Uncoupling Protein 3 in the Rainbow Trout, Oncorhynchus mykiss Sequence, Splicing Variants, and Association with the AvaIII SINE element

  • A rainbow trout uncoupling protein 3 (UCP3) cDNA clone, encoding a 310 amino acid protein, was cloned and sequenced from a liver cDNA library. Two different splice variants designated UCP3-vl and UCP3-v2, were identified through liver cDNA library screening using rainbow trout UCP3 cDNA clone as a probe. UCP3-vl has 3 insertions in the UCP3 cDNA: the first insertion (133 bp), the second (141 bp), and the third (370 bp) were located 126 bp, 334 bp and 532 bp downstream from the start codon, respectively. UCP3-v2 contained a single insertion, identical in sequence and location to the second insertion of UCP3-vl. UCP3, a mitochondrial protein, functions to modulate the efficiency of oxidative phosphorylation. UCP3 has been detected from heart, testis, spinal cord, eye, retina, colon, muscle, brown adipose tissue and white adipose tissue in mammalian animals. Human and rodent UCP3s are highly expressed in skeletal muscle and brown adipose tissue, while they show weak expression of UCP3 in heart and white adipose tissue. In contrast to mammalian studies, RT-PCR and Southern blot analysis of the rainbow trout demonstrated that UCP3 is strongly expressed in liver and heart. UCP3, UCP3-vl, and UCP3-v2 all contain an Ava III short interspersed element (SINE), located in the 3'untraslated region (UTR). PCR using primers from the Ava III SINE and the UCP3 3'UTR region indicates that the UCP3 cDNA is structurally conserved among salmonids and that these primers may be useful for salmonid species genotyping.

  • This study examined the hatching rate of eyed-eggs and growth of fingerlings of USA strain rainbow trout, Oncorhynchus mykiss, which were transplanted on Nov. 17, 2005 from the USA for improvement of breeding. The diameter and weight of eyed-eggs were $4.95{\pm}0.34\;mm$ and $0.07{\pm}0.003\;g$, respectively. The eyed-eggs were hatched in 8 days later and hatching rates were $77.3{\sim}98.6%$. After 56 days from hatching, the fingerlings grew to $4.0{\pm}0.7$ cm and 0.62 g in total length and body weight, respectively. The relationships between total length (TL) and elapsed days (ED) were expressed as TL=0.0366ED+1.8645 ($r^2=0.928$, n=350). The relationships between body weight (BW) and elapsed days (ED) were expressed as following; $BW=0.0638e^{0.0408ED}\;(r^2=0.9917,\;n=350)$. And survival of rainbow trout was $64.5{\pm}1.3%$ in 56 days after hatching.

Determination of the dietary lysine requirement by measuring plasma free lysine concentrations in rainbow trout Oncorhynchus mykiss after dorsal aorta cannulation

  • This study evaluated the dietary lysine requirement by measuring the plasma free lysine concentrations in rainbow trout, Oncorhynchus mykiss after dorsal aorta cannulation. A basal diet containing 36.6 % crude protein (29.6 % crystalline amino acids mixture, 5 % casein and 2 % gelatin) was formulated to one of the seven L-amino acid based diets containing graded levels of lysine (0.72, 1.12, 1.52, 1.92, 2.32, 2.72 or 3.52 % dry diet). A total of 35 fish averaging $512{\pm}6.8g$ ($mean{\pm}SD$) were randomly distributed into seven groups with five fish in each group. After 48 h of feed deprivation, each group of fish was fed one of the experimental diets by intubation at 1 % body weight. Blood samples were taken at 0, 5 and 24 h after intubation. Post-prandial plasma free lysine concentrations (PPlys, 5 h after intubation) of fish fed diets containing ${\geq}2.32%$ lysine were higher than those of fish fed diets containing ${\leq}1.92%$ lysine. Post-absorptive free lysine concentrations (PAlys, 24 h after intubation) of fish fed diets containing 2.32 and 3.52 % lysine were higher than those of fish fed diets containing ${\leq}1.52%$ lysine. The brokenline regression analysis on the basis of PPlys and PAlys indicated that the lysine requirement of rainbow trout could be 2.34 and 2.20 % in diet. Therefore, these results strongly suggested that the dietary lysine requirement based on the broken-line model analyses of PPlys and PAlys could be greater than 2.20 % but less than 2.34 % (corresponding to be $6.01%{\leq},but{\leq}6.39%$ in dietary protein basis, respectively) in rainbow trout. Also, these results shown that the quantitative estimation of lysine requirement by using PPlys and PAlys could be an acceptable method in fish.

  • The meat samples of carp (Cyprinus carpio, Linne) and rainbow trout (Salmo gairdnerii) packaged in aluminum pouches with polyethylene adjuvant were exposed to gamma radiation of doses up to 1.5 Mrad for the purpose of determining optimum dose range required to bring about a significant storage-life extension at refrigerated temperatures. The maximum permissible dose for carp was determined to be 1.5 Mrad and that for rainbow trout 0.2 Mrad, while the optimum dose was 0.25 Mard and 0.05 Mrad, respectively. By irradiating them at each optimum dose, the practical storage-life of carp could be extended from one week to five at both $0^{\circ}\;and\;5^{\circ}C$ and that of rainbow trout from one week to 3-4 weeks at $0^{\circ}C$ and from 3 days to 14 days at $5^{\circ}C$. The carp meat suffered from extensive drip loss during the post-irradiation storage and it could be reduced effectively by dipping the samples into 10% polyphosphate solution prior to the radurization treatment. The rainbow trout was highly radiosensitive, while carp appeard to one of promising species to be radurization treated for the purpose of extending storage-life at refrigerated temperatures.

Marginal fidelity of zirconia core using MAD/MAM system (MAD/MAM을 이용한 치과용 지르코니아 코어의 변연 적합도)

  • Purpose: The purpose of this study was to evaluate the fit of zirconia core using MAD/MAM system comparing to that of conventional metal-ceramic and CAD/CAM system. Materials and methods: Duplicating the prepared resin tooth, 50 improved stone dies were fabricated. These dies are classified as a group of 5 to create the core. The groups were composed of metal-ceramic, $Cercon^{(R)}$, $Ceramill^{(R)}$, $Rainbow^{TM}$, and $Zirkonzhan^{(R)}$. Each core was cemented to stone die, and then, absolute marginal discrepancy was measured with microscope at a magnification of ${\times}50$. Statistical analysis was done with one-way ANOVA test and Tukey's HSD test. Results: The mean absolute marginal discrepancy for metal-ceramic was $51.97{\pm}23.38{\mu}m$, for $Cercon^{(R)}$ was $62.16{\pm}25.88{\mu}m$, for $Ceramill^{(R)}$ was $67.64{\pm}40.38{\mu}m$, for $Rainbow^{TM}$ was $125.07{\pm}42.19{\mu}m$, and for $Zirkonzhan^{(R)}$ was $105{\pm}44.61{\mu}m$. Conclusion: 1. Fit of margin was identified as in the order of metal-ceramic, $Cercon^{(R)}$, $Ceramill^{(R)}$, $Zirkonzhan^{(R)}$, and $Rainbow^{TM}$. 2. Absolute marginal discrepancy of the zirconia core that designed by MAD/MAM system had significant differences in order of $Ceramill^{(R)}$, $Zirkonzhan^{(R)}$, and $Rainbow^{TM}$. 3. The mean absolute marginal discrepancy between $Cercon^{(R)}$ and $Ceramill^{(R)}$ did not show significant differences.

Morphometric Comparison in Rainbow Trout, Oncorhynchus mykiss, Coho Salmon, Oncorhynchus kisutch, and their Allotriploid (무지개송어, 은연어 및 그 잡종3배체의 체형 비교)

  • For identification of morphometric characteristics in rainbow trout, Oncorhynchus mykiss, coho salmon, O. kisutch, and their allotriploid, twenty one body traits were measured, and the ratios of each trait to fork length, length from postorbital to the origin of caudal fin and head length were calculated. The ratios of fin length to fin height in anal, dorsal and adipose fins showed significant differences among these two species and their allotriploid $(p\leq0.01)$. Allotriploid could be distinguished easily from two parents species in the ratios of length of origin of ventral fin to fork length and anal fin length to anal fin height.

Membrane Hyperpolarization Increases cAMP to Induce the Initiation of Sperm Motility in Salmonid Fishes, Rainbow Trout and Masu Salmon

  • Sperm motility of salmonid fishes is suppressed by external $K^{+}$ and initiated by decrease of $K^{+}$ concentration surrounding the sperm. It was shown that the decrease in external $K^{+}$ concentration induced not only the initiation of sperm motility, but also hyperpolarization of the plasma membrane and synthesis of cAMP in the sperm of rainbow trout, steelhead trout, and masu salmon. Inhibitors of $K^{+}$ channels, especially voltage-dependent $K^{+}$ channels, inhibited these three reactions, and the inhibitions were abolished by subsequent addition of a $K^{+}$ ionophore, valinomycin, suggesting that $K^{+}$ efflux through the $K^{+}$ channel contributes to rapid changes in the membrane potential of sperm and cAMP synthesis, thereby resulting in the initiation of sperm motility of salmonid fishes.

  • The wood veneers were clearly rainbow-colored with natural dyes. As shown through Korean-style jacket with stripes of multi-colors beyond traditional obang colors (red, blue, yellow, black and white colors), eco-friendly coloring methods representing Korean colors familiar from old times could be used nobly by coloring natural wood veneer being raw material of wood products. In terms of industrialization, the study to manifest korean color, substituting chemical stains such as dye and pigment, would be necessary. In order to realize this purpose, the study about economical dyeing materials and characteristics, that is, mordant, dyeing and drying techniques showing environment-friendly coloring and high coloration level ought to be followed. In addition to this, investigating discoloration transition by fading test for interior and exterior uses would have to be carried out.
