Fucoidan의 Oxy-radical 포획능 평가 (Evaluation of Oxy-radical Scavenging Capacity of Fucoidan)

  • Algal fucoidan is a marine polysaccharide containing sulfur with a wide variety of biological activities including anti-inflammatory and anti-thrombotic effects. Although antioxidants can inhibit inflammatory signals through inhibiting activator protein-1 and/or nuclear factor-kappaB activation, it is obscure whether fucoidan directly scavenges oxy-radicals or indirectly regulates oxidant production and/or antioxidant defense system. The antioxidant activities of fucoidan against peroxyl radicals, peroxynitrites and hydroxyl radicals were determined by the total oxy-radical scavenging capacity (TOSC) assay. The specific TOSC values of fucoidan against peroxyl radicals, peroxynitrites or hydroxyl radicals were $282{\pm}60$, $43{\pm}1$ or $40{\pm}1\;TOSC/mg/mL$, respectively. These specific TOSC values against peroxyl radicals, peroxynitrites or hydroxyl radicals are 23, 12, or 13% of the specific TOSC values of glutathione, a positive control, respectively. These results suggest that fucoidan has direct oxy-radical scavenging capacity, which may be related with anti-inflammatory effect of fucoidan.

  • Although taurine (2-aminoethanesulfonic acid) can inhibit oxidative stress in both animal and epidemiological studies, it is obscure whether taurine directly scavenges oxy-radicals or indirectly regulates oxidant production and/or antioxidant defense system. The reason for this discrepancy remains unknown but may be due, in part, to the lack of a validated assay system for evaluating oxy-radical scavenging capacity. The antioxidant activities of taurine and hypotaurine (2-aminoethanesulfinic acid), a precursor of taurine, against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay and cell-based assay using H4IIE cells. tert-Butylhydroperoxide or hydrogen peroxide-induced cell toxicity determined by MTT assay was markedly inhibited by 10mM taurine or hypotaurine. The tert-butylhydroperoxide- or hydrogen peroxide-induced changes in oxidative stress markers, such as cellular glutathione and malondialdehyde, were ameliorated by 10mM taurine or hypotaurine. However, specific TOSC values calculated from the slope of the linear regression for taurine against peroxyl radicals, hydroxyl radicals or peroxynitrites were all less than 1 TOSC/mM. On the other hand specific TOSC values for hypotaurine against peroxyl radicals, hydroxyl radicals or peroxynitrites were 48, 2096, or 69 TOSC/mM, respectively. These results suggest that taurine protects cells against oxidative insults, which is not ascribed to directly scavenging activity of taurine against oxy-radicals. These results support the idea that the oxidation state of sulfur in antioxidants may be a determinant of oxy-radical scavenging capacity.

The Scavenger Effects of Various Antioxidants in Cigarette Filters on the Free Radicals in Mainstream Smoke

  • This study was conducted to evaluate the effect of additives (antioxidants for free radicals reduction) in cigarette filter treated with various antioxidants (three types of proanthocyanidins and ascorbic acid) and various concentrations of ascorbic acid and loaded with activated carbon on the delivery of free radicals of mainstream smoke (MS) by ESR. Also, we analyzed Hoffmann's analytes andscavenger activity according to the storage time and in vitro cytotoxicity. The analysis of spin number of vapor and particulate phase free radicals in MS are decreased to $14{\sim}24\;%$ and $16{\sim}40\;%$, respectively. As aresult of antioxidant potential for inactivity of vapor and particulate phase free radicals, natural antioxidants were more effective than ascorbic acid. Based on the result of the analysis of Hoffmann's analytes for various antioxidantstreated cigarette filters during the smoking, cigarette filter treated with ascorbic acid showed the lower amount of the deliveries of hydroquinone, isoprene and quinoline in MS than those treated with the other antioxidants. In the significant t-test on the difference of the cytotoxicity among the various antioxidants treated-cigarette filters, there are no significant differences at the 95 % confidence level. Those results indicated that the antioxidants were useful for reducing free radicals in MS because of the fast reaction between antioxidant and free radicals.

Vibronic Spectroscopy of Jet-Cooled Benzyl-type Radicals Produced from 2-Fluoro-4-Chlorotoluene by Corona Discharge

  • A home-made pinhole-type glass nozzle was employed to generate vibronically excited but jet-cooled benzyl-type radicals from precursor 2-fluoro-4-chlorotoluene with a large amount of carrier gas He, from which the visible vibronic emission spectrum was recorded with a long-path monochromator. From an analysis of the spectrum observed, it was found that two benzyl-type radicals, 2-fluorobenzyl and 2-fluoro-4-chlorobenzyl radicals, were formed from the precursor in corona discharge. The possible pathway for the production of benzyl-type radicals that can explain the spectroscopic observation is herein proposed. In addition, the electronic energy of the $D_1{\rightarrow}D_0$ transition and the vibrational mode frequencies in the $D_0$ state of the 2-fluoro-4-chlorobenzyl radical were determined for the first time.

Spectroscopic Identification of Isomeric Trimethylbenzyl Radicals Generated from 1,2,3,4-Tetramethylbenzene

  • The visible vibronic emission spectrum was recorded from the corona discharge of precursor 1,2,3,4-tetramethylbenzene with a large amount of inert carrier gas helium using a pinhole-type glass nozzle coupled with corona excited supersonic expansion. The spectrum showed a series of vibronic bands in the $D_1{\rightarrow}D_0$ electronic transition of jet-cooled benzyl-type radicals formed from the precursor in a corona excitation. The analysis confirmed that two isomeric radicals, 2,3,4- and 2,3,6-trimethylbenzyl radicals, were produced as a result of removal of a hydrogen atom from the methyl group at different substitution positions. For each isomeric product, the electronic transition and a few vibrational mode frequencies were determined in the ground electronic state.

  • Isomeric trimethylbenzyl radicals were generated and vibronically excited from precursor 1,2,3,5-tetramethylbenzene, isodurene, with a large amount inert carrier gas helium in a corona excited supersonic expansion (CESE) using a pinhole-type glass nozzle. A long-path monochromator was used to record the visible vibronic emission spectra of the jet-cooled benzyl-type radicals in the $D_1{\rightarrow}D_0$ electronic transition. From the analysis of the spectra, we identified the evidence of the presence of three isomeric trimethylbenzyl radicals in the corona discharge, and obtained the electronic energy and a few vibrational mode frequencies in the ground electronic state for each isomer.

  • This study was undertaken to examine the effect of Cheongahwan(CAH), being known to reinforce Kidney-yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the kidney tissues. Alterations in enzyme activities were observed after in vivo treatment in rats. CAH caused a significant increase in the activities of superoxide dismutase (SOD), glutathione peroxidase and glutathione S-transferase. But catalase activity was not significantly altered by CAH. Treatment in vitro of CAH decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that CAH stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of CAH may contribute to prevent the oxygen free radical-induced impairment of cell function.

On n-Amitsur Rings

  • The concepts of an Amitsur ring and a hereditary Amitsur ring, which were introduced and studied by S. Tumurbat in a recent paper, are generalized. For a positive integer n, a ring A is said to be an n-Amitsur ring if γ(A[Xn]) = (γ(A[Xn]) ∩ A)[Xn] for all radicals γ, where A[Xn] is the polynomial ring over A in n commuting indeterminates. If a ring A satisfies the above equation for all hereditary radicals γ, then A is said to be a hereditary n-Amitsur ring. Characterizations and examples of these rings are provided. Moreover, new radicals associated with n-Amitsur rings are introduced and studied. One of these is a special radical and its semisimple class is polynomially extensible.

  • Gas phase and particulate phase radicals in mainstream cigarette smoke were determined Electron Spin Resonance(ESR) spectroscopy. The free radicals in particulate phase have been investigated by benzene extract of Cambridge Filter Pad containing the smoke condensate. Spin trapping method in conjunction with ESR was used to investigate free radicals in the gas phase of cigarette smoke. Several analytical experiments were conducted in order to determine the optimal conditions for maximum signal intensities and reproducibility of results. All the tests were optimized and normalized using the University of Kentucky 2R4F reference cigarette. The optimal conditions were 0.6 mL for analysis volume of ESR, $4{\sim}5\;mL$ for collection volume of spin-adducts, and PBN for quantification of free radicals in gas phase. The radical levels of Kentucky 2R4F cigarettes were found $2.18{\times}10^{14}\;spins/cig.$ and $2.10{\times}10^{15}\;spins/cig.$ in gas phase.

Purification, Chemical Composition, and in vitro Antioxidant Activity of Two Protein-bound Polysaccharides from Rapeseed Meal

  • Crude polysaccharides from rapeseed meal (PRM) were extracted with 0.3% NaOH aqueous solution, followed by further purifications and 2 fractions, namely PRM1 and PRM2, were separated with a DEAE-cellulose DE-52 column. Their primary compositions were analysed and antioxidant activity was determined, including scavenging activity toward superoxide anion radicals, hydroxyl radicals, and nitric oxide radicals, reducing power, and inhibitory effects against the microsomal lipid peroxidation, compared to that of L-ascorbic acid. The results indicated that PRM1 and PRM2 exhibited not only good reducing power and inhibitory effects on the microsomal lipid peroxidation, but also strong scavenging activity toward superoxide anion radicals, nitric oxide radicals, and hydroxyl radicals. In addition, positive correlations were also observed between the superoxide anion radical scavenging activity and the protein contents of the polysaccharides, and the reducing power and the sulfate contents. These findings thus clearly suggest the polysaccharides possess direct and potent antioxidant activity.
