Assessment on Antioxidant Potential and Enzyme Activity of Some Economic Resource Plants

  • The antioxidant potential and enzyme activities in Salicornia herbacea, Corylopsis coreana, Erythronium japonicum, Phragmites communis, Momordica Charantia, Nelumbo nucifera, Salvia plebeia, Portulaca oleracea, Ficus carica, Citrus junos and Cornus officinalis were determined. Their antioxidant activities were measured using DPPH radical scavenging and nitrite scavenging activity. Enzyme activities in investigated plants were evaluated as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX). The DPPH scavenging rate from 100 to 2500 $mgL^{-1}$ was the highest in the flower of Corylopsis coreana. However, it was not detected in most of the samples at concentration below 100 $mgL^{-1}$. The nitrite scavenging activity according to each kind of resource plants was significantly higher in the stem of Corylopsis coreana and leaf of Nelumbo nucifera. The root extract of Erythronium japonicum had the highest SOD enzyme activity of 94.0% while leaf of Salvia plebeia showed the lowest SOD enzyme activity of 30.4%. The activity of CAT and APX showed higher values in the stem of Corylopsis coreana, root of Erythronium japonicum and root of Phragmites communis in comparison with other plants. The activity of POD showed significantly high values in stem of Corylopsis coreana, Momordica Charantia and pericarp of Citrus junos extracts. The antioxidant enzyme activities differ significantly in different plants. In conclusion, we showed that Corylopsis coreana, Erythronium japonicum Cornus officinalis, and Momordica Charantia had the potent biological activities. Therefore, these plant resources showing antioxidant activity could be good materials for development of source of functional healthy food.

In Vitro and Intracellular Antioxidant Activities of Brown Alga Eisenia bicyclis

  • The antioxidant activities of a methanolic extract of Eisenia bicyclis and its organic solvent fractions, including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), n-butanol (n-BuOH), and water ($H_2O$) fractions, were investigated. Scavenging activities against DPPH, hydroxyl, superoxide anion, and peroxynitrite radicals were evaluated using electron spin resonance spectrometry; intracellular reactive oxygen species (ROS) were evaluated by a 2',7'-dichlorofluorescein diacetate assay using RAW264.7 mouse macrophages. The antioxidant activities of the individual fractions were: EtOAc>n-BuOH>$CH_2Cl_2$ >$H_2O$. The EtOAc fraction exhibited strong radical scavenging activity and a significantly reduced ROS level in RAW264.7 cells. Moreover, the phenolic contents of the extract and fractions followed the same order as their radical scavenging activities. Our results indicate that E. bicyclis is a valuable natural source of antioxidants that may be applicable to the functional food industry.

국내산 밤 일부 품종의 기능성 성분분석과 항산화 및 대식세포 활성 (Antioxidative and Macrophage Phagocytic Activities and Functional Component Analyses of Selected Korean Chestnut (Castanea crenata S. et Z.) Cultivars)

  • 국내산 밤 일부 품종(단택, 대보, 석추, 옥광, 병고)의 기능성 성분 분석과 이들 물질들의 항산화력 및 밤 추출물이 대식세포 활성에 미치는 영향력을 알아보았다. 국내산 밤 일부 품종의 내피와 과육에서 coumarin, gallic acid 그리고 catechin이 HPLC에 의해 검출되었다. 밤 내피에 가장 풍부한 기능성 성분은 catechin이고 그 다음 gallic acid와 coumarin 순이었으나, 과육에서는 gallic acid만 검출되었다. 따라서 밤의 기능성 성분인 gallic acid와 catechin의 항산화력을 DPPH radical 소거작용과 SOD 유사활성을 통해 알아보았다. Gallic acid과 catechin을 6.0 mg/100 g 처리하였을 때 DPPH radical 소거작용이 각각 69.4%와 38.3%였으며, gallic acid의 DPPH radical 소거작용은 농도 의존적이었다. Gallic acid를 동일한 농도의 catechin과 비교하면 모든 농도 구간에서 catechin보다 gallic acid가 높은 DPPH radical 소거활성을 나타내었다. 그러나 SOD 유사활성은 gallic acid보다 catechin이 높았다. 더하여 밤의 과육, 노란과육 및 흰과육 추출물들이 대식세포 활성에 미치는 영향력을 알아보았다. 병고 흰과육 추출물은 현저하게 대식세포 활성을 증가시켰고, 병고, 단택, 대보 및 옥광의 노란과육 추출물 역시 아무 것도 처리하지 않은 세포와 비교하여 현저히 대식세포를 활성화시켰다. 노란과육 추출물이 흰과육 추출물보다 더욱 더 대식세포 활성을 증가시켰고, 5품종 중 병고가 대식세포 활성을 가장 증가시켰다.

콩의 폴리페놀, 피테이트 함량과 항산화능과의 관계 (Polyphenol and Phytate Contents and Their Relationship to Antioxidative Activity in Soybeans)

  • In this study we evaluated polyphenol contents, phytate contents, and antioxidant activities (DPPH radical and hydroxyl radical scavenging activity) in several soybean cultivars. The polyphenol contents of the cultivars ranged from 0.0446 mg/g to 0.0652 mg/g, with the highest level found in the WS82 cultivar, whereas and the lowest in Duyukong. The phytate content of the WS2906 cultivar was the highest at 22.28 mg/g whereas Duyukong showed the lowest level. $IC_50$ values (concentration of soybean extract exerting 50% scavenging of radicals) for DPPH radical and hydroxyl radical scavenging activity showed the highest in WS82. A positive correlation between DPPH radical scavenging activity and polyphenol contents was observed in the soybean cultivars, whereas not significant relationship between antioxidant activity and phytate content was found.

Antioxidant Compounds from Distylium racemosum Leaves

  • The leaves of D. racemosum showed strong DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and the order of the radical scavenging activity against DPPH radical is ethyl acetate (EtOAc) fraction>crude extracts>residue fraction>hexane fraction>ether fraction, under the experimental conditions. Since EtOAc fraction has highest antioxidative activity among these fractions, the isolation was performed from the EtOAc fraction of the leaves of D. racemosum and four phenolic compounds were isolated and identified as follows: methyl gallate, kaempferol, quercetin and quercitrin. The free radical scavenging activities of these compounds were 79.9%, 93.1%, 93.6% and 66.7% at 10 ㎍/ml, respectively. The IC50 of compound 1, compound 2, compound 3 and compound 4 were 6.1, 4.1, 3.6 and 6.5 ㎍/ml, respectively. These compounds have higher antioxidative activity compared with reference compounds, ascorbic acid (IC50 = 9.6 ㎍/ml).

동백 유박 에탄올추출물 및 분획물의 항산화 활성 (Anti-oxidant activities of ethanol extract and fractions from defatted Camellia japonica L. seeds)

  • 본 연구는 동백나무(Camellia japonica L.) 유박 추출물 및 분획물의 in vitro 항산화 활성에 대해 조사하였다. 동백나무 유박은 에탄올을 사용하여 추출하였으며, 이 후 부탄올(BuOH), 에틸아세테이트(EtOAc), 클로로포름 및 헥산을 이용하여 분획물을 조제하였다. 동백 유박 추출물 및 분획물의 항산화 활성을 평가하기 위해 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), hydroxyl radical, superoxide anion (O2-) radical 소거능을 측정하였다. 동백 유박 추출물 및 분획물은 농도의존적으로 DPPH, ABTS+ 및 O2- radical 소거능을 증가시켜 항산화 활성을 나타내었다. 특히 동백 유박 BuOH 분획물은 다른 추출물 및 분획물에 비해 가장 높은 radical 소거능을 나타내었다. 동백 유박 BuOH 분획물의 총 폴리페놀 및 플라보노이드 함량은 각각 23.26 mg GAE/g 및 32.39 mg QE/g이었으며, 이는 다른 추출물 및 분획물보다 높은 수치임을 알 수 있었다. 동백 유박 BuOH 분획물 및 EtOAc 분획물의 cameliaside B 함량은 각각 102.37, 165.05 ㎍/g임을 확인하여 다른 추출물 및 분획물에 비해 높은 수치를 나타냄을 알 수 있었다. 따라서 동백 유박 추출 및 분획물 중에서 BuOH 분획물 및 EtOA c 분획물은 다른 추출 및 분획물에 비해 radical 소거능이 우수할 뿐 아니라 항산화 물질 함유량이 높아, 항산화 기능성 소재로 활용될 수 있을 것으로 사료된다.

유산균의 항산화 활성 비교 (Comparative Study of Lactic Acid Bacteria for Antioxidative Activities)

  • Lactic acid bacteria (LAB) are typical probiotic microbes which are used in various industries including fermented foods, feed additives, and pharmaceuticals. The purpose of this study was to compare the ability of antioxidative activities of 23 strains of LAB isolated from cheese, kimchi, yogurt, and etc. LAB were tested for antioxidative activities such as 2,2'- azino-bis(3-ethyl-benzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities and superoxide dismutase (SOD)-like ability. Antioxidative activities were examined different media such as MRS, M17, and 10% skim milk. Culture supernatant in M17 medium showed a higher antioxidative activity than these in MRS and 10% skim milk. Comparison of ABTS radical scavenging ability by the LAB showed a wide variation with a range from 9~93% and SOD-like ability of LAB ranged from 70~78%, respectively. Therfore, some of LAB can play important roles in the protection for oxidative stress and CPP-producing activity of LAB may be a promising material for application in the dairy industry.

전호(Anthriscus sylvestris) 뿌리 정유의 항균 및 항산화 작용 (Antibacterial and Antioxidant Activities of the Essential Oil from the Roots of Anthriscus sylvestris)

  • To develop a new effective and safe natural antibiotics and antioxidant the essential oil was extracted from the roots of Anthriscus sylvestris by steam distillation. Its composition was analyzed by GC-MS. The activities of the essential oil fraction and its main components were evaluated against antibiotic-susceptible and -resistant strains of some food-born bacteria. In addition the synergism was examined with this oil combined with antibiotic by checkerboard titer test. The antioxidant activities were determined by in 1,1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging activity test and reducing power assay. The essential oil fraction of A. sylvestris revealed significant inhibiting activities against antibiotic-susceptible and -resistant species of Vibrio and Shigella with MICs ranged from 1.00~4.00 mg/ml. It showed synergistic or additive effects when it was combined with amphicillin or trimethoprim/sulfamethoxazole (1 : 9). Additionally, the essential oil fraction of A. sylvestris exhibited significant DPPH free radical scavenging activity and the reducing power.

약용식물의 Peroxynitrite와 Hydroxyl radical 소거 활성 (Peroxynitrite and Hydroxyl Radical Scavenging Activity of Medicinal Plants)

  • 약용식물 9종의 물 추출물에 대한 항산화 효과를 탐색하기 위해 in vitro 조건에서 강력한 산화력과 독성을 갖는 $ONOO^-$${\cdot}$OH radical의 소거효과를 측정하였다. 그 결과, 감잎, 구기자, 꾸지뽕나무, 담쟁이덩굴, 마디풀, 비파나무, 으름덩굴, 참취 추출물은 10${\mu}g/ml$ 농도에서 50% 이상의$ONOO^$ 소거효과를 나타냈다. 이러한 $ONOO^-$ 소거효과는 양성 대조물인 penicillamine(94.08${\pm}$3.04)에 비해 낮았지만 비파나무 물 추출물(89.87${\pm}$4.57)이 다른 시료에 비해 가장 강력한 $ONOO^-$ 소거효과를 나타냈다. 또한, 가는잎쐐기풀과 감잎 물 추출물은 1mg/ml 농도에서 양성대조물인 thiourea 보다 효과적인${\cdot}$OH radical 억제효 과를 나타냈다. 결론적으로 9종의 약용식물은 $ONOO^-$${\cdot}$OH radical과 연관된 산화적 스트레스에 의한 세포와 조직의 손상을 억제시킬 수 있는 천연 항산화제로 밝혀졌다.

호박분말 효소가수분해물의 항산화활성 (Antioxidant Activities in Enzymatic Hydrolysates of Pumpkin Powder (Cucurbita spp.))

  • 본 연구는 늙은호박, 단호박 및 애호박을 다양한 탄수화물 분해효소로 소화하여 조제한 효소가수분해물의 총 폴리페놀 함량과 항산화 활성을 측정하였다. 조회분, 조단백질 및 조섬유는 애호박이 높았으며, 탄수화물은 단호박이 높았고, 조지방은 서로 비슷하였다. 애호박 분말의 모든 효소처리군의 총 폴리페놀 함량은 늙은호박과 단호박의 가수분해물에 비하여 좀 더 높았다. 애호박 분말의 AMG와 Termamyl 소화로 조제된 효소가수분해물의 DPPH 유리기 소거활성 또한 늙은호박 및 단호박에 비하여 매우 높게 나타났다. 한편, Visozyme 처리 가수분해물을 제외한 늙은호박과 단호박의 대부분 가수분해물은 애호박보다 더 높은 superoxide anion 소거활성을 나타내었다. 애호박 가수분해물(Ultraflo 예외)의 hydrogen peroxide 소거활성은 다른 호박의 효소 분해물 보다 약간 높았으나, hydroxyl radical 소거활성은 모든 호박 품종에서 14% 미만으로 낮았다. Nitric oxide 소거활성은 단호박 및 애호박의 Viscozyme 분해물에서 매우 효과적이었으며, 다른 효소가수분해물의 경우에도 BHT보다는 낮았으나 $\alpha$-tocopherol보다 높은 활성을 나타내었다.
