Treatment of ramie leaf β-amylase for preliminary purification

  • The thermal properties of ramie leaf ${\beta}$-amylase (RBA) were examined to develop a novel process for enzyme purification. The thermostability of RBA extract prepared from ramie leaf powder was examined at various temperatures. RBA activity decreased slightly, whereas other carbohydrate-active enzymes, such as $\small{D}$-enzyme, were rapidly inactivated during 30 min incubation at $60^{\circ}C$. When the heat-treated extract was incubated with various substrates, maltose was produced exclusively as the major product, whereas the untreated crude extract produced maltose and other maltooligosaccharides. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis, fewer protein bands were observed for the heat-treated extract than the untreated extract, indicating that the thermostable RBA was partially purified and other thermolabile enzymes were eliminated. Thus, the treatment of the RBA extract at $60^{\circ}C$ for 30 min resulted in 5.4-fold purification with a recovery yield of 90%.

Dyeing Properties and Colorfastness of Direct-Dyed Ramie, Flax, and Cotton (모시, 아마, 면의 직접염료 염색에서 염색성과 염색 견뢰도에 대한 연구)

  • In this paper, deying properties and colorfastness of ramie, flax, and cotton fabrics for direct dyes were compared. When dyed in a same dyeing bath ratio, in case of green dyes, colorfastness to light was similar for three fabrics but in case of red dyes, cotton had a lower level. For colorfastness to laundering, three fabrics were similar and there were no consistent differences. The degree of stain about white fabric cotton was highest in case of red dyes. The degree of stain about white fabric wool showed the high resistance of stain and little stain was seen. This result represented that the used dyes were proper direct dyes for cellulosic fibers. For the difference of shade, the cotton was cosistently brighter and the shade of ramie was darker than of cotton and flax. This might mean that optical effects arouse from the comparatively large cross-sectional size of the ramie fiber and its highly ordered structure.

Weaving and Bleaching of Ramie/Silk Mixture Fabrics (모시/실크 교직물의 제작과 표백)

  • The weavability and bleaching of ramie/silk mixture fabrics were examined. When silk fiber was used as warp or weft in the mixture fabrics, the weaving properties became higher than that of ramile fabrics. Especially, the highest weavabiliy of mixture fabrics was obtained when used as warp silk fiber. The whiteness of ramie and tussah fiber treated with hydrogen peroxide, bleaching agent. was increased up to about 80% without significant changes of tenacity and elongation. As the increase of bleaching agent and treatment time, the whiteness of mixture fabrics was increased and the yellowness was decreased. Also, as the whiteness of mixture fabrics increased, the drape stiffness in warp direction was decreased but crease recovery was increased.

  • This study was performed to investigate the effect of Mentha arvensis Herba extract on the treatment of chromaticity and colorfastness. Mentha arvensis Herba has been used as a Korean medicine. It is effective in headache and stress. It is also good as a aphtha and in treating cold. In the long history of Korea, dyeing has been applied for a means representing the grace of natural and inner esthetic consciousness of man. Vegetable dyes give us such great benefits, diversified color, but no pollution. And ramie fabric has distinctive features such as beautiful brilliance, elegance, and strong durability. So, it is regarded as a special product of Korea traditionally. These studies were carried out to treat with acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was died with Mentha arvensis Herba extract. The results of experiment showed as follows: First, the chromophoric degree was the highest in acetate iron but not distinction in another mordants. Second, the light colorfastness was the highest in non treated and dichloride copper, but alum was the lowest. Third, the discoloration was alum and dichloride copper showed first grade in washing colorfastness. Abrasion colorfastness was not significant in this test. According the previous results, Mentha arvensis Herba has an efficiency in removing aphtha and in treating cold. So it is considered that Mentha arvensis Herba can be applied effectively to headache and stress.

Dyeing Property of Bamboo Leaves Extract on Hemp and Ramie Fiber (대나무잎 추출물에 의한 삼베와 모시섬유의 염색성)

  • Bamboo leaves, as natural dye provide unartificial fresh green, are known for expert property at anti-bacterial, anti-aging, anti-oxidation, skin whitening, moisturizing, peeling of horny, deodorizing and some skin disease like as eczema and inflammation without any allergic response. Also, It is quite common to obtain bamboo leaves from our surrounding but most of them were not in use. On purpose of development the way of recycle with common material to dye, bamboo leaves were used to dye on ramie fiber which is valuable for both of apparel and industrial field and investigated their dyeing property. Using extract from bamboo leaves, dyeing property was examined with dyeability, colorimetric property, light fastness and wash fastness under different conditions by extract density, dyeing time, dyeing method and mordants. As results, dyeing was generally colored in yellowish, K/S Value was shown more higher on hemp fiber than ramie. The most optimum dyeing condition was investigated on pre-mordant treatment, dyeing temperature $60^{\circ}C$, dyeing time 30min and extract density 400%. On the treatment with mordant(Al, Fe, Cu), dyebility on both of hemp and ramie fiber were increased by mordant. Especially with Copper(Cu) as mordant, dyebility was highly increased than any other mordants and dyeings showed in deep color. Also on the fastness test which is the most important to natural dyeing, wash-fastness showed good performance with 4-5 grade but light-fastness was above 4 grade on both of hemp and ramie fiber, lower than wash-fastness overall.

Eco-friendly, natural dyeing of linen, ramie, and hemp fabrics using Polygonum tinctoria (친환경을 위한 마직물(아마, 저마, 대마)의 쪽 천연염색에 관한 연구)

  • In this study, natural dyeing using Polygonum tinctoria was performed with linen, ramie, and hemp, which are biodegradable cellulose fibers, considering environmental aspects. In particular, the impacts of alkali NaOH and reducing agent Na2S2O4 were examined, and the possibilities of minimizing the use and reusing the dye were explored. The surface dye concentrations were found to be in the following order: hemp>linen>ramie. With the increase in all additives, the L* value decreased, and the ⊿E and K/S values increased gradually. When Na2S2O4 was 1g/L, the surface color of the dye appeared uniformly from the NaOH concentration of 0.4g/L (pH 10.84). When NaOH was 0.4g/L, the K/S values of linen and ramie increased rapidly after 0.4g/L of Na2S2O4, and hemp maintained a stable color from 0.6g/L of Na2S2O4. With the increase in the dye concentration from 1 to 6g/L, all the fibers were dyed uniformly. The K/S value increased or higher doubled upon repeated dyeing six times for 5 min than when dyed only once for 30 min. Therefore, the linen, ramie, and hemp fibers dyed repeatedly exhibited good washing, rubbing, and colorfastness to perspiration, which was rated between 4 and 4-5, and that to light was rated as 5. Moreover, no discoloration due to sunlight was observed. Finally, linen exhibited a bacterial reduction of 99.9%, thereby indicating its excellent antibacterial property.

Effects of Ramie Leaf according to Drying Methods on Antioxidant Activity and Growth Inhibitory Effects of Cancer Cells (건조방법에 따른 모시잎의 항산화 활성 및 암세포 증식 억제효과)

  • This study was carried out to discriminate the effects of the ramie leaf according to the drying methods (hot air drying and freeze drying) on antioxidative activity in vitro and antiproliferation in human cancer cells. There were no significant differences in total polyphenol content of ramie leaf ethanol extracts depending on the drying methods, but total flavonoid content was significantly higher in hot air dried ramie leaf (HR) than in freeze dried ramie leaf (FR). The DPPH radical scavenging activity of HR and FR ethanol extracts were found to be 77.74%, and 77.29% in 1000 ppm, respectively. Antioxidative index of HR and FR ethanol extracts measured by Rancimat were lower than those in BHT, BHA, and ascorbic acid, but were higher than that in control. The antiproliferation effect of 80% ethanol extracts of HR and FR on liver cancer cell line (H460), stomach cancer cell line (AGS), and lung cancer cell line (A549) were increased with a dose-dependent manner. The cancer cell growth inhibition activities of HR and FR ethanol extracts at the concentration of $800{\mu}g/mL$ showed greater than 80% on Hep G2 and A549 cell line, and greater than 75% on AGS cell line. These results suggest that HR and FR ethanol extracts possess potential antioxidative effect and antiproliferation in human cancer cells, and those activities of ramie leaf ethanol extracts depending on the drying methods were similar.

Dyeability of Ramie Fabrics Using Extract of the Native Plant of Rubia akane Nakai Grown in Korea (한국 자생 꼭두서니 추출물에 의한 모치섬유의 염색성)

  • This study was carried out to investigate into the dyeability of ramie fabrics using the extract of native plant, red-dye madder (Rubia akane Nakai) in different conditions. Surface color of ramie fabrics dyed with extracts from red-dye madder plants showed an order of descent YR. However, it was changed by the different mordants. Surface color of ramie fabrics treated with FeSO$_4$, SnCl$_4$, and tartaric acid was an order of descent Y, and that treated with the ashes of common camellia, and bean straw was an order of descent R. At higher temperatures, surface color and coloring matter concentration of ramie fabrics were as follows that values of a increased, however, values of b decreased and values of L lowered. Coloring matter concentration increased in the order of 7$0^{\circ}C$, 9$0^{\circ}C$, and 6$0^{\circ}C$, and so on. Dyeing colors on the ramie fabrics showed an order of descent Y by pH 4 and 9 in dyestuffs solutions. Coloring matter concentration in terms of K/S values, more or less, was increased at strong acids. Surface color by the concentration of dyestuffs solutions showed an order of descent YR irrelevant to the concentration of dyestuffs solutions. Coloring matter concentration was increased at higher concentrations of dyestuffs solutions. Ten minutes was sufficient for the dyeing of ramie fabrics in terms of surface color and coloring matter concentration.
