A Decision Tree-based Analysis for Paralysis Disease Data

  • Even though a rapid development of modem medical science, paralysis disease is a highly dangerous and murderous disease. Shin et al. (1978) constructed the diagnosis expert system which identify a type of the paralysis disease from symptoms of a paralysis disease patients by using the canonical discriminant analysis. The decision tree-based analysis, however, has advantages over the method used in Shin et al. (1998), such as it does not need assumptions - linearity and normality, and suggest appropriate diagnosis procedure which is easily explained. In this paper, we applied the decision tree to construct the model which Identify a type of the paralysis disease.

  • Clinical and genetic heterogeneity in association with overlapping spectrum is characteristic in pediatric neuromuscular disorders, which makes confirmative diagnosis difficult and time consuming. Considering evolution of molecular genetic diagnosis and resultant upcoming genetically modifiable therapeutic options, rapid and cost-effective genetic testing should be applied in conjunction with existing diagnostic methods of clinical examinations, laboratory tests, electrophysiologic studies and pathologic studies. Earlier correct diagnosis would enable better clinical management for these patients in addition to new genetic drug options and genetic counseling.

A Rapid, Simple Determination of Sulfur-containing Compounds and Methylmalonic Acid on Plasma using GC-MS-SIM for the Diagnosis of Homocysteinemia (혈장 중 황함유 화합물과 메틸말론산의 신속 간편한 분석법 개발; GC-MS-SIM을 이용한 호모시스테인혈증의 진단)

  • Purpose: If early diagnosis is not made, patients with metabolic disorders as homocystinemia rapidly progress to physical defect or mental retardation resulted in storage of the toxic material into the brain. Therefore, it is necessary to develop an analytical method for a rapid screening and/or correct confirmation diagnosis. Methods: The standard solution of sulfur amino acids spiked plasma was subjected to protein precipitation with methanol, and then consecutively derivatized with trimethylsilyl (TMS) and trifluoroacyl (TFA) and determined by GC-MS. The formation of TMS derivative of the hydroxyl and TFA derivative of amino functional group was performed by BSTFA and MBTFA, respectively. Selective ion monitoring (SIM) mode was used for quantification with selected specific ions. Results: A calibration curve on standard spiked pooled plasma showed a linear relationship with correlation coefficient of 0.9936-0.9992 for all compounds over the range of 0.1-300 ng. The precision and accuracy were within S.D. of 1-15% and RSD of 1-15% for intra-day assay at 2 ng/mL, 15 ng/mL and 30 ng/mL. LOD and LOQ was 0.4 ng/mL and 4 ng/mL respectively. Conclusion: A rapid analytical method was developed to quantify sulfur amino acids and methyl malonic acid, after two-step derivatization procedure with good sensitivity and specificity on human plasma. Advantages of a new method are simplicity and rapidity. The method could be useful for routine analysis, diagnosis of homocysteinemia.

  • The enzyme activities of creatine kinase (CK), its isoenzyme MB (CK-MB) and of lactate dehydrogenase isoenzyme 1 (LD-1) have been used for years in diagnosing patients with chest pain in order to differentiate patients with acute myocardial infarction (AMI) from non-AMI patients. These methods are easy to perform as automated analyses, but they are not specific for cardiac muscle damage. During the early 90's the situation changed. First, creatine kinase ME mass (CK-MB mass) replaced the measurement of CK-MB activity. Subsequently cardiac-specific proteins, troponin T (cTnT) and troponin I (cTnI) appeared and displacing LD-1 analysis. However, troponin concentrations in blood increase only from four to six hours after onset of chest pain. Therefore a rapid marker such as myoglobin, fatty acid binding protein or glycogen phosphorylase BB could be used in early diagnosis of AMI. On the other hand, CK-MB isoforms alone may also be useful in rapid diagnosis of cardiac muscle damage. Myoglobin, CK-MB mass, cTnT and cTnI are nowadays widely used in diagnosing patients with acute chest pain. Myoglobin is not cardiac-specific and therefore requires supplementation with some other analyses such as troponins to support the myoglobin value. Troponins are very highly cardiac-specific. Only the sera of some patients with severe renal failure, which requires hemodialysis, have elevated cTnT and/or cTnI without there being any evidence of cardiac damage. The latest studies have shown that elevated troponin levels in sera of hemodialysis patients point to an increased risk of future cardiac events in a similar manner to the elevated troponin values in sera of patients with unstable angina pectoris. In addition, the bedside tests for cTnT and cTnI alone- or together with myoglobin and CK-ME mass can be used instead of quantitative analyses in the diagnosis of patients with chest pain. These rapid tests are easy to perform and they do not require expensive instrumentation. For the diagnosis of patient with chest pain, routinely myoglobin and CK-ME mass measurements should be performed whenever they are requested (24 h/day) and cTnT or cTnI on admission to the hospital and then 4-6 and 12 hours later and maintained less than 10% in imprecision.

  • Recently, human infections caused by severe fever with thrombocytopenia syndrome virus (SFTSV), which can lead to fatality, have dramatically increased in East Asia. With the unavailability of vaccines or antiviral drugs to prevent and/or treat SFTSV infection, early rapid diagnosis is critical for prevention and control of the disease. Here, we report the development of a simple, rapid and sensitive portable detection method for SFTSV infection applying reverse transcription-loop mediated isothermal amplification (RT-LAMP) combined with one-pot colorimetric visualization and electro-free reaction platform. This method utilizes a pocket warmer to facilitate diagnosis in a resource-limited setting. Specific primers were designed to target the highly-conserved region of L gene of SFTSV. The detection limit of the RT-LAMP assay was approximately $10^0$ viral genome copies from three different SFTSV strains. This assay exhibited comparable sensitivity to qRT-PCR and 10-fold more sensitivity than conventional RT-PCR, with a rapid detection time of 30 to 60 minutes. The RT-LAMP assay using SFTSV clinical specimens has demonstrated a similar detection rate to qRT-PCR and a higher detection rate compared to conventional RT-PCR. Moreover, there was no observed cross-reactive amplification of other human infectious viruses including Japanese Encephalitis Virus (JEV), Dengue, Enterovirus, Zika, Influenza and Middle East Respiratory Syndrome Coronavirus (MERS-CoV). This highly sensitive, electro- and equipment-free rapid colorimetric visualization method is feasible for resource-limited SFTSV field diagnosis.

Survey Study about Sasangin's Characteristics of Face, Voice, Skin and Pulse Diagnosis (사상인(四象人)의 안면, 음성, 피부 및 맥진 특성에 관한 설문조사 연구)

Objectives The purpose of this study is to find out the grade of practical use, the important element and the significant characteristics of Sasangin' s face, voice, skin and pulse diagnostic impression, in Sasang constitutional clinical diagnosis. 2. Methods We analysed the survey data about Sasangin' s face, voice, skin and pulse diagnostic impression, drawn up by specialist in Sasang constitutional medicine. 3. Results and Conclusions (1) In the application degree of face feature, the case which it was applied with 20-40% and 40-60% were 16 people(43.2%) respectively. In voice, the case applied with 0-20% was 19 people(51.4%), in skin, 0-20% and 20-40% were 14 people(37.8%) respectively and in pulse diagnosis, 0-20% were 25 people(73.0%). (2) In constitutional diagnosis, the important element of face were 'frontal whole shape', 'whole impression' and 'size and shape of eye, ear, mose and mouth', the important element of voice 'speed of speech', 'purity and impurity' and 'pitch', the important element of skin 'thickness', 'feel of touch' and 'size of skin pores' and the important element of pulse diagnosis 'speed of pulse', 'sinking and floating' and 'weakness and firmness'. (3) The important face characteristics of Taeyangin were 'bright eye', 'broad forehead' and 'strong impression', Soyangin 'protruding forehead', 'thin and small lips', 'narrowing and sharp chin', Taeumin 'thick lips', 'flat face', 'large eye, nose, ear and mouth' and Soeumin 'long and slender face', 'downward slanting eyes' and 'small eye, nose, ear and mouth', The important voice characteristics of Taeyangin were 'loud' and 'clear', Soyangin 'rapid' and 'high-pitched tone', Taeumin 'chick', 'slow' and 'low-pitched tone' and Soeumin 'small and feeble' and 'slow'. The important skin characteristics of Taeyangin were 'thin' and 'white', Soyangin 'thin', 'smooth' and 'elastic', Taeumin 'thick', 'large skin-pore', 'coarse' and Soeumin 'soft', 'thin' and 'subtle skin-pore'. The important pulse characteristics of Taeyangin were 'rapid' and 'large', Soyangin 'rapid' and 'floating', Taeumin 'tense', 'long' and 'solid' and Soeumin 'fine', 'weak' and 'slow'.

  • In this paper, using Covid-19 diagnostic data provided by the Korea Disease Control and Prevention Agency (KDCA), we examine the probability of confirmed cases and the probability of actually being confirmed when the rapid test is negative according to the sensitivity and specificity of the rapid diagnostic kit. When we know the conditional probability of confirmation given a positive test, we induce the relationship between sensitivity and specificity, and compute the actual sensitivity of the rapid diagnosis kit based on the data of KDCA.

  • The diagnosis of tuberculosis is usually established using staining and culturing techniques. Fluorescent stains have improved the sensitivity of direct microscopy. Improved culture media coupled with radiometric means of detecting early mycobacterial growth have shortened the time needed for cultural diagnosis. Rapid immunodiagnostic techniques based on the detection of mycobacterial antigen or of antibodies to theses antigens have not, however, come into widespread clinical use. The DNA or RNA hybridization tests with labeled specific probes which have been described so far are not sensitive enough to be used for clinical speicimens without prior culturing. The advent of the polymerase chain reaction (PCR) has opened new possibilities for diagnosis of microbial infections. This technique has already been applied to a number of microorganisms. In the field of mycobacteria the PCR has been used to identify and to detect DNAs extracted from various mycobacteria. However, despite the extraordinary enthusiasm surrounding this technique and the considerable investiment, PCR has not emerged from the developmental "trenches" in the passed several years. It may be a considerable lenth of time before clinical microbiology laboratories become PCR playgrounds because many details remain to be worked out.

  • In an automated industry PLC plays a central role to control the manufacturing system. Therefore, fault free operation of PLC controlled manufacturing system is essential in order to maximize a firm's productivity. On the contrary, distributed nature of manufacturing system and growing complexity of the PLC programs presented a challenging task of designing a rapid fault finding system for an uninterrupted process operation. Hence, designing an intelligent monitoring, and diagnosis system is needed for smooth functioning of the operation process. In this paper, we propose a method to continuously acquire a stream of PLC signal data from the normal operational PLC-based manufacturing system and to generate diagnosis model from the observed PLC signal data. Consequently, the generated diagnosis model is used for distinguish the possible abnormalities of manufacturing system. To verify the proposed method, we provided a suitable case study of an assembly line.
