Development of PCR-Based Molecular Marker for Detection of Xanthomonas campestris pv. campestris Race 6, the Causative Agent of Black Rot of Brassicas

  • Xanthomonas campestris pv. campestris (Xcc), the pathogen of black rot which is the most destructive disease of Brassica vegetables throughout the world. Here, we reported two novel sequence-characterized amplified region (SCAR) markers (i.e., XccR6-60 and XccR6-67) for the detection of Xcc race 6 via re-alignment of the complete genome sequences of Xcc races/strains/pathovars. The specificity of SCAR primer sets was verified by mean of PCR amplification using the genomic DNA template of Xcc races/strains/pathovars and two other plant infecting bacterial strains. The PCR result revealed that the XccR6-60 and XccR6-67 primer sets amplified 692-bp and 917-bp DNA fragments, respectively, specifically from race 6, while no visible amplification was detected in other samples. In addition, the SCAR primers were highly sensitive and can detect from a very low concentration of genomic DNA of Xcc race 6. However, the complete genome sequence of Xcc race 6 is not yet publicly available. Therefore, the cloning and sequencing of XccR6-60 and XccR6-67 fragments from race 6 provide more evidence of the specificity of these markers. These results indicated that the newly developed SCAR markers can successfully, effectively and rapidly detect Xcc race 6 from other Xcc races/strains/pathovars as well as other plant pathogenic bacteria. This is the first report for race-specific molecular markers for Xcc race 6.

  • 이 실험의 목적은 탄수화물 부하식이요법시의 삼출건비방 투여가 달리기 선수의 뇨중 대사성분변화에 미치는 영향에 대해 관찰 연구한 것이다. 탄수화물 부하식이시 삼출건비탕(蔘朮健脾湯) 투여는 전해질대사 관련 변인에 있어서 $Na^+$, $Cl^-$, $K^+$, $Ca^{+ +}$에 유의성 있는 영향을 미쳤으며, 이는 세포막 투과성의 변화를 의미하는 것으로 $Na^+$-$K^+$ pump의 활성도가 증가하여 운동수행능력을 향상시킬 것으로 기대되나 추후 전해질 대사와 관련하여 renin, aldosterone 등의 호르몬 분비 기전에 관해서도 지속적인 연구가 진행되어야 할 것으로 사려된다.

  • 가잠을 이중교배함으로씨 발생하는 형질을 조사한 결과 다음 결과를 얻었다. 1. 10분간 이중교배한 결과 그 형질이 잠품종에 따라 변화하였다. 그러므로 잠종제조에 있어서 자유교미를 절대적으로 방지하여야 한다. 2. 동품종과 이품종간의 수정력강도에 일정한 경향을 발견하기가 곤란하다. 그리고 먼저 사정된 정자가 뒤에 사정된 정자보다 우위를 차지한다고 보기 곤란하다.

Thoroughbred 경주마의 관절경수술 후 경주복귀율 (2005~2010) (Rate of return to race after arthroscopic surgeries in Thoroughbred racehorses (2005~2010))

  • The racehorses that under arthroscopic surgery due to be injured his limbs were studied during exercise or training at Busan Race Park from 2005 to 2010. Rate of arthroscopic surgical treatments was 1.4% (63/4,642). Affected bones were radius, radial carpal bone, third carpal bone, proximal phalanx, third metacarpal bone, femur, tibia, proximal sesamoid bone and intermediate carpal bone. The lesions were fracture, chip fracture, slap fracture, osteochonrosis, and osteochondrotitis dissencans. Number of patients under arthroscopic surgery were 63. Success horses of returned to racetrack or tried to return to racetrack were 58, and 5 horses were in training or resting at the time of publication. Success horses of returned to their previous use in the patients were 49 horses (84.4%) and no returned to the racetrack were 9 horses (15.6%) in 58 horses.

Race- and Isolate-specific Molecular Marker Development through Genome-Realignment Enables Detection of Korean Plasmodiophora brassicae Isolates, Causal agents of Clubroot Disease

  • Clubroot is one of the most economically important diseases of the Brassicaceae family. Clubroot disease is caused by the obligate parasite Plasmodiophora brassicae, which is difficult to study because it is nonculturable in the laboratory and its races are genetically variable worldwide. In Korea, there are at least five races that belongs to four pathotype groups. A recent study conducted in Korea attempted to develop molecular markers based on ribosomal DNA polymorphism to detect P. brassicae isolates, but none of those markers was either race-specific or pathotype-specific. Our current study aimed to develop race- and isolate-specific markers by exploiting genomic sequence variations. A total of 119 markers were developed based on unique variation exists in genomic sequences of each of the races. Only 12 markers were able to detect P. brassicae strains of each isolate or race. Ycheon14 markers was specific to isolates of race 2, Yeoncheon and Hoengseong. Ycheon9 and Ycheon10 markers were specific to Yeoncheon isolate (race 2, pathotype 3), ZJ1-3, ZJ1-4 and ZJ1-5 markers were specific to Haenam2 (race 4) isolate, ZJ1-35, ZJ1-40, ZJ1-41 and ZJ1-49 markers were specific to Hoengseong isolate and ZJ1-56 and ZJ1-64 markers were specific to Pyeongchang isolate (race 4, pathotype 3). The PCR-based sequence characterized amplified region (SCAR) markers developed in this study are able to detect five Korean isolates of P. brassicae. These markers can be utilized in identifying four Korean P. brassicae isolates from different regions. Additional effort is required to develop race- and isolate-specific markers for the remaining Korean isolates.

Molecular Marker Development for the Rapid Differentiation of Black Rot Causing Xanthomonas campestris pv. campestris Race 7

  • Xanthomonas campestris pv. campestris (Xcc) is a plant pathogen of Brassica crops that causes black rot disease throughout the world. At present, 11 physiological races of Xcc (races 1-11) have been reported. The conventional method of using differential cultivars for Xcc race detection is not accurate and it is laborious and time-consuming. Therefore, the development of specific molecular markers has been used as a substitute tool because it offers an accurate and reliable result, particularly a quick diagnosis of Xcc races. Previously, our laboratory has successfully developed race-specific molecular markers for Xcc races 1-6. In this study, specific molecular markers to identify Xcc race 7 have been developed. In the course of study, whole genome sequences of several Xcc races, X. campestris pv. incanae, X. campestris pv. raphani, and X. campestris pv. vesicatoria were aligned to identify variable regions like sequence-characterized amplified regions and insertions and deletions specific to race 7. Primer pairs were designed targeting these regions and validated against 22 samples. The polymerase chain reaction analysis revealed that three primer pairs specifically amplified the DNA fragment corresponding to race 7. The obtained finding clearly demonstrates the efficiency of the newly developed markers in accurately detecting Xcc race 7 among the other races. These results indicated that the newly developed marker can successfully and rapidly detect Xcc race 7 from other races. This study represents the first report on the successful development of specific molecular markers for Xcc race 7.

  • Concentration of total protein, bilirubin, sodium and potassium in blood sera, and thymol turbidity of race horses, 78 males and 57 females aging from 2 to 5 years, were estimated. The results obtained were as follows: 1. Serum of race horses was low in protein concentrations as mean values of $5.48{\pm}0.77$ g/100ml. 2. Serum bilirubin was in normal range as mean values of total bilirubin, direct bilirubin and indirect bilirubin were $1.05{\pm}0.57$, $0.12{\pm}0.05$ and $0.93{\pm}0.58$ mg/100ml, respectively. 3. Mean value of serum thymol turbidity was $1.41{\pm}0.72$ S-H unit. 4. Mean values of serum sodium and potassium of race horses from Australia were $143{\pm}8.7$ and $3.85{\pm}0.53$ mEq/liter, respectively, and mean values of serum sodium and patassium of race horses from Japan were $142{\pm}9.4$ and $3.58{\pm}0.60$ mEq/liter, respectively.

Characterization of Pyrenophora tritici-repentis (Tan Spot of Wheat) Races in Baltic States and Romania

  • Tan spot, caused by the fungus Pyrenophora tritici-repentis, is economically important foliar disease in Latvia, Lithuania, and Romania; however, race structure from Baltic States and Romania is not known. In this study, we performed genotypic and phenotypic race characterization of a large collection of P. tritici-repentis isolates from these countries to determine race structure and utilize this information for better disease management and breeding wheat for tan spot resistance. We characterized 231 single spore isolates from Latvia (n = 15), Lithuania (n = 107), and Romania (n = 109) for Ptr ToxA and Ptr ToxB genes using two genes specific primers. A subset (139) of 231 isolates were further characterized for their race structure by inoculating them individually on tan spot wheat differentials set. Majority (83%) of the 231 isolates amplified Ptr ToxA gene suggesting prevalence of race 1 and 2. Further, phenotypic characterization of 139 isolates also showed wide prevalence of races 1 (68%), 2 (8%), 3 (11%), and 4 (5%) were also identified from Baltic States as well as Romania. Eighteen of the isolates (13%) did not seem to be of any of the eight known races as they lacked Ptr ToxA gene but they behaved like either race 1 or race 2, suggesting possibility of novel toxins in these isolates as their virulence tools.

한국에 분포한 맥류 줄기녹병균(Puccinia graminis f. sp. tritici)의 생리적 품종 (Physiologic Races of Puccinia graminis f. tritici in Korea)

  • 1. 맥류 줄기 녹병균에(p. graminis f. sp. tritici) 대한 저항성 품종 육종에 기초자료를 얻고자 1971-1972년 2개년간에 한국에 분포한 생리적 품종을 조사하였다. 2. 2개년간 총 55개의 균주를 채점하여 Stakman씨 방법에 따라 접종한 결과 27개 균주가 동정되었는데 그 성적은 다음과 같다. Race 11이 2균주, Race 17과 21은 각각 8균주 Race 34의 4균주 그리고 Race 37과 56이 각각 한 균주였다. 그 나머지 3균주는 Stakman씨의 색인표와 부합치 않으므로 가칭 $K_3K_4$$K_5$라고 명명하였다. 3. 그러므로 맥류줄기녹병균에 대한 생리적 품종의 조사는 저항성 품종 육종과 병행하여 계속 실시되어 야 할 것이다.

Reaction of Global Collection of Rye (Secale cereale L.) to Tan Spot and Pyrenophora tritici-repentis Races in South Dakota

  • Rye (Secale cereale L.) serves as an alternative host of Pyrenophora tritici-repentis (PTR) the cause of tan spot on wheat. Rye is cultivated as a forage or cover crop and overlaps with a significant portion of wheat acreage in the U.S. northern Great Plains; however, it is not known whether the rye crop influences the evolution of PTR races. We evaluated a global collection of 211 rye accessions against tan spot and assessed the diversity in PTR population on rye in South Dakota. All the rye genotypes were inoculated with PTR races 1 and 5, and infiltrated with Ptr ToxA and Ptr ToxB, at seedling stage. We observed 21% of the genotypes exhibited susceptibility to race 1, whereas, 39% were susceptible to race 5. All 211 accessions were insensitive to both the Ptr toxins. It indicates that though rye exhibits diversity in reaction to tan spot, it lacks Ptr ToxA and ToxB sensitivity genes. This suggests that unknown toxins or other factors can lead to PTR establishment in rye. We characterized the race structure of 103 PTR isolates recovered from rye in South Dakota. Only 22% of the isolates amplified Ptr ToxA gene and were identified as race 1 based on their phenotypic reaction on the differential set. The remaining 80 isolates were noted to be race 4. Our results show that races 1 and 4 are prevalent on rye in South Dakota with a higher frequency of race 4, suggesting a minimal role of rye in the disease epidemiology.
