Two-gene sequences and morphology of Gelidium zollingeri (Kutzing) comb. nov. (Gelidiales, Rhodophyta)

  • Porphyroglossum is the last one of nine genera within the family Gelidiaceae that has yet to be analyzed by molecular markers. We analyzed rbcL and cox1 genes from P. zollingeri specimens collected near the type locality in Indonesia and compared them with other gelidioid algae. Thalli are cartilaginous, complanate, and up to 15 cm high. Abundant rhizoidal filaments are concentrated in the medullary layer. Tetrasporangial sori are on small, determinate ramuli. In all gene analyses, P. zollingeri consistently nested within Gelidium. The sister relationship of P. zollingeri to G. floridanum was well resolved. Because Gelidium has priority over Porphyroglossum, a new combination is proposed, viz. Gelidium zollingeri. Network analysis of the four cox1 haplotypes revealed many missing haplotypes, indicating high genetic diversity in the species.

The conspecificity of Pterosiphonia spinifera and P. arenosa (Rhodomelaceae, Ceramiales) inferred from morphological and molecular analyses

  • The genus Pterosiphonia includes twenty-one currently described species of red algae that occur in temperate to tropical regions of the Atlantic and Pacific Oceans. Pterosiphonia spinifera was originally described as Polysiphonia spinifera from Peru and later transferred to Pterosiphonia. Pterosiphonia spinifera has been reported from Peru as Pterosiphonia pennata, which was originally described from the Mediterranean Sea. Recently, Pterosiphonia arenosa was described based on specimens of P. pennata from Korea. We collected P. spinifera along the coast of Peru and P. arenosa near the type locality in Korea. We compared them with the isotype specimens of P. arenosa using both morphological and molecular data. Our morphological observations and our phylogenetic analysis of rbcL sequences demonstrate that P. spinifera and P. arenosa are conspecific and indicate that P. arenosa is a later synonym of P. spinifera. Our study confirms the wide occurrence of P. spinifera in the western and eastern Pacific Ocean.

Miuraea migitae, a new record of the order Bangiales (Bangiophyceae, Rhodophyta) from Korea

  • We found specimens of foliose Bangiales from the subtidal zone of Udo, Jeju Island, Korea. In molecular analyses of rbcL sequences, these Korean specimens were almost identical to Miuraea migitae from Osaka, Japan. In the morphological comparison, Korean specimens were consistent with habitat, color, and vegetative characteristics with the description of M. migitae. This is the first record of M. migitae outside the type locality and Nagasaki in Japan. This study confirms that new or unrecorded species of the order Bangiales may be discovered from subtidal habitats.

한국산 미기록 해조 2종, 흐린깃털말 및 가시모자반 (New Record of Two Marine Algal Species in Korea: Bryopsis triploramosa and Sargassum polyporum)

  • Morphological and molecular information of two marine algae collected from Korea is given. One belonging to a green alga, is characterized by small size up to 4 cm high, much branched thalli on all sides, slender and linear branchlets which is constricted near base, axes without prominent branch scars, chloroplasts with a prominent central pyrenoid. The other, as a brown alga, is distinct from other Korean species in having cylindrical elevated projections at all parts of thallus axis, leaves with slightly serrulate margin together with distinct midrib bearing spines and absence to rarely occurred vesicles. In phylogenetic tree based on rbcL sequences, these two species are nested in the same clade with Bryopsis triploramosa and Sargassum polyporum, respectively. In this study, these two species are newly recorded in the Korean marine algal flora based on the morphological and molecular data.

A novel species Symphyocladia glabra sp. nov. (Rhodomelaceae, Rhodophyta) from Korea based on morphological and molecular analyses

  • Six species of the genus Symphyocladia are currently recognized worldwide, all of which are reported to grow on the Korean coast. We described a new species Symphyocladia glabra sp. nov. based on morphological and molecular evidence. The new species is characterized by mostly an erect and broad thallus, the parallel arrangement of numerous apical cells, completely fused congenital cells, a corticated basal portion of faint midrib with six to eight pericentral cells, rhizoids cutting off from pericentral cells, and absent vegetative trichoblasts. The new species has morphological similarity to S. marchantioides and S. jejuinsula. However, S. glabra sp. nov. is distinguished from S. marchantioides by the corticated lower portion of the thallus and the absence of vegetative trichoblasts, and S. jejuinsula by number of pericentral cells and the fact that the thallus does not taper upward. The phylogeny of rbcL sequences indicated that S. glabra sp. nov. is definitely a separate entity within the genus Symphyocladia.

A new species of marine algae from Korea based on morphology and molecular data: Gelidium palmatum sp. nov. (Gelidiales, Rhodophyta)

  • Two species of the agar-yielding genus Gelidium, G. galapagense and G. isabelae, have previously been reported from Korea but their occurrence has not been confirmed with molecular data. We intensively collected samples of Gelidium from Jeju Island, where the two species were reported, and the southern coast of Korea. Phylogenetic analyses based on cox1 and rbcL sequences revealed that only a single species occurred in Korea. The Korean species was distantly related to G. galapagense and G. isabelae from the Galápagos Islands, and formed a clade with G. microdonticum, G. millarianum, and G. pakistanicum. A new species, G. palmatum, is described for those specimens that were previously recognized as either G. galapagense or G. isabelae from Korea. G. palmatum is small in size (up to 0.7 cm), with compressed, lanceolate axes, irregular, digitate to palmate branches, abundant rhizines in the medulla, tetrasporangial sori without sterile margins, and rounded bilocular cystocarps borne subapically on palmate branchlets.

New Record of a Marine Algal Species, Ahnfeltiopsis linearis (Phyllophoraceae, Gigartinales), in Korea

  • A marine algal species belonging to Gigartinales was collected from Geoje, Korea. This shares the generic features of Ahnfeltiopsis, such as multiaxial thalli with a compact and pseudoparenchymatous medulla, densely cytoplasmic secondary medullary cells around immersed cystocarps with a carpostome, and is distinct from similar species within the genus by a combined feature of small (up to 4 cm tall) and tuft thalli, compressed to subcompressed branches except for ultimate branchlets and base of main axes, cartilaginous in texture, dichotomous branches, rarely produced proliferations, absence of hypha-like filament in the medulla and internal cystocarps with a carpostome. In phylogenetic tree based on rbcL sequence, the Korean species nests in the same clade with Ahnfeltiopsis linearis. The genetic distance between both sequences within the clade was 1.5%, considered to be within the intra-species range for the genus. This morphological and molecular evidence confirms the Korean alga to be identified as A. linearis originally described from California. This is the first record of A. linearis in Korea.

Description of an unrecorded diatom Fragilaria saxoplanctonica Lange-Bertalot & Ulrich (Bacillariophyceae) from Paldang Reservoir in Korea

  • Diatoms are unicellular eukaryotic microalgae, and they are highly diversified in aquatic environments. We describe an unrecorded diatom species Fragilaria saxoplanctonica Lange-Bertalot & Ulrich (Bacillariophyceae) collected from Paldang Reservoir, Korea, on 4 April 2022. The valve was needle shaped and narrowly rectangular, its ends were rounded, and no spines were found on the outline of their valves. The valve was 67.2-70.2㎛ in length and 1.4-2.9㎛ in width. The ratio of width-to-length was 1 : 23.2-50.1. The pattern of striation was alternate or opposite, and the number of striae in 10㎛ was 24-26. Molecular comparisons of the 18S rDNA and rbcL sequences showed that it belonged to the genus Fragilaria. These morphological and phylogenetic results confirmed that our species was F. saxoplanctonica, and it was the first record in Korea.

Adding to the freshwater red algal diversity in North America: Lympha mucosa gen. et sp. nov. (Batrachospermales, Rhodophyta)

  • The strictly freshwater red algal order Batrachospermales has undergone numerous taxonomic rearrangements in the recent past to rectify the paraphyly of its largest genus Batrachospermum. These systematic investigations have led to the description of new genera and species as well as re-circumscription of some taxa. Specimens collected from two locations in the southeastern USA were initially identified as being allied to Batrachospermum sensu lato, but could not be assigned to any recognized species. Representative rbcL (plastid) and COI-5P (mitochondrion) sequences showed these specimens to be similar to each other and not closely matching the previously published sequence data for other Batrachospermum taxa. Comparison of sequence variation and morphology with a broader range of batrachospermalean taxa resulted in the proposal of a new monotypic genus Lympha mucosa gen. et sp. nov. to accommodate these specimens. Lympha mucosa is sister to members of a newly described genus Volatus, but the two genera are easily distinguished based on straight versus curved, twisted or spirally coiled carpogonial branch, respectively. This new taxon has morphological similarities to Batrachospermum sections Turfosa and Virescentia, but can be differentiated based on genetic divergence in rbcL and COI-5P as well as a combination of morphological characters: dense, compressed whorls, axial carposporophytes with a single type of gonimoblast filament; cortication of the main axis closely appressed; and short, straight carpogonial branch arising from the pericentral cell and carpogonia with unstalked, lanceolate trichogynes. This new taxon adds to the freshwater red algal diversity of the southeastern USA, a region already known for biodiversity and high endemism of the aquatic flora and fauna. It is also a relevant new addition to the taxonomic knowledge of the freshwater red algal Batrachospermales.

국내에 유입되는 열대피(Echinochloa colona) 동정: DNA 바코드 중심 (Discrimination of Echinochloa colona (L.) Link from other Echinochloa Species using DNA Barcode)

  • 열대피와 같은 논농사지역의 악성잡초는 국내에 유입될 경우 문제 잡초로 정착될 가능성이 높기 때문에 유입되는 초기에 방제가 될수 있도록 정확한 동정이 되어야한다. 그러나 피속 잡초는 형태적으로 연속변이가 많이 존재하여 종간 구별이 매우 어려운 잡초이다. 본 연구는 미국 NPGS에서 분양받은 열대피와 국내에서 채집한 돌피와 논피를 고등식물 표준바코드 마커 rbcL과 matK를 이용하여 바코드하고, 추가적으로 핵 DNA ITS 부위를 바코드하여 표준바코드 구간과 ITS 구간의 열대피를 동정할 수 있는 능력과 바코드 활용성을 비교하였다. 바코드 결과, rbcL은 0.36%, matK는 0.29%, ITS는 3.2%의 열대피 특이 염기서열이 조사되었고 Neighbor-joining 계통수에서 종별 유집이 뚜렷하게 나타나 표준바코드 마커와 ITS 모두 쉽고 간편하게 열대피를 국내의 돌피, 논피와 동정할 수 있었다. 특히 ITS는 분석구간은 짧지만 열대피를 국내의 논피, 돌피와 정확하게 구분해낼 수 있어서 ITS 단독으로 국내에 유입되는 열대피 동정에 활용할 수 있을 것으로 생각한다.
