The antioxidant activity was analyzed in gamma-irradiated cooking juices. Because the activities of antioxidants have been attributed to various mechanisms, different assay methods including 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP), have been conducted and compared. All of these antioxidant assay showed that the antioxidant activity of cooking juice was increased by gamma-irradiation. To investigate this increase of antioxidative activity, the protein was extracted from cooking juices and its antioxidant activity was measured. From the results, it was thought that the modification of protein in cooking juiced by irradiation caused the increase of antioxidant activity of cooking juice. Therefore, gamma irradiation could be an promising method for a sterilization of cooking juice with increased antioxidant activity.
This study was carried out to assess the effect of radiation on the changes of Hizikia fusiformis cooking juice ethanol extract and to compare the effect of gamma ray and electron beam. On the applying radiation, the dark color of cooking juice became changed with higher brightness and lower redness and yellowness. But, there was no difference between gamma ray radiation and electron beam radiation. 1,1-Diphenyl-2-picryl-hydrazyl radical scavenging activity and tyrosinase inhibitory activity of cooking juice were shown to be increased by radiation independent on the radiation source types. The reason for the increased biological activities was caused by higher content of total phenolic compounds. The results could be applied to investigate the effect of radiation source on the color and antioxidant activity of biomaterials, and it was thought that irradiation could be an promising method for enhancing the biological activity of biomaterials.
Two chitinase producing strains CHI2 and CHI4 were isolated from soybean rhizosphere soil. Both the strains belonged to Lysobacter enzymogenes as indicated by 16S rDNA sequence analysis. Though strain CHI2 and CHI4 produced extracellular chitinase, they differ in their chitinolytic activity. CHI4 produced approximately three times the higher amounts of enzyme than that of CHI2 under specified conditions. CHI2 produced $535.67U\;l^{-1}$ of chitinase after 48 h incubation with a specific activity of $3.91U\;mg^{-1}$ of protein while strain CHI4 produced $1584.13U\;l^{-1}$ of chitinase with a specific activity of $10.88U\;mg^{-1}$ protein. SDS-PAGE analysis indicated that the molecular weight of chitinase enzyme was approximately 45 kDa. A faint band with a molecular weight of 55 kDa reveals the possibility for the presence of another kind of chitin binding protein. Mutant library was developed by exposing the isolates to gamma rays at their $LD_{99}$ value (0.23 kGy). Totally, 11 mutants of CHI2 and CHI4 are reported to have enhanced chitinase activity. Several leaky mutant clones with decreased enzyme activity and a defective mutant (CHI2-M16) with complete loss of chitinase activity were also identified. CHI4-M18, CHI4-M8 and CHI4-M29 showed 78.8, 41.5, and 31.9% increased chitinase activity over wild type CHI4.
In order to evalute the effects of radiation on mammalian neuronal system, we have examined the effect of gamma-ray radiation on the monoamine oxidase (MAO) activity in monoaminergic neurons. Following the whole body irradiation, MAO activity in the rat brain was measured as well as in the liver for the comparative studies between the neuronal and nonneuronal system. The effects of some radiation protectors and sensitizers were also examined in addition to the $O_2$ effect. The results can be summarized as follows. 1) The MAO activity of rat brain was minimally affected by the radiation dose up to 1,700 cGy Radiation dose above 2,500 cGy inhibited the brain MAO activity by no less than $l0\%.$ MAO-A form was found to be particularly sensitive to radiation. The liver MAO was somewhat inhibited (by about $5\%$) but hardly dependent on the dose of radiation. 2) The inhibitory effect on the brain was initiated immediately by the radiation dose of 2,500 cGy. On the contrary, for the liver, the inhibitory effect became apparent only 2 days after irradiation. 3) Two days after a dose of 2,500 cGy, Vmax and Km of the brain mitochondrial MAO decreased. For liver, Vmax decreased while Km increased, which indicates the kinetic patterns for the neuronal and nonneruronal systems are not affected similarly by radiation. 4) The effect of several known radiation protectors and sensitizers on MAO activity was tested ut no definite results were obtained. The level of -SH group increased in some degree upon radiation but not by the compounds. 5) MAO activity was not affected by $O_2$ concentration, while an elevated level of lipid peroxidase was found under the same condition. The results described here indicate that characteristics of MAO, one of the most important central nervous system enzymes, are liable to radiation, which is partially differentiated from the liver MAO. Also indicated are that the -SH groups are hardly related to the effect of radiation but the production of the lipid peroxide seems to be somewhat correlated to the effect of radiation.
Radiation-induced late injury to normal tissue is a primary area of radiation biology research. The present study was undertaken to investigate whether the late effect of the ionizing radiation appears as an age-related oxidative status in the brain. Three groups of 4-month old C57BL/6 mice that were exposed to $^{137}Cs$${\gamma}-rays$ at a single dose (5 Gy) or fractionated doses ($1Gy{\times}5times$, or $0.2Gy{\times}25times$) at 2 months old were investigated for the oxidative status of their brains with both young (2-month) and old (24-month) mice. A significant (p<0.05) decrease in superoxide dismutase (SOD) activity was observed in old mice brains compared with that of the young mice. malondialdehyde (MDA) content was significantly (p<0.05) increased in the old mice brain. However, any significant difference in SOD activity and MDA contents of the irradiated brain was not observed compared to age-matched control group mice. SOD activity and MDA content were observed within good parameters of brain aging and there were no late effects on the age-related oxidative level in the ${\gamma}-ray$ irradiated mice brains.
The activity concentration of primordial radionuclides i.e., $^{238}U$ series, $^{232}Th$ series and $^{40}K$, in soil samples collected from Udagamandalam environment, have been measured by employing NaI (Tl) Gamma ray Spectrometer. The absorbed gamma dose rate has also been simultaneously measured by using both Environmental Radiation Dosimeter at each soil sampling location (ambient gamma dose) as well as from the gamma dose derived from the activity concentration of the primordial radionuclides. The results of activity concentration of each radio nuclides in soil, absorbed dose rate in air due to soil activity and possible cosmic radiation at each location along with human effective dose equivalent for Udagamandalam environment are presented and discussed.
[6]-Gingerol, a major polyphenol of ginger (Zingiber officinale), exhibits a variety of biological properties including anti-oxidant, anti-inflammatory and anti-cancer activity. However, the radioprotective effect of [6]-gingerol is still unknown. The aim of this study was to investigate the radioprotective effect of [6]-gingerol against radiation-induced cell cytotoxicity and oxidative stress in HepG2 cells. [6]-Gingerol pretreatment attenuated radiation-induced cell cytotoxicity caused by 5Gy (half lethal dose, $LD_{50}$ of HepG2 cells). The measurements of superoxide dismutase (SOD) and catalase (CAT) activity were also performed. The results showed that [6]-gingerol pretreatment reduced increasing SOD and CAT activity after exposure of IR, indicating that [6]-gingerol protected oxidative stress by regulating cellular antioxidant enzyme (SOD and CAT) activity. These findings suggest that [6]-gingerol acts as a radioprotector by attenuating cell cytotoxicity and oxidative stress.
A mushroom mutant with increased cellulolytic activity was developed through radiation mutagenesis. The homogenized hypha suspension of Pleurotus florida was exposed to gamma radiation ($^{60}Co$, AECL) at the dose of $LD_{99}$ (0.51 kGy, $D_{10}$; 0.26 kGy). Among 16 mutants, Pf CM4 showed 17.24% more cellulolytic activity than the wild type (p<0.05). It was observed that Pf CM4 can utilize all kinds of carbon sources tested for their mycelia growth. Starch, xylan, and glucose favourably supported the radial mycelia extension. Yeast extract and $NH_4NO_3$ have been recorded as the best organic and inorganic nitrogen sources, respectively. Pf CM4 was found to grow significantly faster, even at high temperature ($30^{\circ}C$), than wild type (p<0.05), and the optimal pH was 5.5~6.5. This study reveals that the mutant Pf CM4 could be employed for the effective recycling of cellulosic wastes, in addition to mushroom farming.

To investigate the antifungal activity related protein in pesticidal bacteria, a bacterial strain LTD was isolated from soil collected at Gimje in Jeonbuk province, Korea, and identified as Bacillus subtilis LTD based on a API50 CHB kit and 168 rDNA sequencing. It has an antifungal activity against 9 plant pathogenic fungi in a paper disc assay. The antifungal activity- deficient mutant, B. subtilis mLTD was induced at a 5 kGy dose of $^{60}Co$ gamma radiation. Using the two-dimensional electrophoresis and the matrix assisted laser desorption ionization time-of-flight mass spectrometry, the comparison analysis of proteins between the wild and mutant were performed. A major intracellular serine proteinase IspA (MW: 32.5 kDa), a NAD (P) H dehydrogenase (MW: 20.0 kDa), and a stage II sporulation protein AA, SpoIIAA (MW: 14.3kDa) were detected only in the B. subtilis LTD. These results suggested that the functions of these proteins found only in the B. subtilis LTD could. be closely related to the antifungal activity against plant pathogenic fungi.

The biological effects of low dose ionizing radiation (LDIR) remain insufficiently understood. We examined for the scientific evidence to show the biological effects of LDIR using radiation-sensitive immune cells. We found that Ikaros protein was responsed to low dose-dependent effects of gamma radiation in IM-9 B lymphocytes. Ikaros encodes zinc finger transcription factors that is important regulators of a hematopoietic stem cells (HSCs) progression to the B lymphoid lineage development, differentiation and proliferation. In this study, we observed that cell proliferation was enhanced from 10% to 20% by LDIR (0.05 Gy) in IM-9 B lymphocytes. The Ikaros protein was phosphorylated in its serine/threonine (S/T) region and decreased its DNA binding activity in the cells exposed to LDIR. We found that Ikaros phosphorylation was up-regulated by CK2/AKT pathway and the residues of ser-304 and ser-306 in Ikaros was phosphorylated by LDIR. We also observed that Ikaros protein was localized from the nucleus to the cytoplasm after LDIR and bound with Autotaxin (ENPP2, ATX) protein, stimulating proliferation, migration and survival of immune cells. In addition, we found that the lysoPLD activity of ATX was dependent on Ikaros-ATX binding activity. These results indicate that the Ikaros is an important regulator of immune activation. Therefore, we suggest that low dose ionizing radiation can be considered as a beneficial effects, stimulating the activation of immune cells.
