RNA Interference of Chitinase Gene in Spodoptera litura (담배거세미나방(Spodoptera litura) Chitinase gene의 RNA interference)

  • RNA interference (RNAi) is the method which controls phenotypes of gene in live cells. Chitinase is the enzyme helping digestion and absorption of old cuticles during the ecdysis of insects. In order to investigate molting-inhibition effect with the chitinase related gene in Spodoptera litura, RNA was extracted from the $5^{th}$ instars. cDNA was synthesized and then we obtained about 700 bp size chitinase. After PCR products were cloned into a pGEM T-easy vector, colonies were picked. DNA was extracted from the colony cultures. EcoR I enzyme was used to check whether PCR products were inserted or not. And then we confirmed vector band of about 3 kb and insert band of about 700 bp. To synthesize the dsRNA, each DNA was cut with Spe I and Nco I enzymes (Circular DNA became lineared DNA). After synthesis of dsRNA, approximately 5 ul dsRNA was injected into the $3^{rd}$ abdominal segment of S. litura $4^{th}$ larvae. The concentration of dsRNA was about $10{\mu}g/{\mu}l$. We confirmed larval-larval molting : there were phenotypically abnormal individuals - for instance malformation, molting inhibition and change of integument color. Pupaadult molting : there were phenotypically abnormal individuals - for instance molting inhibition, change of wings and malformation. Also we could investigate the pupation, emergence and variation about noninjection, treated with DW and dsRNA. Each pupation was non-injection 83.3%, DW 78.3% and dsRNA 66.7%. Each emergence was non-injection 90.0%, DW 72.3% and dsRNA 65.0%. So we considered that chitinase dsRNA induced molting inhibition effect. But each variation was non-injection 8.9%, DW 2.9% and dsRNA 19.2%. Therefore dsRNA group showed the highest variation value. When 18 hours after injecting dsRNA, we could obtain abnormal individual.

  • A survey of biological and chemical characteristics in the middle-northern East Sea of Korea was carried out at 28 stations in October, 1995 on board R/V Tam-Yang. On the basis of the vertical profiles of temperature, salinity and dissolved oxygen, water masses in the study area were divided into 5 major groups; (1) Low Saline Surface Water (LSSW), (2) Tsushima Surface Water (TSW), (3) Tsushima Middle Water (TMW), (4) North Korean Cold Water (NKCW), (5) last Sea Porper Water (ESPW). Other 4 mixed water masses were also observed. It is highly possible that the LSSW which occured at depths of $0\~30m$ in the most southern part of the study area is originated from the Yangtze River (Kiang) of China due to very low salinity $(<32.0\%_{\circ})$ relatively high concentration of dissolved silicate and no sources of freshwater input into that area. Oxygen maximum layer in the vertical profile was located near surface at northern cold waters and became deeper at the warm southern area. Oxygen minimum layer af depths $50\~100m$, which is TMW, were found in only southern area. In the vortical profiles of nutrients, the concentrations were very low in the surface layer and increased drammatically near the thermocline. The highest concentration occurred in the ESPW. The relatively low value of Si/P ratio in the ESPW (13.63) compared to other reports in the East Sea was due to continuous increase of P with depth as well as Si. The N : P ratio was about 6.92, showing that nitrogenous nutrient is the limiting factor for phytoplankton growth. The exponential relationship between Si and P, compared to the linear relationship between N and P, indicates that nitrate and phosphate have approximately the same regenerative pattern, but silicate has delayed regenerative pattern.

Application of Gamma Irradiation for the Microbiological Safety of Fried-Frozen Cheese Ball (냉동치즈볼의 미생물학적 안전성 확보를 위한 감마선 조사기술의 이용)

  • This study was conducted to sanitize fried-frozen cheese ball by gamma irradiation. Total aerobic bacteria and yeasts and molds counts were 4.4 and 2.8 log CFU/g in non-irradiated sample, respectively. Microorganisms were decreased with increase of irradiation dose $(D_{10}=1.25\;kGy)$, and were not detected in samples irradiated at 3 kGy or more $(<10^2\;CFU/g)$. $D_{10}$ value for Escherichia coli (KCTC 1682) was 0.25 kGy. TBA (2-thiobarbituric acid) values were increased as irradiation dose was increased, but there was no significant difference between non-irradiated and irradiated samples at 3 kGy or less (p<0.05). The results of sensory evaluation showed that there was no significant difference between non-irradiated and irradiated samples at 3 kGy or less, the sensory scores were decreased with irradiation at 5 kGy or more (p<0.05). These results indicated that gamma irradiation at 3 kGy was considered to be an effective treatment to ensure the microbiological safety of fried-frozen cheese balls without any sensorial change, even though further studies should be investigated to reduce detrimental effects induced by irradiation.

  • In the analysis of variance between population and between individual trees (families), the fluctuation of values of variances due to sample size, (number of family) was analysed by two different designs, i.e. 2-level nested design with equal sample size and randomized complete block design. The variables were seedling heights and root calipers of 1-0 and 1-1 seedlings of Pinus densiflora S. et Z. The details of three natural stands and their progeny characters were presented in previous reports. 1. In nested design analysis. increase of sample size resulted the decrease of F-values among families in general, however, the F-values among populations showen the increasing tendency. The smaller the sample size, the larger the F-values fluctuation was resulted in general. At the point of beyond sample size 10, however, the fluctuation become to be stabilized. The F-value fluctuation seemed to be more in the case of analysis with random sampling method than with sequentially accumulated sampling method. And also such a tendency was more obvious in smaller sample size than in large one. 2. In R.C.B.D. analysis, the sample size to affirm the family variation was smaller than that for population variations.

A Study of the Vegetation in the Deogyusan National Park - Focused on the deciduous forest at Namdeogyu area - (덕유산국립공원의 식생에 관한 연구 -남덕유지역 낙엽활엽수림을 중심으로-)

  • This study was carried out to classify vegetation structure on the deciduous forest at Namdeogyu area in Deogyusan National Park from May 2007 to September 2008 using the gradient analyses and phytosociological method. The vegetation was classified into Quercus mongolica community(Typical subcommunity, Rhododendron sehlippenbaehii-Q. mongolica subcommunity, Carex lanceolata-Q. mongolica subcommunity), Q. Serrata community(Typical subcommunity, Q. variabilis -Q. serrata subcommunity, Carpinus laxiflora - Q. serrata subcommunity), C. laxiflora community, Fraxinus mandshurica community and Corn us controversa community. Ecological characteristics such as species composition, layer structure, vegetation ratio, and the distribution of individual trees by DBH(diameter at breast height) were significantly different among communities. The order of important value of the forest community with DBH 2cm above plants was Q. mongolica(67.8), F. mandshurica(44.8), Q. serrata(27.5), Acer pseudo-sieboldianum(17.0), C. controversa(l5.9), F. sieboldiana(l1.5), R. schlippenbachii(11.0), C. cordata(11.0), Styrax obassia(10.9), Magnolia sieboldii(10.4) and C. laxiflora(9.8). Distribution of DBH of Q. mongolica had a formality distribution, suggesting a continuous domination of these species over the other species for the time being. In contrast, F. mandshurica appeared limited to the valley of the sheet and a higher frequency of young individuals, suggesting a continuous domination of these species the development of a climax forest terrain. Q. serrata had a higher frequency of young individuals and middle individuals, suggesting a continuous domination of these species over the other species for the time being in contrast. This study examined the correlation between each community and the environment according to DCCA ordination. The Q. mongolica community predominated in the highest elevation habitats which had many C.E.C and T-N. F. mandshurica community predominated in the highest elevation habitats which had many Moisture and EX-Cation. The Q. serrata community and C. laxiflora community mainly occurred in the low elevation habitats which had few C.E.C and T-N. The C. controversa community appeared on the park in the middle habitats.

The Changes of Types of Nutritional Risk and Nutritional Intake according to Depression and Self-esteem among the Elderly in Chunnam Province (전남지역 노인에서 우울 및 자아존중감의 인식정도에 따른 영양위험도 및 영양소 섭취량의 변화 양상)

  • This study was conducted in order to identify the relationship between psychological factors, such as depression and self-esteem, and nutritional status, such as nutritional risk index and nutrient intake, among the elderly in Chunnam Province. The participants were 119 elderly individuals over the age of 65 years who visited the Senior Welfare Center in Chunnam province between January 29 and February 2, 2007. This study was conducted using a structured questionnaire that included, the Center for Epidemiologic Studies Depression Scale (CES-D), General Self Efficacy Scale (GSES), Nutritional Screening Initiative Checklist (NSI checklist), questions pertaining to the general characteristics of the participants and an estimation of nutrient intake using the 24-hour recall method. Data were analyzed by the SPSS program. Analysis of the participant's CES-D scores revealed that 43.7% of the subjects were normal and 56.3% had more than mild depression. The mean GSES score was 45.9 for the entire group of subjects (51.9 for men, 43.6 for women). The mean nutritional risk index value was 4.30 (5.03 for men, 4.01 for women). Analysis of the participant's scores on the NSI checklist revealed that 69.7% of the subjects were normal and 30.3% exhibited a moderate nutritional risk. The CES-D was positively correlated with the NSI checklist (p < 0.05) but negatively correlated with nutrient intake. However, the GSES was negatively correlated with the NSI checklist (p < 0.05), but positively correlated with nutrient intake (P < 0.01 for protein, calcium, phosphorus, zinc etc.). The results of this study indicate that it is necessary to manage psychological factors, including depression and self-esteem, in the elderly in order to decrease their nutritional risk and increase their nutrient intake.

A Study on Trust Transfer in Traditional Fintech of Smart Banking (핀테크 서비스에서 오프라인에서 온라인으로의 신뢰전이에 관한 연구 - 스마트뱅킹을 중심으로 -)

  • In this study, we investigated the effect of offline banking trust on smart banking trust. As influencing factors of smart banking trust, this study compared offline banking trust, smart banking's system quality, and information quality. For the empirical study, 186 questionnaire data were collected from smart banking users and the data were analyzed using Smart-PLS 2.0. As results, it was verified that there is trust transfer in FinTech service, by the significant effect of offline banking trust on smart banking trust. And it was proved that the effect of offline banking trust on smart banking trust is lower than that of smart banking itself. The contribution of this study can be seen in both academic and industrial aspects. First, it is the contribution of the academic aspect. Previous studies on banking were focused on either offline banking or smart banking. But this study, focus on the relationship between offline banking and online banking, proved that offline banking trust affects smart banking trust. Next, it is the industrial contribution. This study showed that offline banking characteristics of traditional commercial banks affect the trust of emerging smart banking service. This means that the emerging FinTech companies are not advantageous in the competition of trust building compared to traditional commercial banks. Unlike traditional commercial banks, the emerging FinTech is innovating the convenience of customers by arming them with new technologies such as mobile Internet, social network, cloud technology, and big data. However, these FinTech strengths alone can not guarantee sufficient trust needed for financial transactions, because banking customers do not change a habit or an inertia that they already have during using traditional banks. Therefore, emerging FinTech companies should strive to create destructive value that reflects the connection with various Internet services and the strength of online interaction such as social services, which have an advantage over customer contacts. And emerging FinTech companies should strive to build service trust, focused on young people with low resistance to new services.

Analysis of Nutritional Compounds and Antioxidant Effect of Freeze-Dried powder of the Honey Bee (Apis mellifera L.) Drone (Pupal stage) (서양종 꿀벌(Apis mellifera L.) 수벌번데기 동결건조 분말의 영양학적 성분 및 항산화 효과)

  • In this study, we analyzed the nutritional ingredients of drone pupae (16th to 20th instar old) to evaluate the value of bee products and provide basic data for product diversification, and the extracts prepared using these pupae were tested for physiological activity. According to the analysis of the general ingredients of the freeze-dried powder of these bee pupae, the moisture, crude protein, crude fat, and crude ash was 1.69 ± 0.07%, 48.52 ± 0.20%, 23.41 ± 0.14%, and 4.05 ± 0.02%, respectively. Vitamin C and vitamin E were 14.92 ± 0.52 mg/100 g and 6.06 ± 0.11 mg α-TE/100 g, respectively. Regarding minerals, the highest content of K (1349.13 ± 34.57 mg/100 g) and P (1323.55 ± 43.85 mg/100 g) was observed and Ca and Fe were 55.43 ± 1.51 mg/100 g and 5.49 ± 0.19 mg/100 g, respectively. The fatty acids of the water extracted freeze-dried pupae powder accounted for approximately 59.62 of saturated fatty acids and 40.38 of unsaturated fatty acids, and high-quality fatty acids such as palmitic acid (C16:0) was 35.49 ± 0.08 and oleic acid (C18:1, n-9) was 35.91 ± 0.22 (g/100 g total fatty acids). The total amino acid content was 38.99 ± 2.63 g/100 g and the free amino acid was a total of 5129.04 mg/100 g, of which 1257.68 mg/100 g was proline and 759.12 mg/100 g glutamic acid. The DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity of the drone pupae extract showed values of 0.8 for distilled water extract, 3.2 for 50% EtOH extract, 6.4 for 70% EtOH extract, and approximately 90% for 32 ㎍/mL for 100% EtOH extract. These results suggest that the main compound contributing to the antioxidant activity is a polar compound, and it is highly likely to be a low-molecular protein or a free amino acid. In conclusion, the honey bee drone pupa is excellent as a food resource and can be utilized as a new functional material for food and functional food.

A Studs on Exposure to Organic Dust and Ammonia in Poultry Confinement Buildings (일부 육용 양계 농업인의 유기먼지와 암모니아 노출에 관한 연구)

  • Objectives: This study was carried out to assess exposure levels of organic dusts and ammonia in poultry farms in Korea. Methods: A total of six poultry farms were investigated. The farms were located in Namwon, Chonlabuk-do and in Kae-San, Chungchongbuk-do. This study consisted of a questionnaire and measuring organic dusts and ammonia. The questionnaire included the characteristics of the farms, work patterns and the tasks of the poultry farms. Results and Conclusions: The farmers raised the chickens 45 times a year and the average number of years in the poultry farm were eight years ranging from 2 to 12 years. They worked for seven days per week and the average hours spent caring the chickens are 6.3 hours per day. The duration of staying in the confinement buildings was 3.3 hours per day. The work time in summer was longest. The feed and the water supply systems were automatic and the control of ventilation windows used "winch curtain" was semiautomatic. They used mechanical ventilation system in winter and used dilution ventilation system in the other seasons. The geometric mean concentration of total and respirable dust sampled in the poultry confinement buildings was 4.0 mg/$m^3$and 0.9 mg/$m^3$ respectively. The ratio of respirable to total dusts range from 9 to 49 percent. There was no sample exceeding the criteria 10 mg/$m^3$ for total dust and 3 mg/$m^3$ for respirable dust in farms. The criteria have been recommended by Korean Ministry of Labor and American Conference of Governmental Industrial Hygienist. The personal respirable dusts measured during a circle work averaged geometric mean concentration 1.4 mg/$m^3$ Two personal samples were exceeded the threshold 3 mg/$m^3$. There was a positive relation between an index and the personal samples of respirable dusts($R^2$=0.98). The index is calculated by multipling the total number of chickens in the farm by the age of the chickens and then dividing by the volume of the confinement building. The geometric mean concentration of area and personal ammonia samples was 23.3 ppm and 22.2 ppm, respectively. Some of the ammonia samples, both area and personal samples, exceeded the short term exposure limit value 35 ppm.

A CYANOACETYLENE STUDY OF THE MOLECULAR DISK IN STAR FORMING REGIONS

  • We have observed dense core around young stellar objects, DR21, S140, Orion-KL, and L1551 using four millimeter-wave transitions of $HC_3N\;J$=4-3, J=5-4, J=10-9, and J=12-11. The spatial distribution of $HC_3N$ emission closely resembles the morphology of the previous CS observations that trace high density gas. These observations reveal the existence of $HC_3N$ dense cores around central IR source, elliptical in shape and almost perpendicular to the CO bipolar outflow axis. Small differences can be explained by that $HC_3N$ molecular line is more optically thin and is seen to be more detailed structure in the neighborhood of central IR sources. In S140 and Orion-KL, massive(${\sim}10\;M_{\odot}$), slowly rotating dense cores lie near at the central IR sources of bipolar outflows. The velocity channel maps of DR21 show that the bipolar outflow gas may have a correlation with the dense core of DR21. We analyzed intensities of the four lines to derive physical conditions in dense core from two methods, LTE and LVG. The column density of $HC_3N$, $N(HC_3N)$, between LTE and LVG calculations agree well with each other. The abundances of $HC_3N$ in each observing source have been estimated using the average values of $n(H_2)$ and $N(HC_3N)$ and assuming the size of dense core. The fractional $HC_3N$ abundances in massive dense cores of DR21, S140, and Orion-KL have a range of $(2-7){\times}10^{-10}$, while that of low mass dense core, L1551, has one order of magnitude greater value of $2{\times}10^{-9}$. This should be considered good agreement with the result by Morris et al.(1976). It may be considered that dense cores of DR21, S140, and Orion-KL may have almost same stage of chemical evolution, and their abundances have a small values relative to that of L1551. The column density $N(HC_3N)$ decreases with increasing distance from the densest part of the cloud, the central infrared source, and have the relation of $N(HC_3N){\varpropto}R^{\alpha}$, where a has a range of 0.65 to 0.89. The values of $n(H_2)$ are not varied with increasing distance from the dense core, and have almost same values. Therefore, it is considered that the dense cores in these regions probably consist of dense clumps in diffuse molecular gas medium, and $n(H_2)$ of each clump is ${\sim}10^5\;cm^{-3}$. Levels in the $T_{ex}$ increases with $n(H_2)$. It is considered that the $HC_3N$ dense cores are not completely thermalized. We examine the relationships between the luminosity of central infrared sources versus mass of the dense cores, and the luminosity of central infrared sources versus molecular hydrogen column density. Luminosities of the central IR sources show good correlation with mass and hydrogen column density of the dense core. Same has been found from CS observations. However, mass and size derived from $HC_3N$ observations are one order of magnitude smaller than those from CS. It can be interpreted that we see more central part of the cloud cores in $NC_3N$ lines than CS lines.
