Optimization of In-line Coagulation/MF Process Using On-line Streaming Potential Measurement (On-line streaming potential 측정에 의한 in-line 약품응집/정밀여과 공정의 최적화)

  • Microfiltration with in-line rapid coagulation for drinking water production was examined. The in-line rapid coagulation was conducted using newly developed mixing device instantaneous flash mixer. The flux decline during membrane filtration was monitored with coagulant dosage varied. Flux decline was minimized at 1.1mg/L of coagulant dosage, where streaming potential of coagulated water was near zero. The optimum dosage for the process control was explained by dimensionless distance (${\kappa}{\times}a$) of particle pairs, obtained from electrophoresis parameter describing electrostatic repulsion relative to Van der Waals energy between particle pairs in the pre-coagulated water.

Improvement of Rapid Sand Filtration to Two Stage Dual Media Filtration System in Water Treatment Plant (정수처리장 사여과지의 이단이중여과재 시스템으로의 개량)

  • This study aimed for evaluating the applicability of the two stage dual media filtration system in field water treatment plant. The field plant of two stage and dual media filtration system was operated for 2 months. Average iron concentrations of the settled water, existing filtered water and second stage filtered water was 0.041 mg/L, 0.007 mg/L and 0.005 mg/L, respectively. Removal efficiency of iron concentration in the second stage is appropriately 35% more than in existing filtered water. Also removal efficiency of residual chlorine in the dual media filtration system is relatively 42.3% more than in existing filtered water due to adsorption of activated carbon, but the removal of ammonia nitrogen by adsorption is insufficient. Average concentrations of THM and chloroform in the settled water are 0.033 mg/L, 0.026 mg/L, respectively and in existing filtered water are 0.023 mg/L and 0.023 mg/L. Average concentrations of THM and chloroform in the dual media filtration system are 0.008 mg/L and 0.013 mg/L. Therefore removal efficiency of THM concentration in second stage is more than 66.4% in existing filtrated water. Also removal efficiency of chloroform in the dual media filtration system is more than 50.0% in existing filtered water because of the adsorption of activated carbon. In this case backwashing period in dual stage system is 4~5 days, but in existing filtration system is 1~2 days.

  • It has been hypothesized that efficient exclusion of methylated retrotransposons and repeated DNA region is one of the rapid and cost-effective approaches for comprehensive gene discovery in large genome size of maize. Three kinds of methylation-sensitive restriction enzymes, HapII, MspI and McrBC, were used to identify the restriction frequency of cytosine methylation sites in maize genome. Roughly 60% of total maize genomic DNA was restricted less than 500bp by McrBC, and the most of restricted small size fraction was composed retrotransposon. In order to validate the efficient construction of gene-rich shotgun library, we compare two gene-rich methyl-filtration shotgun libraries using in vivo and in vitro methyl-filtration system. The size selected DNA fraction by Sau3A-McrBC enzyme treated was very stable and has not appeared modification in E. coli, but most insert DNA size of partially digested with Sau3A were decrease less than 500bp by bacterial methylation-modification system. In compare of retroelements portion, A 44.6% of the sequences were retroelement in unmethyl-filtered library, and the most of them was Copia type, such as Prem, Opie and Ji. The portion of retroelement was drastically decreased to 25% and 20% by in vivo and in vitro filtration system, respectively.

Comparison of Upgraded Methods for Detecting Pathogenic Escherichia coli in Foods Using Centrifugation or Filtration

  • In the present study, centrifugation and filtration pretreatments were evaluated to decrease sample preparation time and to improve the sensitivity and specificity of multiplex polymerase chain reaction (PCR) for the detection of low levels of pathogenic Escherichia coli in various foods. Pathogenic E. coli (E. coli NCCP11142, E. coli NCCP14037, E. coli NCCP 14038, E. coli NCCP14039, and E. coli NCCP15661) was inoculated into pork, beef, and baby leafy vegetables at 1, 2, and 3 Log CFU/g. The samples were shaken 30 times (control), then centrifuged or filtered. DNA extracts from the samples were subjected to PCR using the $Powerchek^{TM}$ Diarrheal E. coli 8-plex Detection Kit. In the pork samples, no E. coli was detected in the control samples, while E. coli were detected in 100% of 3-Log CFU/g inoculated and centrifuged samples, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In the beef samples, all control samples appeared to be E. coli-negative, while E. coli was detected in 50-75% of centrifuged samples, regardless of inoculated level, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In baby leafy vegetables, E. coli were not detected in 25-50% of the control samples, while E. coli were detected in 0-25% of the centrifuged samples, and 75-100% of the filtered samples, depending on the inoculum amount. In conclusion, filtration pretreatment can be used to minimize sample preparation time, and improve the sensitivity and specificity of rapid detection of pathogenic E. coli in various foods.

Improvement of Rapid Sand Filtration to Two Stage Dual Media Filtration System in Water Treatment Plant (정수처리장내 급속모래 여과지의 이단복합여과시스템으로의 개량)

  • This study aimed for developing a two stage dual media filtration system. It has a sand and activated carbon layer above the under-drain system, and a sand layer above the middle-drain system for pretreatment. When retrofitting an old sand filter bed or designing a new one, this technology can substitute the existing sand filter bed without requiring a new plant site. The removal rate of total particle is 93, and 3~7 ${\mu}m$ and 5~15 ${\mu}m$ particles are all 97%. These high removal efficiencies of each pollutant due to adsorption and biological oxidation in activated carbon filter layer. The best backwashing method of two stage dual media filtration system is ascertained by air injection, air + water injection and water injection sequence. In this study, a pilot plant of two stage and dual filtration system was operated for 4 months in water treatment plant. The stability of turbidity was maintained below 1 NTU. The TOC, THMFP and HAAFP were removed about 90% by two stage and dual media filtration system, which is almost 2 times higher than existing water treatment plant.

A Study on the Drinking Water Treatment by Precoat Filtration and Activated Carbon Adsorption Process (규조토여과 및 활성탄흡착 공정을 이용한 용수처리에 관한 연구)

  • I performed the research about the drinking water treatment by precoat filtration and activated carbon adsorption process in the D water treatment plant at Gwangju. D water treatment plant inlet water is supplied from Juam lake in Jeollanamdo. The results are as follows; 1. Element disk used in this experiment are R(pore size $10{\mu}m$), B(pore size $20{\mu}m$). And diatomaceous earth are A(cake pore size $3.5{\mu}m$), B(cake pore size $7{\mu}m$) and C(cake pore size $17{\mu}m$) 2. Filtrate of precoat filter during 30 min are B-C 10.2 > BB 5.7 > R-A 5.4 ($m^3/m^2$). 3. The water quality through B-C+AC and R-A+AC are DOC 1.76 mg/1, 1.288 m/l respectively. 4. total THMs produced by chlorination are $84.2{\mu}g/l$(B-C+AC), $66.11{\mu}g/l$ (R-A+AC), $97{\mu}g/l$ (rapid sand filtration water) respectively. 5. The R-A+AC and B-C+AC process can be substitute of CWTS.

Ammonium Ion Removal and Regeneration for Zeolite Filtration in Drinking Water Treatment (정수처리에서 제올라이트 여과를 이용한 암모니아성질소의 제거와 재생)

  • As the concentration of ammonium nitrogen could be reached 2~3 mg/L in the winter in the river. It was clear that the excessive concentration of chlorinated organics could be produced with the increase of chlorine addition to remove ammonium nitrogen. In the innovative ammonium nitrogen removal process, zeolite adsorption is very efficient as substitute for rapid sand filtration without other adverse quality change in the water. This study is conducted to evaluate the feasibility of ammonium nitrogen removal and regeneration by zeolite adsorption in drinking water treatment. Also, the reuse possibility of zeolite is evaluated to change the removal efficiency of ammonium nitrogen through several times of regeneration. The ammonium nitrogen was not removed in sand filter, but it was almost removed in zeolite filter during 7 days. The sand and zeolite filters have a similar result of turbidity removal. Therefore, zeolite filtration was confirmed the removal of turbidity and ammonium nitrogen as a media. When compared KCl with NaCl as a chemical for zeolite regeneration, it is demonstrated that KCl was more efficient than NaCl in the ability of zeolite regeneration. The adsorption rate of ammonium nitrogen was almost not decreased in the results of several times of regeneration. It is indicated that both zeolite and regeneration solution were possible to reuse without variation of regeneration rate through this study.

Flux Optimization Using Genetic Algorithms in Membrane Bioreactor

  • The behavior of submerged membrane bioreactor (SMBR) filtration systems utilizing rapid air backpulsing as a cleaning technique to remove reversible foulants was investigated using a genetic algorithm (GA). A customized genetic algorithm with suitable genetic operators was used to generate optimal time profiles. From experiments utilizing short and long periods of forward and reverse filtration, various experimental process parameters were determined. The GA indicated that the optimal values for the net flux fell between 263-270 LMH when the forward filtration time ($t_f$) was 30-37 s and the backward filtration time ($t_b$) was 0.19-0.27 s. The experimental data confirmed the optimal backpulse duration and frequency that maximized the net flux, which represented a four-fold improvement in 24-h backpulsing experiments compared with the absence of backpulsing. Consequently, the identification of a region of feasible parameters and nonlinear flux optimization were both successfully performed by the genetic algorithm, meaning the genetic algorithm-based optimization proved to be useful for solving SMBR flux optimization problems.

Removal Efficiency of Cryptosporidium Tracer in Drinking Water Treatment Process (정수처리 공정에서 Cryptosporidium Tracer의 제거효율)

  • In this study, removal efficiencies of treatment processes with C. tracer which is similar to the characteristics of Cryptosporidium were investigated. The highest removal efficiency of C. tracer was 97.16% when the input dose of PACI(Poly aluminium chloride, $Al_2O_3$(10%)) was 10 mg/L. The higher turbidity and SS removal efficiencies were, the more C. tracer cohesion efficiency increased. Also when pH of the raw water was high, removal efficiency of C. tracer increased. As the correlationship($R^2$) between effluent turbidity after coagulation-precipitation and removal efficiency of C. tracer was 0.9506, removal efficiency of Cryptosporidium could be evaluated by effluent turbidity after coagulation-precipitation. Also the range of C. tracer removal efficiency by sand filtration was $94.00{\sim}95.83%$ and the correlationship($R^2$) between effluent turbidity after filtration and removal efficiency of C. tracer was 0.8704. Therefore, when filtration-effluent turbidity is good under the optimized coagulation condition, removal efficiencies of Cryptosporidium by coagulation-precipitation, sand rapid filtration and sand rapid filtration after coagulation-precipitation are estimated as 1.55 log(97.16%), 1.38 log(95.83%) and 2.31 log(99.51%) respectively.

Verification of the Filter Media Applied to Filter Type Facility Considering the Treatment Efficiency Factor (여과형 시설에 적용되는 여재의 처리효율 영향인자를 고려한 실증화 평가)

  • Increased impervious surface caused by rapid urbanization serves to produce the discharge of non-point source pollutants such as total suspended solid (TSS). There exist various methods of removing TSS, including a filtration process using granular media (a well-known method to be practically used after the consideration of removal efficiency, clogging, and backwashing efficiency). To determine the TSS removal capability of the filter, we initially performed lab-scale experiments which assessed flow rates, influent concentrations, permeability co-efficients, the particular shapes of suspended solids and potential clogging, and also evaluated TSS removal efficiency when applied to filtration facility in a pilot-scale. In low filtration flux condition, the removal efficiency of suspended solids was more than 95 %, while decreased to 83% in high filtration flux. Regarding the clogging aspect of the experiment, total cumulative solids were loaded up to $19.15kg/m^2$, and TSS removal efficiency was noted to commence to decrease when the loaded solids exceeded $9.0kg/m^2$. It was also noted, however, that superior efficiency was maintained for six hours. In addition, for pilot-scale experiment, the removal efficiency was still high enough (83.4 %) for the solid concentration of 140 ~ 343 mg SS/L and after backwashing, head loss was recovered to 92 ~ 95 % during two hour filtration. With these results, It was confirmed that lifetime of the filter applied to the test was prolonged due to the high treatment efficiency and good backwashing efficiency for the cumulative solids load.
