Antioxidative Activity of Lichen Thamnolia vermicularis in vitro

  • This study was aimed at evaluating the antioxidant activities of methanol extract of Thamnolia vermicularis. The antioxidant activity, reducing power, superoxide anion radical scavenging and free radical scavenging activities were studied. The antioxidant activity of the extract correlated with its concentration ($0.2{\sim}2\;mg/ml$) in the reaction mixtures containing linoleic acid. Upto 67% of lipid peroxidation was inhibited by 2 mg/ml of the lichen extract. The extract showed strong free radical scavenging activity similar to that of BRA (positive control) in a manlier of concentration dependent. The lichen extract also showed moderate effects on superoxide anoin scavenging activity and reducing power, which was not so effective as that of Quercetin and BHA used as positive controls. This study suggests that T. vermicularis lichen can be used as a novel source of natural antioxidant.

Antioxidant Activities of Ribes diacanthum Pall Extracts in the Northern Region of Mongolia

  • Ribes diacanthum Pall (RDP) is a member of the Saxifragaceae family. The plant is traditionally used in Mongolia for the treatment of various ailments associated with kidney and bladder's diseases, cystitis, kidney stone, and edema. This study was aimed to investigate antioxidant activities of different solvent extracts of whole Pall plants, based on ferric-reducing antioxidant potential (FRAP), 2,2'-azinobis(3-ethybenzothiazoline-6-sulfonic acid) ($ABTS{\cdot}+$) radical scavenging activity, 1,1-diphenyl-2-picrydrazyl ($DPPH{\cdot}$), and hydroxyl (${\cdot}OH$) radical scavenging activities. Additionally, total flavonoids and phenolic contents (TPC) were also determined. The ethyl acetate extract of RDP (EARDP) had a remarkable radical scavenging capacity with an $IC_{50}$ value of 0.1482 mg/mL. In addition, EARDP was shown to be higher in total phenolic and flavonoid contents than the methanol extract of RDP (MRDP). Moreover, the EARDP had the predominant antioxidant capacity, DPPH, hydroxyl, and ABTS radical scavenging activities and ferric reducing power. These results suggest a potential for R. diacanthum Pall extract as a functional medicinal material against free-radical-associated oxidative damage.

삼채부위별 에탄올 추출물의 생리활성탐색 (Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri)

  • 삼채뿌리와 잎이 가진 기능성 식품소재로서의 활용도를 높이고자 다양한 생리활성을 평가하였다. 삼채뿌리의 조사포닌, 폴리페놀, 플라보노이드 함량은 각각 4.28 mg/g, 18.27 mg/g, 0.95 mg/g, 삼채잎의 조사포닌, 폴리페놀, 플라보노이드 함량은 각각 4.17mg/g, 35.91 mg/g, 2.79 mg/g으로 나타났다. 삼채뿌리 및 잎 부위의 70% 에탄올 추출물의 DPPH 라디칼 소거능 $IC_{50}$으로 23.01 mg/mL, 4.82 mg/mL로 나타났으며, hydroxyl radical 소거능 $IC_{50}$ 값이 각각 40.82 mg/mL, 27.24 mg/mL를 나타났다. ${\alpha}$-Amylase 저해 활성은 삼채뿌리 및 잎 부위 에탄올 추출물 10 mg/mL를 처리했을 때 억제활성이 각각 61.38%, 61.01%로 나타났다. 삼채뿌리 및 잎 부위의 아질산염 소거능을 측정한 결과, 삼채추출물의 농도가 증가함에 따라 그 활성 역시 증가하는 경향으로 나타났다. 이상의 결과로 삼채뿌리보다는 삼채잎 부위에 항산화 및 항당뇨 활성이 나타났으며, 삼채잎을 활용한 식품 소재로써 개발할 가치가 매우 높은 것으로 판단된다.

Fucoidan Protects LLC-PK1 Cells against AAPH-induced Damage

  • This study was designed to investigate the protective effect of fucoidan against AAPH-induced oxidative stress in LLC-PK1 cells (porcine kidney epithelial cells). Oxidative stress was induced by exposing of LLC-PK1 cells to the 1 mM 2,2'-azobis(2-amidino propane) dihydrochloride (AAPH) for 24 hr. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in a significant (p<0.05) decrease in cell viability, but fucoidan treatment protected LLC-PK1 cells from AAPH-induced cell damage in a dose dependant manner. To investigate the protective action of fucoidan against AAPH-induced damage of LLC-PK1 cells, we measured the effects of fucoidan on lipid peroxidation and antioxidant enzymes activities of AAPH treated cells as well as scavenging activities on superoxide anion radical and hydroxyl radical. Fucoidan had protective effect against the AAPH-induced LLC-PK1 cellular damage and decreased lipid peroxidation and increased activities of antioxidant enzymes such as superoxide dismutase (SOD) and glutathione peroxidase (GSH-px). Furthermore, fucoidan showed strong scavenging activity against superoxide anion radical. The $IC_{50}$ value of fucoidan was $48.37{\pm}1.54\;{\mu}g/mL$ for superoxide anion radical scavenging activity. The fucoidan also had high hydroxyl radical scavenging activity ($IC_{50}=32.03\;{\mu}g/mL$). These results indicate that fucoidan protects against AAPH-induced LLC-PK1 cell damage by inhibiting lipid peroxidation, increasing antioxidant enzyme activities and scavenging offree radicals.

Quality Evaluation of Cheongpomuk Added with Chlorella Powder

  • This study examined the quality characteristics to evaluate the optimal mixing ratio of a chlorella powder in Cheongpomuk. The total polyphenol and total flavonoids contents of Cheongpomuk and chlorella powder was 279.90 ㎍/mL, 489.50 ㎍/mL and 48.79 ㎍/mL, 687.85 ㎍/mL respectively. The DPPH free radical scavenging activities and ABTS radical scavenging activities of chlorella powder was 65.39%, 91.28% respectively. The L values (58.85~26.23), a values (-0.44~-5.84) of the Hunter's color values decreased and b value (-10.98 ~4.12) increased significantly in proportion to the amount of chlorella powder. The total polyphenol contents (55.00~734.20 ㎍/mL), total flavonoids contents (41.00~88.29 ㎍/mL), DPPH free radical scavenging activities (35.10~65.50%) and ABTS radical scavenging activities (50.29~92.86%) of Cheongpomuk increased significantly in groups of chlorella powder. In the physical properties, the hardness (135.93~103.93 g/㎠), chewiness (6.76~3.76 g), springiness (4.92~4.36 %), and gumminess (147.70~94.23) of Cheongpomuk decreased significantly in proportion to the amount of chlorella powder. The Cheongpomuk containing 15% chlorella powder showed a high score with regard to color, flavor, moistness, chewiness and overall acceptance. The appropriate addition amount of chlorella powder for making Cheongpomuk is 15%. This study indicate that Cheongpomuk treated with chlorella powder had the highest functional component and antioxidant activity.

호도약침액(胡桃藥鍼液)의 유리기(遊離基) 소거(消去)와 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究) (Radical Scavenging and Antioxidant Effects of Juglandis Semen Extract(JSE))

  • This study was performed to determine if Juglandis semen extract(JSE) has free radical scavenging and antioxidant activities. Superoxide anion generation by xanthine oxidase/xanthine and in neutrophils activated by phorbol-12, 13-dibutyrate was inhibited by JSE and its effect was dose-dependent. JSE also inhibited generation of $H_2O_2$ induced by glucose oxidase/glucose and in opossum kidney cells treated with antimycin A. JSE exerted a direct $H_2O_2$ scavenging effect. Exposure of opossum kidney cells to 1mM tBHP caused a significant increase in lipid peroxidation, which was prevented by JSE. JSE also prevented tBHP-induced LDH release. These data suggest that JSE has free radical scavenging and antioxidant activities. However, further studies should be carried out to find the active ingredient(s) of JSE that exerts radical scavenging action.

Optimization and production of protein hydrolysate containing antioxidant activity from tuna cooking juice concentrate by response surface methodology

  • To optimize the hydrolysis conditions in the production of antioxidant hydrolysates from tuna cooking juice concentrate (TC) to maximize the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, TC containing 48.91% protein was hydrolyzed with Alcalase 2.4 L, and response surface methodology (RSM) was applied. The optimum hydrolysis conditions included a 2.2% (w/v) Alcalase concentration and 281 min hydrolysis time, resulting in the highest DPPH radical scavenging activity of 66.49% (0.98 µmol Trolox/mg protein). The analysis of variance for RSM showed that hydrolysis time was an important factor that significantly affected the process (p < 0.05). The effects of different drying methods (freeze drying, hot air drying, and vacuum drying) on the DPPH radical scavenging activity and amino acid (AA) profiles of TC hydrolysate (TCH) were evaluated. Vacuum-dried TCH (VD) exhibited an increase in DPPH radical scavenging activity of 81.28% (1.20 µmol Trolox/mg protein). The VD samples were further fractionated by ultrafiltration. The AA profiles and antioxidant activities in terms of the DPPH radical scavenging activity, 2,2'-azino-bis(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power, and ferrous ion chelating activity were investigated. Glutamic acid, glycine, arginine, and cysteine were the major AAs found in the TCH fractions. The highest DPPH radical scavenging activity was found in the VD-1 fraction (< 5 kDa). The VD-3 fraction (> 10 kDa) exhibited the highest ABTS radical scavenging activity and ferric reducing antioxidant power. The ferrous ion chelating activity was the highest in VD-1 and VD-2 (5 to 10 kDa). In conclusion, this study provided the optimal conditions to obtain high antioxidant activities through TCH production, and these conditions could provide a basis for the future application of TCH as a functional food ingredient.

민들레 물추출물의 항산화 및 자유라디칼 소거활성 (Antioxidative and Free Radical Scavenging Activity of Water Extract From Dandelion (Taruaxacum officinale))

  • 민들레 잎과 뿌리로부터 물추출물을 제조하여 항산화 활성 및 라디칼 소거활성을 분석하기 위하여 linoleic acid에 대한 과산화물 생성저해율, DPPH radical에 대한 소거활성, chemiluminescence를 이용한 hydroxyl radical에 대한 소거활성, EPR을 이용한 superoxide anion radical에 대한 소거활성및 hydrogen peroxide에 대한 소거활성을 측정하였다. 그 결과, 민들레 잎의 물추출물이 뿌리의 물추출물 보다 지방산에 대한 과산화물 생성 저해율이 높았고, DPPH radical, hydroxyl radical, superoxide anion radical 및 hydrogen peroxide에 대한 소거활성 역시 매우 높은 것으로 나타났다.

Antioxidant and Antidiabetic Activities of Aralia elata Seeds

  • Aralia elata seeds were successively extracted with water, methanol, ethanol, acetone and chloroform. The crude extracts were investigated for antioxidant and antidiabetic activities. The antioxidant properties of various extracts were evaluated by antioxidant tests, such as DPPH free radical-scavenging activity, hydroxyl radical-scavenging assay, metal-chelating activity, lipid peroxidation inhibition activity and reducing power assay. The 70% methanol extract exhibited the highest activity in the in vitro models of DPPH free radical-scavenging activity, metal-chelating activity, and reducing power assay. Acetone extract showed good effects on lipid peroxidation inhibition and hydroxyl radical-scavenging assay at a low concentration. In addition, the $\alpha$-glucosidase inhibition assay showed that 70% methanol extract had the highest activity. These results indicate the high possibility of using A. elata seeds for medical application due to their efficient antioxidant properties.

수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해 (Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5))

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.
