NO Formation of the PMA and LPS-activated Rat Kupffer- and Endothelial Cells in vitro (In Vitro에서 PMA와 LPS로 활성화된 흰쥐 간내 Kupffer-와 Endothelial 세포에서의 NO 형성에 관한 연구)

  • The Present study was undertaken to indicate the major source of NO by liver cells in vitro. Even at early stages of induction or low LPS concentrations, NO was produced at high rates by LPS(Lipopolysaccharide) on the isolated rat kupffer cells. PMA(phorbol 12-myristate 13-acetate) induced NO formation at low rates in the same cells. IFN-${\gamma}$ (Interferon-${\gamma}$) alone had not induced NO formation but it stimulated the effects of LPS. Calcium ionophore A23187 caused no stimulatory effect. It suggests that LPS has especially strong NO inducer on the kupffer cells and its mechanism is related to those on macrophage in other organs. In other nonparenchymal liver cells, sinusoidal endothelial cells were not stimulated to produce NO either by inducers of aortic endothelium(A23187, ATP and ADP) or by effectors of macrophages(LPS, IFN-${\gamma}$. This results suggest that rat liver kupffer cells appear to be the major source of NO by liver cells in vitro. But in vivo, liver endothelial cells may still be capable of producing NO. Furthermore, kupffer cells may produce factors that facilitate NO production by the endothelial cells.

A study on the isozyme alterations of lactic dehydrogenase in the tissues of albino rat by the exposure in sulfur dioxide (아황산가스에 노출된 백서조직 lactic dehydrogenase의 isozyme변화에 관한 연구)

  • The isozyme alteration of lactic dehydrogenase in the tissues of albino rat inhaled SO$_{2}$ were studied in vivo and in vitro, with the following results: (1) The H-type of LDH activity relatively dominated in the normal brain, heart and kidney tissues of rat, M-type in the normal lung, liver, and muscle tissues of the animal. (2) When rats inhale SO$_{2}$ in the concentration of 250 ppm, it appears that the M-type tends to predominate in the anaerobic tissues such as liver, kidney and muscle tissues and the H-type in the aerobic tissues such as brain and heart tissues. (3) When 5% SO$_{2}$ is introduced into tissue homogenates, LDH activities in the heart, lung, liver and muscle tissues are increased more than that of introducing room-air only. With sam treatment, LDH activity is decreased in the kidney tissue and no alteration is observed in the brain tissue. (4) Although, after the aeration of SO$_{2}$, the oxygen tension seems to bring decreases in the level of LDH activity in the anerobic tissues such as liver and muscle tissues, while, on the other hand, increases in the level of the activity in the aerobic tissues, such as the brain, heart and lung tissues. (5) Accordinglly, SO$_{2}$ affects LDH activities, its isozyme pattern of each organs, and their metabolic pathway by its absorption of the gas.

3-D OCT Image Reconstruction for Precision Analysis of Rat Eye and Human Molar (쥐 눈과 인간 치아의 정밀한 단층정보 분석을 위한 OCT 3-D 영상 재구성)

  • Optical coherence tomography(OCT) is a high resolution imaging system which can image the cross section of microscopic organs in a living tissue with about $1{\mu}m$ resolution. In this paper, we implement OCT system and acquire 2-D images of rat eye and human molar samples especially in the field of opthalmology and dentistry. In terms of 2-D images, we reconstruct 3-D OCT images which give us another inner structural information of target objects. OPEN-GL reduces the 3-D processing time 10 times less than MATLAB.

Effects of PMSG Treatment on Reproductive Organs and Serum Steroid Hormone Level in Adrenalectomized Immature rats (부신척출 미성숙 흰쥐에 PMSG투여가 생식기관 및 혈청중 Steroid Hormone 농도에 미치는 영향)

  • The purpose of this experiment was to investigate the effects of adrenalectomy and PMSG treatment on reproductive organs and serum steroid hormone level in immature female rats. The animals used in this experiment were 25 days old female rats weighing a, pp.oximately 70g. They were randomly divided into two groups of intact rat group (Int-) and adrenalectomized rat group (Adx-) and each group were subdivided into two groups of Non-PMSG (-Cont) and PMSG treated (-PMSG) group. The rat of PMSG-treated group (-PMSG) was administered subcutaneously with 25 IU PMSG on first day (9 a.m.) after adrenalectomy. The adrenalectomized rat groups were su, pp.ied with saline solution through the experiment period. The rate of ovulation and vaginal opening and reproductive organ weights were observed at 8, 32, 56, 80 and 104 hours after PMSG treatment. At the same time, the serum level of estradiol-17${\beta}$ and progesterone were measured by the radioimmunoassay. The results obtained were as follows: 1. Ovulation was shown at 56 hours after treatment in Int-PMSG group and Adx-PMSG group and Adx-PMSG group. The rate of ovulation was very low in PMSG-treated groups, but it was increased in 80 to 90% at 104 hours after treatment. However, there was no ovulation in Int-Cont group and Adx-Cont group. 2. Vaginal opening was shown at 56 hours after treatment in Int-PMSG group and Adx-PMSG group and a, pp.ared in 80% at 104 hours after treatment. The rate of vaginal opening in PMSG-treated groups was very low, but Int-Cont group and Adx-Cont group had no vaginal opening. 3. The weight of ovary and uterus in two PMSG-treated groups were increased with the elapse of time after treatment and were significantly heavy in all observation time, but changes in Int-Cont group and Adx-Cont group were not recognized. The weights of ovaries and utera in Adx-Cont group were increased with the elapse of time. 4. The level of serum estradiol-17${\beta}$ was remarkably increased in PMSG-treated groups (Int-PMSG and Adx-PMSG groups) compared with Int-Cont and Adx-Cont group, and significant difference was recognized between Non-PMSG group and PMSG-treated group in the experimental period. Especially, the highest levels of Int-PMSG groups and Adx-PMSG groups were shown at 80 and 56 hours after treatment and after ward estradiol-17${\beta}$ levels of PMSG-treated groups were decreased. However, changes of the levels did not a, pp.ared in Non-PMSG groups at 104 hours after treatment. 5. The level of serum progesterone in PMSG-treated groups was significantly increased between 80 and 104 hours after treatment. With the elapse of time, the level was increased in all observed groups except for Int-Cont and Adx-Conx group. And the order from the highest level at 104 hours after treatment was Int-PMSG, Adx-PMSG, Int-Cont and Adx-Cont group.

Study of single dose test of Sweet Bee Venom in rats (Sweet BV의 rat를 이용한 단회 근육시술 독성시험)

  • Objectives: This study was performed to analyse single dose toxicity of pure melittin(Sweet Bee Venom-Sweet BV) extracted from the bee venom by utilizing protein isolation method of gel filtration. Methods: All experiments were conducted at Biotoxtech, a non-clinical studies authorized institution, under the regulations of Good Laboratory Practice (GLP). Six weeks old female Sprague-Dawley rats were chosen for the pilot study and determined 30㎎/㎏ which is 4285 times higher than the clinical application dosage as the high dosage, followed by 15 and 7.5㎎/㎏ as mid and lose dosage, respectively. Equal amount of excipient to the Sweet BV experiment groups was administered as the control group. Results: 1. No mortality was witnessed in all of the experiment groups. 2. Hyperemia and movement disorder were observed around the area of administration in all groups, and higher occurrence in the higher dosage groups. Hyperemia and movement disorder diminished with elapsed time. 3. For the weight measurement, male groups showed larger reduction in weight in accordance with higher dosage. Female groups didn't s how significant changes. 4. To verify abnormalities of organs and tissues, cerebellum, cerebrum, liver, lung, kidney, and spinal nerves were removed and conducted histological observation with H-E staining. No abnormalities were detected in any of organs and tissues. 5. One female rat in the 30㎎/㎏ group had amputated toe near the administered area and histopathological finding was hemorrhage with inflammation. This is presumed as a secondary infection after the administration of Sweet BV. Conclusion: Above findings suggest Sweet BV is relatively s safe treatment medium. Further studies on the subject should be conducted to yield more concrete evidences.

Biodistribution and pharmacokinetic evaluation of Korean Red Ginseng components using radioisotopes in a rat model

  • Background: Although many studies have evaluated the efficacy and pharmacokinetics of Korean Red Ginseng (KRG) components (Rg1, Rb1, Rg3, Rd, etc.), few have examined the in vivo pharmacokinetics of the radiolabeled components. This study investigated the pharmacokinetics of ginsenosides and their metabolite compound K (CK), 20(s)-protopanaxadiol (PPD), and 20(s)-protopanaxatriol (PPT) using radioisotopes in rat oral administration. Methods: Sprague-Dawley rats were dosed orally once with 10 mg/kg of the tritium(3H) radiolabeled samples, and then the blood was collected from the tail vein after 0.25, 0.5, 1, 1.5, 2, 4, 6, 8, 12, 24, 48, 96, and 168 h. Radioactivity in the organs, feces, urine, and carcass was determined using a liquid scintillation counter (LSC) and a bio-imaging analyzer system (BAS). Results and conclusion: After oral administration, as the 3H-labeled ginsenosides were converted to metabolites, Cmax and half-life increased, and Tmax decreased. Interestingly, Rb1 and CK showed similar values, and after a single oral administration of components, the cumulative excretion ratio of urine and feces was 88.9%-92.4%. Although most KRG components were excreted within 96-168 h of administration, small amounts of components were detected in almost all tissues and mainly distributed to the liver except for the digestive tract when observed through autoradiography. This study demonstrated that KRG components were distributed to various organs in the rats. Further studies could be conducted to prove the bioavailability and transmission of KRG components to confirm the mechanism of KRG efficacy.

Gene Expression Patterns of the Endogenous Antioxidant Enzymes in Linuron-Treated Rat Ventral Prostates after Castration

  • Linuron is a pesticide with a weak anti-androgenic property, which impacts male reproductive organs. In this study, to clarify whether linuron affects the cellular antioxidant system of ventral prostate, gene expression patterns of the representative antioxidant enzymes such as glutathione peroxidase (GPx), selenoprotein P (SePP), and superoxide dismutase (SOD) were investigated in the rat ventral prostates exposed to linuron using real-time RT-PCR analyses. Sprague-Dawley rats castrated at 6 weeks old were treated with linuron (25, 50, or 100 mg/kg per oral) daily for 10 days after testosterone propionate administration (0.4 mg/kg) subcutaneously. As compared to normal control animals, mRNA levels of phospholipid hydroperoxide GPx (PHGPx), SePP, and Mn SOD significantly increased in the prostates exposed to linuron (25, 50, and 100 mg/kg). However, cytosolic GPx (100 mg/kg) and Cu/Zn SOD (25, 50, and 100 mg/kg) mRNA levels significantly decreased in the ventral prostates. These results indicate that linuron upregulates the expressions of PHGPx, SePP, and Mn SOD mRNAs, but down-regulates the expressions of cytosolic GPx and Cu/Zn SOD in rat prostates, suggesting that linuron may have dual effects in the cellular antioxidant system of prostate.

Immunological Gharacterization of Inositol(1,4,5) triphosphate 3-Kinase in Rat Tissues (흰쥐 조직에 존재하는 Inositol(1,4,5) triphosphate 3-Kinase의 면역학적 특성)

  • Brain, heart, liver, lung, kidney and thymus etc. 12 organs were removed and homogenized from Dawley-Sprague rats after suffocation. After fractionation of the tissue cytosols, enzymatic activities of the key enzymes in metabolic inositol phosphates cycle, PLC, IPSK and Ins(1,4,5) P35-phosphatase, were measured respectively. Hybridoma monoclones producing anti-lP3K murine monoclonal antibodies were obtained by the fusion of SP2/Ag 0-14 and spleen cells of mouse immunized with purified 53KDa IPSK, screening and cloning procedures. 18 cloned hybridoma cells were obtained, background due to nonspecific binding was very low with 10 clones. These Abs were purified from ascitic fluids by using affi-gel 15, and determined subtype of Abs. When immunoreactivities for rat tissues IP3K were exercised by adding the mixed Abs of 19Gl and 19G2b, they showed an overall similarity with noncompetitive inhibition. Brain tissue has high sensitivity for anti-lP3K Ab, whereas heart tissue has very low activity. In kinetic parameters Km value was 1.58 mM and Vmx value was 5.41umol/min/ml, respectively Only one form of 40 KDa IPSK was detected in heart tissues, however rat brain contains at least three immunologically distinct IP3K (53, 51 and 40 KDa) in western blot analysis. Of them 53 KDa protein was major enzyme in enzymatic activity. Northern blot analysis with 32P-labeled CDNA probe which encodes 1.8 Kb IPSK gene was performed. These results suggest that IPSK are regulated at transcriptional level during rat tissue development.

Reduction of long-term potentiation at Schaffer collateral-CA1 synapses in the rat hippocampus at the acute stage of vestibular compensation

  • Vestibular compensation is a recovery process from vestibular symptoms over time after unilateral loss of peripheral vestibular end organs. The aim of the present study was to observe time-dependent changes in long-term potentiation (LTP) at Schaffer collateral-CA1 synapses in the CA1 area of the hippocampus during vestibular compensation. The input-output (I/O) relationships of fEPSP amplitudes and LTP induced by theta burst stimulation to Schaffer's collateral commissural fibers were evaluated from the CA1 area of hippocampal slices at 1 day, 1 week, and 1 month after unilateral labyrinthectomy (UL). The I/O relationships of fEPSPs in the CA1 area was significantly reduced within 1 week post-op and then showed a non-significant reduction at 1 month after UL. Compared with sham-operated animals, there was a significant reduction of LTP induction in the hippocampus at 1 day and 1 week after UL. However, LTP induction levels in the CA1 area of the hippocampus also returned to those of sham-operated animals 1 month following UL. These data suggest that unilateral injury of the peripheral vestibular end organs results in a transient deficit in synaptic plasticity in the CA1 hippocampal area at acute stages of vestibular compensation.

Single Oral Dose Toxicity Test of Low Molecular Weight Fucoidan in Rats

  • The object of this study was to evaluate the single oral dose toxicity of Low Molecular Weight Fucoidan (LMF) in male and female rats. LMF was administered to female and male SD rats as an oral dose of 2,000, 1,000 and 500 mg/kg (body wt.). Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation organ weight and histopathology of 14 principle organs were examined upon necropsy. As the results, no LMF treatment related mortalities, clinical signs, changes on the body and organ weights, gross and histopathological observations against 14 principle organs were detected up to 2,000 mg/kg in both female and male rats except for some sporadic findings not LMF treatment related toxicological signs. Therefore, $LD_{50}$ (50% lethal dose) and approximate LD of LMF after single oral treatment in female and male rats were considered over 2,000 mg/kg - the limited dosages recommended by KFDA Guidelines [2005-60, 2005], respectively.
