시설원예지에 분포하는 뿌리혹선충의 종류 및 간이 동정법 (Root-knot Nematode Species Distributing in Greenhouses and Their Simple Identification Scheme)

  • 남부 시설재배지에서 서식하는 뿌리혹선충의 종류와 race를 조사하고 그들에 대한 간이동정법을 제시하였다. 남부 시설재배지에는 Meloidogyne arenaria race 2(59%), M. incognita race 1(23%), M. incognita 미동정 race(18%)가 분포하고 있었음, 우점종은 땅콩뿌리혹선충(M. arenaria) race 2 였다. 유충의 길이는 M. arenaria가 411$\mu\textrm{m}$(306-503$\mu\textrm{m}$), M. incognitark 384$\mu\textrm{m}$(312-488$\mu\textrm{m}$)로 두 종의 범위가 겹치고 있어 종 동정에 참고하기 어려웠으며, M. arenaria와 M. incognita의 종 동정에 가장 쉽게 이용할수 있는 특징은 암컷의 머리에 있는 Excretory pore 위치였다.

Pathogenic Diversity of Ascochyta rabiei Isolates and Identification of Resistance Sources in Core Collection of Chickpea Germplasm

  • Ascochyta blight caused by Ascochyta rabiei (Pass.) Lab. (Telomorph: Didymella rabiei) (Kov.) is one of the most important fungal diseases in chickpea worldwide. Knowledge about pathogen aggressiveness and identification resistance sources to different pathotypes is very useful for proper decisions in breeding programs. In this study, virulence of 32 A. rabiei isolates from different part of Iran were analyzed on seven chickpea differentials and grouped into six races based on 0-9 rating scale and susceptibility/resistant pattern of chickpea differentials. The least and most frequent races were race V and race I, respectively. Race V and VI showed highly virulence on most of differential, while race I showed least aggressiveness. Resistance pattern of 165 chickpea genotypes also were tested against six different A. rabiei races. ANOVA analysis showed high significant difference for isolate, chickpea genotypes and their interactions. Overall $chickpea{\times}isolate$ (race) interactions, 259 resistance responses (disease severity ${\leq}4$) were identified. Resistance spectra of chickpea genotypes showed more resistance rate to race I (49.70%) and race III (35.15%), while there were no resistance genotypes to race VI. Cluster analysis based on disease severity rate, grouped chickpea genotypes into four distinct clusters. Interactions between isolates or races used in this study, showed the lack of a genotype with complete resistance. Our finding for virulence pattern of A. rabiei and newly identified resistance sources could be considerably important for integration of ascochyta blight resistance genes into chickpea breeding programs and proper decision in future for germplasm conservation and diseases management.

  • 우리나라에 발생하고 있는 뿌리혹선충의 Race를 조사하고 기후에 따른 종의 분포 및 밀도를 파악하기 위하여 3226포장에서 시효를 채취하여 Baermann's funnel법으로 선충을 추출하여 선충의 형태와 International Meloidogyne Project의 Differenctial Host Test Method로 조사하였다. 우리나라에 있어서 뿌리혹선충의 종류별 분포는 M. hapla가 중부지방에서, M. incognita가 남부지방에서 우점종을 이루었고, M. arenaria는 남부지방에서 검출율이 다소 높았으며 M. javanica는 제주와 남부해안지방을 중심으로 분포하고 있었다. 우리나라에 분포하고 있는 뿌리혹선충의 종별 Race는 M. arenaria에서 Race 1과 Race 2, 그리고 M. incognita에서 Race 1, Race 2 및 Race 3이 알려졌으며, 이 중에 M. arenaria Race 1과 M. incognita Race 3은 우리나라에서 처음으로 보고되었다.

콩 Cyst 선충 Race 5에 대한 저항성 QTL 탐색 (Identification of Quantitative Trait Loci for Resistance to Soybean Cyst Nematode Race 5)

  • 콩 품종 Essex와 PI 437654간 교잡 후 F$_2$유래 F$_3$ 계통들을 재료로 하여 작성된 RAPD 유전자지도상에 cyst 선충 race 5에 대한 저항성 QTLs 분석을 실시한 바 결과를 요약하면 다음과 같다. 1. 회귀분석 결과 26개의 marker들(22 RAPD, 4 RFLP)에서 cyst 선충 race 5 저항성 반응에 대한 유의성이 인정되었다. 2. MAPMAKER /QTL 분석 결과2개의 저항성 QTL들이 탐색되었는데, 이 QTL들은 2개의 linkage groups(LGC-20와 Group 2)에 위치하였다. 3. 탐색된 2개의 QTL들 중 1개는 우성유전, ?고 나머지 하나는 열성유전양상을 나타내었다. 4. 콩 cyst 선충 race 5의 저항성에 대한 유의성이 인정되는 5개의 marker들간 상호작용을 알아보기 위한 다중회귀분석 결과 총 26개의 조합들 중 4개의 marker들(E02$^3$, G$10^1$, W03, pK418C)로 구성된 조합에서 가장 높은 표리적 변이의 값(35.2%)을 나타내었다.

국내 멜론 흰가루병균의 race 동정 및 시판품종의 흰가루병 저항성 판별 (Identification of fungal races that cause powdery mildew in melon (Cucumis melo L.) and selection of resistant commercial melon cultivars against the identified races in Korea)

  • 국내 멜론 및 박과채소 재배농가(안성, 이천, 영암, 창녕, 순천)에서 발병하고 있는 멜론 흰가루병균을 채집한 후 흰가루병균의 형태학적 특성을 이용한 종류 구분 및 멜론 race 판별계통을 이용한 국내에서 발병하고 있는 흰가루병균의 race를 확인하였다. 그 결과 국내 멜론 흰가루병균의 종류는 Podosphaera xanthii (syn. Sphaerotheca fuliginea)였으며, 기존에 보고된 race 1, N2, A와 판별결과가 일치하는 3 종류의 균주가 분리되었다. 또한 기존에 보고된 race (1, N1, N2, A, S, 5)와 검정 결과가 다른 새로운 race의 흰가루병균 2종이 동정되었다. 그리고 5개의 분리 및 동정된 흰가루병균을 이용하여 국내 시판중인 멜론 15품종에 대한 저항성을 조사한 결과, race1에 대해서는 15품종 중 9품종이 저항성을 보였다. Race N2와 A대해서는 소수의 품종에서 저항성을 보였으며, 새로운 race BN968에서는 15품종 모두가 이병성을 나타내었다. 또한 멜론 흰가루병 race 1 저항성 DNA 마커 KPMR1M-1과 KPMR1M-2에 의한 이병성 및 저항성 판별 결과와 race 1 접종 결과는 일치하였다. 따라서 국내 멜론 품종 육종에 있어서 다양한 race 특이적 마커 개발 및 이를 활용한 복합 race 저항성 품종 개발이 요구된다.

Identification of a host range determinant from Ralstonia solancearum race 3

  • Ralstonia solancearum infects many solanaceous plants, however race 3 infects only potato and tomato weakly. To identify genes responsible for race specificity of R. solanacearum, we mobilized genomic library of LSD2029 (race 3) into LSD341 (race 1) and inoculated 1,000 transconjugants into hot pepper. One transconjugant that did not induce wilt symptom in hot pepper was isolated. We found that a cosmid clone, pRSl, conferred avirulence to LSD341. By deletion and mutational analyses of pRSl, we found the 0.9-kb PstI/Hindlll fragment carries avirulence functions. We sequenced the fragment and identified one possible open reading frame, a rsal gene, possibly encoding 110 amino acids. The rsal was preceded with a plant-inducible promoter (PIP) box, indicating that the gene might be regulated by HrpB. Interestingly, the promoter region of the rsal homolog in the strain GM11000 (race 1) did not have the PIP box. Rsal did not show any significant homologies with proteins in the database, indicating th e protein is different from the previously reported avirulence proteins. When we mutated the rsal gene by marker-exchange in LSD2029, the mutant was less virulent in potato.

  • This paper describes how to identify standard numbers and to diagnose defects of the ball bearings. The first stage of the networks is a procedures for identifying standard numbers of the bearings, and the next stage carries out the diagnosis of defects on the outer race and the inner race of bearings. The identification and the diagnosis of bearings were carried out by simulations and experiments.

PCR-Based Assay for Rapid and Specific Detection of the New Xanthomonas oryzae pv. oryzae K3a Race Using an AFLP-Derived Marker

  • We describe the development of a polymerase chain reaction method for the rapid, precise, and specific detection of the Xanthomonas oryzae pv. oryzae (Xoo) K3a race, the bacterial blight pathogen of rice. The specific primer set was designed to amplify a genomic locus derived from an amplified fragment length polymorphism specific for the K3a race. The 1,024 bp amplicon was generated from the DNA of 13 isolates of Xoo K3a races out of 119 isolates of other races, pathovars, and Xanthomonas species. The assay does not require isolated bacterial cells or DNA extraction. Moreover, the pathogen was quickly detected in rice leaf 2 days after inoculation with bacteria and at a distance of 8 cm from the rice leaf 5 days later. The results suggest that this PCR-based assay will be a useful and powerful tool for the detection and identification of the Xoo K3a race in rice plants as well as for early diagnosis of infection in paddy fields.

  • The purpose of this study is to investigate the symbolic meaning in body image and fashion style portrayed in Korea and American movies in aspects of sex, race, nature, and technology. The researcher analyzed movies in Korea and USA, and the results were as follows. In aspects of sex, movies portrayed new masculinity and femininity escaped from previous gender stereotypes, and the clothing styles characterized sex identity and sex roles. In aspects of race, more various races appeared in America movies than Korea's, and the appearance and stereotypes related with race were blurred by the globalization and the change of film market environments. In aspects of nature, Korea movie portrayed the human being as lyrical appearance in nature, and America movie expressed the human as a strong men conquering the nature in adventure movie or super human image combined with nature thing which endowed nature's superior characteristics to human body in fantasy and science fiction movies. In aspects of technology, the human bodies were described as cyborg, alien, or super human, which symbolized the do-identification in digital world, de-boundarization between human and machines, and the human's uncertainty.

Identification of Two New Races of Podosphaera xanthii Causing Powdery Mildew in Melon in South Korea

  • Powdery mildew caused by the obligate biotrophic fungus Podosphaera xanthii poses a serious threat to melon (Cucumis melo L.) production worldwide. Frequent occurrences of the disease in different regions of South Korea hints at the potential existence of several races which need to be identified. The races of five isolates collected from different powdery mildew affected regions were identified based on the pathogenicity tests of these isolates on eight known differential melon cultigens namely, SCNU1154, PMR 45, WMR 29, PMR 5, MR-1, PI124112, Edisto 47 and PI414723. None of the isolates have shown same disease responses to those of the known races tested in this study and in previous reports on these identical differential melon cultigens. This indicates that the tested uncharacterized isolates are new races. Among the isolates, the isolates from Hadong, Buyeo, Yeongam and Gokseong have shown same pathogenicity indicating the possibility of these isolates being one new race, for which we propose the name 'race KN1'. The isolate of Janghueng have also shown unique disease response in the tested differential melon cultigens and hence, we identified it as another new race with a proposed name 'race KN2'. Report of these new races will be helpful in taking effective control measures in prevalent regions and for future breeding programs aimed at developing varieties that are resistant to these race(s).
