A Study on the Immunohistology in Injury Cure of Rat by using InGaAlP Laser Diode (InGaAlP 레이저다이오드를 적용한 Rat의 착상 치유에서 면역조직화학적 연구)

  • The apparatus has been fabricated using the laser diode and microprocessor unit. The apparatus used a InGaAlP laser diode for laser medical therapy and was designed for a pulse width modulation type to increase stimulation effects. To raise the stimulus effect of the human body, the optical irradiation frequency could be set up. The study has executed in-vivo experiment by employing our own developed laser diode irradiation system to investigate the effects of the InGaAlP laser diode irradiation on the wound healing as a preliminary study aimed at the application of InGaAlP laser diode to wound healing of human skin injury. The study cut out whole skin layers of Sprague-Dawley rat on the back part in 1 cm circle and observed developing effects after executing light irradiation for 9 days, and in result it is found that the light irradiation rat showed earlier wound healing than non-irradiation rat during the experimental period. In addition, there are some differences found regarding the healing process between laser diode irradiated rats and non-irradiated ones.

Hair growth effects of Mori Cortex Radicis Mixture on the hair of rat (탈모증에 대한 상백피 복합물의 모발성장 효과)

  • This study was carried out to investigate the hair growth effect of Mori cortex radicis mixture on the hair of rat. Cytarabine(50mg/kg) was injected to eight-day-old rats for 7 days. When the mixture of Mori cortex radicis was administered to the rat by route of skin, it promoted the growth of hair. These data suggest that Mori cortex radicis mixture have an effect on the hair growth in cytarabine-induced alopecia rat.

Effect of Locally Applied Keratinase on Thickness of Rat Skin (국소도포한 각질분해효소가 흰쥐피부의 두께에 미치는 효과)

  • The aim of this study is to elucidate the effect of keratinase on epidermis of rat skin. Twenty-five male Sprague-Dolly rats were used. The hair on the back were removed and $2{\times}2cm$ area was marked. The rats were divided five groups; 1) Control group(Co), 2) Cleansing gel group(Cl), 3) Cleansing gel+keratinase group, 4) Exfoliant gel group(Ex), and 5) Exfoliant gel+ keratinase group(Ex+K). The solutions were applied to the back area twice a day for five days. On fifth day, the skins were harvested, fixed and prepared for histologic sections. The thickness of keratin layer, living epidermis, dermis, and cell layer number of living epidermis were measured. In the group containing keratinase(Cl+K, Ex+K), the thickness of keratin layer and living layer were thinner than other groups. However, there were no significant differences of the cell layer number of living epidermis and thickness of the dermis among the five groups. We think the keratinase may have the effect thinning the keratin layer as well as the thickness of living epidermis, without effecting the living cell and dermal component. The keratinase containing soap may be of benefit to remove the excess keratin layers in human.

  • True collagenolytic enzymes in animal tissues were first demonstrated by Gross and Lapiere (1962), who showed the ability of such an enzyme in the culture medium of living explants of tadpole tissue to degrade a specific substrate of undenatured collagen under physiological conditions. Recently, tumor-associated collagenolytic activity has been demonstrated in human neoplasm and in ascites V Carcinoma. This investigation have been peforme to determine whether or not a collagen lytic enzyme could e found in isolated solid Tawa sarcoma of Donryu female rat obtained the culture medium. The results were as follows. 1. 11.5mg% of hydroxyproline contained in Donryu rat skin collagen, which was extracted by 0.5M acetic acid. 2. Cultivation of solid Tawa sarcoma tissues on reconstituted rat skin collagen gels showed lysis of adjacent gel after 18 hours, and much more extensive lysis after 5 days. 3. Collagen substrate was not attacked by the common proteolytic enzymes, trypsin, pepsin, and pronase.

The Effects of Enhancers on Transdermal Absorption of Ketoprofen Packs (케토프로펜 팩제제에서 경피흡수에 미치는 투과촉진제의 영향)

  • In order to reduce the systemic side effects and gastrointestinal irritation after its oral adminitration, ketoprofen was formulated as water-soluble packs. The effects of fatty acids and fatty alcohols on the penetration of ketoprofen through excised rat skins were evaluated. The role of stratum corneum as a protective barrier was also investigated. Fatty acids and fatty alcohols were generally effective in promoting ketoprofen penetration. The flux of ketoprofen through rat skin was maximized when oleic acid or lauryl alcohol was used as an enhancer. As the concentration of fatty acids and fatty alcohols varied from 0% to 10%, the amounts of ketoprofen penetrated were in direct proportion to that of fatty acids but those had no relationship with that of fatty alcohols. The penetration of ketoprofen through stripped skin was enhanced compared to normal skin irrespective of enhancer type, which indicated that the action site of enhancers would be stratum corneum.

Healing Effect of 635 nm Laser Diode in Skin Injury (635nm 레이저다이오드의 피부 결손 치유 효과)

  • Low level laser therapy has various therapy effects. This paper performed the basic study for fabricating the low level laser therapy apparatus, and one of the goals of this paper was to make this apparatus used handily. The apparatus has been fabricated using the laser diode and microprocessor unit. The apparatus used a 635 nm laser diode for laser medical therapy and was designed for a pulse width modulation type to increase stimulation effects. In this study, the designed device was used to find out how 635 nm laser diode affected the skin injury of SD-Rat(Sprague-Dawley Rat). We divided the participants into two groups; irradiation group which was irradiated 10 minutes a day for 9 consecutive days, and none irradiation group. The results showed that the study group had lower incidence of inflammation and faster recovery, compared with the control group.

  • A new capsaicin analog modified with 4-hydroxyl and alkyl chain of capsaicin was a very potent antiinflammatory analgesic drug and may be clinically useful for those who have rheumatoid arthritis, diabetic neuropathy and cancer. The purpose of this study was to investigate histopathology after short and long term application of poloxamer-based gels, and percutaneous absorption of various topical formulations. Poloxamer-based gel was prepared by cold method using poloxamer 407. The poloxamer gels was applied to dorsal sites of hairless mouse skin during one week or one month for the evaluation of skin irritation. The applied site was then sectioned for histopathologic examination. The topical formulations were also prepared using CMC, HPMC, MC, carbopol and glycerylmono stearate. Skin variation of poloxamer gels was studied using excised hairless mouse, rat, hamster and human penis skin. Franz-type diffusion cells were used far skin penetration of drug against receptor phase filled with about 10$m\ell$ of 0.9% saline solution kept at 32$^{\circ}C$. The concentration of drug was determined by the reverse phased C18, Symmetry HPLC with fluorometeric detector. No skin erythema was observed after dorsal application of poloxamer-based gels for one week or one month. No histopathologic changes was also examined, suggesting no skin toxicity of poloxamer-based gels. The order of flux rate was HPMC > MC ( CMC > poloxamer >> glycerylmono stearate ( carbopol. There was a skin variation of poloxamer gels. The flux rate of poloxamer gels was highest in case of hairless mouse followed by rat, human and hamster skin.   • Rhamnose-rich (RROP-s) and fucose-rich (FROP-s) oligo-and polysaccharides were prepared and extensively characterised by physical and chemical procedures [1,2] and compared to L-fucose. Their biological properties were then studied on human skin fibroblast cell cultures, human skin explant cultures and on hairless rat skin, using a variety of cell-biological, biochemical and computerised morphometrical procedures. Among the most important properties we could establish, the following are of particular interest for the tretment and prevention of age-dependent modifications of human skin (loss of skin-tissue, cells and matrix, wrinkle formation and others) : stimulation of cell proliferation (by $^3$[H]-thymidine incorporation and the MTT test), scavenging of reactive oxygen species (ROS) using several different procedures, and protease (MMP-2 and MMP-9) down-regulation. A topical preparation, using RROP-s and FROP-s, and/or L-fucose, was shown to increase cell proliferation, dermal matrix synthesis, efficient scavenging of ROS-s and to increase also the thickness of dermal tissue when applied for 4 weeks on hairless rat skin, accompanied by the densification of collagen bundles as well as by an increase of elastin synthesis. Using fluorescent labeled FROPs, it could be shown that these oligosaccharides react with cell-membrane receptors and especially with the elastin-laminin-receptor and the fucose-mannose receptor, but they penetrate also in the cell nucleus, suggesting the possibility of a direct action on the regulation of gene expression. When applied to the human skin of a team of voluntary women encompassing all age-groups, the efficiency of FROP-containing preparation could be confirmed using indentometry and computerised evaluation of skin micro-relief, as well as evaluation of periorbital wrinkles. It appears therefore that these preparations correspond to all the requirements of active anti-aging principles.

  • Antisense phosphorothioate oligodeoxynucleotide(PS-ODN) against $TGF-{\beta}$ was developed as scar formation inhibitor. The scar was caused collagen deposition due to overexpression of $TGF-{\beta}$ in wounded skin. The percutaneous absorption of partially modified PS-ODN(25 mer) was investigated for the purpose of its effective delivery. Though PS-ODN has high molecular weight (MW=8,000) and polyanionic charge, it was permeated through skin. The skin permeation of PS-ODN was markedly increased by the removal of stratum corneum and dermis. Moreover, the skin permeation of PS-ODN was decreased in the following order; hairless mouse skin>rat skin>human cadaver skin. Thus, PS-ODN represents a logical candidate for further evalution due to the potential for delivery into the wounded skin.

Cytotoxic Effect of Adriamycin in Cultured Skin Cells of Fetal Rat (백서 태자의 배양 피부세포에서 Adriamycin의 세포독성에 관한 연구)

  • Local extravasation during intravenous administration of adriamycin (doxorubicin HCl) can cause severe skin ulceration and necrosis. To investigate the mechanism of adriamycin-induced skin toxicity, effects of adriamycin on reactive oxygen radical metabolism using cultured skin cells of fetal rat. Adriamycin produced significant release of lactic dehydrogenase from cultured skin cell preparations dose- and time-dependently. The production of superoxide anion in sonicated suspensions of cultured skin cells was significantly increased by adriamycin under the presence of NADPH and NADH. The drug also stimulated malondialdehyde (MDA) production, an index of lipid peroxidation, in NADPH- and NADH-supported cell preparations. The increased production of MDA was significantly inhibited by oxygen radical scavengers (superoxide dismutase, catalase, thiourea) and antioxidants (butylated hydroxytoluene, ${\alpha}-tocopherol$). Treatment of cultured skin cells with 1, 3,-bis (2-chloroethyl)-1-nitrosourea (BCNU), an inhibitor of glutathione reductase, enhanced the lipid peroxidation induced by adriamycin. The present study suggests that lipid peroxidation which is resulted from the stimulated production of reactive oxygen radical causes cellular damage in adriamycin-treated skin cells of rat.
