Role of $17{\beta}$- Estradiol on Brain Atrophy Following Cerebral Infarction (뇌졸중후 뇌위축에 대한 조경론적 접근)

  • Objective : The aim of this study was to investigate the neuroprotection effect of estrogen on brain atrophy following cerebral infarction. Method : All animals in this study were classified into 4 groups; ovariectomy group (OVXgroup), cerebral infarction group (INF group), combination ovariectomy and cerebral infarction group (OVX + INF group), and naturally intact group for control data (NOR group). Cerebral infarction was made by Chen's method with some modification. Ovariectomy was performed by Wayforth's method. Experimental data for each group was collected at 15 days, month, 3 months, and 6 months after starting observation. Serum $17{\beta}-estradiol(E2)$ was determined by radioimmunoassay. Brain volume was measured and calculated with image analysis. Each brain was sliced at intervals of 2mm in chamber after 30 min of freezing in refregerater. Cerebral volume was obtained by sum of volume of each slice level, which was mean $area{\;}{\times}{\;}2mm$. Results : Cerebral ischemia was found to decrease the serum concentration of $17{\beta}-{\;}estradiol(E2)$ and to inhibit the physiologically conpensatary function of the ovariectomized rats. Also we found that deprivation of estrogen have resulted in more severe cerebral atrophy followed by cerebral infarction. Conclusion : It is suggested that estrogen has a neuroprotection effect on cerebral atrophy following cerebral infarction.

  • Osteoporosis means a deficiency in the amount of bone tissue in the skeleton or part of the skeleton. Osteoporosis is a lesion, not a specific disease. 'Osteoporotic' describes the slate of a bone or skeleton at a given time. Osteoporosis may be diagnosed subjectively by visual appraisal, or objectively by measurement of radiographs, sawn bones, or microscopic sections. This study was carried out to make clear of the influence of ovariectomy on time course of fibular osteoporosis in rats. Seven weeks after ovariectomy, osteoporosis was evident, when the size of the bone marrow cavities significantly increased and the width from the bone marrow cavity and cortex significantly decreased than normal.

  • The aim of this investigation was to examine the effects of osteocalcin, bone-specific alkaline phophatase, estrogen, insulin-like growth factor-I (IGF-I), Ca, P and bone mineral density on osteoporosis induced by ovariectomy in rats. Female Sprague-Dawley 30 rats of three-forth's birth, weighing $215{\pm}10g$, were divided into two groups including the sham operation group(5 heads) and ovariectomy group(25 heads). They were fed normal diets for 2 weeks before the experimental operation and for 8 more weeks after operation. The level of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were increased in experimental group, but a little increased in sham operation group at same period. The change of rates of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were significantly higher in experimental group than sham operation group. $Ca^{2+}$ was not changed between two groups and P was significantly decreased in experimental group and Ca/P ratio was higher in experimental group than sham operation group. Body weights were increased in all two groups and growth rate per day was higher in experimental group than sham operation group. However, femur weight I body weight ratio was lower in experimental group than sham operation group.

Difference in Spinal Fusion Process in Osteopenic and Nonosteopenic Living Rat Models Using Serial Microcomputed Tomography

  • Objective : To identify and investigate differences in spinal fusion between the normal and osteopenic spine in a rat model. Methods : Female Sprague Dawley rats underwent either an ovariectomy (OVX) or sham operation and were randomized into two groups: non-OVX group and OVX group. Eight weeks after OVX, unilateral lumbar spinal fusion was performed using autologous iliac bone. Bone density (BD) was measured 2 days and 8 weeks after fusion surgery. Microcomputed tomography was used to evaluate the process of bone fusion every two weeks for 8 weeks after fusion surgery. The fusion rate, fusion process, and bone volume parameters of fusion bed were compared between the two groups. Results : BD was significantly higher in the non-OVX group than in the OVX group 2 days and 8 weeks after fusion surgery. The fusion rate in the non-OVX group was higher than that in the OVX group 8 weeks after surgery (p=0.044). The bony connection of bone fragments with transverse processes and bone formation between transverse processes in non-OVX group were significantly superior to those of OVX group from 6 weeks after fusion surgery. The compactness and bone maturation of fusion bed in non-OVX were prominent compared with the non-OVX group. Conclusion : The fusion rate in OVX group was inferior to non-OVX group at late stage after fusion surgery. Bone maturation of fusion bed in the OVX group was inferior compared with the non-OVX group. Fusion enhancement strategies at early stage may be needed to patients with osteoporosis who need spine fusion surgery.

Effect of Crude Saponin from Korean Red Ginseng on Clinical Chemical Parameters of Ovariectomized Rat (난소절제 흰쥐의 임상화학지수매 미치는 홍삼조사포닌의 영향)

  • This study was carried out to investigate the effect of crude saponin from Korean red ginseng on clinical chemical parameters in ovariectomized rats. The crude red ginseng saponin was prepared by Diaion HP-20 adsorption chromatography and spirit. Tota1 of 50 rats were divided into 5 groups: normal control (NC), sham-ovariectomized (SO), ovariectomized (OR), ovariectomized and saponin treated (OS) and normal control treated with saponin (NS). Saponin was intraperitonally administered for 12 weeks since 1 week before ovariectomy: The body weight of ovariectomized rats showed no significant change but that of NS group showed significant increase when compared with NC group. Platelet counts of serum showed significant increase when treated with saponin regardless of ovariectomy. Triglyceride content of serum in NC group was 152.1 mg/㎗, while that of OR group was decreased to 99.9 mg/㎗ However, when saponin was administered, the content was increased to 138.0 mg/㎗. The weight of spleen also showed significant increase when treated with saponin, while the other organs showed no weight changes. On the other hand, ovariectomy in rats induced decrease in femur weight by 10% when compared with NC group. However, administration of crude saponin in ovariectomized rats recovered the weight of the femur to the similar level of NC (e<0.01 0.05). In addition, femur weight of NS group indicated 10 to 16% higher value than that of NC. These results suggest that Korean red ginseng saponin attenuates phyiological disorders induced by malfunction of ovary.

Effects of High Protein and Calcium Intakes on Calcium Metabolism and Renal Function in Ovariectomized Osteporosis Rat Model (골다공증 모델 흰쥐에서 고수준의 단백질의 칼슘 섭취가 칼슘대사 및 신장기능에 미치는 영향)

  • This study was to investigate interaction between dietary protein and Ca levels in Ca metabolism and renal function in osteporosis rats. Five week-old female rats were fed a low Ca diet for 4 weeks after ovariectomy operation to establish rat models of osteoporosis. The ovariectomized osteoporosis rats were divided into six groups and were fed experimental diets which contained two levels of protein, normal (20%) and high(40%) , and three levels of Ca, low (0.06%), normal (0.47%) and high(0.94%) for 4 weeks , respectively. The ovaricetmized rat model of osteoporosis showed a remarkable decrease in serum Ca concentration, fresh weight and breaking force of femur, Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca at the dietary levels of normal and high levels significantly enhanced Ca bioavailability shown in the above experimental rat models of osteoporosis, regardless of dietary protein levels ; whereas the rats which were fed the low Ca diet demonstrated rather a decrease in its bioavailability. Irrespectively of the dietary Ca levels, the rats which were fed high protein diet exhibited an increase in kidney weight, urinary Ca, volume and hydroxyproline, and glomerular filtration ratio(GFR). The results show that dietary protein and calcium levels affect the renal function and Ca metabolism independently, while the interaction between protein and calcium have not been shown.

Changes of spontaneous pancreatic exocrine secretion during the estrous cycle in rats (흰쥐에서 발정주기에 따른 자발적인 췌장외분비의 변화)

  • Since the role of female sexual hormones on pancreatic exocrine secretion was not fully understood, this study was investigated to clarify the difference of spontaneous pancreatic exocrine responses during the estrous cycle and the roles of ovarian hormones on pancreatic exocrine secretion in the anesthetized female rats. Pancreatic juice was collected from the sequential 15-min samples, and then fluid and protein secretion were measured from the collected samples. The stages of estrous cycle were defined by staining the vaginal smear. The spontaneous pancreatic fluid and protein secretion were significantly increased during the diestrus stage compare to the corresponding value during the estrus stage. In the ovariectomized rat, spontaneous pancreatic exocrine secretion was significantly decreased compare to the value of female rat during the diestrus stage and was restored by subcutaneous injection of progesterone (50 mg/kg). This results suggest that the spontaneous pancreatic exocrine secretion of female rat is fluctuated according to the estrous cycle and progesterone released from ovary could stimulate the spontaneous pancreatic exocrine secretion of female rat.

  • This study was performed to evaluate the effect of HCB on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with HCB, extract of Houyttnia cordata, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of HCB. Osteoporotic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns.

EXPERIMENTAL STUDY OF PERI-IMPLANT TISSUE REACTION IN OVARIECTOMIZED OSTEOPOROTIC RATS (난소절제로 유도된 골다공증 흰쥐에서 implant 주위 조직 반응에 관한 실험적 연구)

  • This study was designed to investigate the peri-implant tissue reaction in ovariectomized osteoporotic female rats, and to evaluate effects of estrogen, calcitonin, parathyroid hormone on the bone - implant interface in osteoporotic rats. 120 Sprague - Dawley rats were used in this experiments. Osteoporosis was induced by bilateral ovariectomy. They were divided 5 groups : sham-operated control group(Sham), ovariectomized group (OVX), OVX and estrogen treated group (OVX+E), OVX and PTH treated group (OVX+PTH), and OVX and calcitonin treated group (OVX+CT). Eight weeks after ovariectomy, two titanium screw implants were inserted into the left tibia of each rat. Eight weeks after the insertion of the implants, the periotest values (PTV) of implant were examined, and the rats were sacrificed, and examined the reaction of bone tissue surrounding the implant both histologically and histomorphometrically. The bone density and ash weight of opposite right tibia were examined. Over 40 rats were fractured on left tibia that was implant inserted. On histologically finding, all groups were osseointegrated well, especially in OVX+PTH group. In OVX group, tibial cortical bone showed many large harversian canal and microfracture lines. The OVX+PTH group showed the lowest mean PTV (-2.33) (p<0.05), and the hightest mean bone - implant contact percentage (89%) (p>0.05). But the OVX+CT group showed the highest mean bone density ($5.45mg/cm^3$) and ash weight (56.12%) (p<0.05). The results indicate that PTH treatment enhances osseointegration of implant in OVX rats, and CT treatment depresses bone turnover and prevent the development of osteopenia in OVX rats.

The Effect of Kamijoaguiem on Osteoporosis Induced by Ovariectomy in Rats (가미좌귀음(加味左歸飮)이 난소적출(卵巢摘出)로 유도(誘導)된 백서(白鼠)의 골다공증(骨多孔症) 미치는 영향(影響))

  • Purpose : This study was peformed to evaluate the effect of Kamijoaguiem(JGE) on the bone mass and its related factors. Methods : We used ovariectomized rat as an estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 7th day after operating ovariectomy, rats were administered with JGE per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of JGE. Results : Bone density, bone ash density, bone calcium, magnesium and phosphorus was decreased in osteoporotic rats. JGE improved the decreased bone density, bone ash density and the decreased bone magnesium, but JGE didn't improve the decreased bone calcium and phosphorus in osteoporotic rats. Osteocalcin in serum and hydroxy-proline excretion in urine were increased in osteoporotic rats. Their levels were decreased when JGE was administered. ALP activity in serum was increased in osteoporotic rats. JGE didn't induce any significant changes. JGE showed significant increase in serum calcium level, total protein level, albumin level, BUN level, serum LDH activity. JGE didn't show significant increase in serum T-cholesterol density, triglyceride density, HDL-cholesterol density. JGE didn't show significant increase in RBC number, hemoglobin level, platelet number, hematocrit level. JGE showed inhibitory effect on the degradation of bone-matrix in osteoporotic rats, in histological examination to Hematoxylin-eosin stain. Conclusion : JGE might improve bone density due to inhibition of bone resolution in osteoporotic rats. It suggest that JGE may be useful prescription in osteoporosis.
