방사선 방어효과 예측 가능한 면역증강 인삼 다당체의 활성인자 (Representative Parameter of Immunostimulatory Ginseng Polysaccharide to Predict Radioprotection)

  • 암 환자를 대상으로 방사선 치료빈도가 높아짐에 따라 다양한 작용 기전을 바탕으로 한 방사선 방어제 개발에 많은 노력을 경주하고 있다. 본 연구에서는 조혈, 면역세포의 증식과 다양한 싸이토카인의 분비를 증가시킴으로써 방사선 방어효능이 보고된 바 있는 인삼 다당체를 대상으로 화학적 구성성분과 각종 면역활성 지표 및 방사선 방어효능을 확인하고 가장 상관관계가 좋은 인자를 추출해내고자 하였다. 면역학적 활성으로는 임파구 증식능, 대식세포의 산화질소 분비능, AK세포 활성능을 검사하였으며 방사선 방어효과는 endogenous colony-forming unit(e-CFU)로 측정하였다. 인삼으로부터 추출된 다당체의 구성성분 중 galactose함량이 증가할수록 면역증강작용이 우수하였으며 임파구 증식능이 방사선 방어효과와 밀접한 관계가 있는 것으로 나타났다.

방사선조사 동물모델 이용 면역기능개선-생약복합물 헤모힘(HemoHIM)개발 (Development of A New Herbal Composition HemoHIM as An Immune-Improving Agent Using Irradiated Animal Models)

  • A new herbal composition. HemoHIM, was developed using irradiated animal models and was successfully applied as an immune-improving agent. In a view that the protection and recovery of immune, hematopoietic and self-renewal tissues are essential for radioprotective agents, HemoHIM was developed based on a novel combination of three edible herbs (Angelica Radix, Cnidii Rhizoma. Paeonin Radix) that meet all those requirements. HemoHIM significantly protected the immune and hematopoietic system and enhanced their recovery in y-irradiated mice. For the application of HemoHIM as a health functional food and a supplementary agent for the cancer patients, the efficacy of HemoHIM to improve the immune functions was further evaluated in immune-depressed animals and humans. Animal studies demonstrated that HemoHIM significantly improved the immune functions in cyclophosphamide-treated mice, aged mice, and dexamethasone-treated mice. In human studies, HemoHIM enhanced the immune activity and cytokine secretion in sub-healthy volunteers, and alleviated the severe leukocyre depression in cancer patients during radiation and chemotherapy. Based on these results, HemoHIM was approved by Korea FDA as a material of health functional food for immune function improvement and will be commercially available soon. This case of HemoHIM research and development suggested that irradiated animals can be good models for biological degenerations such as immune depression, self-renewal tissue damage, and aging for the development of biological modulators.

흰쥐의 자궁에 대한 애엽-모링가 혼합추출물의 방사선 방호효과 (Radiation Protection Effect of Mixed Extracts of Artemisia asiatica Nakai and Moringa oleifera Lam on Rats Uterus)

  • 본 연구의 목적은 항산화 식품으로 알려진 애엽과 모링가를 적절한 비율로 혼합하여 제조한 추출물의 방사선방호제 개발 가능성을 검토하였다. 애엽-모링가 혼합 추출물을 암컷 SD Rat 2주간 경구 투여 후 7 Gy 방사선을 조사하여 1일 7일, 21일 후에 혈구 성분의 변화, SOD활성 및 자궁의 조직 변화를 관찰하였다.AM + IR Group이 IR Group에 비하여 백혈구(p<0.05) 및 혈소판(p<0.05) 수치 회복 경향이 더 높은 것으로 확인하였다. 또한 SOD 활성을 증가시켰으며 자궁 조직에서는 세포사가 감소한 것을 확인 하였다.이 결과를 토대로 A와 B의 혼합추출물은 방사선 피폭으로 인한 혈구 및 자궁피해를 감소시킬 수 있는 방사선 방호제로서 유용할 것으로 기대된다.

타우린의 뼈 형성 작용과 관련질환의 경감 (Taurine in Bone Formation and Alleviation of Its Diseases)

  • Taurine, amino acid, chemically known as 2-amino ethane sulphonic acid was discovered more than two hundred years ago from ox bile. it is widely distributed in both mammals and nonmammals. It is found in considerably high amount in hUl11an: a normal adult of 70 kgs contains about 70 grams of taurine. Taurine with this much concentration, is involved in almost all life processes. Its deficiency causes several abnormalities in major organs like brain, eye and heart. Taurine-bone interaction is latest addition to its long list of actions. In bone cells, taurine is also found in high concentration. Taurine is found to help in enhancing the bone tissue formation which is evidenced by increased matrix formation and collagen synthesis. Besides stimulating the bone tissue formation, it also inhibits the bone loss through inhibiting the bone resorption and osteoclast formation. Thus, taurine acts as a double agent. In addition to these two major actions of taurine in bone, it also has beneficial effect in wound healing mld bone repair. Taurine possess radioprotective properties, too. As it is a naturally available molecule, it can be used as a preventive agent. Taurine has a potential to replace bisphosphonates which are currently in use for the inhibition of bone loss but this needs in depth study. As taurine is involved in bone formation and inhibition of bone loss, a detailed study can make it a single marker of bone metabolism. All these taurine-bone interaction is a symbol of their deep involvement but still require further extension to make taurine as a choice for tile sound bone health.

Protective effect of the methanol extract of Polyopes lancifolia (Harvey) kawaguchi et wang against ionizing radiation-induced mouse gastrointestinal injury

  • The radioprotective efficacy of a methanol extract of the red algae Polyopes lancifolia (Harvey) kawaguchi et wang (mPL) was evaluated in mice subjected to total-body gamma irradiation. mPL protection against radiation-induced oxidative stress was examined by histological evaluation of intestinal crypt-cell survival and liver activities of the antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT). mPL (100 mg/kg body weight) administered intraperitoneally at 24 h and 1 h prior to irradiation protected jejunal crypt cells from radiation-induced apoptosis (p < 0.01). The pretreatment of mPL attenuated a radiation-induced decrease in villous height (p < 0.05), and improved jejunal crypt survival (p < 0.05). The dose reduction factor was 1.14 at 3.5 days after irradiation. Treatment with mPL prior to irradiation resulted in significantly higher (p < 0.01) levels of SOD and CAT activities, compared to those levels of irradiated control mice with vehicle treatment. These results suggest that mPL is a useful radioprotective agent capable of defending intestinal progenitor cells against total-body irradiation, at least in part through mPL antioxidative activity.

Effect of Ginseng on Calretinin Expression in Mouse Hippocampus Following Exposure to 835 MHz Radiofrequency

  • Exponential rise in the use of mobile communication devices has generated health concerns due to radiofrequency (RF) exposure due to its close proximity to the head. Calcium binding proteins like calretinin regulate the levels of calcium ($Ca^{2+}$) which plays an important role in biological systems. Ginseng is known for maintaining equilibrium in the human body and may play a beneficial radioprotectant role against electromagnetic field (EMF) exposure. In the present study, we evaluated the radioprotective effects of red ginseng (RG) extract in a mouse model. Calretinin (CR) expression was measured using a free-floating immunohistochemical method in the hippocampus of mice after 835 MHz EMF exposure for 5 h/d for 5 d at specific absorption rate=1.6 W/kg for the different experimental groups. The control animals were treated with NaCl while the experimental animals received 10 mg/kg ginseng, or 30 mg/kg; EMF exposed mice were also treated with NaCl, 10 mg/kg ginseng (E10), or 30 mg/kg (E30). Decreases in CR immunoreactivity (IR) along with loss of CA1 and CA3 interneurons and infragranular cells were observed in the ENaCl group while such losses were not observed in the E10 and E30 groups. CR IR significantly increased in the RG-treated group compared to control and EMF-exposed groups treated with NaCl. The study demonstrates that RG extract can serve as a radioprotective agent that maintains $Ca^{2+}$ homeostasis and prevents neuronal loss in the brain hippocampal region caused by RF exposure.

흰쥐의 전립선에 대한 셀레늄(Se)의 방사선 방호효과 (Radiation Protection Effect of Selenium on the Rat's Prostate)

  • 첨단 의료 장비의 보급으로 의료 분야의 방사선 활용도가 증가하면서 천연물을 활용한 방사선 방호제 연구는 사회적으로 중요한 과제가 되고 있다. 천연물인 셀레늄(Se)이 전립선에서 높게 발현되며 전립선 세포에 필수적인 역할을 한다는 것으로 알려져 있다. 전립선 조직을 대상으로 셀레늄에 의한 방사선 방호 효과를 연구하기 위하여 10 Gy의 방사선을 조사 시킨 후 1, 7, 21일 기간에 따른 혈구성분 및 항산화효소(Superoxide Dismutase; SOD) 활성 변화와 조직학적 변화를 관찰하였다. 방사선조사군(Rad)에 비해 셀레늄 투여 후 방사선조사군(Se+Rad)에서 조혈면역계의 손상을 경감시키는 유의한 방호 효과가 있었다(p<0.05). 셀레늄이 항산화효소인 Superoxide Dismutase(SOD)의 활성을 증가시키는 유효한 성분이며, 방사선 조사에 의한 전립선비대증의 발현을 억제하는 효과가 있음을 확인하였다. 셀레늄이 부득이하게 수반되는 방사선 피폭으로 인한 전립선 관련 질병의 예방과 방사선 방호제로서 유용성이 있을 것이라 사료된다.

  • A series of organosulfur compounds were synthesized with the aim of developing chemopreventive compounds active against hepatotoxicity and chemical carcinogesis. 2-(Allylthio) prazine (2-AP) was effective in inhibiting cytochrome P450 2E1-mediated catalytic activities and protein expression, and in inducing microsomal epoxide hydrolase and major glutathione S-transferases. 2-AP reduced the hepatotoxicity caused by toxicant sand elevated cellular GSH content. Development of skin tumors, pulmonary adenoma and aberrant crypt foci in colon by various chemical carcinogens was inhibited by 2-AP pretreatment. Anticarcinogenic effects of 2-AP at the stage of initiation of tumors were also observed in the aflatoxin B1 ($AFB_1$)-induced three-step medium-term hepatocarcinogenesis model. Reduction of $AFB_1$-DNA adduct by 2-AP appeared to result from the decreased formation of $AFB_1$-8,9-epoxide via suppression of cytochrome P450, while induction of GST 2-AP increases the excretion of glutathione-conjugated $AFB_1$ . 2-AP was a radioprotective agent effective against the lethal dose of total body irradiation and reduced radiation-induced injury in association with the elevation of detoxifying gene expression. 2-AP produces reactive oxygen species in vivo, which is not mediated with the thiol-dependent production of oxidants and that NF-KB activation is not involved in the induction of the detoxifying enzymes. the mechanism of chemoprotection by 2-AP may involve inhibition of the P450-mediated metabolic activation of chemical carcinogens and enhancement of electrophilic detoxification through induction of phase II detoxification enzymes which would facilitate the clearance of activated metabolites through conjugation reaction.

감마선을 조사한 마우스의 조혈 및 소장줄기세포에 대한 fucoidan의 방호효과 (Radioprotective effect of fucoidan against hematopoietic and small intestinal stem cells of γ-ray irradiated mice)

  • We investigated the potential of fucoidan for its ability to provide protection from gamma rayinduced damage. In our results, the fucoidan significantly improved the counts of endogenous colony forming unit to $9.5 {\pm} 1.5$, from $5.5 {\pm} 2.5$ compared with un-treated irradiated control group at 10 day after 7 Gy whole body irradiation. After 2 Gy irradiation, fucoidan treatment attenuated the percent of tail DNA of splenocytes, parameters of DNA damage, from $30.17 {\pm} 1.7%$ to $13.67 {\pm} 2.81%$ 2.81% by comet assay and also accelerated the proliferation of splenocytes, compared with un-treated irradiated control group by 3Hthymidine incorporation assay. Furthermore, fucoidan decreased the number of apoptotic fragments per intestinal crypt by 31.8% at 1 days after 2 Gy irradiation. These results indicated that the fucoidan significantly improved the hematopoietic recovery, prevented the DNA damage in immune cells and enhanced their proliferation, which had been suppressed by ionizing radiation. in addition, fucoidan rescued intestinal cells from radiation-induced apoptosis. Thus, this study raises the possibility of using fucoidan as adjuvant therapeutic agent after radiotherapy.

Curcumin Attenuates Radiation-Induced Inflammation and Fibrosis in Rat Lungs

  • A beneficial radioprotective agent has been used to treat the radiation-induced lung injury. This study was performed to investigate whether curcumin, which is known to have anti-inflammatory and antioxidant properties, could ameliorate radiation-induced pulmonary inflammation and fibrosis in irradiated lungs. Rats were given daily doses of intragastric curcumin (200 mg/kg) prior to a single irradiation and for 8 weeks after radiation. Histopathologic findings demonstrated that macrophage accumulation, interstitial edema, alveolar septal thickness, perivascular fibrosis, and collapse in radiation-treated lungs were inhibited by curcumin administration. Radiation-induced transforming growth factor-${\beta}1$ (TGF-${\beta}1$), connective tissue growth factor (CTGF) expression, and collagen accumulation were also inhibited by curcumin. Moreover, western blot analysis revealed that curcumin lowered radiation-induced increases of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), TNF receptor 1 (TNFR1), and cyclooxygenase-2 (COX-2). Curcumin also inhibited the nuclear translocation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) p65 in radiation-treated lungs. These results indicate that long-term curcumin administration may reduce lung inflammation and fibrosis caused by radiation treatment.
