REAL ROOT ISOLATION OF ZERO-DIMENSIONAL PIECEWISE ALGEBRAIC VARIETY

  • As a zero set of some multivariate splines, the piecewise algebraic variety is a kind of generalization of the classical algebraic variety. This paper presents an algorithm for isolating real roots of the zero-dimensional piecewise algebraic variety which is based on interval evaluation and the interval zeros of univariate interval polynomials in Bernstein form. An example is provided to show the proposed algorithm is effective.

해녀콩(canavalia lineata) 뿌리혹으로부터 공생균주 Rhizobium spp.의 분리 (Isolation of Symbiotic Rhizobium spp. Strain from Root Nodule of Canavalia lineata)

  • The root nodule of Canavalia lineta was classified as a determinate nodule and the symbiont as a Rhizobium-bacteriod based on their morphological characteristics. Isolated encosymbiont was similar both to R. leguminosarum and R. meliloti in its peritrichous arrangement of flagella and some of the physiological characteristics. Compared to control plants, Canavalia seedlings inoculated with the isolate grew normally due to induced root nodules, confirming isolate's infectivity and effectivity. Characteristics of the reisolated endosymbiont from induced root nodule were identical to those of the first isolate, indicating the nodules were induced by the first isolate. From these results, it was confirmed that Rhizobium strain isolated from the root nodules of Canavalia lineata was a real symbiont, and was named Rhizobium sp. SNU003.

Isolation, Characterization, and Use for Plant Growth Promotion Under Salt Stress, of ACC Deaminase-Producing Halotolerant Bacteria Derived from Coastal Soil

  • In total, 140 halotolerant bacterial strains were isolated from both the soil of barren fields and the rhizosphere of six naturally growing halophytic plants in the vicinity of the Yellow Sea, near the city of Incheon in the Republic of Korea. All of these strains were characterized for multiple plant growth promoting traits, such as the production of indole acetic acid (IAA), nitrogen fixation, phosphorus (P) and zinc (Zn) solubilization, thiosulfate ($S_2O_3$) oxidation, the production of ammonia ($NH_3$), and the production of extracellular hydrolytic enzymes such as protease, chitinase, pectinase, cellulase, and lipase under in vitro conditions. From the original 140 strains tested, on the basis of the latter tests for plant growth promotional activity, 36 were selected for further examination. These 36 halotolerant bacterial strains were then tested for 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity. Twenty-five of these were found to be positive, and to be exhibiting significantly varying levels of activity. 16S rRNA gene sequencing analyses of the 36 halotolerant strains showed that they belong to 10 different bacterial genera: Bacillus, Brevibacterium, Planococcus, Zhihengliuella, Halomonas, Exiguobacterium, Oceanimonas, Corynebacterium, Arthrobacter, and Micrococcus. Inoculation of the 14 halotolerant bacterial strains to ameliorate salt stress (150 mM NaCl) in canola plants produced an increase in root length of between 5.2% and 47.8%, and dry weight of between 16.2% and 43%, in comparison with the uninoculated positive controls. In particular, three of the bacteria, Brevibacterium epidermidis RS15, Micrococcus yunnanensis RS222, and Bacillus aryabhattai RS341, all showed more than 40% increase in root elongation and dry weight when compared with uninoculated salt-stressed canola seedlings. These results indicate that certain halotolerant bacteria, isolated from coastal soils, have a real potential to enhance plant growth under saline stress, through the reduction of ethylene production via ACC deaminase activity.

Conservative approach of a symptomatic carious immature permanent tooth using a tricalcium silicate cement (Biodentine): a case report

  • The restorative management of deep carious lesions and the preservation of pulp vitality of immature teeth present real challenges for dental practitioners. New tricalcium silicate cements are of interest in the treatment of such cases. This case describes the immediate management and the follow-up of an extensive carious lesion on an immature second right mandibular premolar. Following anesthesia and rubber dam isolation, the carious lesion was removed and a partial pulpotomy was performed. After obtaining hemostasis, the exposed pulp was covered with a tricalcium silicate cement (Biodentine, Septodont) and a glass ionomer cement (Fuji IX extra, GC Corp.) restoration was placed over the tricalcium silicate cement. A review appointment was arranged after seven days, where the tooth was asymptomatic with the patient reporting no pain during the intervening period. At both 3 and 6 mon follow up, it was noted that the tooth was vital, with normal responses to thermal tests. Radiographic examination of the tooth indicated dentin-bridge formation in the pulp chamber and the continuous root formation. This case report demonstrates a fast tissue response both at the pulpal and root dentin level. The use of tricalcium silicate cement should be considered as a conservative intervention in the treatment of symptomatic immature teeth.

시스템 결함원인분석을 위한 데이터 로그 전처리 기법 연구 (A Study on Data Pre-filtering Methods for Fault Diagnosis)

  • High performance sensors and modern data logging technology with real-time telemetry facilitate system fault diagnosis in a very precise manner. Fault detection, isolation and identification in fault diagnosis systems are typical steps to analyze the root cause of failures. This systematic failure analysis provides not only useful clues to rectify the abnormal behaviors of a system, but also key information to redesign the current system for retrofit. The main barriers to effective failure analysis are: (i) the gathered data (event) logs are too large in general, and further (ii) they usually contain noise and redundant data that make precise analysis difficult. This paper therefore applies suitable pre-processing techniques to data reduction and feature extraction, and then converts the reduced data log into a new format of event sequence information. Finally the event sequence information is decoded to investigate the correlation between specific event patterns and various system faults. The efficiency of the developed pre-filtering procedure is examined with a terminal box data log of a marine diesel engine.

조경 설계에서 전원 이상의 전통과 그 이면 (The Pastoral Ideal and its Legacy to Landscape Design)

  • This paper examines the pastoral ideal and its legacy to landscape design. We have had the routine convention of experiencing natural beauty from the picture-like nature. We, who would liken a scenic view to a picture, often equate natural beauty with superficial representations of nature shown in pastoral literature and traditional landscape paintings: the lush of towering trees, the field of endless green, the soft babbling flow of the river and the crisp clear sky. It is not a portrayal of nature as it is, but in fact a conceptual expression of its ideal form. The ideological root of that natural beauty can be retraced to the pastoral ideal, so eagerly longed for by people of the western world. A pastoral landscape graciously marked with leisurely peace and subtle harmony is what we identify as beautiful nature. In fact, however, it is no more than an artificial refuge in contextual isolation from its surrounding, and a by-stander's nature seen purely through the eyes of the outsider. The tradition of pastoralism, reaching its peak in the 18th century with its English Landscape Garden style, was transplanted into the real world through the practice of landscape architecture. Landscape design in such a form is just a static means of decoration devoid of meaning and process. And we simply identify the green ornamentation with nature.

한국에 유통중인 신선편이 채소류의 미생물 품질 및 병원성 세균의 오염도 조사 (Microbial and Pathogenic Contamination of Ready-to-eat Fresh Vegetables in Korea)

  • 신선 농식품은 일반적으로 가열하지 않고 직접 신선한 상태로 섭취하기 때문에 병원성 미생물에 오염되어 있을 경우 식중독을 일으킬 수 있다. 이에 본 연구는 한국에 유통되는 총 20종의 채소에서 총균수, 대장균군, 대장균, 효모, 곰팡이 및 병원성 세균(S. aureus, L. monocytogenes, B. cereus, Salmonella, E. coli O157:H7, C. sakazakii, Shigella, Campylobacter)을 측정하여 신선농식품의 미생물 오염실태를 조사하였다. 채소의 총균수와 대장균군의 분포는 각각 3.74-8.04 log CFU/g, 0.16-5.02 log CFU/g 수준이였으며 이중에서 미나리, 새싹, 숙주, 도라지에서 가장 높은 수준의 미생물이 관찰되었다. 유기농과 비유기농의 상추와 깻잎의 오염도를 관찰했을 때, 두 군의 일반 미생물 수준에는 유의적 차이가 나타나지 않았다. 신선 농식품에 오염된 병원성 세균을 조사하였을 때, 선택배지를 이용한 분리법에서 L. monocytogenes가 배추, 양상추, 오이, 콩나물에서 검출되었고, B. cereus는 상추, 깻잎, 당근, 미나리, 새싹에서 검출되었으며, Campylobacter의 경우는 20종 모두에서 한번 이상 검출되었다. 이중 API kit로 동정한 결과 총 48개의 시료가 양성으로 판명되었으며 마지막으로 real-time PCR법과 16S rRNA sequencing법을 이용하여 확인한 결과 깻잎, 당근, 미나리, 새싹에서 발견된 B. cereus만이 양성으로 확인되었다. 따라서 본 연구결과로부터 몇몇 신선 농식품에서 B. cereus가 검출되었으며 비교적 높은 총균수와 대장균군의 수준을 확인할 수 있었다. 또한 본 연구결과 분리방법에서 선택배지나 API kit를 이용하였을 때 몇몇 병원성 세균에 대해서는 맞지 않는 양성결과가 확인되었으므로 이에 신선 농식품에서 보다 정확하게 분리할 수 있는 방법의 개발이 필요할 것으로 사료된다.

STUDIES ON THE TISSUE CULTURE OF PANAX GINSENG

  • Unlike the tissue culture in animals and human being, in higher plants various parts of the plant are cultured for varied purposes, and they are named variously depending on which parts are used as explants or what purposes they are cultured for. Followings are some of the names of culture used frequently: organ culture, tissue culture, callus culture, single cell culture, meristem culture, mericlone culture, ovary culture, ovule culture, embryo culture, endosperm culture, anther culture, pollen culture, protoplast culture, etc.. As the names of the culture indicate, in some kinds of culture the explants used for culture are actually not tissues, but organs, single cells, or protoplasts. It seems, however, convenient to call all of the above-mentioned cultures grossly as tissue culture. Several kinds of tissue culture were attempted using Panax ginseng as material and some of the results were summarized below. 1. Callus culture After dormancy of the sed was broken, whole embryo or parts (hypocotyl, cotyledon and epicotyl) of partly grown embryo were cultured in the media supplemented with growth regulators. Rapid swelling occurred in a few weeks, but most of the swelling was observed only in the basal part of epicotyl, changes in the other parts of embryo appearing in much later stages. The swelling or increase in size, however, was resulted not from the divisions of cells, but from the mere expansion of cell. Real calli were formed about two months after inoculation of explants. Callus tissues developed from cortex, pith, and vascular bundle in the cases of hypo- and epicotyl, from mesophyl tissue in the case of cotyledon. Shoots developed more easily from cotyledons regardless of whether they are detached from or attached to the embryo proper. 2. Culture in the Knudson C medium When cotyledons, detached from or attached to the embryo proper, were cultured in the growth regulator-free Knudson C medium comprision only several kinds of mineral compounds and sucrose, shoot primordium or callus developed profusely and finally plantlets were produced directly from shoot primordium or indirectly through callus. In this medium epidermal cells as well as mesophyl cells of the cotyledon became meristematic and divided, changing into multinucleate cells or multicellular bodies, developing eventually into either shoot primordia or calli. 3. Anther culture Anthers were cultured in the media supplemented with various growth regulators applied singly or in combinations. Callus was formed mostly in the connective tissue of anther. Cells of anther wall layers changed in appearance, but no division occurred. Microspores of all stages in development were not changed, ruling out the possibility that microspore-originated callus might be formed. 4. Isolation of protoplast Protoplasts were isolated from young root, leaf, and epicotyl, using 0.7M D-mannitols as osmoticum and using macerozyme and cellulase respectively for maceration and digestion of the cell wall. Production in large number of naked intact protoplast was rather difficult as compared with other plant species. Fusion of protoplasts occurred infrequently mainly due to the fewer number of naked protoplasts in the solution.
