Characteristics of bioethanol production using sweet sorghum juice as a medium of the seed culture (단수수 착즙액이용 배양종균의 바이오에탄올 생산 특성 연구)

  • Sweet sorghum [Sorghum bicolor (L)] is one of the major crops for biofuels such as sugarcane and sugar beet which raw materials rich in saccharide. Sweet sorghum juice was extracted from the stem. It's composed of fermentable sugars such as glucose, fructose and sucrose. Ethanol from the extracted sweet sorghum juice can be easily produced by yeast fermentation process. Sweet sorghum juice is consisted of not only sugars but also various nutrients like nitrogen and phosphate. For commercial production of bioethanol, seed culture is one of the important parts of fermentation, so that optimal culture medium should be selected for the reduction of processing costs. In this study, sweet sorghum juice was estimated as a culture medium for seed culture of cellulosic bioethanol. For the comparison of cultures with various substrates, it used YPD including each 5 g/L yeast extract and peptone, sweet sorghum juice and hydrolyzed Miscanthus was taken part in the culture with 2%, 5% and 10% sugar conditions. Based on media of YPD and sweet sorghum juice, cell-mass concentration was obtained maximum more than $2.5{\times}10^8CFU/mL$ after 24 h of cultivation. Consequently sweet sorghum juice is suitable for the cell culture with more than $1.0{\times}10^8CFU/mL$ after 12 h of cultivation. This can be used as a culture medium for the cellulosic bioethanol industry.

Experimental Studies on the Optimum Pasteurization Condition of the Cow's Milk Produced in Korea IV.   • The results of experiment to review the optimum pasteurization condition for the raw milk produced in Korea by using UHT procedure of $100-145^{\circ}C$, the changes in chemical composition, microbiological aspects and the keeping quality of the heat treated milk are summarized as follows: 1. In UHT milk sterilized at $100-145^{\circ}C$, the pH value decreased from 6.55 to 6.33 but protein, fat, lactose and ash did not show significance changes while casein nitrogen and non-protein nitrogen increased but non-casein nitrogen and filterable nitrogen decreased. 2. Calcium content decreased gradually from 119.8 mg/100 g of raw milk to 75.75 mg/100 g at $145^{\circ}C$ as the heat treatment increased and vitamin C decreased rapidly from 1.37 mg/100 ml to 0.82 mg/ 100 ml while artificial digestibility increased from 14.07% of raw milk to 26.0% as the heat treatment increased. 3. As the heat treatment increased, microorganism counts decreased to $0.5{\times}10^2/ml$ and were not found above $135^{\circ}C$ - coliforms and psychrotrophic bacteria from $100^{\circ}C$ thermoduric bacteria, thermophiles, mould and yeast from $125-130^{\circ}C$. Heat treatment above $135^{\circ}C$ showed 100% sterilization effect. 4. The result of preservation test for heat treated milk did not show any significant changes in titratable acidity and general composition at $4^{\circ}C$, $25^{\circ}C$ and $37^{\circ}C$ up to 15 days. Viable bacteria counts, coliforms and psychrotrophic bacteria were not found but loss of vitamin and increase in viable bacteria counts appeared after 20 days.

  • The raw milk produced in Korea was heated at $70^{\circ}C$, $75^{\circ}C$, $80^{\circ}C$, $85^{\circ}C$, $90^{\circ}C$, $95^{\circ}C$ and $100^{\circ}C/15sec.$. The changes in chemical composition and microbiological aspects of the milk were summarized as following results: 1. In high temperature pasteurized milks as the heat treatment increased, pH value decreased but protein, fat, lactose and ash did not show significant changes in their contents while casein nitrogen and non-protein nitrogen increased but non-casein nitrogen and filterable nitrogen decreased in their contents. 2. Calcium content of raw milk decreased from 119.79mg/100g to 111.86mg/100g at $75^{\circ}C$ and to 106.24mg/100g at $100^{\circ}C$. Vitamin C decreased from $1.37mg/100m{\ell}$ of raw milk to $1.15mg/100m{\ell}$ at $75^{\circ}C$ and $0.94mg/100m{\ell}$ at $100^{\circ}C$. Artificial digestibility increased as the heat treatment got higher. 3. Viable bacteria counts decreased from $9.0{\times}10^3/m{\ell}$ at $75^{\circ}C$ to $3.4{\times}10^2/m{\ell}$ at $100^{\circ}C$. Coliforms were not found at $70^{\circ}C$ and thermoduric bacteria, thermophiles, psychrotrophic bacteria, mould and yeast decreased rapidly as the heat treatment increased. 4. The results of Keeping quality test for high temperature pasteurized milk showed that the' milks preserved at $25^{\circ}C$ and $37^{\circ}C$ were clotted just after 1 day but the milk preserved at $4^{\circ}C$ showed good shelf life which did not have any deterioration in titratable acidity, microorganisms and com positions.

Changes in Various Quality Characteristics of Short-ripened Salami During Storage at Chilled or Room Temperatures (단기숙성 살라미의 냉장 및 실온저장 중 품질 특성 변화)

  • This study investigated the microbial contamination levels of raw meats used for short-ripened salami and changes in the microbial and physico-chemical properties of the product during storage at 10 and $25^{\circ}C$ for up to 120 days. The microbial counts of raw meats ranged between 2 and 4 Log CFU/g. Frozen-thawed sow meat showed higher total aerobe and Enterobacteriaceae counts than fresh chilled pork and pork back fat. Staphylococcus aureus was found in all raw materials except fresh chilled pork samples, and Clostridium perfringens was detected in a sample stored for 21 days at $25^{\circ}C$. The counts of total aerobes, lactic acid bacteria and Staphylococcus spp. decreased more rapidly at $25^{\circ}C$ than at $10^{\circ}C$ when the storage time was extended. The growth of Enterobacteriaceae, Pseudomonas spp., Clostridium spp., yeast, and mold were restricted to levels below 2 Log CFU/g during storage. The contents of salt, water, crude protein, crude fat, and ash of salami samples were 3.4, 33.4, 30.8, 32.7, and 4.3%, respectively, which were not affected by storage time or temperature. The pH value of the salami was initially 4.79 and increased to 5.02 and 5.26 after 120 days of storage at 10 and $25^{\circ}C$, respectively, whereas the water activity values decreased from an initial value of 0.91 to 0.90 and 0.88 after 120 days at 10 and $25^{\circ}C$, respectively. The TBA and VBN values increased slowly during storage. The redness value of the salami samples stored at $25^{\circ}C$ decreased more significantly than the samples stored at $10^{\circ}C$. With increased storage time, the values for the rheological characteristics of the salami in terms of hardness, brittleness, elasticity, cohesiveness, gumminess, and adhesiveness tended to decrease more remarkably at $25^{\circ}C$ than at $10^{\circ}C$. Based on sensory evaluation scores, it appears that short-ripened salami is no longer acceptable after 90 days at $10^{\circ}C$ and 30 days at $25^{\circ}C$.

Effects of Different Cultivars and Milling Degrees on Quality Characteristics of Barley Makgeolli (보리의 품종 및 도정률이 막걸리의 품질 특성에 미치는 영향)

  • The purpose of this study was to evaluate the effects of cultivar and milling degree of barley on quality characteristics of Makgeolli as well as compare differences between barley Makgeolli and rice Makgeolli. Saessal-bori groups (Ss-4, Ss-12, and Ss-18) showed dry lees contents of 92.3 g, 69.4 g, and 63.8 g, respectively, whereas Huinchalssal-bori groups (Hcs-6, Hcs-14, and Hcs-20) showed contents of 62.3 g, 42.2 g, and 32.2 g. There were significant differences in quality characteristics between milling degrees and cultivars (P<0.05). The moisture, ash, crude protein, and crude fat contents of raw materials decreased with elevated milling degree. Especially, ash content of raw materials had a direct effect on Makgeolli. The pH, total acidity, and amino acidity milling with elevated decreased degree. There were no differences in total sugar or alcohol content in Makgeolli according to milling degree of barley, whereas there were significant differences between cultivars. Barley Makgeolli showed total sugar and alcohol contents of $10.7{\sim}11.8^{\circ}Brix$ and 14.07~15.07%, respectively, which were significantly lower than $12.0{\sim}12.2^{\circ}Brix$ and 17.27~17.77% in rice Makgeolli (P<0.05). Differences in colors of raw barley according to milling degree had effects on chromaticity of Makgeolli; as milling degree increased, L and b values increased. Lactic acid bacteria counts were 7.21, 6.99, and 6.67 log CFU/mL in Ss-4, Ss-12, and Ss-18, respectively, as well as 6.14, 5.39, and 5.65 log CFU/mL in Hcs-6, Hcs-14, and Hcs-20, which suggests significant reductions with increased milling degree (P<0.05). The same trend was observed in yeast as a key quality of Makgeolli. Suspension stability differed depending on milling degree, so it is expected that suspension stability can be improved by adjusting milling degree.

Study on the Quality of Market Dahi Compared to Laboratory Made Dahi

  • Dahi is yoghurt like fermented dairy products available in Bangladesh made by traditional ways from raw milk. The aim of the present study was to check and compare the present quality status of the market dahi in reference to laboratory made dahi. For this, dahi was prepared in the laboratory (A) and collected from Kishoregonj district (B), Gazipur district (C), Bogra district (D) and Dhaka district (E and F). Consumer acceptance quality was judged on 100 point by a judge panel and chemical and microbiological quality test were done by established methods. 'A' type dahi was superior followed by E, D, F, B and C considering the consumer acceptance quality parameter. From chemical test, it appears that, B possess the lowest pH ($3.75\;{\pm}\;0.05$) and highest was in C ($4.46\;{\pm}\;0.15$). Total solids content of F type dahi ($318.40\;{\pm}\;4.44\;g/kg$) was highest and fat ($52.00\;{\pm}\;2.00\;g/kg$), protein ($44.33\;{\pm}\;2.00\;g/kg$) and ash ($10.76\;{\pm}\;0.31\;g/kg$) content were also higher in F type dahi. E and A type dahi were also with considerable figure in chemical quality parameter. Dahi B was inferior in respect of microbiological quality- total viable count (TVC) (${\times}\;10^5\;cfu/ml$) content was $94.00\;{\pm}\;4.58$, yeast (cfu/ml) content was $183.33\;{\pm}\;15.28$ and mold (cfu/ml) content was $53.33\;{\pm}\;15.28$. The TVC (${\times}\;10^5\;cfu/ml$), yeast (cfu/ml) and mold (cfu/ml) content of A type dahi were $39.67\;{\pm}\;4.51$, $50.00\;{\pm}\;10.00$ and $20.00\;{\pm}\;10.00$, respectively. Considering all the three major quality aspects dahi A was superior than others followed by F, D, E, C and B.

Evaluation of Microbially Ensiled Spent Mushroom (Pleurotus osteratus) Substrates (Bed-Type Cultivation) as a Roughage for Ruminants (균상재배 느타리버섯부산물 생균발효사료의 반추동물 조사료원으로서의 가치 평가)

  • An in situ ruminal disappearance trial and an in vivo sheep metabolism trial were conducted to evaluate the nutritional value of spent mushroom substrate (SMS, originated from Pleurotus osteratus bed-type cultivation). The raw SMS was ensiled (ESMS) for 30 days with 5% (w/w, DM basis) molasses, 0.5% (v/w) yeast (Saccharomyces cerevisiae) and 0.5% (v/w) lactic acid bacteria (Lactobacillus plantarum). Two ruminally cannulated Holsteins (average BW 620 kg) were used to evaluate in situ disappearance. Six sheeps (average BW 48 kg) were fed, in $3{\times}3$ Latin square design, rice straw alone (Control), 25% (ESMS-25) and 50% (ESMS-50) of rice straw were replaced with ensiled SMS. For an in situ trial, ruminal DM and neutral detergent fiber (NDF) disappearance of SMS were increased after ensiling (P<0.05). For a sheep trial, protein and fiber (NDF, acid detergent fiber, crude fiber) digestibilities were decreased (P<0.05), crude ash digestibility was increased (P<0.001), and nitrogen retention was not affected (P>0.05) as rice straw was replaced with ensiled SMS. Ruminating time was decreased by an average of 28% by feeding ensiled SMS (P<0.05). Ensiled SMS (Bed-type cultivation) had 76% of energy value of rice straw. Consequently, ensiled SMS (Bed-type cultivation, 100% cotton waste) could be used as a roughage source appropriate for maintenance type rations for ruminants.

Quality Characteristics of Beef Rib Aged in Fermented Sauce Prepared by Zygosaccharomyces rouxii Cultivation (Zygosaccharomyces rouxii를 이용하여 제조한 발효양념으로 숙성시킨 소 갈비육의 품질 특성)

  • This study was performed to evaluate the effect of the fermented beef rib sauce (FBS) on the change of quality characteristics and sensory properties of aged and seasoned beef rib with FBS. The FBS was manufactured by fermentation with Zygosaccharomyces rouxii Y-80 yeast in raw sauce ingredients. The decreasing changes of pH of seasoned beef rib with FBS showed a similar pattern as compared with seasoned beef rib by non-fermented beef rib sauce (NFBS) during aging periods. The cooking loss of the seasoned beef rib with FBS and NFBS exhibited minor changes, respectively. Water holding capacity of the seasoned beef rib with FBS was nearly unchanged, but that of the seasoned beef rib with NFBS increased from 83% to 91% after 5 day aging. Shear forces of the seasoned beef rib with FBS were lower than those of the seasoned beef rib with NFBS. The yellowness and lightness of the seasoned beef rib with FBS decreased as compared with the seasoned beef rib with NFBS, but redness of the seasoned beef rib with FBS was more stable than that of the seasoned beef rib with NFBS. The sensory evaluation indicated that color, aroma and overall acceptability of the seasoned beef rib with FBS were scored relatively higher than those of the seasoned beef rib with NFBS.

Two-Stage Fed-Batch Culture of Candida magnoliae for the Production of Erythritol using an Industrial Medium (산업용 배지를 이용한 Candida magnoliae의 2단계 유가식 배양에서 에리스리톨의 생산)

  • Experiments were carried out to select an industrial nitrogen source and optimize erythritol production by Candida magnoliae in fed-batch culture. Among the industrial nitrogen sources tested, light steep water (LSW) was found to be the best nitrogen source for producing erythritol, based on erythritol yield and raw material price. The maximum erythritol concentration obtained a 131.6 g/L, with a 52.6% yield and 0.52 g/L-hr productivity from a 250 g/L glucose and 43.3 mL/L LSW in batch culture. Two-stage fed-batch culture was chosen to improve the volumetric productivity and the yield of erythritol. High cell density culture in cell growth stage was achieved by batch type culture containing 100 g/L glucose and 500 mL/L LSW. The cell concentration was 71.0 g/L after 23 hours of culture. Erythritol productivity was decreased by increasing glucose concentration in the production stage. But 37.3% of the maximum erythritol yield was obtained with 185.5 g/L of erythritol and 1.66 g/L-hr of productivity when 820 g of glucose powder was directly added to a concentration of 450 g/L glucose in production stage.

Changes in Quality Characteristics and Antioxidant Activity of Bitter Melon (Momordica charantia L.) Pickle during Ageing (여주 피클 숙성 중 품질 특성 및 항산화 활성 변화)

  • Bitter melon (Momordica charantia L.) has various biological functions, including anti-diabetic, anti-cancer, anti-inflammatory, anti-viral, and antioxidant activities. However, the development of foods using bitter melon (BM) is unexplored due to its bitter taste. In this study, BM pickle was prepared, and changes in quality characteristics and antioxidant activity during ageing were determined. After 4 weeks of ageing, pH levels, salinities, soluble solids, and reducing sugars of BM pickle decreased, whereas acidities increased. In addition, total viable, lactic acid bacteria, and yeast viable cell numbers greatly increased until 1 week, after which they slightly decreased. Levels of soluble phenolics increased during ageing, antioxidant activity increased accordingly. Crude protein, K, and P contents of pickles were lower than in raw material of BM, whereas crude fat and Na contents increased. Corn silk extracts were added to BM pickle during ageing in order to improve the bitter taste and flavor. The best sensory quality was obtained by adding 0.25% corn silk extracts.
