b] indole (Trp-P-1). In spore rec-assay using Bacillus subtilis $H17(rec^+)$ and $M45(rec^-),$ crude comfrey extract showed strong antimutagenic effects on MNNG in the concentration of $40{\mu}l/disc$(p<0.01). In the Ames test using Salmonella typhimurium TA98 and TA100, the crude comfrey extract suppressed about 43% and 52% in the mutagenesis induced by $B{(\alpha)}P.$ However, the heated comfrey extract strongly suppressed about 75% and 76% in the mutagenesis in Salmonella typhimurium TA98 and TA100 induced by Trp-P-1(p<0.01).

  • Antimutagenic effects of boiled water extract and tannin from persimmon leaves were studied by using Ames test, spore rec assay and SOS chromotest. Strong antimutagenic activities toward aflatoxin B1(AFB1), dimethyl-aminobiphenyl(DMAB), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO) were observed when boiled water extract and tannin from the persimmon leaves were added in the Salmonella typhimurium TA 100. In spore rec assay using Bacillus subtilis H17($rec^{+}$) and M45($rec^{-}$), boiled water extract and tannin from the persimmon leaves considerably inhibited the mutagenesis induce by MNNG. In SOS chromotest using Escherichia coli PQ37, these samples also exhibited strong antimutagenic activity toward 4-NQO. The tannin was more effective than boiled water extract of persimmon leaves in the antimutagenicity tests.

  • Mutangen sensitive hyper-rec type musN mutants were assigned into the UvsC group which contains genes involved in recombination and mutation. however, phenotypic properties of musN mutants were very different from those found in uvsC mutant strains which are rec- and lack UV-induced mutation. musN was not a mutator like uvsC. In addition, selenate resistant mutations in musN were induced sililar to those in wild types by UV irradiation. Wild type levels of UV-sensitivity in dividing cells of musN also differ from the uvsC phenotypes. These indicate that the UvsC group has branched pathways.

Characteristics and Pathogenicity of Host Range Expanded Recombinant Viruses in Insect Cells (숙주범위가 넓어진 재조합 바이러스의 세포주에서의 특성 및 병원성)

  • To use recombinant viruses with wider host range as viral insecticides, we investigated the characteristics and pathogenicity of host range expanded recombinant viruses in insect cells. We compared host range expanded recombinant viruses, RecS-B6 and RecB-8, constructed by cotransfection of Autographa californica nuclear polyhedrosis virus (AcNPV) and Bombyx mori NPV (BmNPV), to host range expanded AcNPV, Ac-BH, by substitution of the 0.6 Kb fragment of the BmNPV helicase gene. Restriction endonuclease profiles of RecS-B6 and RecB-8 DNAs were different from those of parent viruses. Nucleotide sequence analysis of the 0.6 Kb region in the putative helicase gene of RecS-B6 and RecB-8 showed that their structures were identical to the counterpart region of BmNPV. Comparison of viral replication of these recombinant viruses in Sf-21 and BmN-4 cells showed that Ac-BH, compared to wild type viruses, replicated well in BmN-4 cells but poorly in Sf-21 cells. In contrast, RecS-B6 and RecB-8 replicated relatively well in both cells compared to parent viruses. These results may imply that random genomic recombinant viruses, RecS-B6 and RecB-8, possess better potential as viral pesticides than helicase-mediated recombinant virus, Ac-BH.

Molecular Biological Characterization of Recombinant Baculovirus with an Expanded Host Range (숙주범위가 넓어진 유전자 재조합 핵다각체병 바이러스의 분자생물학적 특성)

  • To investigate the host range determining factors of nuclear polyhedrois virus (NPV), Autographa california NPV and Bombyx mori NPV were coinfected into the two different cell lines, BmN-4 and Sf-9. The recombinant baculoviruses, RecS-A6 and RecB-727 which have an expanded host range, were isolated from Sf-9 and BmN-4 cell lines, respectively. The molecular biological characteristics of the recombinant baculoviruses were investigated. The pathogenicity of RecB-727 was similar to that of wild type BmNPV, while the pathogenicity of RecS-A6 was relatively lower than that of wild type BmNPV. The restriction enzyme digestion patterns of parental viruses and recombinant viruses showed that the recombinant virus has an expanded host range by genetic recombination. Southern blot analysis revealed that the p10 gene of RecB-727 was derived from AcNPV genomic DNA, while RecS-A6 has p10 gene of BmNPV in a viral genome. To investigate the host range expansion mechanism of recombinant baculovirus, HindIII-SacI 0.6 kb DNA fragments of RecS-A6 and RecB-727 were cloned and sequenced. The results showed that of wild type BmNPV helicase gene, suggesting that the expanded host range of recombinant baculoviruses was due to the insertion of BmNPV helicase gene into AcNPV viral genome.

  • PCR-based Weissella species-specific detection method was developed to apply for the discrimination of Korean and Chinese kimchi by detecting a Weissella species only found in Korean or Chinese kimchi. PCR primers were designed from the species-specific sequence in the recN gene of each species. The primers allowed the species-specific detection and identification of nine species in the genera Weissella, and were successfully applied to the detection of W. cibaria, W. confusa, W. koreensis, and W. soli in kimchi with 20 ng template DNA. W. cibaria, W. confusa, and W. koreensis were detected from the Korean kimchi samples tested but W. soli was not detected. However, the four species were detected from Chinese kimchi samples. PCR-based W. soli-specific detection could not be perfectly applied as the Chinese kimchi discriminating method but has significance as an approach to evaluate the potential of scientific verification method based on the difference of microbial community.

Isolation and Characterization of Lactic Acid Bacteria Producing Antimutagenic Substance from Korean Kimchi (김치로부터 항돌연변이 물질을 생산하는 유산균의 분리 및 특성)

  • Various lactic acid bacteria were isolated from Korean Kimchi in order to study their antimutagenic substances. Ames test using Salmonella typhimurium TA98 and TA100 showed the strain KLAB21 to have the highest antimutagenic activity among the 230 isolated strains against MNNG (N-methyl-N'-nitro-N-nitrosoguanidine), NPD (4-nitro-O-phenylenediamine), NQO (4-nitrosoquinoline-1-oxide) and AFB1 (aflatoxin B1). The strain was identified as Lactobacillus plantarum based on its morphological, cultural and physiological characteristics. Antimutagenic activity of L. plantarum KLAB21 was found in culture supernatant suggesting the bacterium secrete antimutagenic substance in the media. No mutagenic activity was found in the culture supernatant. The isolated strain L. plantarum KLAB21 showed much higher antimutagenic activity than L. plantarum IAM1261 which is being used industrially for fermented milk production. The antimutagenic activity of L.plantarum KLAB21 was reconfirmed by the spore-rec assay using spores of Bacillus subtilis H17($Rec^+$) and M45($Rec^-$).

Antimutagenic Effect of Korean Mistletoe Extracts (겨우살이 추출물의 항돌연변이 효과)

  • This study was carried out to investigate mutagenecity and antimutagenic effects from crude extract, heating extract and alcohol extract of Korean mistletoe(Viscum album L.) on the bacterial short-term tests, such as Ames test, spore rec-assay, SOS spot test and SOS chromotest by using several kinds of mutagens. In the Ames test, each extract did not show any mutagenesis, but each extract showed inhibitory effects of 80∼95% and 70∼94% against mutagenesis induced by 3-amino-1, 4-dimethyl-5H-pyrido[4,3-b] indole(Trp-P-1) and 2-aminofluorene(2-AF) in Salmonella typhimurium TA98, respectively. In th spore rec-assay, mistletoe ectracts showed antimutagenic effect with inhibiton zone in the range of 5∼11mm against mutagenicity induced by mitomycin C(MMC, 18mm) and N-methyl-N'-nitro-N-nitrosoguanidne (MNNG, 24mm), respectively. The heating and alcohol extracts in the SOS chromotest showed 96% and 70% inhibition against benzo-α-pyrene[B(α)P] and Trp-P-1 induced mutagenesis, respectively.

Influence of Electronic-cigarette Smoke on Cardiac Autonomic Nerve Responses in Comparison with Conventional-cigarette Smoke (전자담배흡연이 심장자율신경조절에 미치는 반응: 궐련담배와의 비교 검증)

  • This study aims to observe changes in heart-rate variability (HRV) indices induced by e-cigarette and conventional-cigarette smoking and to compare the differences in acute cardiac autonomic regulation. All participants (n=41) were exposed to both e-cigarette smoke (ES) and conventional cigarette smoke (CS) in a randomized crossover trial. HRV analysis was performed during each smoking session based on a recorded r-r interval 10 minutes before smoking and at specified recovery periods (REC1, 0-5 min; REC2, 5-10 min; REC3, 10-15 min; REC4, 15-20 min; REC5, 20-25 min; and REC6, 25-30 min). ES led to a significantly increased cardiac sympathetic index (LF/HF ratio) compared with the baseline, and it shifted the sympathovagal balance toward sympathetic predominance, including reduction in the complexity of the interbeat interval (SampEn). In REC1 after ES, only decreases of parasympathetic indices such as rMSSD, pNN50, HF, and SD1 were indicated. CS sessions produced not only an increased LF/HF ratio during smoking and recovery periods (REC1 and REC4) but also enhanced sympathetic predominance on autonomic balance during smoking and recovery periods (REC1, REC2, and REC4). In the CS trials, parasympathetic indices of time and non-linear analysis (rMSSD, pNN50, and SD1) were decreased during smoking and in REC1 to REC5. SampEn was also reduced during smoking and REC1 to REC4. Acute sympathoexcitatory effects induced by e-cigarette use produced statistically significant results. Parasympathetic withdrawal after smoking suggests that e-cigarettes may cause increased cardiovascular risk.

Characterization of Recombinant Baculovirus Ewpressing Polyhedrin Gene of Bombyx mori and Autographa californica Nuclear Polyhedrois Virus (두 종류의 다각체 단백질 유전자를 발현하는 유전자 재조합 핵다각체병 바이러스의 특성)

  • Recombinant baculoviruses having expanded host range were selected by coinfection of Autographa california NPV and Bombyx mori NPV into Sf-9 and BmN-4 insect cell lines. In order to determine the polyhedra morhplogy of RecS-A6, one of a recombinant baculovirus, polyhedra of RecS-A6 produced in insect cells were observed by phase contrast microscope and scanning electron microscope. The results revealed that the recombinant baculovirus had a various polyhedra morphology which was different from its parental viruses, suggesting that the various morhpology of recombinant baculovirus with an expanded host range was due to the genetic recombination of viral genome. To analyze the genomic recombinantion of the recombinant baculoviruses, genomic DNAs of two parent viruses and RecS-A6 were digested with restriction endonuclease and subjected to agarose gel electrophoresis. Southern blot analysis revealed that RecS-A6 has two polyhedrin gene of AcNPV and BmNPV in a viral genome. Polyhedral protein of recombinant baculovirus was analysed by SDS-PAGE. The result showed that molecular weight of polyhedral protein of RecS-A6 containing two polyhedrin gene of AcNPV and BmNPV was as the 31 kDa band of AcNPV and 30 kDa band of BmNPV.
