This study was carried out of to determine effects of high voltage pulsed galvanic stimulation on the soleus. target muscle of the sciatic nerve, of white rat two condition. The coditions included normal, and denervated muscle. The histochemical, ultrastructural observations were made. The following results were obtains. 1. The histochemical observations found the inflammatory cells between muscle bundle and muscle fiber since 1-week control group. In addition, nucleus located in the muscle fiber was frequently observed. 2. The experimental group showed a similar phenomenom to the normal muscles in terms of glycogen granules in the 1-week group, where as fiber were not distinguishable in4-weeks group which indicated that the degenerative changes had occured. 3. The NADH-TR reaction showed that the red muscle slightly increased in the 2-weeks group, and the distinguished was impossible the red fiber 4. The ultrastructures of the muscles in both groups were severely bend, and a number of vacuoles were observed due to the destruction of mitochondria..

The effects of insulin and exercise on glucose uptake of skeletal muscle were investigated in soleus muscle isolated from low dose streptozotocin induced diabetic rats in vitro. Glucose uptake was assessed by measuring $^3H$-methylglucose uptake in vitro. Basal glucose uptake in diabetes was reduced by approximately one-third of the control value($5.6{\pm}0.73{\mu}Mol$/g/20min. in diabetes versus $8.4{\pm}0.77$ in control, P<0.01). There was also a significant decrease(P<0.01) in glucose uptake of diabetes at physiologic insulin concentration ($200{\mu}IU$/ml) by 40% ($6.1{\pm}1.20$ versus $10.0{\pm}0.81$). Furthermore, maximal insulin($20000{\mu}IU$/ml)-stimulated glucose uptake was 36% lower in diabetes as compared with control($7.3{\pm}1.29$ versus $11.4{\pm}1.29$, P<0.01). In contrast, exercise(1.0km/hr, treadmill running for 45min.) effect on glucose uptake was so dramatic in diabetes that glucose uptake at basal state was 8.4+1.09 and insulin stimulated-glucose uptake were $10.2{\pm}1.47$ and $11.9{\pm}1.64$, in 200 and $20000{\mu}IU$/ml added insulin, respectively. These results suggest that insulin insensitivity develops in skeletal muscle after 2 weeks of streptozotocin-induced diabetes, but these insensitivity was recovered significantly by single session of running exercise.
Purpose: The purpose of this study was to compare the effects of anorexia induced by consecutive low-dose and high-dose of cisplatin (CDDP) on the hindlimb muscles of rats. Methods: Male Sprague-Dawley rats were assigned to three groups: Control group (C) received a saline (the same dose and duration as the low CDDP group), the high-dose cisplatin (High CDDP) group received a single 5 mg/kg dose of cisplatin, the consecutive low-dose cisplatin (Low CDDP) group had 1 mg/kg of cisplatin administered for five consecutive days. On the 8th day the soleus and gastrocnemius muscles were dissected. Body weight, food intake, activity, muscle weight, Type I, II fiber cross-sectional area (CSA) of the dissected muscles were measured. Results: Body weight, food intake, muscle weight and Type I, II fiber CSA of the High CDDP and Low CDDP groups were significantly less than the C group. The High CDDP group showed significant decreases, compared to the Low CDDP group, in body weight, food intake, activity score, muscle weight and Type I, II fiber CSA. Conclusion: Hindlimb muscle atrophy occurs due to anorexia induced by both consecutive low-dose and high-dose cisplatin. The muscle atrophy induced by consecutive low-dose cisplatin is less apparent than high-dose cisplatin.
The effects of dietary supplementation of Eleutherococcus senticosus, taurine and carnitine on maximal endurance exercise performance along with other related parameters were evaluated in rats that underwent aerobic exercise training for 6 weeks. Thirty-two male rats (4 weeks old) were randomly divided into 4 groups, and fed experimental diets and/or aerobic exercise trained according to the protocol: SC (sedentary control group), EC (exercise-trained control group), EE (exercise-trained Eleutherococcus senticosus-supplemented group), and EETC (exercise-trained Eleutherococcus senticosus, taurine and carnitine-supplemented group). The food efficiency ratio of EC rats was significantly lower than the value for SC rats (p < 0.01). Exercise-trained control animals (92 $\pm$ 8.8 min) could run significantly longer until exhausted on the treadmill than sedentary control rats (11 $\pm$ 0.8 min) (p < 0.001). Animals fed an Eleutherococcus senticosus-supplemented diet, and an Eleuthherococcus sonticosus, taurine and carnitine- supplemented diet while undergoing aerobic exercise training for 6 weeks exhibited, respectively, 8 and 5 minutes longer running performance until exhausted than the rats fed the control diet. The gastrocnemius muscle glycogen concentration of the rats, measured at 48 hours post maximal exercise performance test, was 43% higher in EC rats than the value for SC rats (p < 0.05), but was not different among EC, EE, and EETC rats. The mitochondrial citrate synthase activity of the soleus muscle was significantly higher in EC rats compared to the value for SC rats (p < 0.01), and showed a tendency to increase, without statistical significance, in EE or EETC rats compared to the value for EC rats. These results indicate that aerobic exercise training for 6 weeks significantly improved maximal exercise performance, muscle glycogen content along with citrate synthase activity, which are important in the energy metabolism of muscle under aerobic exercise. Dietary supplementation of Eleutherococcus senticosus in rats while undergoing aerobic exercise training improved maximal endurance exercise performance without significantly affecting muscle glycogen content and enzyme activities involved in energy metabolism during exercise. Taurine and carnitine supplementation failed to show an additive effect on maximal endurance exercise performance when consumed along with Eleutherococcus senticosus.

The effect of persistant mild hyperglycemic hyperinsulinemia on the development of the insulin resistance in rats was studied in vivo. Also, the characteristics of the insulin resistance compared with the insulin resistance of STZ diabetic rats. Persistant mild hyperglycemic hyperinsulinemic rat model was produced by ingestion of glucose polymer for 8 days. The glucose disappearance and infusion rate was measured by hyperinsulinemic euglycemic clamp technique at steady state of blood glucose and insulin levels. The clamped level of blood glucose was 100 mg/dl, and the clamped levels of insulin were $70{\mu}U/ml$ (physiologic condition) and $3000{\mu}U/ml$ (supramaximal condition). Hepatic glucose producticon rate was calculated using measured data. And the glycogen synthetic capacity of skeletal muscle(soleus) and liver was measured after 2 hours of hyperinsulinemic euglycemic clamp study. The glucose disappearance and glucose infusion rate in glucose polymer group was decreased in the both physiological and supramaximal insulin level compared to the rate of the normal control group. The rate of STZ diabetic group wase lowest at supramaximal insulin level among two another experimental groups. The hepatic glucose production rate of glucose polymer group was decreased compared to normal control but increased in STZ diabetic group. The glycogen synthetic capacity of skeletal muscle and liver of glucose polymer group was not significantly different from normal control group, but it was markdly decreased in STZ diabetic group. These results suggest that persistant mild hyperglycemic hyperinsulinemia may induce insulin resistance, but glycogen synthetic capacity is intact.
As a central component of a novel protein kinase cascade, the activation of the mitogen-activated protein (MAP) kinase cascade has attracted considerable attention. We sought to determine the effect of exercise and diet on the activation of the extracellular-signal regulated protein kinase (ERK) 1/2 and the p38 MAP kinase pathways in rat soleus muscle. Forty-eight Sprague-Dawley rats were assigned to one of two dietary conditions: high-carbohydrate (CHO) or high-fat (FAT). Animals having each dietary condition were further divided into one of three subgroups: a sedentary control group that did not exercise (NT), a group that performed 8 weeks of treadmill running and was sacrificed 48 h after their final treadmill run (CE), and a group that was sacrificed immediately after their final routine exercise training (AE). A high-fat diet did not have any significant effect on phosphorylated and total forms of ERK 1/2 or p38 MAP kinase. In chronically trained muscle that was taken 48 h after the last training, phosphorylated ERK 1/2 significantly increased only in the FAT but not in the CHO groups. In the case of total ERK 1/2, it increased significantly for both groups. In contrast, both phosphorylated and total forms of p38 MAP kinase decreased markedly compared to sedentary muscle. In muscle that was taken immediately after a last bout of exercise, phosphorylated ERK 1/2 increased in both groups but statistical significance was seen only in the CHO group. Total ERK 1/2 in acutely stimulated muscle increased only in the CHO-AE group even though the degree was much lower than the phosphorylated status. Muscle that was taken immediately after the routine training increased in phosphorylation status of p38 MAP kinase for both dietary conditions. However, statistical significance was seen only in the CHO group owing to a large variation with FAT. In conclusion, a high-fat diet per se did not have any notable effect versus a high-carbohydrate diet on MAP kinase pathways. However, when diet (either CHO or FAT) was combined with exercise and/or training, there was differentiated protein expression in MAP kinase pathways. This indicates MAP kinase pathways have diverse control mechanisms in slow-twitch fibers.
In the present study, all of the treadmill exercise-trained SHR expressed clear adaptive changes such as reduced resting heart rate and blood pressures, LPOA, homocysteine Therefore, treadmill exercise was sufficient to induce physiological adaptation in the SHR. Endurance training is known to induce physiological cardiac hypertrophy, while hypertension induces patho logical cardiac hypertrophy that increases cardiomyocyte apoptosis. The pathological adaptation to pressure overload has also been associated with a further increase in the expression of several marker genes including cardiomyocyte ET-1 in the SHR, but not in the exercise-trained SHR. Additionally, there is an increase in the endothelial nitricoxide synthases (eNOS) protein expression of soleus, gastrocnemius, and extensor digitorum longus muscle in the exercise-trained SHR but not in the SHR in the present study. Thus, compared to pathological adaptation to pressure overload, physiological adaptation to exercise training is associated with distinct alterations in cardiac and molecular phenotypes. based on these results, exercise training improves hypertension by cardiovascular regulating genes and hemodynamic parameters.
The purpose of this study was to investigate the effects of glucuronic acid on antioxidative defense system and recovery of muscle fatigue in rat artier aerobic exercise. Sprague-Dawley male rats weighing 150 $\pm$ 10g were randomly assigned to one normal(N) group and three exercise training groups. Exercise training groups were classified into glucuronic acid free intubation group(T group), 250mg glucuronic acid/kg bw intubation group(TU group), and 500 mg glucuronic acid/kg bw intubation group(2TU group) according to glucuronic acid supplementation level. The glucuronic acids were administered to rats by oral intubation before exercise training. The experimental rats in exercise training groups(T, TU and 2TU) were exercised on glucuronic acid supplementation or rats in normal group were confined in cage for 4 weeks. And rats were sacrificed with an overdose of pentobarbital injection just after running. Liver xanthine oxidase(XOD) activities were not significantly different among four groups. The activity of superoxide dismutase(SOD) in T group was no significant difference from N group, but those of TU and 2TU groups were increased by 9% and 18%, respectively, compared with that of T group. Liver glutathione peroxidase(GSHpx) activites of T and TU groups showed a similar tendency to that of normal group, but increase 17% in 2TU group compared with normal group. The ratio of GSH/GSSG in liver of T group was lower than that of normal group, but those of TU and 2TU groups were a similar tendency to that of normal group. Contents of thiobarbituric acid reactive substance(TBARS) in T group was increased by 47%, compared with that of normal group but those of TU group and 2TU group were lower 27% and 35%, respectively, compared with that of T group. The contents of glycogen in soleus muscle significantly lower in all three trained exercise groups than that of normal group, but there were no significant differences among the trained exercise groups. Contents of hepatic glycogen in T group were decreased 27% compared with those of normal group while those of TU and 2TU groups were the same as normal group levels. The contents of serum lactic acid in T group were increased 240% of normal group, but hose of TU and 2TU groups were decreased 38%, 39%, respectively, by glucuronic acid supplementations, compared with that of T group. In conclusion, the effects of glucuronic acids in exercise training rats would appear to reduce peroxidation of tissue as an antioxidative defense mechanism and promote recovery of muscle fatigue.
This study investigated the effect of endurance exercises on blood pressure, angiogenesis, and the vascular endothelial growth factor (VEGF) expression in the skeletal muscle of spontaneously hypertensive rats (SHR). Five week old SHRs and Wistar-Kyoto rats (WKY) were randomly divided into 3 groups: Wistar-Kyoto rats (WKY, n=9), SHR control (SHR-C, n=9), and SHR endurance exercise training (SHR-E, n=9). Endurance exercise training was performed on a treadmill (12-20 m/min, 0% grade, 60 min/day, 5 days/week, 16 weeks). Systolic blood pressure was monitored with the tail-cuff method. The expression of VEGF protein and capillary density were identified using western blotting and H&E staining in the soleus muscle, respectively. Systolic blood pressure was reduced by endurance exercise in SHR ($p$ <0.05). The capillary density of skeletal muscles in SHR-C was lower than in WKY ($p$ <0.05), but it was recovered by endurance exercise training (SHR-E) compared to SHR-C ($p$ <0.05), and VEGF protein was also increased by endurance exercise training compared to SHR-C ($p$ <0.05). These data suggest that the enhancement of capillary density via an increase of VEGF expression in skeletal muscles by endurance exercise training could be an important factor to inhibit blood pressure elevation in SHR.
The purpose of this study was to investigate that the effect of dietary fatty acid composition on pro- and macro-glycogen utilization and resynthesis. The analyses were further extended for different muscle fibers (type I, type II, & type IIb) as well as tissues (i.e., liver & heart). Total one hundred sixty Sprague-Dawley rats were used, and rats were randomly allocated into four experimental groups: animals fed standard chow diet (n=40), animals fed saturated fatty acid diet (n=40), animals fed monounsaturated fatty acid (n=40), and animals fed polyunsaturated fatty acid (n=40). Animals in each groups were further divided into five subgroups: sacrificed at REST (n=8), sacrificed at immediately after 3 hr swim exercise (P-0HR, n=8), sacrificed at one hour after 3 hr swim exercise (P-1HR, n=8), sacrificed at four hour after 3 hr swim exercise (P-4HR, n=8), and sacrificed at twenty-four hour after 3 hr swim exercise (P-24HR, n=8). Soleus (type I), red gastrocnemius (type IIa), white gastrocnemius (type IIb), liver, and heart were dissected out at appropriated time point from all animals, and were used for analyses of pro- & macro-glycogen concentrations. After 8 weeks of dietary interventions, there was no significant difference in body mass in any of dietary conditions (p>.05). After 3 hr swim exercise, blood lactate level was higher compared to resting conditions in all groups, but it was returned to resting value after 1 hr rest (p<.05). Free fatty acid concentration was higher in all high fat fed groups(regardless of fatty acid composition) than CHOW consumed group. At rest, pro- & macro-glycogen concentration was not different from any of experimental groups (p>.05). Regardless of forms of glycogen, the highest level was observed in liver (p<.01), and most cases of supercompensation after 3hr exercise observed in this study were occurred in CHOW fed tissues. Except heart muscle, all tissues used in this study showed that pro- and macro-glycogen concentration was significantly decreased after 3 hr exercise. Based on these results, two conclusions were made: first, there is no different level of glycogen content in various tissues regardless of types of fatty acids consumed and second, the highest mobilization rate would be demonstrated from CHOW fed animals compare to animals that consumed any kinds of fatty acid diet if prolonged exercise is applied.
