Scintillation Proximity Assay (섬광근접측정법)

  • Scintillation proximity assay (SPA) is a unique type of radioimmunoassay and makes it possible to use radioisotopes for monitoring binding reactions continuously without separation procedure. Microbeads containing a fluorophor are covalently linked to antibody or receptor. When a radiolabeled antigen or ligand is added it binds to the beads and the emitted short range electrons, excite the fluorophor in the beads. The light emitted can be measured in a scintillation counter. $^3H$ or $^{125}I$ has been used for SPA. The sensitivities achieved with SPA are comparable to the sensitivities of other procedures. SPA is applicable to immunology, receptor binding, monitoring interactions of biomolecules and study for the kinetics of interaction between receptors and ligands.

  • Utilizing the commercial radioimmunoassay kit, the author assayed HGH and evaluated the problems of the method. The method had the sensitivity of 0.5 mug/ml degree and could determine the plasma HGH concentration directly without the help of plasma extraction. Also it was specific for the HGH, when tested by a dilution method using test utilizing the plasma of the acromegalic patient. The author also obtained the $^{125}I$ labelled HGH of specific activity of $156.3{\mu}Ci/{\mu}g$, performing the chloramine-T method.

  • The established methods in the residue analysis of endosulfan require an extensive sample clean-up prior to quantification by relatively complex equipment. A radioimmunoassay(RIA) provides a simple procedure with theoretically higher sensitivity and specificity necessitating only a minimum of sample clean-up. Endosulfan-specific antibodies were developed in rabbits by using a bovine serum albumin(BSA) conjugate wherein the alcohol form of endosulfan was multiply bound to the protein via succinylation. Produced antibodies showed the high titers to endosulfan-BSA(1 : 32,000). An RIA method was developed in water and carp by using $^{14}C-labeled$ endosulfan as a tracer. The lowest detection amount of endosulfan was 1 ng in the liver, kidneys, gut and water samples, and 3 ng in the whole body sample of carp without any clean-up, corresponding to 0.1 ppb of endosulfan.

  • The radioimmunoassay of human thyrotropin was performed in various thyroid states, utilizing the anti-h-T.S.H. antibody and purified human thyrotropin supplied from National Institute of Arthritis and Metabolic Diseases, Bethesda, Ma., U.S.A., and human thyrotropin standard-A obtained from National Institute for Biologic Standards, Mill Hill, London, England. $^{131}I$ labelled h-TSH was prepared after the Chloramine-T method of Greenwood et al. This double antibody system had a assay sensitivity of about $1.0{\mu}U/ml$ of plasma HTS-A and could detect the plasma h-TSH level in the euthyroid patients. Plasma h-TSH level of the normal 26 Korean was $1.1{\pm}0.83{\mu}U/ml$, and that of the 8 hypothyroidisms were 8.3 to $67.5{\mu}U/ml$. In hyperthyroidisms, no cases showed the plasma h-TSH levels over $1.0{\mu}U/ml$. Between the hypothyroidism and euthyroidsm, no overlap is noticed on plasma h-TSH levels. A case of transient hypothyroid state identified by determination of plasma h-TSH level is presented. These results revealed that the radioimmunoassay of h-TSH in plasma could be a sensitive method to diagnose the hypothyroidsm, if not caused by a pituitary disease.

  • A radioimmunoassay technique has been developed for the quantitative analysis of Abscisic acid (ABA). The antibody, obtained by immunizing rabbits against a conjugate of ABA with human serum albumin, had a high affinity (Ka=3.28x10$\^$13/l/mol) for ABA. The use of $^3$H-labelled ABA as tracer and of dextran-coated charcoal for separation of free ABA from antibody-bound ABA permitted detection of as little as 0.5x10$\^$-12/ mol ABA. The measuring range extended to 14x10$\^$-12/ mol. Because of the high specificity of this immunoassay, no extract purification steps were required prior to analysis. And then, only 2 hr in radioimmunoassay was required to ABA analysis. From these results, it is suggested using this assays that more than hundreds samples can be assayed sensitive and simple per day for ABA.
