ULTRASTRUCTUAL ANALYSIS OF THE FIBROUS LAYER OF CONDYLE IN THE RAT TEMPOROMANDIBULAR JOINT WITH AGEING (가령에 따른 흰쥐 하악과두 섬유층의 미세구조 및 교원원섬유의 변화)

  • The fibrous layer of mandibular condyle of the neonatal, 1-, 7-, 14-, 27-, 55-days and 1 year old rats were examined in the electron microscope with particular attention to the ultrastructure and diameter of collagen fibrils. In the 1-day rats, most of the cells of the fibrous layer were undifferentiated mesenchymal cells and fibroblasts with rough a little developed rough endoplasmic reticulum(RER) and golgi apparatus(GA). In 7-, 17 and 27-days old rats, most of the fibroblast showed well developed GA and RER with widely distended cisternae containing granular materials. In many of these cells contained intracytoplasmic filaments among the cytoplamic organelle. In 55-day and 1-year old rats, three types of cells were observed, ie, cells containing well developed cytoplasmic organelle presumed to be involved in the collagen fibril synthesis, cells showing well developed lysosomes, golgi apparatus, mitochondria and short cytoplasmic process presumed to be involved in the active resorption of the injured collagen fibrils or cellular debris, cells containing many intracytoplasmic filaments and a little organelle presumed to be cells of inactive state. The average diameters of collagen fibrils were similar in 1- and 7-day old rats as $38.48{\pm}3.81nm$, $38.06{\pm}3.86nm$. That was thickest in 14 days old rats as $50.21{\pm}3.93nm$ among experimental groups. They were gradually thinner in 27-, 55-day rats as $40.05{\pm}2.52nm$, $43.63{\pm}1.20nm$ and thinnest in 1-year old rats as $37.38{\pm}2.17nm$. The distribution pattern of diameters of collagen fibrils were unimordal with peak of 30-60nm in rats from 1-day to 17-day old. With aging from 27-day to 1 year old rats, collagen fibril diameters showed wide distribution pattern and percentage of thin collagen fibrils increased. These results may show the functional adaptation of fibrous layer of mandibular condyle to the increased mechanical forces with aging.

THE REACTION OF BONE REGENERATE TO THE VARIOUS FORCE RATIO AND PERIODS ON DISTRACTION OSTEOGENESIS WITH COMBINED DISTRACTION FORCE AND COMPRESSION FORCE (수축력과 신장력을 병용한 골신장술에서의 다양한 힘의 비와 부여시기에 따른 신연골 반응)

  • The purpose of this study was to identify the effectiveness of the modified distraction osteogenesis (DO) method with the concept of overdistraction and compression stimulation which have been previously suggested by the authors in 2002 and to explore the optimal distraction-compression ratio and appropriate latency period for the compression force application during consolidation. The experimental specimens were assessed with radiography, histologic findings, and dual energy x-ray absorptiometry (DEXA) after the conventional DO method and the modified DO technique had been applied on rat mandibles. Total 60 rats were used for the study. In experimental group of 54 adult rats, mandibular osteotomies between the first and second molar areas were performed and customized external distractors were applied. The surgeries on 6 rats of control group also were done with same osteotomy technique and DO device application. Final amount of distraction was set-up as 2 mm on both groups. But, in a experimental group of 54 rats, distraction osteogenesis with a compression force were performed with the different distraction-compression ratio and variant latency periods for compression. The three ratio-subgroups were made as distraction 4 mm group with compression 2 mm, distraction 3 mm group with compression 1 mm, and distraction 2.5 mm group with compression 0.5 mm. In addition, The three subgroups with 3, 7, 11 days latency period prior compression were allocated on each ratio-subgroups. Total 9 subgroups consisted of 6 rats on each subgroup. In control group of 6 rats, conventional distraction technique were routinely performed. The rats of control groups were sacrificed on postoperative 3, 6 weeks after 2 mm distraction. The rats in the experimental groups also were sacrificed on the same euthanasia days of control groups to compare the wound healing. Final available specimens were 55 rats except 5 due to osteomyelitis, device dislodgement. Distraction-compression combined group on 6 weeks generally had showed increased bone mineral density than the same period group of conventional distraction technique on the DEXA study. More matured lamellar bone state and extended trabecular pattern in the combined group than those of control group were also observed in the histologic findings on 6 weeks. In the distraction-compression combined groups, the bone density of 2.5 mm distraction subgroups with 0.5 mm compression showed the highest value on the DEXA study among various force ratio groups. In the distraction-compression combined groups, the bone density of 3 day latency period subgroups showed the highest value on the DEXA study among various latency period groups for the compression application. From this study, we could deduce that 1/5 force ratio for the compression versus distraction, 3 day latency period prior compression application would be the most effective condition if modified distraction osteogenesis technique might be applicable. The modified DO method with a compression force may improve the quality of bone regenerate and shorten total treatment period in comparison with conventional DO technique clinically.

Biochemical and Nutritional Effects of Rice, Barley, and Millet on Rats (백미대맥(白米大麥) 및 속(粟) 사료(飼料)가 실험용(實驗用) 백서(白鼠)에 미치는 영양학적(營養學的) 및 생물화학적(生物化學的) 연구(硏究))

  • Dividing into six(6) experimental groups; Stock Diet(SD) Group, Rice Diet(RD) Group, Rice Diet supplemented with Barley(RD-BD) Group, Barley Diet(BD) Group, Rice Diet supplemented with Millet(RD-MD) Group, and Millet Diet(MD) Group, biochemical and nutritional effects of rice, barley, and millet on rats are studied. The grains used in this experiment are polished ones and purchased from market. Six(6) weaning male albino rats are housed into the individual cages in each group and fed with each experimental diet and water ad libitum for 21 weeks. The composition of the experimental diets are shown in Table 1. At the end of experiment, the appearences of rats are observed and the bloods are sampled by heart puncture method following ether anesthesia for determinations of serum protein and serum cholesterol. And the livers are sampled immediately right after heart after heart puncture for determinations of liver weight, liver nitrogen, liver fat, and liver glycogen. And also liver tissues are observed histologically. The result of growth experiment is shown in Table 2 and Figure 7. The results of blood analysis are shown in Table 3 and the results of liver analysis are shown in Table 4. And the histological observations of liver tissue are shown in Figures 1, 2, 3, 4, 5, and 6. According to the above results, it could be summarized as follows; 1) The normal appearences are observed at SD Group and BD Group. Alopecia is observed at RD, RD-­BD, RD-MD, and MD groups and the wet appearences of hair are observed at RD-MD and MD groups. The color of hair is changed to yellowish white at RD, RD-MD groups. The maximum growth inhibition, is observed at MD group and the growths of RD, RD-BD, BD, and RD-MD groups are significantly lower than that of SD group. The growth of RD group is lower than those of RD-BD and BD groups, though this difference is not significant. 2) At the blood analysis, the highest serum protein content is shown at SD group and the lowest serum protein is observed at MD group. The serum cholesterol contents are not significantly different among the six(6) experimental groups. 3) The highest liver weight is observed at SD group and the lowest weight is shown at MD group. However, oppositely the highest liver weight per 100 grams of body weight at MD group and the lowest liver weight per 100 grams of body weight at SD group are observed as shown in Table 6. 4) The differences of liver nitrogen contents of RD, RB-BD, BD, RD-MD, and MD Groups are not significant except SD group showing the highest content. 5) The significant highest content of liver fat is observed at RD group and the lowest content of liver fat is shown at SD group. And no significant differences in contents of liver glycogen are observed among the six(6) experimental groups. 6) No significant abnormal histological changes of liver tissue are observed at SD, RD-BD, and BD groups. However, the fat metamorphosis at peripheral zone of liver tissue is observed at RD Group and the some histological abnormal changes are observed at RD-MD and MD groups. According to the above summaries, the followings might be estimated and recommended; 1) The BD group shows histological normality of liver tissue, improvement of liver fat and liver nitrogen, normality of appearence, and better growth compared with RD Group. This could be indicated that barley is nutritionally and biochemically better than rice as the feeding diet on rat. And it might be considered that there might be the species speciality for nutritionary evaluation of food. 2) Recently, Marshall indicated that the high consumption of carbohydrate causes fatty liver and Kondo reported that the higher consumption of rice causes the more heart disease and the shorter life span. Considering these reports, the higher consumption of rice as a main dish in our country should be considered as an important national nutritional problem. And this might be improved by supplementing with barley or other grains to rice.

Effect of Cynanchi wilfordii Radix Extracts on Lipid Compositions and Blood Pressure in Spontaneously Hypertensive Rats (백하수오(Cynanchi wilfordii Radix) 추출물이 선천성 고혈압 흰쥐의 지질성분 및 혈압에 미치는 영향)

  • The purpose of this study was to observe the effects of Cynanchi wilfordii Radix extracts (CRE) on the improvement of the lipid compositions and blood pressure level in spontaneously hypertensive rats (SHR) fed an experimental diet for 5 weeks. The rats were divided into 3 groups: a control group, a 0.5% CRE treated group, and a 1% CRE treated group. Consumption of CRE extract for 5 weeks in SHR significantly suppressed blood pressure rise with aging (p<0.05). After eating the experimental diets, the triglycerides in serum was significantly lower in the CRE group than that in the control group. The fasting glucose levels of the 0.5% and 1% CRE group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR (HDL-cholesterol/total cholesterol) of the SHR in the 0.5% and 1% CRE diet were significantly increased compared to the control diet. Thus, long term consumption of CRE might be beneficial in lowering high blood pressure and the improvement of lipid metabolism in SHR rats.

  • Objectives: To find out the effects GGTl, an antiobestic drug widely used in clinics, has on the blood-antiobestic index and the toxicity index using the data from the hGHTg obese male rats. We looked closely into both of the two indices because GGTl antiobestic effect can happen not only by pharmacological action, but also by its toxicity. Also, we verified the difference in effect between GGTl and reductil (sibutramine), which has been approved by the FDA of the United States. Methods: After performing the experiments for 8 weeks on the hGHTg obese male rats divided into three groups: the control group, the GGTl group, and the reductil (RD) group, we anesthetized the rats with Diethyl ether and took a 3ml blood sample from the heart. Then, after coagulating the blood in room temperature by using the plasma separator, we centrifuged it for 25 minutes in 3,000rpm using the high-speed refrigerated centrifuge. We kept the separated plasma in a deep freezer at $-80^{\circ}C$, and repeatedly measured the antiobestic index and the toxicity index twice using the hematology biochemistry analyzer. Also, in order to judge the indirect toxicity index, we separated liver from kidney and observed them. Results: When we looked at the results of the analysis of covariance on the measuring elements related to the antiobestic index (TC, HDL, LDL, TG, and GLU), there was no significant difference among the groups in all measuring elements. Also, the results of the analysis of covariance on the two roups (RD group and GGTl group) showed that the p-values had no significant difference under the level of significance 0.05. When we looked at the result of the analysis of covariance on the measuring elements related to the toxicity index (GOT, GPT, GGT, CREA, UA, ALB, and TP), we could see that the p-values in GPT, ALB, and TP have a significant difference among the groups. Also, the results of the analysis of covariance about the measuring elements related to the toxicity index on both groups, RD group and GGTl group, showed no significant difference in the p-values of all of the measuring elements in the two groups, RD and GGTl group. Conclusions: In conclusion, through this experiment, the safety of GGTl has been approved, and although the verification on its medical effect has not been clearly approved, when we consider the fact that it belongs to the same group as reductil, an antiobestic drug approved by the FDA of the United States, we could indirectly verify that GGTl has an antiobestic effect. We believe that when doing a sample design for a future experiment, it needs to be performed on a greater sample size based on the power analysis that needs to be performed primarily in experiments, and a more accurate verification is needed through more systematic experiment plans.

Effect of Peking-Duck By-Product Extracts Supplemented with Medicinal Herbs on Serum Heavy Metal Levels and Blood Parameters of Rats Exposed to Lead and Mercury (한약재를 첨가한 오리부산물 추출액이 납과 수은에 노출된 흰쥐 혈청의 중금속 및 혈액지표에 미치는 영향)

  • This experiment was planned to develop a functional supplement by food resources to prevent and lessen the deleterious effects caused by environmental pollutants such as polluted food, air, water and heavy metals. The goal of this study was to investigate the effects of peking-duck extracts supplemented with six kinds of medicinal herbs (DJ) on the intoxication of lead and mercury in rats. Sprague-Dawley rat weighing $150g\pm15g$ g, were randomly assigned to 5 groups, basal diet only (NCG), heavy metal without DJ injection (HCG), heavy metals and DJ (3 mg/mL) injection (HMLD), heavy metal and DJ (30 mg/mL) injection (HMMD), heavy metal and D] (300 mg/mL) injection (HMHD). Mecury (Hg) and lead (Pb) injected at the level of 50 ppm for 17 days. Also DJ oral feeding was continued for 31 days. The result of this study were as follows; Food intake and body weight gain in heavy metal administered groups were lower than those of control group (NCG). The activities of GOT, GPT and BUN level were significantly reduced in DJ-treated groups as compared to HCG. DJ was shown to suppress the accumulation of Hg and Pb in serum. The results suggest that DJ might have protective effect on Hg and Pb intoxication.

Effect of γ-PGA (Poly-γ-Glutamic Acid) Supplement on Calcium Absorption and Bone Metabolism in Rats (γ-PGA(Poly-γ-glutamic acid) 보충이 흰쥐의 칼슘 흡수율 및 골대사에 미치는 영향)

  • This study was Conducted to investigate the effect of ${\gamma}-PGA\;({\gamma}-poly\;glutamic\;acid)$ on Ca absorption and bone metabolism in rats. Weaned 4-week old male rats were fed Ca-deficient diets for 3 weeks after the adjustment period. Rats were divided into 6 groups and were fed experimental diets for four weeks. Experimental groups were basal (Ca deficient), control (Ca diet: Ca 0.45%), CP1(Ca 0.45%+casein phosphopeptide 1%), PG1(Ca 0.45%+gamma poly glutamic acid 1%), CPG (Ca 0.45%+casein phosphopeptide 1%+gamma poly glutamic acid 1%) and PG3(Ca 0.45%+gamma poly glutamic acid 3%). Though daily Ca intake and food intake of experimental groups showed no significant difference that of control group. The values of fecal Ca excretion and urinary Ca excretion in groups fed ${\gamma}-PGA$ were significantly lower than that in tile control group. The values of Ca absorption in groups fed ${\gamma}-PGA$ were significantly higher than that in the control group. The levels of femur Ca in ${\gamma}-PGA$ supplemented group were significantly increased compared to the control group. Also, breaking force of femur in ${\gamma}-PGA$ supplemented group showed about 40% increase compared to the control group. These results show that ${\gamma}-PGA$ supplement could be helpful to increase Ca absorption as well as to intensify the femur strength and to increase the Ca content of femur in rats.

Effect of Meal Pattern on Lipogenesis and Lipogenic Enzyme Activity in Rat Adipose Tissue Fed High Fat Diet (식이급여형태가 고지방식이를 급여한 흰쥐 지방조직의 지방합성 및 지방합성 효소활성에 미치는 영향)

  • This study was undertaken to investigate the effects of meal pattern on lipogenesis and activities of lipogenic enzyme in rats epididymal and mesenteric adipose tissues. A high fat diet was fed either ad libitum or in 1 meal during the last 3 h of the dark cycle for 4 weeks. Lipogenesis was measured as glucose conversion to total lipid and activities of glucose-6-phosphate dehydrogenase (G6PDH), 6-phophogluconate dehydrogenase (6PGDH) and NADP-malate dehydrogenase (ME) were determined by measuring NADPH production. Lipoprotein lipase (LPL) activity and serum lipoprotein concentrations were also measured. Meal-fed (3 h) rats had a decreased food intake, body weight and carcass fat compared with rats fed ad libitum. The serum triglyceride concentration of meal-fed rats tended to be higher than that of the ad libitum rats. However, there were no differences between meal-fed group and ad libitum group in serum concentrations of HDL-cholesterol, LDL-cholesterol and total-cholesterol. Rates of lipogenesis in both epididymal and mesenteric adipose tissues were significantly higher in the meal-fed group than that in the ad libitum group. In addition, meal-fed group showed higher G6PDH, 6PGDH and LPL activities in both epididymal and mesenteric adipose tissues, but exerted no significant effect on ME activity. These results suggest that meal-fed rats compared with ad libitum rats have marked lipogenic capacity, although such elevation probably does not result in increase in carcass fat concentration. Thus, meal-fed diet can be an important determinant of the alterations in adipose lipid metabolism.

Effects of Dietary Protein and Energy on the Growth and Body Composition of Growing Rats (단백질과 에너지 수준이 흰쥐의 성장 및 체조성에 미치는 영향)

  • In order to investigate the effect of dietary protein and energy on growing female and male rats, Sprague-Dawley 90 female rats and 54 male rats of 3 weeks old weighing approximately 70-80g and 65-75g, respectively, were subjected to feeding trials for 8 weeks and then subsquently to metabolic trials for 2 weeks. Three dietary energy levels (3200, 3600, 4000 kcal ME/kg) were employed and each energy level contained three protein levels (15, 25, 35% of 3600 kcal ME/kg) and three fat levels (10, 20, 40% of 3600 kcal ME/kg) by addition of an appropriate amount of carbohydrate and the following results were obtained. The body weight gain of female rats was highest for LPHE ration but that of male rats was highest for LPME ration. The weight gains both of female and male rats were not affected by the level of protein. Food efficiencies both of female and male rats was affected by the level of protein, whereas that of male rats was not. Protein efficiencies of female and male rats were highest at low protein level and tended to decrease as the level of protein increased, but that of female rats was highest at high energy level, while that of male rats was highest at medium energy level. The analysis of the body composition after feeding trials for 8 weeks has shown that the contents of body water and protein were not affected by protein level both in female and male rats. The content of body fat increased remarkably as the protein and energy levels increased in case of female rats, but it was not affected by the protein and energy levels in case of male rats. From the above-mentioned experimental results it may be con eluded that the best formula of diet of growing female rats may be composed of low protein (13%) and high energy levels (4000 kcal/kg) whereas that for male rats may be composed of low protein (13%) and medium energy levels (3600 kcal/kg), since all the efficiencies of food, protein and energy have shown to be best at these levels.

Effect of treadmill exercise on autophagy related protein expression in the cardiac muscle of high-fat diet fed rats (트레드밀 운동이 고지방 식이 쥐 심근세포의 자가포식 관련 단백질 발현에 미치는 영향)

  • The purpose of this study was to investigate the influence of obesity on the expression of autophagy-related proteins in cardiac muscle. To this end, obesity was induced in rats through 20 weeks of high-fat diet, and the animals were then subjected to 8 weeks of treadmill exercise. Subsequently, the expression of proteins that regulate the induction of autophagy, formation of autophagosome, and fusion of autophagosome and lysosome was confirmed. Obesity was induced in the experimental animals (SD rats) through 20 weeks of high-fat diet (carbohydrate: 20%, fat: 60%, and protein: 20%), and they were subsequently subjected to 8 weeks of treadmill exercise (5 days/week, 30 min/day, 5 minutes; 8m/min, 5 minutes; 11m/min, 20 minutes; 14m/min). The experimental groups comprised the normal diet control group (ND-CON, n=10), high-fat diet comparison group (HFD-CON, n=10), and high-fat exercise group (HFD-TE, n=10). Oral glucose tolerance test was conducted before and after 8 weeks of treadmill exercise, and the area under the curve (AUC) was calculated. Through fasting insulin and fasting glucose levels, HOMA-IR, which is an index of insulin resistance, and abdominal visceral fat/body weight (AVF/BW) were calculated for comparison. Moreover, autophagy-related proteins were analyzed from cardiac tissue to investigate the effects of exercise training. Obesity was successfully induced in the HFD-CON group through long-term high-fat diet, and the HFD-CON group had higher body weight, AUC, HOMA-IR, and AVF/BW compared to the ND-CON group. The HFD-TE group, which underwent 8 weeks of treadmill exercise, showed improvements in AUC, HOMA-IR, and AVF/BW. Although the body weight tended to decrease as well, there was no statistically significant difference. mTOR and AMPK, which are involved in the induction of autophagy, both decreased in obesity but increased upon exercise. Beclin-1, BNIP3, ATG-7, p62, and LC3, which are related to the formation of autophagosomes, all increased in obesity and decreased after exercise. Cathepsin L and LAMP2, which regulate the fusion of autophagosome and lysosome, both decreased in obesity and increased upon exercise. Physical activity, including treadmill exercise, was found to induce normal autophagy and improve pathological phenomena observed in metabolic diseases. Therefore, the findings suggest the need to consider treadmill exercise as a primary means to achieve effective prevention and treatment of cardiac diseases.
